ABSTRACT

The genus Psidium, belonging to the family Myrtaceae, encompasses several plant species rich in essential oils. Several studies involving different research areas have shown promising results for essential oils, which has attracted interest from diverse sectors, including pharmaceutical, food, and cosmetic industries. Thus, the objective of this study was to evaluate the biological activity of the essential oil extracted from Psidium bahianum leaves (EOPb) collected in an Atlantic Forest remnant in the northern coast of the state of Bahia, Brazil. EOPb was extracted by steam distillation and analyzed through proton nuclear magnetic resonance (1H-NMR). The free radical scavenging activity was assessed using the DPPH methodology (analyzing the scavenging of the stable free radical 1,1-diphenyl-2-picrylhydrazyl). Antimicrobial disk diffusion assays were conducted; toxicity was determined through assays on sheep erythrocytes and Artemia salina. 1H-NMR evaluation showed the presence mainly of monoterpenes and sesquisterpenes. The percentage of antioxidant activity was 18.03±2.53 53 for 125 125 µg mL-1. Antimicrobial assays showed that the essential oil at a concentration of 10 mg mL-1 inhibited the growth of the microorganisms Staphylococcus aureus, Bacillus subtilis, B. cereus, Micrococcus luteus, Escherichia coli, Aspergillus niger, Candida albicans, and C. glabrata. The toxicity results indicated that concentrations of EOPb at 10, 100, 250, and 500 µg mL-1 were not toxic to sheep erythrocytes. The toxicity assay on A. salina showed that the lethal concentration for 50% of individuals (LC50) within 24 and 48 hours were 371.48 and 302 µg mL-1 of EOPb, respectively, which were considered moderately toxic, whereas the LC50 within 72 horas was 33.11 µg mL-1, considered highly toxic. These findings are promising, as the essential oil from P. bahianum leaves showed antimicrobial activity against both Gram-positive and Gram-negative bacteria and fungi. The data obtained for the essential oil toxicity in A. salina could be tested on different tumor cell lines.

ABSTRACT

The indiscriminate use of allopathic drugs has selected resistant bacterial and fungal populations which represents a severe public health problem worldwide. On the other hand, plants are in a prominent position due to the capability to synthesize structurally complex bioactive metabolites that can be an alternative to resistant microorganisms' control. In this work, we evaluated the chemical composition and the antimicrobial, antioxidant, and cytotoxic potential of the fractionated extract of C. savannarum in ethyl acetate. The extract of C. savannarum was divided into 12 fractions that were submitted to phytochemical screening, minimum inhibitory concentration (MIC), reduction of 1,1-diphenyl-2-picrylhydrazine (DPPH), and hemolytic activity of sheep erythrocytes assays. During the investigation, all extract fractions presented alkaloids, triterpenoids, steroids, and phenolic compounds in qualitative analyses, while in the quantitative evaluation, we observed the presence of both phenols and flavonoids in these fractions. Among the fraction, the highest phenolic content was observed in the Cs23-24 fraction (2.480 mg EAG/g), while the Cs31-34 fractions presented the highest amount of flavonoid (182.25 µg EQ/100 mg). Nine of the 12 fractions of the moss species' extract showed antimicrobial action Against Gram-positive bacteria: Bacillus cereus, Staphylococcus aureus, Micrococcus luteus, Bacillus subtilis; Gram-negative bacteria: Pseudomonas aeruginosa; and also antifungal activity against Candida albicans and Candida glabata. The cytotoxic assay demonstrated that the tested fractions did not induce hemolysis at concentrations 10 and 100(µG/ML). In the antioxidant evaluation, the Cs55-69 fractions were the ones that presented the highest scavenging activity (57, 0%) followed by the Cs45-54 fraction (42.7%). Overall, the evaluation of the biological potential of the fractionated extracts of Campylopus savannarum showed promising data, in the search for natural antimicrobial compounds.

