ABSTRACT

BACKGROUND: The prevalence of overweight and obesity among adolescents in Saudi Arabia has been progressively increasing. Obesity is associated with an increased risk of various morbidities and mortality. Identifying the factors that contribute to obesity in this age group is crucial for implementing targeted prevention measures. AIM: The aim of this study was to identify risk factors for overweight and obesity among adolescents aged nine to 17 years residing in Tabuk City, Saudi Arabia. METHODS: A case-control study was conducted during the 2021-2022 academic year at Alabnaa Schools in Tabuk City, Saudi Arabia. The study included overweight/obese individuals (cases, n = 125) and normal-weight individuals (controls, n = 201) who were selected based on their body mass index and classified according to the World Health Organization's reference for defining overweight and obesity in individuals aged five to 19 years. Data were collected from both groups using a self-administered questionnaire. RESULTS: The study analyzed 125 overweight/obese students and 201 normal-weight students who were matched for sex and age (p > 0.05). Logistic regression analysis identified several risk factors for overweight or obesity among adolescents. A family history of obesity was found to be associated with a 5.735 times increased likelihood of obesity (95% CI: 3.318-9.912, p < 0.001). Another significant contributing risk factor for obesity was frequent consumption of four or more meals per day (adjusted odds ratio: 3.091, 95% CI: 1.094-8.736, p = 0.033). Students who used electronic devices for more than five hours were 2.422 times more likely to exhibit obesity (p = 0.006). CONCLUSIONS: Certain factors may increase the risk of overweight or obesity in adolescents aged nine to 17 years. These factors include frequent eating, prolonged use of electronic devices, family history of obesity, and the misconception that obesity is not an illness. Tailored school health programs are needed to improve students' healthy lifestyles and eating behaviors, minimize sedentary entertainment and use of electronic devices, and engage children in physical activity.

ABSTRACT

In this work, readily achievable synthetic pathways were utilized for construction of a library of N/S analogues based on the pyrazolopyrimidine scaffold with terminal alkyl or aryl fragments. Subsequently, we evaluated the anticancer effects of these novel analogs against the proliferation of various cancer cell lines, including breast, colon, and liver lines. The results were striking, most of the tested molecules exhibited strong and selective cytotoxic activity against the MDA-MB-231 cancer cell line; IC50 1.13 µM. Structure-activity relationship (SAR) analysis revealed that N-substituted derivatives generally enhanced the cytotoxic effect, particularly with aliphatic side chains that facilitated favorable target interactions. We also investigated apoptosis, DNA fragmentation, invasion assay, and anti-migration effects, and discussed their underlying molecular mechanisms for the most active compound 7c. We demonstrated that 7c N-propyl analogue could inhibit MDA-MB-231 TNBC cell proliferation by inducing apoptosis through the regulation of vital proteins, namely c-Src, p53, and Bax. In addition, our results also revealed the potential of these compounds against tumor metastasis by downregulating the invasion and migration modes. Moreover, the in vitro inhibitory effect of active analogs against c-Src kinase was studied and proved that might be the main cause of their antiproliferative effect. Overall, these compelling results point towards the therapeutic potential of these derivatives, particularly those with N-substitution as promising candidates for the treatment of TNBC type of breast cancer.

ABSTRACT

Illicit drug use has become a global issue, posing significant health, socioeconomic, and cultural risks. The study examined essential trace metals: selenium, zinc, and copper in blood concentrations, as well as in serum and scalp hair samples, from 240 male drug-abuse subjects/patients aged 18-45, categorized into three age groups. The study compared 45 healthy subjects of the same age group using an acid digestion method supported by a microwave oven during sample preparation. The technique of atomic absorption spectrometry was employed to identify essential and toxic elements, utilizing certified reference materials for accuracy. According to a recent study, plasma zinc and selenium concentrations in drug abusers are lower than those in referent subjects, potentially increasing vulnerability to infection due to poor nutritional status or other contaminants.

