ABSTRACT

OBJECTIVE: Rat liver fatty acid binding protein (FABP) is believed relevant to understanding of homeostasis in lipid metabolism and lipid related diseases. Relatively little is known about endocrine control of FABP production. Thus, we examined endocrine effects on its hepatic content. METHODS/RESULTS: Hypophysectomy of 300-325 g males caused statistically significant drops of FABP levels averaging 62.2% and 67.0%, expressed g/liver or 100 g/body weight, 30-52 days after surgery. Cortisol administration (3.8 mg/kg, daily, 32-36 days) did not significantly alter this effect of hypophysectomy. Recombinant human growth hormone (GH, 2.0 U/kg, b.i.d, 17-20 days) greatly decreased the effect of hypophysectomy on FABP but had no effect in intact males. Supporting the control of FABP content by GH, FABP levels decreased significantly in 12-13 and 16-22 month old males, but not in growing, 4-6 or 10-11 month old males. FABP levels in 12-13.5 month old females also dropped significantly compared to 4-6 month old females. DISCUSSION: The importance of the data to metabolism, growth, and aging is discussed.

ABSTRACT

A 43-year-old man with dysphagia and a tendency to aspirate was found to have squamous cell carcinoma of the esophagus. Curative surgery was planned and preoperative computed tomography of the chest and abdomen was ordered. A ward nurse administered Gastrografin according to a "standing" order, 4 hours before the CT was to be done. The patient aspirated about 50 mL of Gastrografin, and went into cardiorespiratory arrest caused by pulmonary edema. He sustained severe brain damage and died. This is a first report of lethal aspiration of Gastrografin, given in preparation for CT. We advise alerting nurses who administer Gastrografin, especially to patients with dysphagia or impaired consciousness about the grave consequences that can result if the contrast agent is aspirated by the patient. We further advise that responsibility for using contrast agents in radiologic procedures be assumed by the radiologist and not by medical house staff.

ABSTRACT

Sulfation of hepatotoxic monohydroxy bile salts is viewed as an important detoxification mechanism. Bile salts are bound by fatty acid binding protein (FABP) with decreasing affinity as the extent of their hydroxylation increases. This binding has the potential to interfere with sulfation of monohydroxy bile salts and to augment their toxicity. FABP inhibits monohydroxy bile salt sulfation via bile salt sulfotransferases BST 1 and 2. With BST 1, the main BST, we obtained a maximal reduction of sulfation by 42.8 +/- 8.1%, using 10 microM glycolithocholate as substrate. FABP had no effect on sulfation of either 10 microM glycodeoxycholate or glycochenodeoxycholate. FABP may therefore specifically alter hepatotoxicity of lithocholate and its metabolites.

ABSTRACT

Feeding rats a diet deficient in choline results in fatty liver within 1 d. We studied the effect of short-term (1-3 d) choline deficiency on rat liver Z protein (fatty acid-binding protein). Groups of three females Sprague-Dawley rats were fed ad libitum a purified diet lacking choline and L-methionine or were supplemented with 0.2% choline chloride and 0.82% L-methionine. Animals were killed after 1, 2 or 3 d of consuming control or experimental diets and hepatic Z protein was prepared. Z protein in livers from experimental and control rats were estimated with the fluorescent probe dansylamino undecanoic acid. The corresponding fatty acid-binding activity was also determined. One day of choline-deficient diet increased Z protein concentration threefold, reaching a plateau on the second and third day. Fatty acid-binding activity of Z protein remained unchanged.

Z protein or fatty-acid-binding protein is abundant in the cytosol of many cell types including liver cells. It is considered to play an important role in intracellular transport and metabolism of long-chain fatty acids and other organic anions. We studied the role of Z protein in the metabolism of prostaglandin E1 (PGE1). Binding of tritiated prostaglandin E1 to this fatty-acid-binding protein (Z protein) purified from rat liver was determined. The binding of [3H]prostaglandin E1 to Z protein is rapid, saturable and reversible. Scatchard analysis of [3H]PGE1 binding to Z protein showed a single class of binding sites with a dissociation constant (Kd) of 37 nM. The binding capacity is 110 nmol/mg Z protein. Optimal [3H]PGE1 binding occurred at pH 7.4. The presence of 3 mM MgCl2 stimulated the prostaglandin E1 binding to Z protein. Competition experiments show that the binding of this autacoid to Z protein is highly specific. It could not be displaced by other prostaglandins (PGA1, PGA2, PGE2, PGB1, PGI2, PGD2, PGF2 alpha, and 6-keto PGF1 alpha). Z protein might be involved in the metabolism of prostaglandins in the cytosol.

ABSTRACT

Report on two cases of jaundice developing shortly after ingestion of a new laxative, Normolaxol (quinolylmethylenediphenol). Clinical and morphological aspects of the acute cholestasis are described and discussed regarding differential diagnosis and pathogenesis.
