ABSTRACT

From 2006 through 2014, we conducted seroepidemiological surveys on non-human primates and sloths to investigate the possible circulation of arboviruses in Bahia Atlantic Forest, Brazil. We collected a total of 196 samples from 103 Leontopithecus chrysomelas, 7 Sapajus xanthosternos, 22 Bradypus torquatus and 7 Bradypus variegatus. Serum samples were tested using neutralization test and hemagglutination inhibition test to detect total antibodies against 26 different arboviruses. The overall prevalence of arboviruses was 36.6% (51/139), with the genus Flavivirus having the highest prevalence (33.1%; 46/139), followed by Phlebovirus (5.0%; 7/139), Orthobunyavirus (4.3%; 6/139) and Alphavirus (0.7%; 1/139). Monotypic reactions suggest that the wild animals were exposed naturally to at least twelve arboviruses. Added results from the neutralization test, animals were exposed to thirteen arboviruses. Most of these viruses are maintained in transmission cycles independent of human hosts, although antibodies against dengue virus serotypes 1, 2 and 3 were found in this study. To our knowledge, this is the first study reporting exposure to arboviruses in L. chrysomelas, S. xanthosternos and B. torquatus. Our results also highlight that the Southern Bahia Atlantic Forest has a variety of vertebrate hosts and potential vectors, which may support the emergence or re-emergence of arboviruses, including those pathogenic to humans.

ABSTRACT

Saponins are high molecular weight glycosides which are known for their broad range of biological activities. In case of Maesa lanceolata, a tree growing in African countries, the maesasaponins showed virucidal, haemolytic, molluscicidal and anti-angiogenic activity. Since the different activities are dependent on the structure of the saponins, a method was developed and validated for the analysis of the individual saponins in this plant. Since the saponins were only present in small amounts, it was necessary to develop a very sensitive analytical method. For the fast and sensitive analysis of the extracted and purified plant samples ultra-performance liquid chromatography was coupled to a triple quadrupole mass spectrometer for MS/MS detection. A method in positive ESI mode, using sodium acetate in the mobile phase, was developed. The sodium adduct ion was selected as the precursor ion since it provided better sensitivity and a better, more stable fragmentation compared to the deprotonated and protonated ions. The intensity of the signal obtained by fragmentation of the sodium adducts of the saponins, was optimized by the addition of different concentrations of sodium acetate to the mobile phase. Reference standards were not available for all 14 saponins. Therefore, a relative MS/UV response was calculated allowing the estimation of the saponins in real samples. alpha-Hederin was used as external standard. The method was linear over the investigated concentration range with a good correlation coefficient (>0.99). The intra- and inter-day precisions were below 15% for most maesasaponins with the exception of maesasaponin II, which showed a precision within 20%. The recoveries of the spiked pure compounds maesasaponin IV.1 and VII.1 were 96.6% and 85.5%, respectively. The validated method can be applied in the investigation of the content of 14 saponins in transgenic and non-transgenic plant material of M. lanceolata.

ABSTRACT

In Callitrichidae, reproduction in subordinate females is generally inhibited but occurs in rare cases, possibly in association with the presence of an unrelated male, important food resources or low dispersal opportunities. This study investigates the occurrence of groups with multiple breeding females in captive golden-headed lion tamarins (Leontopithecus chrysomelas), the factors leading to their formation and the consequences for the group. Information obtained from studbook data on the world captive population during 1984-1998 revealed that polygynous groups in captivity are very rare: only 7 cases were discovered. Family groups in which daughters started breeding with a related male were larger than average, had a high number of sexually mature sons and eldest offspring that were well past the age of sexual maturity. Following a breeding attempt, severe aggression frequently occurred, especially if the infants were liveborn. Polygynous groups composed of two related females and an unrelated male tended to remain stable for a longer period than families with breeding daughters. Competition for infant care is probably an important factor determining whether the polygynous situation can persist and for how long.

ABSTRACT

The effect of posture on hand preferences was examined in an experiment with 5 bonobos (Pan paniscus). To obtain a food reward, the animals had to adopt 1 out of 7 different postures. These postures represented an increasing problem to the maintenance of body equilibrium. It was expected that an increasing demand for equilibrium maintenance would elicit individual preferences and a population-level bias. All animals showed an increasing trend toward left-handedness while shifting to a bipedal posture from a seated posture by way of a quadrupedal posture. The importance of bipedalism in the evolution of left- and right-handedness is discussed briefly.
