ABSTRACT

The single-nucleotide polymorphisms (SNPs) of the growth hormone (GH) gene could be related to growth traits, particularly in farm animals. This study aimed to identify the SNPs of the GH gene (A781G and A1575G) in Black Bengal (BB) goats in Thailand. Seventy-seven BB goats of both sexes were recruited, and their genotypes were identified. Preweaning growth at birth (weight, W0; height, H0; length, L0; and chest girth, C0) and at 10 weeks postpartum (W10, H10, L10, and C10), including average daily gain (ADG) at 0-4 weeks (ADG0-4W), 4-8 weeks (ADG4-8W), and 8-12 weeks (ADG8-12W), was compared among the different genotypes in goats born from twin litter-size dams. The results showed one genotype, CC, for A1575G and three genotypes, AA, AB, and BB, for A781G. The AA gene had significantly higher W10 than AB (p < 0.05) and BB (p < 0.05). The AA had significantly higher L10 than AB (p < 0.05), while C10 was only higher in male goats (p < 0.01). The ADG4-8W of the AA genotype was significantly higher than the BB genotype (p < 0.01). We came to the conclusion that A781G is associated with growth traits during the preweaning period, while the AA genotype showed better performance than the other genotypes.

ABSTRACT

Background and Aim: Climate change challenges with incremental sea level cause saltwater intrusion, which has affected the quality of freshwater and groundwater in coastal provinces, particularly the Mekong River Delta provinces of Vietnam. Interestingly, Bach Thao goats are predominant in this area and well adapted to saline water under tropical conditions. Therefore, this study investigated the salt tolerance threshold of Bach Thao goats drinking diluted seawater (DSW). Materials and Methods: The experiment was performed using seven Bach Thao male goats (20.60 ± 1.12 kg) and divided into two phases. In Phase 1 (control, C), all goats were provided fresh water (FW) for 7 days from two identical buckets, and daily water intake (WI) was recorded from both buckets. In Phase 2 (preference test, PT), each goat was provided FW from the first bucket and DSW from the second bucket or vice versa. The concentration of DSW for the preference test was 0.5%-2.0%. Results: Body weight and dry matter intake showed no differences according to DSW consumption; however, WI was significantly lower during Phase 2 (p < 0.05), which was due to the lower WI with 1.5% and 2% of DSW (p < 0.05). Goats showed a similar preference for fluid intake between FW and DSW at 0.0%-1.0% levels and began avoiding DSW at 1.5% and rejected at 2.0% of DSW. Goats consuming 1.5% of DSW showed increased respiration rate from 13:00 to 19:00 h and rectal temperature at 13:00 h (p < 0.05). Conclusion: Goats can tolerate up to 1.0% of DSW. Shifting to FW activated aversive drinking to 1.5% and 2.0% of DSW. This behavioral response was prominent at 0.5% DSW. Moreover, goats that drank 1.5% of DSW had decreased thermoregulation.

ABSTRACT

Obesity has been linked to metabolic syndrome, type 2 diabetes, and non-alcoholic fatty liver disease (NAFLD). Obesity causes a decrease in growth hormone (GH) levels and an increase in insulin levels. Long-term GH treatment increased lipolytic activity as opposed to decreasing insulin sensitivity. Nonetheless, it is possible that short-term GH administration had no impact on insulin sensitivity. In this study, the effect of short-term GH administration on liver lipid metabolism and the effector molecules of GH and insulin receptors were investigated in diet-induced obesity (DIO) rats. Recombinant human GH (1 mg/kg) was then administered for 3 days. Livers were collected to determine the hepatic mRNA expression and protein levels involved in lipid metabolism. The expression of GH and insulin receptor effector proteins was investigated. In DIO rats, short-term GH administration significantly reduced hepatic fatty acid synthase (FASN) and cluster of differentiation 36 (CD36) mRNA expression while increasing carnitine palmitoyltransferase 1A (CPT1A) mRNA expression. Short-term GH administration reduced hepatic FAS protein levels and downregulated gene transcription of hepatic fatty acid uptake and lipogenesis, while increasing fatty acid oxidation in DIO rats. DIO rats had lower hepatic JAK2 protein levels but higher IRS-1 levels than control rats due to hyperinsulinemia. Our findings suggest that short-term GH supplementation improves liver lipid metabolism and may slow the progression of NAFLD, where GH acts as the transcriptional regulator of related genes.

