ABSTRACT

The prominent role of the P-element induced wimpy testis (PIWI)-interacting RNA (piRNA) pathway in animals is to silence transposable elements and maintain genome stability, ensuring proper gametogenesis in gonads. GASZ (Germ cell protein with Ankyrin repeats, Sterile alpha motif, and leucine Zipper) is an evolutionarily conserved protein located on the outer mitochondrial membrane of germ cells and plays vital roles in the piRNA pathway and spermatogenesis in mammals. In the model insect Drosophila melanogaster, GASZ is essential for piRNA biogenesis and oogenesis, whereas its biological functions in non-drosophilid insects are still unknown. Here, we describe a comprehensive investigation of GASZ functions in the silkworm, Bombyx mori, a lepidopteran model insect, by using a binary transgenic CRISPR/Cas9 system. The BmGASZ mutation did not affect growth and development, but led to sterility in both males and females. Eupyrene sperm bundles of mutant males exhibited developmental defects, while the apyrene sperm bundles were normal, which were further confirmed through double copulation experiments with sex-lethal mutants, which males possess functional eupyrene sperm and abnormal apyrene sperm. In female mutant moths, ovarioles were severely degenerated and the eggs in ovarioles were deformed compared with that of wild type (WT). Further RNA-seq and RT-qPCR analysis revealed that amounts of piRNAs and transposon expression were dysregulated in gonads of mutants. In summary, this study has demonstrated vital roles of BmGASZ in gametogenesis through regulating the piRNA pathway in B. mori.

ABSTRACT

Objective: To evaluate the effectiveness of scapular stabilization exercises (SSE) in the treatment of subacromial pain syndrome (SAPS). Methods: Clinical randomized controlled trials (RCTs) on SSE in the treatment of SAPS were searched electronically in PubMed, Science Direct, Cochrane Central Register of Controlled Trials (CENTRAL), EBSCOhost, Physiotherapy Evidence Database (PEDro), Web of Science, and other databases from 2000 to 2022, supplemented by manual search. Final RCTs were selected based on inclusion and exclusion criteria, and the Physiotherapy Evidence Database scale was used to evaluate the methodological quality of the study. A meta-analysis was conducted on data using the RevMan5.4 software. Results: Eight RCTs involving 387 participants were included. The meta-analysis showed that the experimental group (SSE) had greater improvements in the Visual Analog Scale score [Weighted Mean Difference (WMD) = -0.94, 95% CI (-1.23, -0.65), p < 0.001] and the Shoulder Pain and Disability Index score [WMD = -10.10, 95% CI (-18.87, -1.33), p = 0.02] than the control group (conventional physical therapy). However, range of motion (ROM) was not found to be greater in the experimental group than in the control group. Conclusion: Existing evidence moderately supports the efficacy of SSE for reducing pain and improving function in SAPS, without significant improvement in ROM. Future research should focus on larger, high-quality, standardized protocols to better understand SSE's effects across diverse SAPS populations, treatment, and outcome measures. ABSTRACT

CONTEXT: Evidence linking glucose metabolism status with brain macro- and microstructure is limited and inconsistent. OBJECTIVE: We aim to investigate the associations of glucose metabolism status with brain macrostructure and microstructure, including brain volumes, subcortical gray matter volumes, and white matter microstructural metrics. METHODS: This study enrolled 29 251 participants from the UK Biobank. Glucose metabolism status was classified into normal glucose metabolism (NGM), prediabetes, type 2 diabetes (T2D) with HbA1c <7%, and T2D with HbA1c ≥7%. Brain macrostructural metrics included volumes of total and subcortical gray matter, white matter, white matter hyperintensity (WMH), cerebrospinal fluid, and brain stem. Brain microstructural metrics included fractional anisotropy (FA) and mean diffusivity in white matter tracts. Multivariable linear regression models were used to estimate ß values and 95% CI. RESULTS: After multivariable adjustment including demographic and lifestyle factors, medical history, and total intracranial volume, those with prediabetes had smaller total and subcortical gray matter volumes than participants with NGM, while atrophy of total and subcortical gray matter was more pronounced in those with T2D (all P trend < .05). Moreover, participants with T2D had larger volumes of white matter and WMH (both P trend < .05). For brain microstructure, participants with prediabetes had lower FA values in commissural fibers (ß -0.04; 95% CI -0.08, -0.003). Global and tract-specific microstructural abnormalities of white matter were observed in participants with T2D, especially for T2D with HbA1c ≥ 7% (all P trend < .05), except for FA values in projection fibers. CONCLUSION: These findings suggest that interventions for hyperglycemia at an earlier stage may help protect brain health.

