ABSTRACT

The author has presented the results of electron-microscopic and electron-histochemical investigations of the lysosomal apparatus in human platelets under the action of certain agents (acetylsalicylic acid, vincristine) and in pathologic states (acute leukemia, myeloproliferative diseases, autoimmune thrombocytopenic purpura). Interrelation has been recorded between ultrastructural changes in the lysosomal apparatus and parameters of platelet functional activity. Biogenesis of lysosomal enzymes has been considered basing on molecular biology of the cell.

ABSTRACT

Electron-microscopic and electron-histochemical data have been presented on platelet ultrastructural shifts in patients with idiopathic thrombocytopenic purpura 1 h, 1, 4 and 8 days after one injection of vincristine. Vincristine induces depolymerization of microtubules+, appearance of crystalloid structures in platelet cytoplasm alters the biogenesis of lysosomal enzymes; for the first time the activity of acid phosphatase has been shown in cisterns of the dense tubular system. Electron-microscopic signs of thrombocytopoiesis stimulation are revealed on day 4 after the injection. The mechanism has been shown of platelet number growth. The duration of the effect of a single low dose of vincristine is eight days, that has been evidenced by the state of the microtube structure.

ABSTRACT

Data are presented on yak platelet ultrastructure (in comparison with human platelets), and on acetylsalicylic acid influence on the platelet ultrastructure and function. The following species features of this animals are associated with their adaptation to high-altitude hypoxia: organization of alpha-granules, mitochondria, the contractile system (microtubes, F-actin), ovoid form of platelets. Acetylsalicylic acid influences the yak platelet plasma membrane inducing fragmentation, which is attended by a rise in the total number of platelets, by the appearance of a great number of microforms, and by the reduction of functional activity. The above changes remained up to 11 days from the beginning of the drug action.

ABSTRACT

Electron microscopical and electron histochemical studies and functional investigation of platelets have been undertaken in 10 normal individuals and 20 patients with ischaemic heart disease and angina pectoris of effort before and after acetylsalicylic acid (ASA) administration during 3 days. In ischaemic heart disease changes in the ultrastructure of platelets have been recorded, pointing to their activation. After ASA administration there was decrease of functional activity, due to structural changes in the plasma membrane, lysosome membranes, fragmentation of platelets, enhanced process of autophagia, decrease of enzymatic activity of structure organelles of platelets.
