ABSTRACT

Bipolar disorder has been associated with a decrease in hippocampal size, and lithium appears to reverse this neuroanatomical abnormality. The objective of this work was to evaluate, at a cellular level, the size of both cell body and nucleus of pyramidal neurons located throughout the Cornu Ammonis (CA1 to CA4 regions). To perform this duty, we used 16 rats that were randomized into two groups: control and dietary lithium-treated. After one month, they were sacrificed and their brains removed for histopathological analysis. Serial photos of the entire Cornu Ammonis were taken and, after dividing them into 4 regions of interest, we measured the cell body and nucleus on each pyramidal neuron belonging to the first 5 photos of each region of interest. As a result of this histological analysis, cell body area and nuclear area were significantly larger in the experimental group in a specific area of the Cornu Ammonis that could correspond to CA2 or the transition between CA1 and CA2. These results suggest that the effect of lithium is not homogeneous throughout the hippocampus and allows directing future studies to a specific area of this structure.

ABSTRACT

The control of pre-analytical-factors in human biospecimens collected for health research is currently required. Only two previous reports using post-mortem brain samples have tried to address the impact of cold-ischemia on tissue pH. Here we report pH variations according to time (third-order polynomial model) in mice for liver, kidney and lung samples. Tissue alkalosis in cold-ischemia time may be an underlying mechanism of gene expression changes. Therefore, tissue-pH regulation after organ removal may minimize biological stress in human tissue samples.

ABSTRACT

El presente trabajo se realizó en el HNERM, entre Setiembre de 1996 a Julio de 1998. Se realizaron 61 procedimientos quirúrgicos en animales de experimentación, 39 en canes y 22 en cerdos. De estos procedimientos 14 fueron transplante ortotópico de hígado. (TOH), 7 en canes y 7 en cerdos. Durante la fase anhepática se utilizó en 6 animales el bypass veno-venoso con bomba centrífuga sin heparinización sistémica y en 8 se utilizó el bypass veno-venoso pasivo con heparinización sistémica. Los animales fueron sacrificados 2 horas después de concluído el procedimiento, para precisar el estado de la anastomosis y la presencia o no de trombos.

ABSTRACT

This study was carried at at the Hospital Nacional Edgardo Rebagliati Martins, from September 1996 to July 1998. 61 surgical procedures were performed in experimental animals, 39 in dogs and 22 in pigs. During the anhepatic phase, we used veno-venous bypass with centrifugal pumps and without systemic anticoagulation drugs in 6 animals, and passive venovenous bypass with systemic anticoagulation drugs in 8 animals. Experimental animals were sacrified two hours after the procedures were concluded in order to assess the status of vascular anastomosis and thrombus formation.

ABSTRACT

An enzyme-linked immunosorbent assay (ELISA) was performed for diagnosis of paratuberculosis in goats, using as antigen a protoplasmic extract (PPA-3). The test was developed on the basis of the results obtained with two serum reference pools, positive and negative respectively. To avoid day-to-day variations, dilutions of the positive serum pool were included in each plate to obtain an arbitrary system, transforming absorbance into immunoglobulin (Ig) G anti-Mycobacterium paratuberculosis units. The ELISA was used on sera of two reference groups of animals. One group consisted of 35 goats suspected of being infected with paratuberculosis, which was confirmed by histological findings and isolation of M. paratuberculosis. The negative group consisted of 61 healthy goats from a farm free of paratuberculosis. The test showed a sensitivity of 100% and a specificity of 91.8%. Absorption of sera with a Mycobacterium phlei suspension did not modify either the sensitivity or the specificity of the test. Sera from the negative group were analysed by Western blotting, and four of them recognized two fractions with a molecular weight of 17.3 and 28.1 kDa.

ABSTRACT

The effect of increasing doses of 6-methyl-prednisolone on the ingestion-microbicidal function activity of dog neutrophils was studied. Histochemical and spectrophotometrical NAT reduction techniques, together with a direct overall method using C. albicans as a test micro-organism were carried out. Samples were analysed 2 and 48 h after inoculation of different doses. The histochemical NAT technique was not very precise, but quantification of responses using the spectrophotometric method revealed that at 48 h.p.i. neutrophil reducing activity declines continuously. At 2 h.p.i. in the first few days of the experiment (i.e. at lower doses), NAT reduction increased, but subsequently fell back to O.D. levels below those recorded at 48 h.p.i. Ingestion and destruction of C. albicans followed the same pattern as that recorded for NAT activity. Simple and differential leukocyte counts performed in all samples revealed leukocytosis (with neutrophilia and lymphopaenia) at 2 h.p.i. This returned to normal levels in the intervals between inoculations.
