ABSTRACT

Insitu stabilization and phytoextraction are considered as two convenient and effective technologies for the remediation of toxic elements (TEs) in soils. However, the effectiveness of these two remediation technologies together on the bioavailability and phytoextraction of TEs in field trials has not been explored yet. Specifically, the remediation potential of fly ash (FA; as stabilizing agent) and ryegrass (as a TE accumulator) intercropped with a target crop for soil polluted with multiple TEs has not been investigated yet, particularly in long-term field trials. Therefore, in this study, a six-month combined remediation field experiment of FA stabilization and/or ryegrass intercropping (IR) was carried out on the farmland soils contaminated with As, Cd, Cr, Cu, Hg, Ni, Pb and Zn where Zanthoxylumbungeanum (ZB) trees as native crops were grown for years. The treatments include soil cultivated alone with ZB untreated- (control) and treated-with FA (FA), produced by burning lignite in Shaanxi Datong power plant, China, soil cultivated with ZB and ryegrass untreated- (IR) and treated-with FA (FA + IR). This was underpinned by a large-scale survey in Daiziying (China), which showed that the topsoils were polluted by Cd, Cu, Hg and Pb, and that Hg and Pb contents in the Zanthoxylumbungeanum fruits exceeded their allowable limits. The TEs contents in the studied FA were lower than their total element contents in the soil. The DTPA-extractable TEs contents of the remediation modes were as follows: FA < FA + IR < IR < control. Notably, TEs contents in the ZB fruits were lowest under the FA + IR treatment, which were decreased by 27.6% for As, 42.3% for Cd, 16.7% for Cr, 30.5% for Cu, 23.1% for Hg, 15.5% for Ni, 33.2% for Pb and 38.1% for Zn compared with the control treatment. Whereas the FA + IR treatment enhanced TEs contents in ryegrass shoots and roots, and the TEs contents in ryegrass shoots were below their regulatory limits for fodder crops. The findings confirmed that the combined remediation strategy, i.e., FA (with low content of TEs) stabilization effect and intercropping of ZB (target crop) and ryegrass (accumulating plant) could provide a prospective approach to produce target plants within safe TEs thresholds with greater economic benefits, while remediating soils polluted with multiple TEs and mitigating the potential ecological and human health risk. Those results are of great applicable concern.

ABSTRACT

BACKGROUND: Chronic inflammation is considered the most critical predisposing factor of hepatocellular carcinoma (HCC), with inflammatory cell heterogeneity, hepatic fibrosis accumulation, and abnormal vascular proliferation as prominent features of the HCC tumor microenvironment (TME). Cancer-associated fibroblasts (CAFs) play a key role in HCC TME remodeling. Therefore, the level of abundance of CAFs may significantly affect the prognosis and outcome in HCC patients. METHODS: Unsupervised clustering was performed based on 39 genes related to CAFs in HCC identified by single-cell RNA sequencing data. Patients of bulk RNA were grouped into CAF low abundance cluster and high abundance clusters. Subsequently, prognosis, immune infiltration landscape, metabolism, and treatment response between the two clusters were investigated and validated by immunohistochemistry. RESULTS: Patients in the CAF high cluster had a higher level of inflammatory cell infiltration, a more significant immunosuppressive microenvironment, and a significantly worse prognosis than those in the low cluster. At the metabolic level, the CAF high cluster had lower levels of aerobic oxidation and higher angiogenic scores. Drug treatment response prediction indicated that the CAF high cluster could have a better response to PD-1 inhibitors and conventional chemotherapeutic agents for HCC such as anti-angiogenic drugs, whereas CAF low cluster may be more sensitive to transarterial chemoembolization treatment. CONCLUSIONS: This study not only revealed the TME characteristics of HCC with the difference in CAF abundance but also further confirmed that the combination therapy of PD-1 inhibitors and anti-angiogenic drugs may be more valuable for patients with high CAF abundance.

