ABSTRACT

The aim of this research was to assess the validity and reliability of a newly developed scoring tool, designed for monitoring youth soccer players during match-play performance to support coaches/scouts with the talent identification process. The method used to design the Hull Soccer Behavioural Scoring Tool comprised of a five-stage process of (i) conducting an initial literature review to establish content validity (ii) gaining content validity through a cross sectional online survey (iii) establishing face validity via expert coach feedback (iv) conducting inter-rater reliability tests and (v) intra-rater reliability tests. In stage two, twenty-two soccer academy practitioners completed an online survey, which revealed that player behaviours such as resilience, competitiveness, and decision making were all valued as the most important behavioural characteristics by practitioners (90.9%), whilst X-factor was valued as least important by a significant amount (27.2%). Stages three to five of the testing procedure included a sample of four academy coaches not involved in the preceding stage. Twenty male collegiate soccer players (under-16 to under-18) involved in the study took part in four versus four small-sided games (SSG) in a 'round-robin' tournament across three weeks which accumulated 14 SSG's, 100-140 minutes of playing time and 70-98 individual player grades. Two of the four academy coaches watched the SSG's and used the Hull Soccer Behavioural Scoring Tool to assess live evidence of desirable player behaviours, which was subsequently followed by retrospective video analysis for intra-rater reliability testing. The remaining two academy coaches watched the same SSG retrospective video footage to test for inter-rater reliability. Reliability results revealed an acceptable level of agreement with scores between 81.25%-89.9% for inter-rater whilst intra-rater provided scores between 80.35%-99.4%. Preliminary evidence here suggests that the Hull Soccer Behavioural Scoring Tool is both a valid and reliable method to assess desirable player behaviours during talent identification processes. Thus, youth soccer practitioners and researchers should seek to test and further validate the tool in order to confirm its utility as a means of measuring behavioural characteristics of youth soccer players.

ABSTRACT

This study aimed to identify professional soccer practitioners' perceptions of the application of performance analysis technology within a single academy club. Secondary aims were to understand the importance that practitioners place on monitoring technical and tactical player characteristics, current practices, and barriers to implementing wearable technology. Utilising a mixed method design, forty-four professional soccer academy practitioners (Age = 32 ± 5.8; Years of experience = 8.5 ± 6.2) completed an online survey intended to examine present trends, professional practices, and perceptions regarding the monitoring of technical and tactical metrics. Frequency and percentages of responses for individual items were calculated. Subsequently, eleven participants who were directly involved with the monitoring of players were recruited to participate in a semi-structured interview. Interview data was transcribed and analysed using a combination of deductive and inductive approaches to identify key themes. The main findings across both phases of the study were that (1) technical and tactical metrics are monitored more frequently in matches (Technical: 89%; tactical: 91%) than training (Technical: 80%; Tactical 64%), predominantly due to time constraints and staffing numbers. Accordingly, practitioners believe that it would be beneficial to have an automated way of tracking technical (79%) and tactical (71%) metrics and would consider using a foot-mounted IMU to do so (technical (68%) and tactical (57%)). (2) Monitoring technical and tactical metrics is beneficial to assist with player development and to enrich feedback provision (3) Key stake holders, coaches and players should be informed of the relevance and rationale for monitoring. (4) For successful implementation and continued uptake, the information delivered needs to be both meaningful and easy to understand. Findings suggest that although participants appreciate the need to collect technical and tactical metrics, they are keen to ensure that wearable-derived data does not replace experiential and contextual knowledge. Accordingly, practitioners need to work closely with coaches to determine the contexts in which metrics may or may not prove useful. However, as the sample comprised of participants from a single academy, further studies including more practitioners are warranted. Likewise, future research could also extend to include academy soccer players perceptions too.