ABSTRACT

Manchar Lake, Pakistan's biggest lake in the arid zone, faces human-induced salinity issues. This study investigated its effects on the multifaceted ecosystem services, including serving as a source of drinking and irrigation water and aquatic health through assessing fish diversity and characteristics. Analyses of 189 water samples from 21 sites revealed spatiotemporal variations in major ions contributing to lake water salinity. The study assessed water suitability for drinking and agriculture using the water quality index (WQI), sodium adsorption ratio (SAR), magnesium hazard (MH), sodium percent (Na%), and Kelly's ratio (KR). The WQI, ranging from 141 to 408, indicated that the lake water was unfit for drinking. In some seasons, such as the pre-monsoon period, the lake water was deemed unsuitable for irrigation due to high SAR values (18 ± 4 g/L, average ± standard deviation), consistently rising MH values exceeding 66 in all seasons and elevated sodium percentages surpassing 66% in both the pre-monsoon and monsoon seasons. The KR remained acceptable (averaging 0.8 to 2.5) in all seasons. Fish health in highly saline conditions was assessed using data from interviews, focus group discussions, and fish sampling (1684 fish from 10 sites). Results depicted that high salt contamination severely impacted fish length and weight. The study found low richness (Simpson's biodiversity: 0.697 and Shannon Weaver: 1.51) and evenness (Pielou's index: 0.48) among the fish populations. Since 1998, Manchar Lake has seen a decline in fish varieties from 32 to 23, with changes in fish species' feeding habits. To improve lake water quality, the study recommends diverting saline water to the sea before and after the monsoon season while utilizing freshwater from alternative sources to fill any water deficit.

ABSTRACT

Phenylpyrazolo[3,4-d]pyrimidine is considered a milestone scaffold known to possess various biological activities such as antiparasitic, antifungal, antimicrobial, and antiproliferative activities. In addition, the urgent need for selective and potent novel anticancer agents represents a major route in the drug discovery process. Herein, new aryl analogs were synthesized and evaluated for their anticancer effects on a panel of cancer cell lines: MCF-7, HCT116, and HePG-2. Some of these compounds showed potent cytotoxicity, with variable degrees of potency and cell line selectivity in antiproliferative assays with low resistance. As the analogs carry the pyrazolopyrimidine scaffold, which looks structurally very similar to tyrosine and receptor kinase inhibitors, the potent compounds were evaluated for their inhibitory effects on three essential cancer targets: EGFRWT, EGFRT790M, VGFR2, and Top-II. The data obtained revealed that most of these compounds were potent, with variable degrees of target selectivity and dual EGFR/VGFR2 inhibitors at the IC50 value range, i.e., 0.3-24 µM. Among these, compound 5i was the most potent non-selective dual EGFR/VGFR2 inhibitor, with inhibitory concentrations of 0.3 and 7.60 µM, respectively. When 5i was tested in an MCF-7 model, it effectively inhibited tumor growth, strongly induced cancer cell apoptosis, inhibited cell migration, and suppressed cell cycle progression leading to DNA fragmentation. Molecular docking studies were performed to explore the binding mode and mechanism of such compounds on protein targets and mapped with reference ligands. The results of our studies indicate that the newly discovered phenylpyrazolo[3,4-d]pyrimidine-based multitarget inhibitors have significant potential for anticancer treatment.

