ABSTRACT

The mitochondrial protein mitoNEET is a type of iron-sulfur protein localized to the outer membrane of mitochondria and is involved in a variety of human pathologies including cystic fibrosis, diabetes, muscle atrophy, and neurodegeneration. In the current study, we found that isoliquiritigenin (ISL), one of the components of the root of Glycyrrhiza glabra L., could decrease the expression of mitoNEET in A375 melanoma cells. We also demonstrated that mitoNEET could regulate the content of reactive oxygen species (ROS), by showing that the ISL-mediated increase in the cellular ROS content could be mitigated by the mitoNEET overexpression. We also confirmed the important role of ROS in ISL-treated A375 cells. The increased apoptosis rate and the decreased mitochondrial membrane potential were mitigated by the overexpression of mitoNEET in A375 cells. These findings indicated that ISL could decrease the expression of mitoNEET, which regulated ROS content and subsequently induced mitochondrial dysfunction and apoptosis in A375 cells. Our findings also highlight mitoNEET as a promising mitochondrial target for cancer therapy.

ABSTRACT

Osteosarcoma (OS) is the most common primary bone malignancy in children and adolescents, the pathogenesis of which remain largely unknown. Small ubiquitin-like modifier (SUMO)-Specific Protease 2 (SENP2) has been reported to serve as a tumor suppressor in hepatocellular carcinoma cells. The aim of the present study was to investigate the critical role of SENP2 in OS cells. Using reverse transcription-quantitative polymerase chain reaction and western blot assays, it was observed that SENP2 was significantly downregulated in clinical OS tissues compared with adjacent normal samples. Ectopic expression of SENP2 resulted in the suppression of proliferation, migration and invasion in OS cells, whereas SENP2 knockdown by CRISPR-Cas9-based gene editing had the opposite effect. SENP2 is associated with the proteasome-dependent ubiquitination and degradation of SRY-box-9 (SOX9). SOX9 silencing impaired SENP2-depletion-induced accelerated cell growth and migration. Together, these results suggest that SOX9 is a critical downstream effector of the tumor suppressor SENP2 in OS.

ABSTRACT

Changes in pig fertility have occurred as a result of domestication, but are not understood at the level of genetic variation. To identify variations potentially responsible for prolificacy, we sequenced the genomes of the highly prolific Taihu pig breed and four control breeds. Genes involved in embryogenesis and morphogenesis were targeted in the Taihu pig, consistent with the morphological differences observed between the Taihu pig and others during pregnancy. Additionally, excessive functional non-coding mutations have been specifically fixed or nearly fixed in the Taihu pig. We focused attention on an oestrogen response element (ERE) within the first intron of the bone morphogenetic protein receptor type-1B gene (BMPR1B) that overlaps with a known quantitative trait locus (QTL) for pig fecundity. Using 242 pigs from 30 different breeds, we confirmed that the genotype of the ERE was nearly fixed in the Taihu pig. ERE function was assessed by luciferase assays, examination of histological sections, chromatin immunoprecipitation, quantitative polymerase chain reactions, and western blots. The results suggest that the ERE may control pig prolificacy via the cis-regulation of BMPR1B expression. This study provides new insight into changes in reproductive performance and highlights the role of non-coding mutations in generating phenotypic diversity between breeds.

ABSTRACT

OBJECTIVE: To develop a Jingcen DY-1 type spraying tanker for Oncomelania hupensis snail control and evaluate its effect of field application as well as the cost. METHODS: The currently available tractor was used as a vector, and the mechanical and electrical equipments and containers were integrated with shafts, pipelines and electric lines to produce a spraying tanker for snail control, with the functions of carrying people and molluscicides, generating electric power and getting water, mixing stocking solutions, adjusting molluscicide solutions evenly, and spraying drugs. The volume of the molluscicide solution, flow rate of water injection, and the flow rate, range and advance speed of the spray gun were tested, and the solution concentrations of molluscicide in the tanker and at the muzzle of the spray gun at different time were detected. Meanwhile, the molluscicidal effect and cost of the spraying tanker were analyzed by the field test. RESULTS: The volume of the liquid storage pot of the Jingcen DY-1 type spraying tanker was 1 800 L, the flow rate of water injection was 400 L/min, the flow rate and the spray range of the standard spray gun were 110-200 L/min and 19.70-23.50 m, respectively, the efficiency of drug spraying of the spraying tanker was 6 000 m/h, and the ratio of spray width (m) to march speed (m/min) was 1:200. When 5 min post mother liquid recirculating, the average concentration of the molluscicide at the upper-, middle- and lower-layers of the liquid storage pot was (1030.39 ± 43.00) mg/L, with a variation coefficient of 4.17%. The average concentration of the molluscicide in the spraying process (spraying for 2, 4, 6, 8, 9 min) was (953.00 ± 68.87)mg/L, with a variation coefficient of 7.22%. The concentration of the residual drug in the liquid storage pot post spraying was 1 000.43 mg/L, which reached the effect concentration for snail control. After spraying for 7 days in the field, the average density of living snails reduced by 88.20% as compared to that before spraying, and the adjusted mortality of snails was 87.65%. The unit cost of Jingcen DY-1 spraying tanker was 0.086 7 Yuan/m². which reduced by 58.20% as compared to that of the conventional spraying tanker. CONCLUSIONS: Jingcen DY-1 type spraying tanker for snail control which integrates various equipments together can effectively control the concentration and dose of the molluscicide, and the machine is labor-saving, efficient, economic and well adapted, and is worthy to be widely applied.