ABSTRACT

The primary aims of this study were to examine the application of maturity status bio-banding within professional soccer academy programmes and understand the methods employed, the intended objectives, and the potential barriers to bio-banding. Using a mixed method design, twenty-five professional soccer academy practitioners completed an online survey designed to examine their perceptions of the influence of maturation on practice, their perceptions and application of bio-banding, and the perceived barriers to the implementation of this method. Frequency and percentages of responses for individual items were calculated. In the next phase of the study, seven participants who had experience with, or knowledge of, the bio-banding process within an academy youth soccer setting were recruited to complete a semi-structured interview. Interview data was transcribed and analysed using a combination of deductive and inductive approaches to identify key themes. The main findings across the two phases of the study were that [1] there is consensus among the practitioners that the individual effect of maturation impacts their ability to accurately assess the soccer competencies, [2] the majority (80%) of the sample had implemented bio-banding, with practitioners showing a clear preference for using the Khamis and Roche method to bio-band players, with the greatest perceived benefit being during maturity-matched formats, specifically for late or post-PHV players, [3] Practitioners perceived that bio-banding enhances their ability to assess academy soccer players, and [4] practitioners who have used bio-banding believe that the method is an effective way of enhancing the perception of challenge thereby providing a number of psycho-social benefits. Findings suggest that a collaborative and multi-disciplinary approach is required to enhance the likelihood of bio-banding being successfully implemented within the typical training schedules across the adolescent phase of the player development pathway.

ABSTRACT

BACKGROUND: Both maturity and relative age selection biases are entrenched within professional academy soccer programmes. Lay opinion, and that of some scholars, holds that relative age effects exist as a product of advanced biological maturity, that is relatively older players succeed as a consequence of the physical and athletic advantages afforded by earlier maturation. There is, however, a growing body of evidence to suggest that this is not the case, and that relative age and maturation should be considered and treated as independent constructs. PURPOSE: To avoid a disconnect between contemporary academic evidence and practitioner practice, the aim of this commentary is to provide a discussion of pre-existing and new evidence relating to maturity and relative age selection biases in soccer. It is hoped that this commentary will provide an overview of new insight regarding the differences between the two selection phenomena and enable practitioners who are responsible for the (de)selection of academy soccer players for talent development programmes to make more informed decisions regarding their retention/selection strategies.

ABSTRACT

The aims of the current study were to investigate the use of dRPE with academy soccer players to: 1) examine the effect of bio-banded and non-bio-banded maturity groups within SSG on players dRPE; 2) describe the multivariate relationships between dRPE measures investigating the sources of intra and inter-individual variation, and the effects of maturation and bio-banding. Using 32 highly trained under (U) 12 to U14 soccer players (mean (SD) age 12.9 (0.9) years, body mass 46.4 (8.5) kg and stature 158.2 (14.9) cm) academy soccer players from two English professional male soccer academies. Players were categorised according to somatic maturity status using estimated percentage of adult stature attainment, with players randomly assigned into teams to play 4v4 SSG. The study used a repeated measures design, whereby the selected players participated within 6 bio-banded (maturity matched [pre-PHV Vs pre-PHV and post-PHV vs post PHV] and miss-matched [pre-PHV vs post-PHV] and 6 mixed maturity SSG at their respective clubs. Using mixed and fixed effect regression models, it was established hat pre-PHV players exhibited higher dRPE compared with their post-PHV counterparts. Mixed bio-banded games reported higher dRPE outputs overall. Variation in dRPE measures across a series of bio-banded games are caused by both between and within sources of variation in relatively equal amounts. Across a series of bio-banded games, the four dRPE measures do not provide unique information, and between variation is best expressed by one or two highly correlated components, with within variation best explained by a single equally loaded component. Using a bio-banding SSG design study, we have shown that pre-PHV players report higher subjective measures of exertion than post-PHV players during. Additionally, when evenly mixing players based on measures of maturation, higher measures of perceived exertion were generally reported.