ABSTRACT

The wastewater containing urea-formaldehyde (UF) and melamine-formaldehyde (MF) from the medium-density fiberboard (MDF) lamination factory disposed into the waterbodies adversely affects human health and aquatic life. Therefore, its treatment before discharge is necessary. Researchers have used various techniques to treat this type of wastewater in the past, but none have tried electrochemical (EC). However, EC can potentially remove pollutants such as chemical oxygen demand (COD), total organic carbon (TOC), formaldehyde (FA), total nitrogen (TN), nitrogen nitrate (NO3-N), and other hydrocarbons. Hence, this study uses the EC technique to treat wastewater containing UF and MF with aluminium electrodes. The experiments were run in batch mode with a 250 mL working volume in a 500 mL Pyrex glass beaker using a variable DC power supply (0-30 V and 0-5 A). The impacts of various parameters, including reaction time (RT) 30-240 min, current density (CD) 8.66-51.94 mA/cm2, inter-electrode distance (IED) 1-2 cm, and mixing speed in the range of 60-120 rpm were examined to achieve the best pollutant removals. The best removal percentage was reached at the optimized conditions of 150 min RT, 43.28 mA/cm2 CD, 1.5 cm IED, and 80 rpm: 81.1% TOC, 61.5% COD, 76.7% TN, 28.3% NO3-N, and 55.2% FA. During the EC process, electrodes and energy consumption were estimated at around 2.367 (g/L) and 0.18 (kWh/L), respectively. A kinetic analysis was also carried out to determine the pollutant's removal trend. This study concluded that the pseudo-first-order kinetic model was the best fit for removing TOC and FA with regression coefficients of 0.96 and 0.83, respectively.

ABSTRACT

Genome-wide association studies significantly increased the number of hypertension risk variants; however, most of them focused on European societies. There is lack of such studies in developing countries, including Pakistan. The lack of research studies and the high prevalence of hypertension in the Pakistani community prompted us to design this study. Aldosterone synthase (CYP11B2) was thoroughly studied in different ethnic groups; however, no such study has been conducted in the Pashtun population of Khyber Pakhtunkhwa, Pakistan. In essential hypertension, the aldosterone synthase gene (CYP11B2) plays a significant role. Aldosterone synthesis is affected by both hereditary and environmental factors. Aldosterone synthase (encoded by the CYP11B2 gene) controls the conversion of deoxycorticosterone to aldosterone and, thus, has genetic influences. Polymorphisms in the CYP11B2 gene are linked to an increased risk of hypertension. Previous research on the polymorphism of the aldosterone synthase (CYP11B2) gene and its relationship to hypertension produced inconclusive results. The present study investigates the relationship between CYP11B2 gene polymorphism and hypertension in Pakistan's Pashtun population. We used the nascent exome sequencing method to identify variants associated with hypertension. The research was divided into two phases. In phase one, DNA samples from 200 adult hypertension patients (of age ≥ 30 years) and 200 controls were pooled (n = 200/pool) and subjected to Exome Sequencing. In the second phase, the WES reported SNPs were genotyped using the Mass ARRAY technique to verify and confirm the association between WES-identified SNPs and hypertension. WES identified a total of eight genetic variants in the CYP11B2 gene. The chi-square test and logistic regression analysis were used to estimate the minor allele frequencies (MAFs) and chosen SNPs relationships with hypertension. The frequency of minor allele T was found to be higher in cases compared to the control (42% vs. 30%: p = 0.001) for rs1799998 of CYP11B2 gene, while no significant results (p > 0.05) were observed for the remaining SNPs; rs4536, rs4537, rs4545, rs4543, rs4539, rs4546 and rs6418 showed no positive association with HTN in the studied population (all p > 0.05). Our study findings suggest that rs1799998 increases susceptibly to HTN in the Pashtun population of KP, Pakistan.