ABSTRACT

Modification of cellular and immunological events due to porcine reproductive and respiratory syndrome virus (PRRSV) infection is associated with pathogenesis in lungs. PRRSV also causes female reproductive dysfunction and persistent infection which can spread to fetus, stillbirth, and offspring. In this study, changes in cellular and innate immune responses to PRRSV type 1 or type 2 infection, including expression of PRRSV mediators, mRNA expression of Toll-like receptors (TLRs) and cytokine, and cytokine secretion, were examined in primary porcine glandular endometrial cells (PGE). Cell infectivity as observed by cytopathic effect (CPE), PRRSV nucleocapsid proteins, and viral nucleic acids was detected as early as two days post-infection (2 dpi) and persisted until 6 dpi. A higher percentage of CPE and PRRSV-positive cells were observed in type 2 infections. PRRSV mediator proteins, CD151, CD163, sialoadhesin (Sn), integrin and vimentin, were upregulated following type 1 and type 2 infection. CD151, CD163 and Sn were upregulated by type 2. In both PRRSV types, mRNA expression of TLR1 and TLR6 was upregulated. However, TLR3 was upregulated by type 1, but TLR4 and TLR8 mRNA and protein were downregulated by type 2 only. Interleukin (IL)-1ß, IL-6 and tumor necrotic factor (TNF)-α were upregulated by type 2, but IL-8 was upregulated by type 1. Both PRRSV type 1 and 2 stimulated IL-6 but suppressed TNF-α secretion. In addition, IL-1ß secretion was suppressed only by type 2. These findings reveal an important mechanism underlying the strategy of PRRSV infection in the endometrium and associated with the viral persistence.

ABSTRACT

Background and Aim: The behavioral mechanism of stray dog occurrence is associated with domestication process. This study aimed to investigate the population and demographic relationship of stray dogs from our ecological habitat. We tested whether baited enclosure method could be used as an operant conditioning treat-ment to activate habituation behavior in stray dogs. Materials and Methods: The first investigation determined the population and demographic characteristics of stray dogs in the metropolitan city of Bangkok using the mark and recapture procedure. In the second investigation, a large cage equipped with a digital camera was used as the feeding and habituation area. Food was provided at four corners for 2 h. The approach behaviors and eating patterns were recorded during this period for 7 days. Results: The average number of stray dogs calculated within each cluster was 24 ± 6 dogs. For the natural small habitat, the density of stray dogs was 662 dogs per km2. This indicated that the number of dogs is underestimated using the mark and recapture procedure because of undetected puppies and shifts in the sex ratio in adult dogs. In the second investigation, we demonstrated that food was a potent positive reinforcer for stray dogs. The average onset of cage entry after offering the food was 17 min. The onset of cage entry and the reduction in the first meal duration suggested that the habituation process could be achieved within 1 week. Conclusion: The results revealed the possibility of using a large cage as a novel enclosure for food offered as the positive reinforcer for TNR program trapping procedures. We suggest that this humane trapping procedure could be used to activate habituation behavior in stray dogs.

ABSTRACT

High ambient temperature (HTa) causes acid-base imbalance and systemic oxidative stress, and this may indirectly affect the mammary gland. Furthermore, HTa induces intracellular oxidative stress, which has been proposed to affect cell metabolism directly. We previously showed in dairy goats that the negative effect of HTa was compromised by enhancing heat dissipation during a high dietary cation and anion difference (DCAD) regimen. Moreover, high-dose vitamin C or ascorbic acid (AA) supplements have been used to manage oxidative stress in ruminants. The present study hypothesized that high DCAD and AA supplements that could alleviate the HTa effect would influence the milk synthesis pathway and mammary gland function. The results showed that goats fed with high DCAD had higher blood pH than control goats in the 4th week. The high dose of AA supplement decreases urine pH in the 8th week. The percent reduction of urine pH from the AA supplement was significant in the DCAD group. The high-dose AA supplement decreased plasma glutathione peroxidase activity and malonaldehyde. This effect was enhanced by a high DCAD supplement. In addition, supplementation with AA increased milk protein and citrate and decreased milk FFA. These alterations indicate the intracellular biochemical pathway of energy metabolism and milk synthesis. It can be concluded that a high DCAD regimen and AA supplement in dairy goats fed under HTa could influence the milk synthesis pathway. The evidence suggests that HTa decreases mammary gland function by modification of acid-base homeostasis and oxidative stress.