Plants possess a renewable source of metabolites with enormous chemical structural diversity, which may have potential therapeutic relevance. Furthermore, this chemical diversity favors the possibility of finding new and different chemical constituents with antimicrobial, antioxidant and anti-tumor activities. This work analyzed preliminary phytochemical profiles and evaluated the antimicrobial, antioxidant and cytotoxic activities of hexane extracts of leaves of ten species of the family Melastomataceae. Phytochemical screening was performed using staining methods while total phenols and flavonoids were quantified by spectrophotometry. Antimicrobial activity was evaluated using the disk diffusion method. Antioxidant activity was determined by the 2,2-diphenyl-1-picrylhydrazil (DPPH) method. Toxicity was recorded using the lethality test with Artemia salina Leach (1819). Cytotoxic activity of the extracts was assessed in vitro with acute monocytic leukemia cells (THP-1). Phytochemical analysis detected the presence of tannins, terpenes, steroids, polyphenols and flavonoids and the absence of alkaloids. Clidemia capitellata (Bonpl.) D. Don had the greatest amount of polyphenols (205.95 mg/g ± 4.14) while Clidemia hirta (L.) D. Don had the highest content of total flavonoids (143.99 mg/g ± 4.18). The hexane extracts did not show antimicrobial activity nor toxicity against Artemia salina. The extract of Tibouchina francavillana Cogn. was the most active in sequestering the DPPH radical. The extracts showed cytotoxicity in THP-1 cells with the appearance of apoptotic bodies and cell death. The extracts of Miconia amoena, Clidemia sericea and Clidemia capitellata are non-toxic against Artemia salina and induce the formation of apoptotic bodies and cell death of the THP-1 lineage.


Os vegetais apresentam uma fonte renovável de metabólitos com enorme diversidade química estrutural, os quais podem apresentar potencial relevante na terapêutica, aumentando as possibilidades de encontrar novos e diferentes constituintes químicos com atividades antimicrobiana, antioxidante e antitumoral. Este trabalho analisou o perfil fitoquímico preliminar e as atividades antimicrobiana, antioxidante, citotóxica dos extratos em hexano das folhas de dez espécies da família Melastomataceae. A triagem fitoquímica foi executada utilizando métodos de coloração e quantificação de fenóis e flavonoides totais por espectrofotometria. A atividade antimicrobiana foi realizada pelo método de difusão em disco. A atividade antioxidante foi determinada pelo método 2,2-difenil1-picrilhidrazila (DPPH). A toxicidade foi registrada utilizando o ensaio de letalidade com Artemia salina Leach (1819). A atividade citotóxica dos extratos foi realizada in vitro com células leucêmicas monocítica aguda (THP-1). A análise fitoquímica detectou a presença de taninos, terpenos, esteroides, polifenóis, flavonoides e ausência de alcaloides. A maior quantificação de polifenóis foi da Clidemia capitellata (Bonpl.) D. Don (205,95 mg/g ± 4,14) e o extrato de Clidemia hirta (L.) D. Don apresentou maior teor de flavonoides totais (143,99 mg/g ± 4,18). Os extratos hexânicos não demostraram atividade antimicrobiana e nem toxicidade frente à Artemia salina. O extrato de Tibouchina francavillana Cogn. foi o mais ativo no sequestro do radical DPPH. Os extratos apresentaram citotoxicidade em células THP-1, com visualização de corpos apoptóticos e morte celular. Os extratos de Miconia amoena, Clidemia sericea e Clidemia capitellata são atóxicos contra Artemia salina e induzem a formação de corpos apoptóticos e morte celular da linhagem THP-1.