OBJECTIVES: Various techniques for dorsal hand vein (DHV) pattern extraction have been introduced using small datasets with poor and inconsistent segmentation. This work compared manual segmentation with our proposed hybrid automatic segmentation method (HHM) for this classification problem. METHODS: Manual segmentation involved selecting a region-of-interest (ROI) in images from the Bosphorus dataset to generate ground truth data. The HHM combined histogram equalization and morphological and thresholding-based algorithms to localize veins from hand images. The data were divided into training, validation, and testing sets with an 8:1:1 ratio before training AlexNet. We considered three image augmentation strategies to enlarge our training sets. The best training hyperparameters were found using the manually segmented dataset. RESULTS: We obtained a good test accuracy (91.5%) using the model trained with manually segmented images. The HHM method showed slightly inferior performance (76.5%). Considerable improvement was observed in the test accuracy of the model trained with the inclusion of automatically segmented and augmented images (84%), with low false acceptance and false rejection rates (0.00035% and 0.095%, respectively). A comparison with past studies further demonstrated the competitiveness of our technique. CONCLUSIONS: Our technique can be feasible for extracting the ROI in DHV images. This strategy provides higher consistency and greater efficiency than the manual approach.

ABSTRACT

Coronary Artery Diseases (CAD) remains the top among Non-communicable Diseases (NCDs). Variations in Apolipoprotein E (APOE) and Paroxonase 1 (PON1) have been associated with Myocardial Infarction (MI) in several populations. However, despite the high prevalence of CAD, no such study has been reported in the Pashtun ethnic population of Pakistan. We have conducted a two-stage (i.e., screening and validation) case-control study in which 200 cases and 100 control subjects have been recruited. In the first stage, Whole Exome Sequencing (WES) was used to screen for pathogenic variants of Myocardial Infarction (MI). In the second stage, selected variants of both APOE and PON1 genes (rs7412, rs429358, rs854560, and rs662) were analyzed through MassARRAY genotyping. Risk Allele Frequencies (RAFs) distribution and association of the selected SNPs with MI were determined using the Chi-square test and logistic regression analysis. WES identified a total of 12 sequence variants in APOE and 16 in PON1. Genotyping results revealed that APOE variant rs429358 (ɛ4 allele and ɛ3/ɛ4 genotype) showed significant association in MI patients (OR = 2.11, p value = 0.03; 95% CI = 1.25-2.43); whereas no significant difference (p˃ 0.05) was observed for rs7412. Similarly, the R allele of PON1 Q192R (rs662) was significantly associated with cases (OR = 1.353, p value = 0.048; 95% CI = 0.959-1.91), with particular mention of RR genotype (OR = 1.523, p value = 0.006; 95% CI = 1.087-2.132). Multiple logistic regression analysis showed that rs429358 (C allele) and rs662 (R allele) have a significantly higher risk of MI after adjustment for the conventional risk factors. Our study findings suggested that the rs429358 variant of APOE and PON1 Q192R are associated with MI susceptibility in the Pashtun ethnic population of Pakistan.

ABSTRACT

An artificial neural network (ANN) is a biologically inspired computational technique that imitates the behavior and learning process of the human brain. In this study, ANN technique was applied to assess the gasification of municipal solid waste (MSW) with the aim of enhancing the H2 production. The experiments were conducted using a horizontal tube reactor under different parameters: temperatures, MSW loadings, residence times, and equivalence ratios. The input and output variables (released gases) were tested and trained using back-propagation algorithm, and the data distribution by K-fold contrivance. The values of the training (80% data) and validation (20% data) dataset were found satisfactory. The values of regression coefficient (R2) for the training phase were lied between 0.9392 and 0.9991, and 0.9363 and 0.993824 for the testing phase. Whereas; the values of root mean square error (RSME) for the training phase were lied between 0.4111 and 0.8422, and between 0.1476 and 0.7320 for the testing phase. Higher H2 production of 42.1 vol% was produced at the higher reaction temperature of 900 °C with LHV of 11.2 MJ/Nm3. According to the tar analysis, the dominant compounds were aromatics (17 compounds) followed by polycyclic aromatic, phenyl, aliphatic, aromatic heterocyclic, polycyclic, and aromatic ketone compounds.

