ABSTRACT

The myeloma surface proteome (surfaceome) determines tumor interaction with the microenvironment and serves as an emerging arena for therapeutic development. Here, we use glycoprotein capture proteomics to define the myeloma surfaceome at baseline, in drug resistance, and in response to acute drug treatment. We provide a scoring system for surface antigens and identify CCR10 as a promising target in this disease expressed widely on malignant plasma cells. We engineer proof-of-principle chimeric antigen receptor (CAR) T-cells targeting CCR10 using its natural ligand CCL27. In myeloma models we identify proteins that could serve as markers of resistance to bortezomib and lenalidomide, including CD53, CD10, EVI2B, and CD33. We find that acute lenalidomide treatment increases activity of MUC1-targeting CAR-T cells through antigen upregulation. Finally, we develop a miniaturized surface proteomic protocol for profiling primary plasma cell samples with low inputs. These approaches and datasets may contribute to the biological, therapeutic, and diagnostic understanding of myeloma.

ABSTRACT

Neoleucinodes elegantalis (Guenée) is an oligophagous species of plants in the Solanaceae family that has a broad geographical distribution in the tropical zones of South America. It is the most important insect pest of naranjilla (Solanum quitoense Lamarck), a crop grown in threatened areas of the tropical old-growth forest in Ecuador. In this study, two host-specific populations of N. elegantalis were collected from infested fruit of naranjilla and tree tomato (Solanum betaceum Cavanilles) in different locations. Sexually virgin adult insects (93 females and 103 males) were dissected to extract their genitalia to measure 12 morphological variables in females and six in males, resulting in six and four informative variables respectively. Using univariate and multivariate analysis of variance, it was found that the Solanaceous host was the main factor differentiating the area measurements of the seventh abdominal segment and ostium bursae in female genitalia, and cornuti length in male genitalia. Principal components generated with these measurements were employed in a logistic regression model for the classification of the Solanaceous host. Female genitalia of individuals from S. betaceum showed significantly larger ostium bursae relative to female genitalia from S. quitoense. For males, individuals collected from S. betaceum showed longer cornuti length than samples collected from S. quitoense. The results suggest genotypic differentiation according to the Solanaceous host or phenotypic plasticity in N. elegantalis. Further molecular and bio-geographical studies are needed to properly differentiate N. elegantalis populations that would help in the control of this pest.

ABSTRACT

Se presenta el caso clínico de un paciente que presenta estenosis esofágica tras ingesta de caústicos, con disfagia a líquidos por lo que se realizó esofagectomía transhiatal con técnica de tubo gástrico como alterna-tiva para reemplazo esofágico con ascenso mediastinal y yeyunostomía de alimentación, luego del procedimiento presenta fístula a nivel de la anastomosis proximal cervical y derrame pleural que se drena mediante avenamiento pleural bilateral, se maneja de manera conservadora sin desfuncionalización la fistula cervical mediante dren semirígido, con cierre espontaneo y se da de alta al paciente luego de tolerar dieta.


The clinical case of a patient with esophageal stenosis after caustic ingestion is presented with dysphagia to liquids, so a transhiatal esophagectomy was performed with gastric tube technique alternatively for esophageal replacement mediastinal rise and feeding jejunostomy. After the procedure, it has a fistula in the proximal anastomosis cervical level and a pleural effusion is drained through bilateral pleural drainage. It is managed conservatively without disfunctionalization of the cervical fistula by semi-rigid drain with spontaneous closure and the patient is discharged after tolerated diet.

ABSTRACT

La presente investigacion tuvo como objetivos determinar el estado actual de las capacidades fisicas y del perfil antropometrico en los estudiantes de la Institucion Educativa Rafael J. Mejiadel municipio de Sabaneta, departamento de Antioquia, Colombia, y determinar la relacion entre estas variables. Se selecciono aleatoriamente una muestra estratificada de 207 alumnos (99 ninos 44%; 108 ninas, 56%) de un total de 455 escolares de la Institucion Educativa. Seaplicaron los procedimientos de medicion y de evaluacion propuestos por J uregui y Ordonez(1993). Los resultados mostraron que una considerable proporcion de la muestra estuvo porencima del percentil 75 para las pruebas antropometricas y de los pliegues subcut neos, y pordebajo del percentil 25 en las capacidades fisicas. Dichos resultados permiten concluir que existe una tendencia de estos ninos hacia la obesidad y el sedentarismo.


This study aims to determine the current state of Physical skills and anthropometric profile, and the relation between these variables, in students of the Rafael J. Mejia School, in Sabaneta. Astratified simple of 207 students (99 boys, 44% 108 girls, 56%) was randomly selected from atotal of 455 students. We applied measurement procedures and evaluation proposed by J uregui and Ordo¤ez (1993). The results showed that a considerable proportion of the sample was above the 75th percentile for anthropometric tests and skinfolds; below the 25th percentile in physical abilities. This suggests that there is a tendency in these children to obesity and physical inactivity.