ABSTRACT

OBJECTIVES: This study aimed to (i) establish the concurrent validity and intra-unit reliability of a foot-mounted inertial measurement unit for monitoring soccer technical actions, (ii) quantify the within-microcycle inter-positional differences in the technical actions of professional soccer training, and (iii) determine the influence of drill category on the technical actions of professional soccer training. METHODS: Twenty-one professional soccer players' technical performance data (ball touches, releases, ball touches per minute, releases per minute), collected during training sessions throughout twenty-four weekly microcycles, were analysed using general linear modelling. RESULTS: The inertial measurement unit exhibited good concurrent validity (PA = 95.1% - 100.0%) and intra-unit reliability (PA = 95.9% - 96.9%, CV = 1.4% - 2.9%) when compared with retrospective video analyses. The most ball touches (X‾ = 218.0) and releases (X‾ = 110.8) were observed on MD - 1, with MD - 5 eliciting the highest frequency of ball touches (X‾ = 3.8) and releases (X‾ = 1.7) per minute. Central midfielders performed the most ball touches (X‾ = 221.9), releases (X‾ = 108.3), ball touches per minute (X‾ = 3.4) and releases per minute (X‾ = 1.6). Small-sided games evoked more ball touches (X‾diff = 1.5) and releases per minute (X‾diff = 0.1) than previously reported in match-play. The fewest ball touches (X‾ = 1.2) and releases per minute (X‾ = 0.5) were observed during tactical drills. CONCLUSION: The results of this study provide a novel understanding of the within-microcycle, inter-positional and drill category differences in the technical actions performed by professional players during training.

ABSTRACT

Relative age selection bias persists within all major soccer leagues and youth soccer academies across the globe, with the relative age effect (RAE) being typically characterized as the over selection of relatively older players (who have sometimes also been shown to be early maturing). The aim of this study was to examine if a new allocation method (i) eliminates the RAE, and (ii) reduces the presence of any additional maturity-related differences in anthropometric and physical fitness characteristics which may exist between players within the same selection category. In the first phase, 1,003 academy soccer players [under (U) 9-16] from 23 UK professional soccer clubs were sampled and a clear RAE per birth quarter (Q) was observed for the overall sample (Q1 = 45.0% vs. Q4 = 9.8%) as well as for the different age categories. Using the newly suggested reallocation method, youth players were divided by allocation date which was defined as the midway point between the chronological age and the estimated developmental (ED) birthdate. Stature was used as an anthropometric reference point on the P50 of the growth curve to determine the developmental birthdate for this new method. After the reallocation of the players using ED, the distribution of players was more equally spread (Q1 = 25.3%, Q2 = 25.6%, Q3 = 22.4%, Q4 = 26.7%). After reallocation, the mean delta stature was reduced by 16.6 cm (from 40.3 ± 7.08 to 23.7 ± 4.09 cm, d = 2.87). The mean delta body mass difference after allocation was reduced by 6.7kg (from 33.2 ± 6.39 to 26.5 ± 4.85 kg, d = 1.18). The mean age difference increased from 1.8 to 3.9 years. A total of 42.7% of the sample would have been reallocated to a different age group compared to the current one. After reallocation, 45% of the anthropometric and physical fitness comparisons showed reductions in the within-group variation expressed as a percentage of coefficient of variation (CV%). The U10 players demonstrated the largest reduction in CV% (-7.6%) of the anthropometric characteristics. The U10 squad also showed the largest reduction in CV% for various physical fitness characteristics (-7.5%). By both eliminating the RAE and reducing temporary maturity-related anthropometric and physical fitness differences, soccer academies across the world may diversify and increase the size of the talent pool both for clubs and national youth teams. In conclusion, this study provides further evidence that the newly proposed allocation method shows the potential to remove the RAE and to create a more "leveled playing field" by reducing the within-group variation of anthropometric and physical fitness characteristics affording relatively younger, and eventually, late-maturing players the opportunity to develop their talent fairly.