ABSTRACT

Background and Aim: In our previous study, we observed that a high dietary cation and anion difference (DCAD) of 40 mEq/kg dry matter (DM) in the diets of lactating dairy goats increased heat dissipation. In the present study, we believe that the level of DCAD fed to non-lactating and non-pregnant goats was twice as high as that fed to lactating goats in our previous study. This increase could have resulted in a greater water balance due to increased intake of water and unchanged urinary excretion. Therefore, this study aimed to determine the behavioral and heat dissipation effects of a dietary shift from low to high DCAD levels in dairy goats under tropical conditions. Materials and Methods: Seven non-lactating and non-pregnant crossbred goats were used in this study. All animals were initially fed a low DCAD (15 mEq/100 g DM) diet from days 0-6 and then switched to a high DCAD (89 mEq/100 g DM) diet from day 7 (high DCAD-7) to day 18 (high DCAD-18). Results: The results revealed that a high DCAD increased DM intake from days 13-18 (p < 0.05). The larger daily meal size associated with the high DCAD-18 group was due to increased daytime meal sizes, not nighttime when compared to the low DCAD group. Dietary cation and anion difference supplementation did not affect daily water intake; however, drinking patterns differed between the low DCAD group and the high DCAD-7 group from 07:00 to 09:00 and during nighttime. Similarly, daily urine volume was unaffected by DCAD supplementation, but urinary patterns differed between the low DCAD and high DCAD-18 groups. The daily water balance remained unchanged across all treatments, yet, a higher morning water balance was observed in the high DCAD group. The high DCAD diet led to an increase in respiration rate and rectal temperature compared to the low DCAD diet. Conclusion: The observed eating, drinking, and urinary patterns collectively suggested that high DCAD supplementation mitigates the effects of heat stress in non-lactating goats fed at high ambient temperatures.

Goats can suffer from intermittent heat stress in high ambient temperature (HTa) conditions, which causes sporadic respiratory hypocapnia. Obstructive urolithiasis is a common urological problem in goats. Sandy uroliths can be partially relieved by urine acidification with short-term ammonium chloride (NH4Cl) treatment. However, the outcome of urine acidification and the physiological responses to short-term NH4Cl supplementation under respiratory hypocapnia of HTa have rarely been reported. The present study investigated the effect of NH4Cl supplementation that produced a low dietary cation-anion different (l-DCAD) diet on acid-base balance and renal function under HTa conditions. The first experiment investigated the physiological responses to natural HTa to prove whether the peak HTa during the afternoon could induce HTa responses without a change in the plasma cortisol. The partial pressure of CO2 also tended to decrease during the afternoon. The second experiment examined the short-term effect of l-DCAD under HTa conditions. Although the blood pH was within the normal range, there was a clear acid-base response in the direction of metabolic acidosis. The major responses in renal function were an increase in tubular function and acid excretion. With a comparable level of creatinine clearance, the fractional excretions (FE) of chloride and calcium increased, and the FE of potassium decreased. Acid excretion increased significantly in the l-DCAD group. We conclude that under HTa conditions, the tubular excretion of electrolytes and acids was the major response to acid loading without changing the filtration rate. The l-DCAD formulation can be used to acidify urine effectively.

Background and Aim: The high salinity of drinking water has been a significant problem of the Mekong Rivers Delta. Animals drinking high salinity water altered feed and water intake (WI), urinary electrolytes excretion, and productivity. This study aimed to evaluate the effects of high salinity in drinking water on drinking and eating behaviors and kidney function in crossbred goats. Materials and Methods: The experiment was completely randomized with two treatments: freshwater (0%, seawater [SW0]) and water high in salinity (1.5%, SW1.5) from diluted SW, with five replicates (five animals per treatment). This experiment lasted 3 weeks: the 1st week for the pre-treatment period and the 2nd-3rd weeks for the post-treatment. Dry matter intake (DMI) and WI were recorded every day, while urine volume (UV) was determined from day 8 to day 21. Blood and urinary samples were collected on days 6, 14, and 21 of the study for electrolytes and creatinine analysis. Results: The results demonstrated that both DMI and WI were affected by SW1.5 (p<0.05). Goats drinking from SW1.5 had lower DMI during D19-21, and the ratio of DMI/WI was significantly different during D16-21 (p<0.05). Interestingly, the UV from SW1.5 was higher than that from SW0 during D13-21 (p<0.05). Although the body weights (BW) of both groups were similar (p>0.05), the weight gain observed in the SW1.5 group tended to decrease (p=0.056) at the 2nd week. The concentration of electrolytes in blood did not differ between the groups (p>0.05). In contrast, the concentration and excretion of Na+ and Cl- in urine increased in SW1.5 goats at D14 (p<0.05), while creatinine levels in the blood remained normal (p>0.05). Conclusion: The study concluded that crossbred male goats can tolerate 1.5% saline water from diluted SW for 2 weeks. The high salinity in water influences drinking and eating behavior in growing goats. However, the adaptive mechanism by increasing urine output and reducing the reabsorption of Na+ and Cl- in the kidney is the key function and works faster than behavioral responses. The kidney apparently drives drinking behavior during high salinity water consumption.