ABSTRACT

Background: It is important to assess the proliferation of endometrial carcinoma (EC) noninvasively using imaging methods. This prospective diagnostic study investigated the value of biexponential and stretched exponential models of intravoxel incoherent motion (IVIM) and dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) in predicting the Ki-67 status of EC. Methods: In all, 70 patients with EC underwent pelvic MRI. The diffusion coefficient (D), pseudo diffusion coefficient (D*), perfusion fraction (f), distributed diffusion coefficient (DDC), water molecular diffusion heterogeneity index (α), volume transfer constant (Ktrans), rate transfer constant (Kep), and volume of extravascular extracellular space per unit volume of tissue (Ve) were compared. The area under the receiver operating characteristic (ROC) curve (AUC) was used to quantify diagnostic efficacy. Multivariate logistic regression and bootstrap (1,000 samples) analyses were used to establish and evaluate, respectively, the optimal model to predict Ki-67 status. Results: D, Ktrans, and Kep were lower while α was higher in the high-proliferation group as compared with low-proliferation group (all P values<0.05). D and Kep were independent predictors of Ki-67 status in EC, and the combination of these parameters had optimal diagnostic efficacy (AUC =0.920; sensitivity 85.71%; specificity 89.29%), which was significantly better than that of D (AUC =0.753; Z=2.874; P=0.004), α (AUC =0.715; Z=3.505; P=0.001), Ktrans (AUC =0.808; Z=2.741; P=0.006), and Kep (AUC =0.832; Z=2.147; P=0.032) alone. The validation model showed good accuracy (AUC =0.882; 95% confidence interval 0.861-0.897) and consistency (C-statistic =0.902). D, Kep, Ktrans, and α showed a slightly negative (r=-0.271), moderately negative (r=-0.534), slightly negative (r=-0.409), and slightly positive (r=0.488) correlation with the Ki-67 index, respectively (all P values <0.05). Conclusions: IVIM- and DCE-MRI-derived parameters, including D, α, Ktrans, and Kep, were associated with Ki-67 status in EC, and the combination of D and Kep may serve as a superior imaging marker for the identification of low- and high-proliferation EC.

ABSTRACT

Corilagin is a naturally occurring water-soluble retrogallic acid tannin, which can be extracted from many kinds of plants. Known at present, it is the main effective ingredient of Phyllanthus urinaria L., Geranium wilfordii Maxim., Phyllanthus matsumurae Hayata, and Trifolium repens L. It also exists in Phyllanthus emblica L., Dimocarpus longan Lour., Canarium album (Lour.) Raeusch., and Terminalia chebula Retz. It can participate in a variety of signaling pathways in vivo and has multiple biological activities, including antitumor, anti-microbial, anti-oxidation, anti-inflammation, hepatoprotective, anti-allergy, anti-proliferation and so on. Given the limited efficacy of first-line treatments for many diseases such as oncology, chronic liver disease, and rheumatic immune system diseases, and the potential for adverse effects to outweigh the therapeutic effects, attention is being focused on alternative treatments, and natural plant extracts are a natural target for alternative treatments, as natural substances tend to have low toxicity to normal tissues. Some proprietary Chinese medicines containing corilagin have been used in clinical applications, being clinically applied to treat chronic liver disease, viral hepatitis B, rheumatoid arthritis and other diseases. This paper reviews the extraction, determination, distribution and harvesting, pharmacokinetics, biological activity, safety assessment of corilagin and its application in clinical practice.

ABSTRACT

It is of great significance to explore the remediation pattern in actual heavy metal (HM) contaminated sites. The field trial was carried out to research the remediation effect of biochar near a lead-zinc smelter in Feng County, China, under the rotation condition of different crops. This kind of cultivation mode is very representative in northern of China. And the pattern of production and restoration is suitable for scarce land resources and large food demand in China. The changes of soil physiochemical properties with the biochar addition, crop growth and the accumulated HMs by crops were focused on. The results showed the biochar application was excellent in improving soil nutrient elements and crop growth. The contents of TK were more obvious than those of TN and TP, with an increase of 2.6%-28.2% compared with the controls (without the addition of biochar). The yield of first season crops, i.e., soybean and corn, increased by 30%-42% and 34%-61%, respectively, and the second season crops, i.e., rape and wheat, with the increment of 25%-41% and 9%-29%, respectively. The availability forms of Cd and Pb decreased by 1.07-10.0% and 2.92-8.35%, respectively. While the improvement on the status of the HMs accumulated by crops was disappointing. The contents of HMs accumulated by crops increased to varying degrees (e.g., Pb and Zn in root, Cu and Pb in grain, and Cd in stems and leaves). Moreover, the concentrations of HMs in seeds of crops were higher than the limited levels given by the Chinese directive. Considering the results of the study and food safety, it is suggested to change the nature of the land around the smelter into woodland or construction land to prohibit the cultivation of food crops in this area.