ABSTRACT

The aim of this study was to assess the measurement properties of external training load measures across three formats of standardised training games. Eighty-eight players from two English professional soccer clubs participated in the study spanning three consecutive seasons. External training load data was collected from three types of standardised game format drills (11v11, 10v10, 7v7+6) using Global Positioning Systems. For each external training load metric in each game format, the following measurement properties were calculated; coefficient of variation (CV%) to determine between- and within-subject reliability, intraclass coefficient correlation (ICC) to determine test-retest reliability, and signal-to-noise ratio (SNR) to determine sensitivity. Total distance (TD) and PlayerLoad™ (PL) demonstrated good sensitivity (TD SNR = 1.6-4.6; PL SNR = 1.2-4.3) on a group level. However, a wide variety of within-subject reliability was demonstrated for these variables (TD CV% = 1.7-36.3%; PL CV% = 4.3-39.5%) and corresponding intensity measures calculated per minute. The percentage contribution of individual planes to PL showed the lowest between-subject CV% (CV% = 2-7%), although sensitivity varied across formats (SNR = 0.3-1.4). High speed running demonstrated poor reliability across all three formats of SSG (CV% = 51-103%, ICC = 0.03-0.53). Given the measurement properties of external training load measures observed in this study, specifically the within-subject variation, reliability across trials of standardised training games should be calculated on an individual level. This will allow practitioners to detect worthwhile changes across trials of standardised game format drills. Such information is important for the appropriate implementation of training and monitoring strategies in soccer.

ABSTRACT

A small evidence base supports the use of virtual reality in professional soccer, yet there is a lack of information available on perceptions and desire to use the technology from those employed at professional soccer clubs. Therefore, the aim of the study was to compare and quantify the perceptions of virtual reality use in soccer, and to model behavioural intentions to use this technology. This study surveyed the perceptions of coaches, support staff, and players in relation to their knowledge, expectations, influences and barriers of using virtual reality via an internet-based questionnaire. To model behavioural intention, modified questions and constructs from the Unified Theory of Acceptance and Use of Technology were used, and the model was analysed through partial least squares structural equation modelling. Respondents represented coaches and support staff (n = 134) and players (n = 64). All respondents generally agreed that virtual reality should be used to improve tactical awareness and cognition, with its use primarily in performance analysis and rehabilitation settings. Generally, coaches and support staff agreed that monetary cost, coach buy-in and limited evidence base were barriers towards its use. In a sub-sample of coaches and support staff without access to virtual reality (n = 123), performance expectancy was the strongest construct in explaining behavioural intention to use virtual reality, followed by facilitating conditions (i.e., barriers) construct which had a negative association with behavioural intention. Virtual reality has the potential to be a valuable technology within professional soccer although several barriers exist that may prevent its widespread use.

ABSTRACT

Despite various solutions proposed to solve the relative age effect (RAE), it is still a major problem confounding talent identification and selection processes. In the first phase, we sampled 302 under 7-21 academy soccer players from two Belgian professional soccer clubs to explore the potential of a new approach to solve the inequalities resulting from relative age- and maturity-related bias. This approach allocates players into four discrete quartile groups based on the midway point of their chronological and estimated developmental (ED) birth dates (calculated using the growth curves for stature of Belgian youth). With the use of chi square analyses, a RAE was found (p < 0.01) for the overall sample (Q1 = 41.4% vs. Q4 = 14.9%) that completely disappeared after reallocation (Q1 = 26.5%; Q2 = 21.9%; Q3 = 27.5%; Q4 = 24.2%). According to the new allocation method, the stature difference was reduced, on average, by 11.6 cm (from 24.0 ± 9.9 to 12.4 ± 3.4 cm, d = 1.57). Body mass difference between the two methods was 1.9 kg (20.1 ± 11.3-18.2 ± 13.1 kg, respectively, d = 0.15). The new method created a maximum chronological age difference of 1.9 vs. 0.8 years for the current method. With the use of this method, 47% of the players would be reallocated. Twenty-three percent would be moved up one age category, and 21% would be moved down. In the second phase, we also examined 80 UK academy soccer players to explore if reallocating players reduces the within-playing group variation of somatic and physical fitness characteristics. The percentage coefficient of variation (%CV) was reduced (0.2-10.1%) in 15 out of 20 metrics across U11-U16 age categories, with the U13 age category demonstrating the largest reductions (0.9-10.1%) in CV. The U12 and U13 age categories and associated reallocation groupings showed trivial to small (ES = 0.0-0.5) between-method differences and trivial to moderate (ES = 0.0-1.1) differences within the U14-U16 age categories. A reduction in RAE may lead to fewer dropouts and thus a larger player pool, which benefits, in turn, talent identification, selection, and development.