The study aims to investigate the relationships between colostral concentrations of insulin-like growth factor 1 (IGF-1), immunoglobulin G (IgG) and vitamin A (Vit A) and growth (body weight and average daily gain) in Black Bengal (BB) and its crossbred. The colostrum from dams (n = 16) was collected at parturition to measure the concentrations of IGF-1, IgG and Vit A. The kid weight at birth (W-0), day 14 (W-14) and day 28 (W-28) were measured and the average daily gain during day 1-14 (ADG1-14) and day 14-28 (ADG14-28) were calculated. The average concentrations of IGF-1, IgG and Vit A in colostrum were 504.6 ± 74.9 ng/ml, 9.7 ± 0.6 mg/ml and 549.1 ± 72.5 µg/100 g, respectively. The average body weight of kids at birth, day 14 and 28 were 1.72 ± 0.08, 2.95 ± 0.11 and 3.94 ± 0.13 kg respectively. Kid's breed, IGF-1, IgG and Vit A had significant positive effects on ADG14-28 while parity, litter size and sex had no effect. The growth factors that were classified into 2 classes based upon the mean values of colostral contents in all kids showed that the kids receiving the higher concentrations of IGF-1, IgG and Vit A in colostrum had higher body weight gain than those receiving the lower concentrations (92.1 ± 7.8 vs. 59.8 ± 5.7 g/day; p = 0.002, 88.3 ± 7.8 vs. 60.3 ± 6.1 g/day; p = 0.009 and 91.1 ± 6.8 vs. 56.7 ± 5.8 g/day; p < 0.001 respectively). It is concluded that IGF-1, IgG and Vit A concentrations in colostrum of dams were associated with increased kid's body weight gain at the end of first month in BB and BB crossbred goats.

ABSTRACT

High ambient temperature (HTa) is an important environmental factor influencing food intake (FI). We previously demonstrated that low-degree HTa exposure decreased FI earlier than activated physiological responses, and this effect was related to the median preoptic nucleus (MnPO) and arcuate nucleus (Arc) connection. The present study refines the condition of low-degree HTa exposure and focuses on the mechanism of Arc neural activation. We demonstrated in the first experiment that with the usual ambient temperature (Ta) at 23 °C, the low degree HTa condition is at a 7 °C temperature difference and with 90 min exposure. Rats exposed to this short-term low-degree HTa had significantly lower 1-h FI than those exposed to control Ta (CTa) without differences in rectal temperature and hematocrit. Under nonfeeding conditions, HTa could enhance c-Fos at the Arc without the activation of proopiomelanocortin (POMC) neurons. Under feeding conditions, HTa could enhance both c-Fos and POMC at Arc. In addition, the number of c-Fos and POMC colocalizations in the HTa group was higher than that in the CTa group. Finally, intracerebral preinfusion with a subthreshold dose of the melanocortin antagonist SHU9119 reversed the effect of low-degree HTa exposure on FI. Therefore, we conclude that the effect of short-term low-degree HTa exposure on FI in rats is mediated in part by activation of POMC neurons at the Arc. The results partially support the hypothesis that Arc is a crucial hypothalamic nucleus for the effect of low-degree HTa exposure on FI.