ABSTRACT

We report the case of successful use of cefiderocol (FDC) in a Carbapenemase Producing K. pneumoniae (CPKP) post-surgical meningitis in a 44-year-old man treated with antimicrobial therapy and external ventricular drainage (EVD). The patient was known for being colonised by CPKP; for this reason, therapy with ceftazidime/avibactam (CZA) plus fosfomycin and linezolid was started. After an initial response a CZA resistant CPKP strain was isolated from CSF culture, so the antibiotic therapy was modified to FDC with trimethoprim/sulfamethoxazole for 14 days, and EVD was replaced. A complete recovery was obtained. This is the first case report describing FDC administration in CPKP meningitis.

ABSTRACT

Cadmium is found in many underdeveloped countries' aquatic bodies. Therefore, contaminated water should be treated before consumption; henceforth, efficient and customized point-of-use filtration is foreseeable. Traditionally, carbon-based sorbents have been utilized for such treatments, but alternative sources are also being investigated. Hydrochars made from mango peels using a thermal activation process were employed as an adsorbent instead of activated carbon in this investigation. The prepared material was porous with active surface functionalities, and the interaction of cadmium with the surface was possibly ion-exchange in nature. The performance of a material for a candle water filtering system with a 2.5 cm internal diameter and a 30.48 cm column height was determined using the parameters acquired by the Thomas model. The material was found to be highly efficient at 453.5 L/min/Filter water, whereas 31670.6 L/min/Filter can be treated if the break point and exhaustion point are considered, respectively, as the candle replacement time. These findings indicate that activated hydrochar might be a suitable sorbent for removing cadmium ions from contaminated water.

ABSTRACT

The heavy metals, such as Cr(VI), Pb(II) and Cd(II), in aqueous solutions are toxic even at trace levels and have caused adverse health impacts on human beings. Hence the removal of these heavy metals from the aqueous environment is important to protect biodiversity, hydrosphere ecosystems, and human beings. In this study, magnetic Nickel-Ferrite Nanoparticles (NFNs) were synthesized by co-precipitation method and characterized using X-Ray Diffraction (XRD), Energy Dispersive Spectroscopy (EDS) and Field Emission Scanning Electronic Microscopy (FE-SEM) techniques in order to confirm the crystalline structure, composition and morphology of the NFN's, these were then used as adsorbent for the removal of Cr(VI), Pb(II) and Cd(II) from wastewater. The adsorption parameters under study were pH, dose and contact time. The values for optimum removal through batch-adsorption were investigated at different parameters (pH 3-7, dose: 10, 20, 30, 40 and 50 mg and contact time: 30, 60, 90, and 120 min). Removal efficiencies of Cr(VI), Pb(II) and Cd(II) were obtained 89%, 79% and 87% respectively under optimal conditions. It was found that the kinetics followed the pseudo second order model for the removal of heavy metals using Nickel ferrite nanoparticles.

ABSTRACT

Traditional agricultural practices, extensive use of inputs, and abrupt changes in climate have been of great concern to agriculture production around the world, especially in developing countries. Therefore, it is very vital to adopt and expand Climate-Smart agricultural (CSA) practices. By the cross-sectional data of 350 cotton farmers from major cotton-growing districts of Punjab Pakistan, adoption of CSA practices such as irrigation and soil and crop management practices is evaluated, and factors which affect farmer adoption decision and its impact on poverty, income, and yield are estimated by using logistic regression and propensity score matching (PSM) respectively. The results found that education, access to credit, tubewell ownership, farming experience, and access to extension services positively influenced farmers' adoption behavior. Further, PSM results revealed that adoption of CSA practices is economical, financially, environmentally desirable, and pro-poor. According to these findings, ultimately adoption would help in reducing the negative impact of climate change on the cotton crop by ensuring profits, removing the barriers in the adoption, disseminating the information about CSA, and strictly enforcing the regulations for CSA.