ABSTRACT

Supramolecular isomerism for coordination networks refers to the existence of different architectures having the same building blocks and identical stoichiometries. For a given building block, different arrangements can lead to the formation of a series of supramolecular isomers. Two one-dimensional Co(II) coordination polymers based on N,N'-bis(pyridin-3-yl)oxalamide (BPO), both catena-poly[[[dichloridocobalt(II)]-bis[µ-N,N'-bis(pyridin-3-yl)oxalamide-κ(2)N:N']] dimethylformamide disolvate], {[CoCl2(C12H10N4O2)2]·2C3H7NO}n, have been assembled by the solvothermal method. Single-crystal X-ray diffraction analyses reveal that the two compounds are supramolecular isomers, the isomerism being induced by the orientation of the dimethylformamide (DMF) molecules in the crystal lattice.

ABSTRACT

Currently, the design and construction of an intelligent stimuli-responsive gel system is still a significant challenge. We present here a new gel system from which the formation of heat-set gels, conventional gels and irreversible heat-set gels can be achieved in aromatic solvents. This gel system is based on two-component gelators containing a succinamic acid derivative (SAD) and a primary alkyl amine (R-NH2). With the increase of temperature to 85 °C, a rarely reported reversible heat-set gel (gel formation with the increase of temperature) is afforded. Upon addition of fatty acids into two-component gelators, a conventional gel (gel formation with the decrease of temperature) is formed. When the fatty acid is replaced with dicarboxylic acid, a new heat-set gel is generated, which is irreversible and thermally super-stable. X-ray diffraction analysis reveals that the formation of a reversible heat-set gel relies on electrostatic interactions, hydrogen bonds, and hydrophobic interactions. These two-component gelators show a perfect gel system for the formation of diverse gels including heat-set gels, conventional gels and irreversible heat-set gels. The tunable strategy demonstrated in this letter may provide a new way for creation of more functional gels in gel science.

ABSTRACT

An extremely stable hydrogen-bonded organic framework, HOF-8, was fabricated. HOF-8 is not only thermally stable but also stable in water and common organic solvents. More interestingly, desolvated HOF-8 exhibits high CO2 adsorption as well as highly selective CO2 and C6H6 adsorption at ambient temperature.

ABSTRACT

In the title polymer, {[Cu(C(8)H(12)N(2)O(6)P(2))]·2H(2)O}(n), the geometry of the five-coordinate Cu(II) ion can best be described as slightly distorted square-pyramidal formed by one N and two O atoms of an N(CH(2)PO(3)H)(2) group and one N atom from a pyridine ring. The elongated apex of the pyramid is occupied by one O atom from a third diphospho-nate ligand. The inter-connection of Cu(2+) ions by the diphospho-nate ligands results in the formation of a double-chain array along the b axis, in which the two sub-chains are inter-locked by pairs of PO(3) groups. The outside of each sub-chain is decorated by other PO(3) groups. These double chains are further assembled into a three-dimensional supra-molecular architecture via a large number of O-H⋯O hydrogen bonds between the phospho-nate groups and lattice water mol-ecules.

ABSTRACT

OBJECTIVE: To evaluate the effect of surgical manipulation on the dissemination of cancer cells into blood circulation in patients with gastric cancer and to analyze its risk factors. METHODS: This study included 45 consecutive patients with gastric cancer undergoing curative resection and 13 control cases (10 healthy persons and 3 patients with peptic ulcer receiving gastrectomy). Peripheral blood was obtained preoperatively and just after surgical manipulation. The mRNA levels of carcinoembryonic antigen (CEA) from the blood samples were assayed by reverse transcription-polymerase chain reaction(RT-PCR) and compared between the 2 groups. RESULTS: CEA mRNA was negative in all control cases. Of the 45 gastric cancer patients, the preoperative positive rate of CEA mRNA was 8.9%, while the postoperative positive rate was 48.9%, which was significantly higher than that of preoperation (P=0.000). Multivariable Logistic regression analysis showed that operative duration (P=0.014) and tumor depth (P=0.010) were independent risk factors for cancer cell dissemination. Furthermore, the operative duration in patients with positive postoperative CEA mRNA was markedly longer than that in patients with negative postoperative CEA mRNA (P=0.000), and positive rate of postoperative CEA mRNA in advanced gastric cancer was higher compared with that in early gastric cancer (P=0.034). CONCLUSIONS: Surgical manipulation of curative gastrectomy can provoke dissemination of cancer cells into blood circulation, and the operative duration and tumor invasion depth may be 2 of the risk factors for cancer cell dissemination.

ABSTRACT

The low-molecular-weight chitosan-Ce(III) complexes were synthesized at pH value 1-2 and characterized by UV spectrum, fluorescence spectrum, FTIR spectrum and 1H NMR spectrum. Its coordination mechanism was also investigated. The results showed that Ce(III) was coordinated with -OH group instead of -NH2 group of low-molecular-weight chitosan.

ABSTRACT

AIM: To study the alkaloid constituents of the root of Lindera angustifolia Cheng. METHODS: The constituents were isolated and purified by column chromatography and the structures were characterized by spectral analysis. RESULTS: Seven aporphine alkaloids, laurotetanine (I), N-methyllaurotetanine (II), boldine (III), isoboldine (IV), norboldine (V), N-ethoxycarbonyllaurotetanine (VII) and a quaternary isoquinoline alkaloid, magnocurarine (VI), were isolated and identified. The structure of VII was further identified by semi-synthesis with I as starting material. CONCLUSION: All compounds were obtained from this plant for the first time and compound VII was found as a naturally occurring compound for the first time.