ABSTRACT

BACKGROUND: Noninvasive identification of the histological features of endometrioid adenocarcinoma is necessary. This study aimed to investigate whether amide proton transfer-weighted imaging (APTWI) and multimodel (monoexponential, biexponential, and stretched exponential) diffusion-weighted imaging (DWI) could predict the histological grade of endometrial adenocarcinoma (EA). In addition, we analyzed the correlation between each parameter and the Ki-67 index. METHODS: A total of 90 EA patients who received pelvic magnetic resonance imaging (MRI) were enrolled. The magnetization transfer ratio asymmetry [MTRasym (3.5 ppm)], apparent diffusion coefficient (ADC), diffusion coefficient (D), pseudo-diffusion coefficient (D*), perfusion fraction (f), distributed diffusion coefficient (DDC), and water molecular diffusion heterogeneity index (α) were measured and compared. Correlation coefficients between each parameter and histological grade and the Ki-67 index were calculated. Statistical methods included the independent samples t test, Spearman's correlation, and logistic regression. RESULTS: MTRasym (3.5 ppm) [(3.72%±0.31%) vs. (3.27%±0.48%)], f [(3.15%±0.36%) vs. (2.69%±0.83%)], and α [(0.89±0.05) vs. (0.81±0.09)] were higher and ADC [(0.82±0.08) vs. (0.89±0.10) ×10-3 mm2/s], D [(0.67±0.09) vs. (0.81±0.11) ×10-3 mm2/s], and DDC [(1.04±0.09) vs. (1.13±0.13) ×10-3 mm2/s] were lower in high-grade EA than in low-grade EA (P<0.05). MTRasym (3.5 ppm) and D were independent predictors for the histological grade of EA. The combination of MTRasym (3.5 ppm) and D were better able to identify high- and low-grade EA than was each parameter. MTRasym (3.5 ppm) and α were moderately and weakly positively correlated, respectively, with histological grade and the Ki-67 index (r=0.528, r=0.514, r=0.395, and r=0.367; P<0.05). D was moderately negatively correlated with histological grade and the Ki-67 index (r=-0.540 and r=-0.529; P<0.05). DDC was weakly and moderately negatively correlated with histological grade and the Ki-67 index, respectively (r=-0.473 and r=-0.515; P<0.05). ADC was weakly negatively correlated with histological grade and the Ki-67 index (r=-0.417 and r=-0.427; P<0.05). f was weakly positively correlated with histological grade and the Ki-67 index (r=0.294 and r=0.355; P<0.05). CONCLUSIONS: Our study found that both multimodel DWI and APTWI could be used to estimate the histological grade and Ki-67 index of EA, and the combination of high MTRasym (3.5 ppm) and low D may be an effective imaging marker for predicting the grade of EA.

ABSTRACT

BACKGROUND: The transformation of hepatic stellate cell (HSC) to myofibroblast is a key event during liver fibrogenesis. However, the differentiation trajectory of HSC-to-myofibroblast transition and the switching genes during this process remains not well understood. METHODS: We applied single-cell sequencing data to reconstruct a single-lineage pseudotime trajectory of HSC transdifferentiation in vitro and analyzed the gene expression patterns along the trajectory. GeneSwitches was used to identify the order of critical gene expression and functional events during HSC activation. RESULTS: A novel cell state during HSC activation was revealed and the HSCs belonging to this state may be an important origin of cancer-associated fibroblasts (CAFs). Combining single-cell transcriptomics with GeneSwitches analyses, we identified some distinct switching genes and the order at which these switches take place for the new state of HSC and the classic culture-activated HSC, respectively. Based on the top switching genes, we established a four-gene combination which exhibited highly diagnostic accuracy in predicting advanced liver fibrosis in patients with nonalcoholic fatty liver disease (NAFLD) or hepatitis B (HBV). CONCLUSION: Our study revealed a novel cell state during HSC activation which may be relevant to CAFs, and identified switching genes that may play key roles in HSC transdifferentiation and serve as predictive markers of advanced fibrosis in patients with chronic liver diseases.

ABSTRACT

Rapid composting by black soldier fly larvae (BSFL) may be insufficient to maturation and humification of composting and further composting is necessary. The purpose of this study was to explore cornstalk addition on toxic metals (Cu, Zn, Pb and Cd), toxic metals resistance bacterial (TMRB) destiny and their relationship with physicochemical factors during BSFL manure composting. High-throughput sequencing was performed by six treatments, namely T1 to T6, where T1 to T3 were BSFL manures from chicken, pig and dairy manure, respectively, and T4 to T6 were same manures and utilized cornstalk to adjust C/N to 25. The results showed that cornstalk amendment could enhance the toxic metals immobilization rate compared to control treatments in the ultimate product. TMRB indicated that the major potential hosts bacteria were Firmicutes, Bacteroidota, Proteobacteria, Acidobacteriota and Actinobacteriota, and the sum relative abundance were 63.33%, 90.62%, 83.62%, 69.38%, 50.66% and 90.52% in T1 to T6 at the end of composting. Bacteria diversity and heat map revealed composting micro-ecology with additive cornstalk to remarkably effect main resistant bacterial distribution via adjusting environmental factors and potential hosts bacterial. Finally, T5 treatment was able to greatly decrease the TMRB abundance, and improve the ability of composting and ultimate product quality.