ABSTRACT

Biological maturation can be defined as the timing and tempo of progress to achieving a mature state. The estimation of age of peak height velocity (PHV) or percentage of final estimated adult stature attainment (%EASA) is typically used to inform the training process in young athletes. In youth soccer, maturity-related changes in anthropometric and physical fitness characteristics are diverse among individuals, particularly around PHV. During this time, players are also at an increased risk of sustaining an overuse or growth-related injury. As a result, the implementation of training interventions can be challenging. The purpose of this review was to (1) highlight and discuss many of the methods that can be used to estimate maturation in the applied setting and (2) discuss the implications of manipulating training load around PHV on physical development and injury risk. We have provided key stakeholders with a practical online tool for estimating player maturation status (Supplementary Maturity Estimation Tools). Whilst estimating maturity using predictive equations is useful in guiding the training process, practitioners should be aware of its limitations. To increase the accuracy and usefulness of data, it is also vital that sports scientists implement reliable testing protocols at predetermined time-points.

ABSTRACT

Research has demonstrated that induced mental fatigue impairs soccer-specific technical, tactical and physical performance in soccer players. The findings are limited by the lack of elite players and low ecological validity of the tasks used to induce mental fatigue, which do not resemble the cognitive demands of soccer. The current study collected survey data from English academy soccer players (n = 256; age groups - U14 - U23), with questions comprising of five themes (descriptors of physical and mental fatigue, travel, education, match-play and fixture congestion). The survey consisted of multiple choice responses, checkboxes and blinded/unblinded (for duration based questions) 0-100 arbitrary unit (AU) slider scales. Listening to music (81.6% of players), using social media (58.3%) and watching videos (34.3%) were the most common pre-match activities. Pre-match subjective mental fatigue was low (18.7±18.8 AU), and most frequently reported at the end of a match (47±26 AU) and remained elevated 24-hours post-match (36±27 AU). Travel (29±24 AU), fixture congestion (44±25 AU) and education (30±26 AU) demonstrated a low to moderate presence of subjective mental fatigue. These findings provide an overview of activities performed by English academy soccer players pre-match, and demonstrate that mental fatigue is experienced as a result of match-play.

ABSTRACT

The purpose of the study was to examine the perspectives of both academics and practitioners in relation to forming applied collaborative sport science research within team sports. Ninety-three participants who had previously engaged in collaborative research partnerships within team sports completed an online survey which focused on motivations and barriers for forming collaborations using blinded sliding scale (0-100) and rank order list. Research collaborations were mainly formed to improve the team performance (Academic: 73.6 ± 23.3; Practitioner: 84.3 ± 16.0; effect size (ES = 0.54), small). Academics ranked journal articles' importance significantly higher than practitioners did (Academic: Mrank = 53.9; Practitioner: 36.0; z = -3.18, p = .001, p < q). However, practitioners rated one-to-one communication as more preferential (Academic: Mrank = 41.3; Practitioner 56.1; z = -2.62, p = .009, p < q). Some potential barriers were found in terms of staff buy in (Academic: 70.0 ± 25.5; Practitioner: 56.8 ± 27.3; ES = 0.50, small) and funding (Academic: 68.0 ± 24.9; Practitioner: 67.5 ± 28.0; ES = 0.02, trivial). Both groups revealed low motivation for invasive mechanistic research (Academic: 36.3 ± 24.2; Practitioner: 36.4 ± 27.5; ES = 0.01, trivial), with practitioners have a preference towards 'fast' type research. There was a general agreement between academics and practitioners for forming research collaborations. Some potential barriers still exist (e.g. staff buy in and funding), with practitioners preferring 'fast' informal research dissemination compared to the 'slow' quality control approach of academics.