This study aimed to evaluate aspects of host immune response using an experimental infection model of Corynebacterium pseudotuberculosis (CP) in C57/Black6 wild-type and knockout for nitric oxide (KO-NO) mice. 28 mice were evaluated: 4 wild-type controls; 10 wild-type infected with CP; 4 KO-NO controls; 10 KO-NO infected with CP. Infection procedures were carried out by intraperitoneal inoculation using 107. Infected C57/Black6 KO-NO mice began to die after the 5° day post-inoculation, up until the 14º day. Neutrophils were found in increased numbers in the infiltrate of KO-NO murine peritoneal cavities. Examination of splenic tissue revealed an accumulation of lymphocytes, predominantly CD8 T-cells, in experimental animal groups. KO-NO animals were found to have a predominance of granulomas 7 days post-inoculation, primarily in the lymph nodes. In addition, greater amounts of bacteria were recovered from the mesenteric lymph nodes of KO-NO mice. There was no statistically significant difference in the levels of total IgG and its subclasses 14 days post-inoculation between KO-NO and wild groups. The results suggest the importance of nitric oxide in the process of controlling CP infection, as KO-NO animals were observed to be markedly more affected by infection with this bacterium.(AU)


O objetivo deste estudo foi avaliar os aspectos da resposta imune do hospedeiro, mediante o uso de um modelo experimental de infecção de Corynebacterium pseudotuberculosis (CP) em camundongos C57/Black6 do tipo selvagem e em C57/Black6 knockout para o óxido nítrico (KO-NO). Foram avaliados 28 camundongos: quatro controles de tipo selvagem; 10 do tipo selvagem infectados com CP; quatro controles KO-NO; e 10 KO-NO infectados com CP. A infecção foi realizada via intraperitoneal, usando-se 107. Os animais C57/Black6 KO-NO infectados começaram a vir a óbito no quinto dia pós-inoculação, o que aconteceu até o 14º dia. Um número maior de neutrófilos foi encontrado na sua cavidade peritoneal. O exame do baço revelou um acúmulo de linfócitos, predominantemente células T CD8, nos grupos de animais experimentais. Nos animais KO-NO, foi observada a presença de granulomas, sete dias pós-inoculação, principalmente nos gânglios linfáticos. Além disso, uma maior quantidade de bactérias foi detectada dos linfonodos mesentéricos desses animais. Não houve diferença estatisticamente significante nos níveis séricos IgG total e em suas subclasses aos 14 dias pós-inoculação nos grupos KO-NO e selvagem. Os resultados obtidos sugerem a importância do óxido nítrico no processo de controle da infecção por CP.(AU)

This study aimed to evaluate the variation in the levels of proline, oxidative metabolism and photosynthetic pigments in plants of Pitcairnia encholirioides grown in vitro under different conditions and after acclimatization. The analyses were performed after 150 days of in vitro cultivation in MS media supplemented with 10 µM GA3 or 0.2 µM NAA, sucrose at 15 or 30 g L-1, in test tubes which allowed gas exchange or in a hermetically sealed system, and 180 days after acclimatization. The in vitro maintenance in hermetically sealed flasks, with GA3 and 15 g L-1 sucrose had adverse metabolic effects, which was demonstrated by the lower proline and photosynthetic pigments accumulation and by the increase in antioxidant enzymes activities. After acclimatization, differences for proline and photosynthetic pigments were no longer found and the enzymatic activities ranged unevenly. The results suggest that the in vitro cultivation in media with 0.2 µM NAA and 30 g L-1 sucrose, in test tubes capped with closures which allowed gas exchange, is more suitable for micropropagation of P. encholirioides, providing a prolonged maintenance of in vitro cultures and plantlets with superior quality for ex vitro development.