Species belonging to genus, Thysanoplusia Fabricius of the subfamily Plusiinae which are polyphagous in nature and pest of vegetables, foods, legumes, fodder, fruits, ornamental plants and cotton crops. Samples were collected from different localities of district Bahawalpur. For collection, comprehensive and comparative surveys were carried out during 2017-18 on taxonomic account of species of the genus Thysanoplusia Fabricius and resulted identified one species Thysanoplusia orichalcea (Fabricius) first time from Pakistan. Morphological characters viz., vertex, frons, labial palpi, antennae, compound eyes, ocelli, proboscis, wing venation, male and female genital characteristics were used for the identification and classification. Dichotomous keys and photographs are also provided. There is hardly any substantial research work on taxonomic studies of subfamily Plusiinae Pakistan. So to fill this gap the present proposal was designed to study the diversity of Noctuid moths from Pakistan and very fruitful results have been obtained.

ABSTRACT

BACKGROUND: Creating and fostering innovation has long been a main concern of different organisational leaders, regardless of the size, nature, or structure of the organisation. Transformational leadership positively affects innovative work behaviour but understanding underlying mechanisms through which transformational leadership influences innovative work behaviour is critical. AIMS: The purpose of this research was to examine the mediating effect of trust in leader and the moderating effect of thriving on the relationship between transformational leadership and nurses' innovative work behaviour. METHODS: The sample of this cross-sectional, correlational study consisted of 326 nurses nested in 39 work groups, who were selected, on the basis of the convenience sampling method, from among the nurses working at 21 public hospitals in Pakistan. The data of the study were collected between May and July of 2018. RESULTS: Results showed that transformational leaders instilled trust within subordinate nurses, which acted as a mediator in predicting their innovative work behaviour. Further, thriving moderated the relationship between trust in leader and nurses' innovative work behaviour. Findings also indicated that thriving nurses exhibited a greater degree of innovative work behaviour when they trusted their head nurses. CONCLUSIONS: Healthcare policies and strategies are needed to facilitate the implementation of transformational leadership by providing healthcare leaders with the appropriate environment.

ABSTRACT

Medicines are often consumed concurrently with food; sometimes to improve its absorption and efficacy. However, certain foods may modify the function of drug metabolizing enzymes or transport mechanisms that are crucial determinants of systemic drug availability. Extensive work has been reported on certain juices like grapefruit that affects the bioavailability of more than 60 medications. However, relatively less work has been reported on certain other commonly used fruit juices, especially in Pakistan, such as mango, strawberry, apple, banana, pomegranate and grape etc. Present review has taken an account of the current work done in this area.

ABSTRACT

There are numerous pygmy grasshoppers (Tetrigidae) that exhibit leaf-like appearance. Leaf-mimic species can be found mainly in the subfamily Cladonotinae (tribes Cladonotini, Xerophyllini). Two leaf-mimic pygmy grasshopper species found in India, Pakistan (Oxyphyllum pennatum Hancock, 1909) and Borneo (Paraphyllum antennatum Hancock, 1913) are the only macropterous leaf-like species of Asia and were traditionally assigned to subfamily Cladonotinae. Our study present new records of these species, as well as updated descriptions. Oxyphyllum pennatum is recorded for the first time from Chhattisgarh (Central India), and furthermore, records from Pakistan were reviewed and confirmed. Paraphyllum antennatum is reported from a few localities in the mountains of Borneo (East Malaysia). New records found in online social media (Flickr, iNaturalist) were implemented. Brachypronotal specimens of P. antennatum are reported. After comparison with Cladonotini and Xerophyllini members, we conclude that Oxyphyllum and Paraphyllum should not be regarded Cladonotinae members. Traditional assignment was based on superficial resemblance to leaf-like Cladonotinae genera (e.g. Phyllotettix, Hymenotes, Holoarcus, Xerophyllum). However, comparison of morphological characters reveals that Oxyphyllum shares morphology with Tetriginae genera, while Paraphyllum shares morphology with Asian Metrodorinae. Here, we assign the genus Oxyphyllum to Tetriginae and the genus Paraphyllum to Metrodorinae.