ABSTRACT

This study assessed the contribution of relative age, anthropometry, maturation, and physical fitness characteristics on soccer playing position (goalkeeper [GK], central-defender [CD], lateral-defender [LD], central-midfield [CM], lateral-midfielder [LM], and forward [FWD]) for 465 elite-youth players (U13-U18's). U13-14 CD were relatively older than LD and CM (likely small effects). CD and GK were generally taller and heavier (likely small to very-likely moderate effects) than other players at each developmental stage and were advanced maturers at U13-14 (very-likely small to likely moderate effects). GK had inferior agility (very-likely small to likely moderate effects), endurance (very-likely small to likely moderate effects), and sprint capacities (likely small-moderate effects) vs. outfield positions at U13-14, but deficits in anaerobic phenotypes were diminished in U15-16 and U17-18. Position specific fitness characteristics were distinguished at U15-16 (likely small) and U17-18 (likely moderate), where LM were faster than their central counterparts. In summary, relative age, maturation and anthropometric characteristics appear to bias the allocation of players into key defensive roles from an early development stage, whereas position-specific physical attributes do not become apparent until the latter stages of talent development in outfield players. Given the inter-individual trajectories of physical development according to biological maturation, playing position allocation might be considered 'plastic' by selectors, until complete-maturity is achieved.

The relative age effect (RAE) and its relationships with maturation, anthropometry, and physical performance characteristics were examined across a representative sample of English youth soccer development programmes. Birth dates of 1,212 players, chronologically age-grouped (i.e., U9's-U18's), representing 17 professional clubs (i.e., playing in Leagues 1 & 2) were obtained and categorised into relative age quartiles from the start of the selection year (Q1 = Sep-Nov; Q2 = Dec-Feb; Q3 = Mar-May; Q4 = Jun-Aug). Players were measured for somatic maturation and performed a battery of physical tests to determine aerobic fitness (Multi-Stage Fitness Test [MSFT]), Maximal Vertical Jump (MVJ), sprint (10 & 20m), and agility (T-Test) performance capabilities. Odds ratio's (OR) revealed Q1 players were 5.3 times (95% confidence intervals [CI]: 4.08-6.83) more likely to be selected than Q4's, with a particularly strong RAE bias observed in U9 (OR: 5.56) and U13-U16 squads (OR: 5.45-6.13). Multivariate statistical models identified few between quartile differences in anthropometric and fitness characteristics, and confirmed chronological age-group and estimated age at peak height velocity (APHV) as covariates. Assessment of practical significance using magnitude-based inferences demonstrated body size advantages in relatively older players (Q1 vs. Q4) that were very-likely small (Effect Size [ES]: 0.53-0.57), and likely to very-likely moderate (ES: 0.62-0.72) in U12 and U14 squads, respectively. Relatively older U12-U14 players also demonstrated small advantages in 10m (ES: 0.31-0.45) and 20m sprint performance (ES: 0.36-0.46). The data identify a strong RAE bias at the entry-point to English soccer developmental programmes. RAE was also stronger circa-PHV, and relatively older players demonstrated anaerobic performance advantages during the pubescent period. Talent selectors should consider motor function and maturation status assessments to avoid premature and unwarranted drop-out of soccer players within youth development programmes.