Resumo Este trabalho objetivou avaliar a contribuição da prolina, do metabolismo oxidativo e dos pigmentos fotossintéticos na propagação in vitro e aclimatização de Pitcairnia encholirioides, uma bromélia criticamente ameaçada de extinção. As análises foram realizadas após 150 dias de cultivo in vitro em meio MS suplementado com 10 µM de GA3 ou 0,2 µM de ANA, 15 ou 30 g L–1 de sacarose, em tubos de ensaio que permitiam trocas gasosas ou em sistema hermeticamente vedado, e também 180 dias após aclimatização. A manutenção in vitro em frascos hermeticamente fechados, com GA3 e 15 g L–1 de sacarose apresentou efeito metabólico adverso, demonstrado pelo menor acúmulo de prolina e pigmentos fotossintéticos e também pelo aumento das atividades de enzimas antioxidantes. Após aclimatização, as diferenças para prolina e pigmentos fotossintéticos não foram mais encontradas e as atividades enzimáticas variaram de maneira desuniforme. Os resultados sugerem que o cultivo in vitro em meio com 0,2 µM de ANA e 30 g L–1 de sacarose, em tubos fechados com tampas que permitem trocas gasosas, é mais adequado para a micropropagação de P. encholirioides, proporcionando uma manutenção prolongada das culturas in vitro e plântulas com qualidade superior para o desenvolvimento ex vitro.

ABSTRACT

Dams are of paramount importance to a wide variety of human services and many of their environmental problems are known; however, there are few studies in the world addressing the impacts on the native vegetation previously distant from water bodies which became close to the lakeshore created by a dam. Thus, this paper aims to analyze the responses of a dry forest to a dam after 15 years. For this, 20 random samples of 40 trees were made, 10 close to the lakeshore and 10 distant from it, by applying the central square point method. Close to the dam, we found higher values regarding basal area, number of trees, number of evergreen trees, and zoochoric syndrome, but there were lower values of Shannon's diversity index. Therefore, the impacts of the dam after 15 years caused several changes to the tree community. The greater basal area close to the dam suggests that water deficit during the dry season was decreased and plants have thicker trunks. On the other hand, this sector had much more zoochoric syndrome and a larger number of evergreen trees than plots which are distant from water, suggesting changes with regard to the community's ecological functions. Furthermore, structural floristic data shows that the sector close to the dam is less similar to other deciduous forests within the same geographical region than the sector distant from water, thus providing evidence of the impacts of dams on the tree community.

The comparison of species SLA showed significant differences among the populations of understories under different disturbance regime, showing a decrease in SLA with an increase in the disturbance intensity. Similar results were found for the SLA of understories communities (SLAU), corroborating our hypothesis. The correlation between a reduction in species SLA and in SLA of understory with an increase in disturbance intensity, contradicted the trend observed in the literature for the community as a whole. This study highlights the importance of the evaluation of SLA in understories, as an indicator of the successional stage of communities.

A área foliar específica (SLA) tem sido relacionada a distúrbios ambientais, apresentando uma correlação positiva entre a intensidade de perturbação e a SLA da comunidade vegetal. Estes estudos, no entanto, avaliaram as respostas da comunidade vegetal como um todo, negligenciando os atributos por espécies, tais como a posição vertical no estrato florestal. Considerando a importância da SLA para entender os processos ecológicos das florestas, este estudo teve como objetivo determinar a influência do regime de perturbação na SLA de espécies de sub-bosque, cuja hipótese é que, ao contrário de comunidades como um todo, um aumento na intensidade de perturbação implica na diminuição da SLA de espécies de sub-bosque. Este estudo foi realizado em nove sub-bosque de florestas estacionais no Brasil. As espécies mais abundantes foram selecionados e suas SLA foram avaliadas. A variabilidade de SLA entre as populações em diferentes florestas foi analisada pelo teste t de Student. O SLA dos sub-bosque (SLAu) também foi comparado por uma adaptação do índice de média ponderada da comunidade. A comparação de SLA das espécies mostraram diferenças significativas entre as populações de sub-bosque sob um regime de distúrbios diferentes, mostrando um decréscimo na SLA com um aumento na intensidade de perturbação. Resultados semelhantes foram encontrados para o SLA dos sub-bosque (SLAu), corroborando nossa hipótese. A correlação entre a redução no SLA espécies e SLA do sub-bosque com um aumento na intensidade de perturbação contradiz a tendência observada na literatura para a comunidade como um todo. Este estudo destaca a importância da avaliação de SLA em sub-bosque, como um indicador do estágio sucessional das comunidades.