Background and Aim: Activation of breathing, the hypothalamic-pituitary-adrenal (HPA) axis, and plasma antioxidant defense are adaptive mechanisms in lactating dairy goats fed during the summer season. However, an excess of these responses can interfere with the gas exchange. This study aimed to investigate the effect of natural high ambient temperature (HTa) on blood gas parameters and their relation to the HPA axis and antioxidant defense. Materials and Methods: Six mid-lactating goats were included in this study and were fed in individual pens for 2 weeks. The data on ambient conditions, physiological responses, and blood chemistry were measured for two sampling days (D7 and D14), 1 week apart during the late summer season. On this two-sampling day, the main physiological responses to HTa, including respiration rate (RR), rectal temperature (Tr), blood gas, and blood chemistry, were measured in the morning and afternoon. Results: Goats from both D7 and D14 increased RR and Tr significantly according to morning and afternoon periods. In addition, goats were at the hypocapnia stage during afternoon panting without a change in blood pH and bicarbonate levels. Interestingly, HTa-induced hypocapnia was not accompanied by an increase in plasma cortisol levels. Finally, ΔTa was negatively correlated with changes in glutathione peroxidase activity. Conclusion: The natural HTa (ΔTa; 5-8°C) in this study activated evaporative heat dissipation and was high enough to induce respiratory hypocapnia. Importantly, this ΔTa did not activate the HPA axis but was correlated with a change in antioxidant defense. Therefore, under natural HTa in tropical conditions, respiratory hypocapnia is the first line of physiological response in goats within a specific range of natural ΔTa (5-8°C).

BACKGROUND AND AIM: Metabolism and environment are closely related. Under high ambient temperature (HTa), dairy cows may have different energy metabolism during summer and winter. The present study was carried out to investigate the effect of HTa on the milk yield and blood concentration of beta-hydroxybutyrate (BHBA) and glucose at the herd level. MATERIALS AND METHODS: One large dairy farm in Thailand with more than 100 crossbred Holstein cows milked each month was selected. The first experiment was performed on non-lactating cows to determine the normal daily concentrations of blood BHBA and glucose. Under the HTa condition, there was no significant change in blood BHBA and glucose concentrations. The second experiment was performed using a prospective cohort clinical design to demonstrate the seasonal effect on milk yield and blood BHBA as an indication of energy metabolism at the herd level. RESULTS: The temperature and humidity index for the winter (78.1±0.5) and summer (83.4±0.7) periods differ significantly. The average milk yield during the winter period was 17.8% higher than during the summer period. The reduction of body condition score (BCS) during early lactation was significant in the winter cows. Both higher milk yield and lower BCS in the winter cows suggested a state of negative energy balance. However, there was no difference in blood BHBA and glucose concentrations between winter and summer cows. The effect of HTa on insulin signaling appeared to be a counterbalancing factor for the ketogenic status. Based on the present results, it would be interesting to further investigate the incidence of subclinical and clinical ketosis in a dairy farm under tropical conditions. CONCLUSION: The present experiment revealed that HTa during summer decreased milk yield in dairy cows fed under tropical conditions. Higher milk yield in winter caused a greater reduction of BCS and suggested a greater negative energy balance. However, there was no seasonal effect on blood BHBA and glucose concentrations.

BACKGROUND AND AIM: Milk synthesis by the mammary gland is negatively influenced in part by high ambient temperature (AT). This study aimed to clarify the pathway of intramammary glucose utilization involved in mediating lactose synthesis during treatment with somatotropin under housing with misters and fans. MATERIALS AND METHODS: A single subcutaneous injection of 500 mg of recombinant bovine somatotropin (rbST) was administered 3 times once every 14 days to 87.5% crossbred Holstein cattle in early-/mid-/late lactation, under housing in a normal shaded barn and in a shaded barn with a mist-fan cooling system. RESULTS: The milk yields of the cooled cows tended to increase compared with those of uncooled cows and exhibited more potentiated effects in response to rbST treatment, coinciding with increases in mammary plasma flow and glucose uptake, but not in the mammary extraction of glucose. Treatment with rbST in the cooled cows resulted in a greater increase in the milk glucose concentration and a greater decrease in the milk glucose-6-phosphate concentration at all stages of lactation. CONCLUSION: rbST treatment exerted its galactopoietic action more by local intramammary factors than by other extramammary factors at a low AT and the synergistic effect between rbST treatment and low AT increased the availability of intramammary glucose transport in activating the process of lactose synthesis.