BACKGROUND: As most sport drinks contain some form of non-nutritive sweetener (e.g. aspartame), and with the variation in blood glucose regulation and insulin secretion reportedly associated with aspartame, a further understanding of the effects on insulin and blood glucose regulation during exercise is warranted. Therefore, the aim of this preliminary study was to profile the insulin and blood glucose responses in healthy individuals after aspartame and carbohydrate ingestion during rest and exercise. FINDINGS: Each participant completed four trials under the same conditions (45 min rest + 60 min self-paced intense exercise) differing only in their fluid intake: 1) carbohydrate (2% maltodextrin and 5% sucrose (C)); 2) 0.04% aspartame with 2% maltodextrin and 5% sucrose (CA)); 3) water (W); and 4) aspartame (0.04% aspartame with 2% maltodextrin (A)). Insulin levels dropped significantly for CA versus C alone (43%) between pre-exercise and 30 min, while W and A insulin levels did not differ between these time points. CONCLUSIONS: Aspartame with carbohydrate significantly lowered insulin levels during exercise versus carbohydrate alone.

ABSTRACT

Although a considerable amount of literature exists on the ergogenic potential of ingesting sodium bicarbonate (NaHCO3) before short-term, high-intensity exercise, very little exists on optimal loading times before exercise. The purpose of this study was to determine the influence of NaHCO3 supplementation timing on repeated sprint ability (RSA). Eight men completed 3 (randomized and counterbalanced) trials of ten 10-second sprints separated by 50 seconds of active recovery (1:5 work-to-rest) on a nonmotorized treadmill. Before each trial, the subjects ingested 0.3 g·kg(-1) body weight of NaHCO3 at 60 (H1), 120 (H2), or 180 (H3) minutes before exercise. Additionally, the subjects were assessed for any side effects (gastrointestinal [GI] discomfort) from the NaHCO3 ingestion via a visual analog scale (VAS). Blood buffering was assessed using a 2-way analysis of variance (ANOVA) with repeated measures, whereas repeated sprint performance and GI discomfort were assessed via a 1-way ANOVA with repeated measures. Blood-buffering capacity was not different at preexercise times (HCO3(-) [millimoles per liter] H1: 30.2 ± 0.4, H2: 30.9 ± 0.6, H3: 31.2 ± 0.6; p > 0.74). Average speed, average power, and total distance covered progressively declined over the 10 sprints; however, there was no difference between conditions (p > 0.22). The incidence of GI discomfort was significantly higher (p < 0.05) from preingestion at all time points with the exception of 180 minutes, whereas severity was only different between 90 and 180 minutes. Ingestion times (between 60 and 180 minutes) did not influence the blood buffering or the ergogenic potential of NaHCO3 as assessed by RSA. However, VAS scores indicated that at 180 minutes postingestion, an individual is less prone to experiencing significant GI discomfort.

ABSTRACT

BACKGROUND: Transmissible Spongiform Encephalopathies (TSEs) are a group of progressive fatal neurodegenerative disorders, triggered by abnormal folding of the endogenous prion protein molecule. The encoding gene is a major biological factor influencing the length of the asymptomatic period after infection. It remains unclear the extent to which the variation between quantitative trait loci (QTLs) reported in mouse models is due to methodological differences between approaches or genuine differences between traits. With this in mind, our approach to identifying genetic factors has sought to extend the linkage mapping approach traditionally applied, to a series of additional traits, while minimising methodological variability between them. Our approach allows estimations of heritability to be derived, as well as predictions to be made about possible existence of genetic overlap between the various traits. METHODOLOGY/PRINCIPAL FINDINGS: Our data indicate a surprising degree of heritability (up to 60%). Correlations between traits are also identified. A series of QTLs on chromosomes 1, 2, 3, 4, 6, 11 and 18 accompany our heritability estimates. However, only a locus on chromosome 11 has a general effect across all 4 models explored. CONCLUSIONS/SIGNIFICANCE: We have achieved some success in detecting novel and pre-existing QTLs associated with incubation time. However, aside from the general effects described, the model-specific nature of the broader host genetic architecture has also been brought into clearer focus. This suggests that genetic overlap can only partially account for the general heritability of incubation time when factors, such as the nature of the TSE agent and the route of administration are considered. This point is highly relevant to vCJD (a potential threat to public health) where the route of primary importance is oral, while the QTLs being sought derive exclusively from studies of the ic route. Our results highlight the limitations of a single-model approach to QTL-mapping of TSEs.