ABSTRACT

RESUMO Euphorbia tirucalli Linneau, conhecida popularmente como "avelós", "cachorro pelado" ou "pau-pelado", é utilizada na medicina popular para o tratamento de câncer, antibiótico e antivirótico. As plantas medicinais são, frequentemente, utilizadas com o intuito de substituir ou auxiliar as terapias convencionais no tratamento de várias doenças, inclusive para evitar os malefícios causados por radiações ultravioletas. Com isso, o objetivo desta pesquisa foi avaliar o potencial fotoprotetor e a triagem fitoquímica do extrato etanólico das partes aéreas de avelós. A atividade fotoprotetora foi avaliada por meio de ensaios in vitro segundo o método adaptado de Mansur, utilizando extrato etanólico das partes aéreas. Os resultados obtidos mostraram que o estudo fitoquímico evidenciou a presença de açúcares redutores, alcaloides, fenóis, flavonoides, taninos, saponinas, esteroides e triterpenóides na fração etanólica. O extrato apresentou capacidade antioxidante (EC50) de 16,98 ± 0,35 µg.mL-1, teor de compostos fenólicos de 96,18 ± 1,33 mg EqAG/g e flavonoides de 63,57 ± 2,74 mg EqC/g que podem favorecer a atividade fotoprotetora. Avaliando o fator de proteção solar, pode-se observar que todas as concentrações testadas (0,01 a 0,1 µg.mL-1) apresentaram potencial de fotoproteção da radiação ultravioleta com variação de FPS de 6,05 a 19,84, respectivamente. Esses resultados sugerem que o extrato etanólico de Euphorbia tirucalli Linneau apresenta possível efeito fotoprotetor


ABSTRACT Euphorbia tirucalli Linneau, popularly known as "avelos", "naked cock" or "naked dog", is used in popular medicinein order to treat cancer, antibiotic and antiviral diseases. Herbal medicines are often employed in order to assist or replace conventional therapies for the treatment of several diseases, including the prevention of the damage caused by ultraviolet radiation. Thus, the aim of this research was to evaluate the photoprotective potential and phytochemical screening of the ethanol extract of the aerial parts of avelos. The photoprotective activity was assessed by in vitrotrials according to the method adapted from Mansur, using the ethanol extract of the aerial parts. The results showed that the phytochemical study revealed the presence of reducing sugars, alkaloids, phenols, flavonoids, tannins, saponins, steroids and triterpenoids in ethanolic fraction. The extract showed antioxidant activity (EC50) of 16,98 ± 0,35 µg.mL-1, phenolic contents of 96,18 ± 1,33 mg EqAG/g and 63,57 ± 2,74 flavonoids from mg EqC/g that can advantage the photoprotective activity. Assessing the sun protection factor, it can be observed that all the tested concentrations (0,01 to 0,1 µg.mL-1) showed potential for photoprotection from ultraviolet radiation with a range of 6,05 to 19,84 FPS, respectively. These results suggest that the ethanol extract of Euphorbia tirucalli Linneau presents possible photoprotective effect

Phase diagrams of ionic solutions of the ionic liquid C(18)mim(+)NTF(2)(-) (1-n-octadecyl-3-methyl imidazolium bistrifluormethylsulfonylimide) in decalin, cyclohexane and methylcyclohexane are reported and compared with that of solutions of other imidazolium ionic liquids with the anions NTF(2)(-), Cl(-) and BF4(-) in arenes, CCl(4), alcohols and water. The phase diagrams are analysed presuming Ising criticality and taking into account the asymmetry of the phase diagrams. The resulting parameters are compared with simulation results for equal-sized charged hard spheres in a dielectric continuum, the restricted primitive model (RPM) and the primitive model (PM) that allows for ions of different size. In the RPM temperature scale the critical temperatures vary almost linearly with the dielectric permittivity of the solvent. The RPM critical temperatures of the solutions in non-polar solvents are very similar, somewhat below the RPM value. Correlations with the boiling temperatures of the solvents and a dependence on the length of the side chain of the imidazolium cations show that dispersion interactions modify the phase transition, which is mainly determined by Coulomb forces. Critical concentrations, widths of the phase diagrams and the slopes of the diameter are different for the solutions in protic and aprotic solvents. The phase diagrams of the solutions in alcohols and water get a lower critical solution point when represented in RPM variables.