The present study aims to investigate the composition including concentrations of IGF-1, IgG and Vit A in colostrum and their effects by litter size and goat parity in 3 groups of goats; Black Bengal (BB), Saanen (SA) and their crossbred (BBSA). Thirty-eight goats were used (23 BB, 7 BBSA and 8 SA). The composition (fat, protein, lactose and total solid; TS) in colostrum (Day 0; D0) and milk (Day 4; D4 and Day 7; D7) were measured. The IGF-1, IgG concentrations were analysed in some samples collected at D0, D4 and D7 while Vit A was analysed only in colostrum. The results showed that colostrum components were similar among experimental groups. However, the colostral IGF-1 concentration of BBSA (983.0 ± 163.6 ng/mL) was higher than that of BB (340.7 ± 85.5 ng/mL, p < 0.01) and SA (417.1 ± 93.9 ng/mL, p < 0.01). The colostral IgG concentration of BB (8.2 ± 0.9 mg/mL) was lower than that of BBSA (12.9 ± 1.7 mg/mL, p < 0.05) and SA (12.9 ± 1.0 mg/mL, p < 0.01). Colostral Vit A concentration in BBSA (787.2 ± 152.6 µg/100 gm) was higher than that in BB (388.9 ± 84.3 µg/100 gm, p < 0.05) but was not different from SA (522.8 ± 96.9 µg/100 gm). Colostrum from all groups contained higher protein and TS but was lower in lactose concentration than milk. The IGF-1 and IgG concentrations in colostrum were much higher than in milk both D4 and D7 (p < 0.001). Additionally, litter size had no effects on colostrum contents but colostrum from goats with a higher parity number had higher IgG concentration. It is concluded that colostrum from BBSA may be superior when fed to BB newborn goats in terms of higher IGF-1, IgG and Vit A contents. Moreover, colostrum from goats with a high parity number contained more IgG content.

ABSTRACT

The objective of the present study was to determine the factors that influenced growth performance of the goat kids of Black Bengal (BB), Saanen (SA), and their crossbred F1 (male Bengal × female Saanen [BBSA] and male Saanen × female Black Bengal [SABB]). Data for 674 kids were analyzed from 316 litters and 134 does. All kids were weekly measured on their characteristics (body weight, length, height at the withers, and chest girth) from birth to 11 weeks old. The kid's breed and sex, litter size, and season of kidding influenced birth weight and other characteristics through the experiment. The SA and BBSA kids showed similar performance, which were higher than BB and SABB kids. Male kids had higher performance than female kids, and kids from a single litter showed the highest performance. Kids born during rainy season showed lower performance than those born in hot and cool seasons. In conclusion, the crossbred BBSA is superior to SABB or BB to raise in tropical climate Moreover, sex, litter size, and kidding season also affected growth performance during the preweaning period up to 11 weeks old.

ABSTRACT

BACKGROUND AND AIM: Immunoglobulin G (IgG) concentration is high in goat colostrum, particularly in the first few hours after parturition, and this is important for the kid's immunity and growth. IgG levels vary depending on several factors, including breed, disease status, colostrum management, handling, and collection time postpartum. A handheld optical refractometer, an affordable instrument that is simple to use in the field, is used widely in dairy farms to measure total solids. However, it can also be applied to estimate colostrum IgG content on the basis of comparison with standard measurement methods, usually radial immunodiffusion. Studies comparing %Brix values in relation to IgG concentration measured using enzyme-linked immunosorbent assay (ELISA) in goats are limited. The present study aimed to evaluate the use of a handheld optical Brix refractometer for the measurement of IgG concentration in goat colostrum, compare results with those using ELISA, and estimate the %Brix cutoff value equating to low-quality colostrum. MATERIALS AND METHODS: Colostrum samples were collected on day 0 from 21 goats (nine Black Bengal, six Saanen, and six of their crossbred offspring) and were frozen. Subsequently, they were analyzed for IgG concentration using a goat-specific ELISA test and Brix percentage using a handheld refractometer. The optimum %Brix cutoff value for the evaluation of colostrum quality was evaluated. RESULTS: The mean IgG concentration and %Brix in colostrum were 10.60±0.64 mg/mL and 25.0±0.9, respectively. There was a significant (p<0.01) correlation between %Brix and IgG concentration. For an IgG concentration of 6.9 mg/dL, the cutoff value for %Brix was 18.5, equating to high specificity (100%) but low sensitivity (50%). A higher %Brix cutoff value of 21.5 showed high specificity (95%) and high sensitivity (100%). CONCLUSION: A Brix refractometer can be used to estimate goat colostrum quality with a proposed %Brix cutoff value of <18.5%-21.5% for poor-quality colostrum.

