ABSTRACT

After the first report of Undaria pinnatifida in north Portugal (between 1999 and 2007), a rapid spread of this species could be expected due to the presence of a stable population and the favourable environmental conditions proposed by distribution models. However, field surveys showed that U. pinnatifida was not present in most of the rocky shores in north Portugal. It seems that U. pinnatifida cannot outcompete native species outside of marinas in north Portugal. The only population in natural rocky shores was found in Buarcos, where this species was frequent. This study provides density data of U. pinnatifida that will be useful in the future to monitor changes on its abundance and distribution in the centre and south of Portugal.

ABSTRACT

The objective of the present study was to evaluate the genetic variability in micropropagated plantlets of ornamental pineapple, after the fourth period of subculture. The basal culture medium consisted of MS salts, vitamins, 3% sucrose, liquid formulation, supplemented with 6-benzylaminopurine (BAP) at concentrations of 0.125, 0.25, 0.5, 1.0, and 2.0 mg/L. The addition of BAP influenced the occurrence of genetic variation revealed using random amplified polymorphic DNA (RAPD) markers. Of a total of 520 primers tested, 44 were selected and amplified; 402 monomorphic bands (97.2%) and 18 polymorphic bands (2.8%) resulted among regenerated plantlets. The polymorphic fragments were produced by 12 primers (OPA-01, OPA-20, OPB-01, OPB-19, OPC-19, OPF-13, OPL-17, OPM-13, OPP-16, OPT-07, OPV-19, and OPX-03). Among the primers that identified polymorphism, OPA-01, OPA-20, OPB-19, OPC-19, OPL-17, OPP-16, and OPX-3 each showed, one polymorphic band and OPF-13 amplified a maximum of three bands. In this study, the RAPD technique was effective in showing the occurrence of somaclonal variations that occur during the micropropagation process of ornamental pineapple cultivation in BAP-supplemented medium, and it is possible to detect the presence of genetic variation in early stages of plant development.

ABSTRACT

The garlic cultivars grown in Brazil evolved from somatic mutations and clone selection by breeding programs and by the introduction of germplasm from other countries. Morphological characters have been used to differentiate these cultivars. Two hundred and six random amplified polymorphic DNA markers were utilized for a diversity analysis of the 17 most planted garlic cultivars in Brazil. Bootstrap analysis showed that the number of markers was efficient and sufficient to obtain a coefficient of variation of 10%. Similarity varied between 16 and 98% and cluster analysis showed that, in general, genetic similarities correlate with morphological characters of the cultivars and production cycle variation. High bootstrap values at most of the nodes supported the dendrogram stability. The grouping of most varieties agreed well with previous reports based on morphological characters. As a vegetative-propagated species, viral diseases are a key problem regarding production and quality of the bulbs, causing gradual loss of yield and decrease in storage capacity. To improve the health quality of garlic seed, a virus-free stock of garlic cloves of the Amarante cultivar was obtained. The ability to distinguish garlic cultivars to detect varietal mixing after in vitro multiplication is extremely important, since correct identification is not possible until bulbs are produced. Random amplified polymorphic DNA markers were also used to differentiate cultivars while they are in vitro and not amenable to morphological discrimination. No difference was identified between the fingerprints of the virus-free or of the infected bulks of Amarante, showing that there was no clove mixing in the handling of material in the clonal multiplication phase.

ABSTRACT

Water used in hydrotherapy units of Nova Iguaçu and Nilópolis, Rio de Janeiro State, Brazil, was microbiologically analyzed. Thirty samples (5ml each) were weekly collected from September 2001 to June 2002 before the beginning and after the end of activities in the units. For analysis, routine techniques were used, which showed the presence of Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella spp, Candida spp, Penicillium spp, Epidermophyton spp, Aspergillus fumigatus, Aspergillus niger, Aspergillus spp, Cephalosporium spp, Cladosporium spp, Trichophyton rubrum, and Trichophyton spp. Results indicated a need for improving hygienic conditions, suggesting that water might be a contamination source in the evaluated units

ABSTRACT

Extensive calcification of the heart, involving predominantly the right ventricle and interventricular septum, was observed in a 3-h-old baby. The aetiology of the condition was attributed to infection by Toxoplasma gondii, based on the presence of several visceral lesions allied to the identification of the microorganism. The case is reported in view of the rarity of the type of the heart lesion. It is considered that calcification of the myocardium is secondary to vascular, inflammatory or toxic condition. The factors implicated in the pathogenesis of this unusual myocardial lesion are discussed.