ABSTRACT

Corynebacterium pseudotuberculosis is the etiologic agent of caseous lymphadenitis, a disease that affects goats and sheep, and can cause severe economic losses. In this study, four different antigenic extracts were obtained from the attenuated strain T1, which was isolated in the state of Bahia (Brazil). Forty-four Canindé breed goats were divided in five groups, each receiving a different antigen solution and saline buffer as a control. The humoral response was monitored through the identification of specific IgG by indirect ELISA and Western Blotting, and the production of IFN-gamma was followed in order to observe the activation of cellular response. After twelve weeks of antigen inoculation, the animals were challenged with 2 x 10(5)CFU of a wild strain, also isolated in Bahia, and necropsy was performed on all animals twelve weeks afterwards. It was observed that the attenuated bacteria gave a protection of 33.3%, in addition to the weak humoral response elicited. Animals inoculated with secreted antigen associated with Freund's incomplete adjuvant and oligodeoxynucleotide containing unmethylated CpG dinucleotides (CpG ODN) showed a strong humoral response, but this inoculation could not prevent the spread of challenge bacteria in the majority of animals. These results demonstrate the immunogenic potential of the attenuated T1 strain in the development of a vaccine against caseous lymphadenitis in goats.

The present study has examined the effects of arborinine, an alkaloid obtained from Erthela bahiensis, a Brazilian plant popularly used as diuretic, antidiabetic, antithermic and expectorant, on the viability and function of immune system cells in vitro using a murine model. Rat spleen and thymus cells were cultured with 10 nM, 1 microM, 10 microM of the drug in the presence or absence of pokeweed (PWM), lipopolysaccharide (LPS), or concanavallin (ConA) mitogens. Cellular proliferation was analyzed by H3-thymidin uptake after 48 and 72 hr. Our results showed an inhibitory effect of arborinine on splenocytes proliferation under ConA or PWM stimulation and increased apoptosis on splenocytes and thymocytes stimulated with PWM in 24 hr. A decrease was observed on Interferon gamma (IFN-gamma) production by ConA- or LPS-stimulated splenocytes in 48 hr and 72 hr and ConA- or PWM-stimulated thymocytes in 72 hr. In contrast, an increase on lymphoproliferation was observed on LPS-stimulated splenocytes and ConA- or PWM-stimulated thymocytes in 48 hr. On this period, apoptosis decreased on LPS- or PWM-stimulated splenocytes and IFN-gamma production increased in PWM stimulated thymocytes. Arborinine also induced a decrease on Interleukin-10 production by splenocytes and thymocytes stimulated with ConA or PWM. There was no significant variation on the necrosis rate of the cells treated with arborinine or any change on their viability or function values in the absence of mitogenic stimulus.

ABSTRACT

Corynebacterium pseudotuberculosis is the causal agent of caseous lymphadenitis, a chronic illness that attacks goats and sheep characterized by pyogranulomas formation in lymph nodes and organs. Regarding the current knowledge of the pathogenesis of the caseous lymphadenitis, there is evidence that besides the humoral response the induction of a durable cellular response is fundamental for its control. In this sense, research on antigens of C. pseudotuberculosis that are capable to inducing cellular immunity is an important step for the development of diagnosis tests and more efficient vaccines. In the present study, the interferon-gamma production in cultures of whole blood from infected goats stimulated with secreted bacterial antigen or somatic antigen were used to evaluate the cellular response. The results demonstrated a significant difference in the ability of the two antigens to induce a cellular response. That is, IFN-gamma production was high with cells from infected animals in response to the secreted antigen while IFN-gamma production was low when somatic antigen was used. The concomitant use of these antigens with PWM also showed differences. That is, the secreted antigen increased the IFN-gamma production induced by PWM, while the somatic antigen seems not to have altered the response to PWM.