ABSTRACT

The mammary gland is a privileged organ for mammals. Because of their high capacity for milk synthesis, dairy ruminants have been distributed throughout the world. In tropical areas, dairy animals face high ambient temperatures (HTa). The indirect effect of HTa on milk synthesis is mediated in part by a reduction in feed intake. The current experiment focused on the direct natural effect of HTa on mammary function. Multiparous Saanen goats were used in this study. The physiological responses for HTa were evaluated from the control period during the winter and from the natural HTa during the summer. Milk samples were collected for isolation of the goat milk cells to study the expression of the ß-1,4 galactosyltransferase (ß-GALT1), Akt, and heat shock protein 70 (HSP70) genes. Although goats in the summer maintained rectal temperature and plasma cortisol levels similar to those observed in the winter, the higher respiratory rate and lower feed intake and milk yield (MY) from the goats in the summer indicated that the goats in the summer were exposed to a higher degree of HTa. This was supported by the significantly higher level of plasma glutathione peroxidase (GPX) activity. Moreover, the relative expression levels of ß-GALT1 and Akt were not different. The relative expression of HSP70 during the summer was significantly higher than what was observed in cells isolated in the winter. In conclusion, the HTa effect on MY during the summer was related to its indirect effect on feed intake. The direct HTa effect might be related to HSP70 gene expression in goat milk cells and to plasma GPX activity. However, the natural HTa did not affect the expression of Akt or ß-GALT1.

ABSTRACT

In general, goat mammary epithelial cells (MECs) are cultured in vitro under 37 °C. We demonstrated previously that goat MECs differentiate under 37 °C although their body temperature is approximately 39 °C. This study aimed to investigate the influence of 39 °C on lactogenic differentiation of goat milk-derived MECs. The results revealed that HSP70 gene was significantly elevated at 1 h after an exposure to 39 °C but declined at 48 h thereafter. Oxidative stress status was not significantly affected by 39 °C. Expressions of CSN2, ß-GALT1, α-LA, and Akt genes tended to increase after the differentiation under 39 °C. Secretion of lactose under 39 °C was not significantly lower than 37 °C. In conclusion, incubation temperature at 39 °C does not dramatically affect lactogenic function of goat milk-derived MECs.

ABSTRACT

OBJECTIVE: This study was carried out to determine the effects of elevated dietary cation and anion difference (DCAD) on dry matter intake (DMI) and ruminal fermentation pattern in lactating dairy goats under tropical conditions. METHODS: Ten dairy goats were divided into two groups of five animals each. The groups received diets at different DCAD levels, either a control diet (22.81 mEq/100 g dry matter [DM], DCAD-23) or a DCAD-39 diet (39.08 mEq/100 g DM, DCAD-39). After parturition, DMI and water intake were recorded daily. Ruminal fluid and urine were collected, and nutrient digestibility measurements were carried out at 8th weeks postpartum (PP-8). Blood samples were collected at PP-4 and PP-8 to measure plasma leptin. RESULTS: Dry matter intake/body weight (DMI/BW) at PP-8 of the animals fed the DCAD-39 diet was significantly higher than those fed with DCAD-23 diet (p<0.05). Animals fed with DCAD-39 consumed more water than those fed DCAD-23 over 24 h, particularly at night (p<0.05). Ruminal pH, acetate concentration, and urinary allantoin excretion increased with the DCAD-39 diet, whereas ruminal butyrate concentration was lower with the DCAD-39 diet. On the other hand, other ruminal parameters, such as total volatile fatty acid concentration, propionate molar proportion and acetate/propionate average ratio, were not affected by increased DCAD supplementation. Apparent digestibility was improved by increased DCAD supplementation. Plasma leptin concentration was higher with DCAD supplementation. CONCLUSION: When feeding goats with DCAD-39 under tropical conditions, an increase in DMI was associated with improved apparent digestibility of nutrients, ruminal fermentation and microbial protein synthesis. An increase in plasma leptin concentration could not explain the effect of high DCAD on DMI.