ABSTRACT

Significance: Performance during risky decision making is one of the essential cognitive functions that is impaired in several psychiatric disorders including addiction. However, the cognitive mechanism and neural correlates underlying risky decision making in chronic pain patients are unclear. To our knowledge, this study is among the first to construct computational models to detect the underlying cognitive process of chronic pain patients during risky decision making. Aim: This study aimed at inspecting the significantly abnormal risky decision-making patterns of chronic pain patients and its neuro-cognitive correlates. Approach: In this case-control study, 19 chronic pain patients and 32 healthy controls (HCs) were included to measure the risky decision making in a balloon analogue risk task (BART). Optical neuroimaging using functional near-infrared spectroscopy, together with computational modeling, was carried out to systematically characterize the specific impairments based on BART. Results: Computational modeling findings on behavioral performance demonstrated that the chronic pain patient group exhibited significant deficits in learning during BART (p<0.001), tending to make decisions more randomly without deliberation (p<0.01). In addition, significant brain deactivation alternation in the prefrontal cortex (PFC) during the task was detected for the patient group compared with that from the control group (p<0.005). Conclusions: Long-term aberrant pain responses significantly disrupted the PFC function and behavioral performance in chronic pain patients. The joint behavioral modeling and neuroimaging techniques open a new avenue for fully understanding the cognitive impairment and brain dysfunction of risky decision making associated with chronic pain.

ABSTRACT

To upgrade the algae pyrolytic oil, the influence of algae components on co-pyrolysis with LDPE were studied, with Spirulina platensis (SP), Nannochloropsis sp. (NS) and Enteromorpha Prolifera (EP) as typical algae samples, as they are enriched with proteins, lipids and carbohydrate, respectively, especially, the N and O transformation behavior during the co-pyrolysis was studied in depth. During co-pyrolysis, the interaction on products depended on the components of algae. EP and SP were prior to form CO2, rather than CO. For pyrolytic oil, co-pyrolysis effectively inhibited the formation of N- and O-compounds, but promoted the generation of long-chain alcohol and formic/acetic ester. And the obvious decrease of N and O content in co-pyrolytic oil was observed. However, the rich lipids in NS resulted in the improvement of N yield in pyrolytic oil during co-pyrolysis.

ABSTRACT

A modified signal processing and target fusion detection method based on the dual platform cooperative detection model is proposed in this paper. In this model, a single transmitter and dual receiver radar system is adopted, which can form a single radar and bistatic radar system, respectively. Clutter suppression is achieved by an adaptive moving target indicator (AMTI). By combining the AMTI technology and the traditional radar signal processing technology (i.e., pulse compression and coherent accumulation processing), the SNR is improved, and false targets generated by direct wave are suppressed. The decision matrix is obtained by cell averaging constant false alarm (CA-CFAR) and order statistics constant false alarm (OS-CFAR) processing. Then, the echo signals processed in the two receivers are fused by the AND-like fusion rule and OR-like fusion rule, and the detection probability after fusion detection in different cases is analyzed. Finally, the performance of the proposed method is quantitatively analyzed. Experimental results based on simulated data demonstrate that: (1) The bistatic radar system with a split transceiver has a larger detection distance than the single radar system, but the influence of clutter is greater; (2) the direct wave can be eliminated effectively, and no false target can be formed after suppression; (3) the detection probability of the bistatic radar system with split transceivers is higher than that of the single radar system; and (4) the detection probability of signal fusion detection based on two receivers is higher than that of the bistatic radar system and single radar system.

ABSTRACT

To improve the quality of the oil produced from microalgae, the co-pyrolysis of low-density polyethylene (LDPE) and Nannochloropsis sp. (NS) in a fixed bed reactor was investigated at different mixing ratios. Co-pyrolysis improved the gas yield, and the lower heating value of the gas products increased obviously with an increase in the LDPE amount. Furthermore, co-pyrolysis promoted the generation of CH4 and C2+, especially C2H4, with the maximum C2+ yield (84.86 mL/g) obtained with 75% LDPE. Meanwhile, the amounts of oxygenous and nitrogenous compounds in the liquid products decreased rapidly with LDPE addition. The aliphatic hydrocarbon content of the liquid products increased from 22.63% for NS pyrolysis to 77.4% with 25% LDPE. During co-pyrolysis with LDPE, O tended to evolve as H2O and CO (rather than as CO2 for NS pyrolysis) and N was more likely to be released into gas products, which enhanced the quality of the pyrolysis oil.

ABSTRACT

In various applications of computed tomography (CT), it is common that the reconstructed object is over the field of view (FOV) or we may intend to sue a FOV which only covers the region of interest (ROI) for the sake of reducing radiation dose. These kinds of imaging situations often lead to interior reconstruction problems which are difficult cases in the reconstruction field of CT, due to the truncated projection data at every view angle. In this paper, an interior reconstruction method is developed based on a rotation-translation (RT) scanning model. The method is implemented by first scanning the reconstructed region, and then scanning a small region outside the support of the reconstructed object after translating the rotation centre. The differentiated backprojection (DBP) images of the reconstruction region and the small region outside the object can be respectively obtained from the two-time scanning data without data rebinning process. At last, the projection onto convex sets (POCS) algorithm is applied to reconstruct the interior region. Numerical simulations are conducted to validate the proposed reconstruction method.