ABSTRACT

Neospora caninum, is a coccidian protozoan known as a major cause of bovine abortion and canine neuropathies. The aim of the present study was to develop a reliable and quick test to detect antibodies to N. caninum in dog sera. Sixty-five serum samples from dogs, including 35 positive and 30 negative for N. caninum antibodies were used for standardization of the test. In parallel, immunoreactivity of the sera to Toxoplasma gondii antigens was investigated using a passive agglutination test. A dot-ELISA test, using soluble extract of N. caninum tachyzoites on nitrocellulose ester membranes, was developed and standardized. SDS-PAGE and complementary analysis of reactivity by Western blotting were used for the characterization of the immunoreactive fractions of all tested sera. The sensitivity and specificity of the dot-ELISA were 94 and 73%, respectively, compared to IFAT at a cut-off of 1:50, and 87 and 100% compared to IFAT at a cut-off of 1:25. Among the sera that tested positively for both IFAT and dot-ELISA, only 8.6% were reactive to T. gondii. The most immunoreactive fractions in Western blots were the 14-, 33-, 42- and 55 kDa bands, with percentages of 42, 60, 42 and 37%, respectively. The 60 kDa band showed a non-specific reaction in 43% of neosporosis-negative animals by both dot-ELISA and IFAT. These results indicate that the dot-ELISA using N. caninum antigen present good sensitivity and specificity, and might be used as a screening test to detect antibodies to N. caninum in dogs.

ABSTRACT

The three-phase partitioning (TPP) technique was used upstream to isolate/concentrate secreted proteins from Corynebacterium pseudotuberculosis cultured in a complex liquid medium. Several parameters of the TPP technique (15, 30, or 60% ammonium sulfate concentration; 4.0, 5.5, or 7.0 pH; and primary (n) or tertiary (t)-butanol solvent isomer) were varied to determine the optimal recovery of serologically and cellularly immunoreactive extracted proteins. A TPP extraction made with 30% ammonium sulfate and an initial pH of 4.0 gave the best humoral and cellular immunoreactivity of caseous lymphadenitis infected goats. In particular, two immunogenic secreted (16 and 125 kDa) proteins, which had not been found by other extraction methods, were identified.

ABSTRACT

Corynebacterium pseudotuberculosis is the cause of caseous lymphadenitis (CLA) in small ruminants, a chronic granulomatous disease that provokes significant zootechnics losses to ovine and goat breeders in northern Brazil. The present work was conducted to analyse aspects of humoral and cellular immune responses after experimental infection. Eight goats were infected intradermally with a single dose of virulent C. pseudotuberculosis strain and specific IgG, interferon-gamma (IFN-gamma) production as well as IgG avidity and antigens pattern recognition dynamics against an excreted-secreted antigen were recorded during 20 weeks. At the end of the follow-up, animals were slaughtered and necropsied. Although no animals showed apparent clinical signs of infection at the end of the trial, IFN-gamma response, even more so than the humoral response, differentiated animals into two groups of high or medium/low response. The time-course of IFN-gamma production presented a short-lived primary response on day 5 after infection of animals of both groups, and a strong and long lasting secondary response starting on day 16 after infection in the high response group. The indirect ELISA used was able to detect a positive antibody titre between 6 and 11 days after infection in the two groups. IgG avidity index oscillated initially between 15 and 45%, and showed approximately 5% units increment during the 20 follow-up weeks. With only one individual exception, the qualitative antigens pattern recognition showed on day 11 after infection remained constant through the experiment. IgG avidity is highly correlated with IgG production, but could not be related with specific immunodominant bands. Both humoral and cellular responses kinetics presented a similar pattern of activation/deactivation but necropsy results suggested that the IFN-gamma test would be a very specific marker of CLA status.