ABSTRACT

Land surface albedo plays a crucial role in the land surface energy budget and climate. This paper identified the spatiotemporal variations of surface albedo on the Tibetan Plateau (TP) from 1982 to 2015, and quantified the relationships between the spatial and temporal patterns of the albedo and associated influencing factors (snow cover, vegetation, and soil moisture) on the seasonal and interannual basis using satellite products and reanalysis data. It was determined that the albedo presented a distinct spatial variability, with high values in mountainous areas and low values on the southeastern TP. Spatially, average albedo exhibited a positive correlation with snow cover and negative correlations with vegetation and soil moisture. Average albedo over the whole TP had a clear seasonal cycle with a peak in winter and a minimum value in summer, which is dictated by seasonal changes in snow and vegetation covers. Annual average albedo exhibited a weakly downward trend, which was mainly contributed by a significant decrease in summer, pointing to the important role in vegetation dynamics for temporal change of the albedo. On the regional basis, interannual variation of albedo was more responsive to snow cover over the snow- and vegetation-coexisting area than the snow-covered area, and to changes in Normalized Difference Vegetation Index (NDVI) over the vegetation-covered area than the snow- and vegetation-coexisting area; albedo had a weakly negative correlation with soil moisture over bare soil. Furthermore, our results indicated that snow cover was the dominant factor for albedo change on mountainous areas, and vegetation change predominated the variation of albedo on the eastern, southern, and northwestern TP. Specifically, variations in snow cover contributed more than those of vegetation to the interannual albedo variation over the Three Rivers Headwater Region. These results would be beneficial for better understanding the climate and eco-environment changes over the TP.

ABSTRACT

Tree-rings of Picea crassifolia from high (3000 m), middle (2750 m) and low (2500 m) altitudes were collected from the Xiying River basin in the eastern Qilian Mountains, with the standard chronology being established using dendrochronological methods. Results of the correlation analysis between tree-ring width index and temperature and precipitation during different periods showed that radial growth of P. crassifolia at different altitudes responded differently to climate warming in the eastern Qilian Mountains. Water and heat availability were the main limiting factors for the radial growth of P. crassifolia in the eastern part of Qilian Mountains. The growth limiting factors at different altitudes were generally the same. Before the prominent warming (1961-1986), radial growth of P. crassifolia at the three sampling altitudes was significantly negatively correlated with mean maximum temperature in July-August of the previous year and August of the current year, and significantly positively correlated with the relative humidity in June of the current year. After the prominent warming period (1986-2014), tree growth at high, middle, and low altitudes remained significantly negatively correlated with air temperature, changed from non-significantly negative to significantly positive correlation with relative humidity in February of the current year, and changed from significantly positive to non-significantly negative correlation with precipitation and relative humidity in June. Warming caused a slowing growth of P. crassifolia tree-ring at all altitudes, with the highest altitude being the most sensitive. Drought stress caused by climate warming might be the main reason for the changes of radial growth of P. crassifolia.

BACKGROUND: Endometrial carcinoma (EC) risk stratification is generally based on histological assessment. It would be beneficial to perform risk stratification noninvasively by MRI. PURPOSE: To investigate the application of amide proton transfer-weighted imaging (APTWI), monoexponential, biexponential, and stretched exponential intravoxel incoherent motion (IVIM), and diffusion kurtosis imaging (DKI) for the evaluation of risk stratification in early-stage EC. STUDY TYPE: Prospective. POPULATION: Eighty patients with early-stage EC (47 classified as low risk, 20 as medium risk, and 13 as high risk by histological grade and International Federation of Gynecology and Obstetrics stage). FIELD STRENGTH/SEQUENCE: T1-weighted imaging, T2-weighted imaging, IVIM, APTWI, and DKI MRI at 3 T. ASSESSMENT: The magnetization transfer ratio asymmetry (MTRasym [3.5 ppm]), apparent diffusion coefficient (ADC), diffusion coefficient (D), pseudo diffusion coefficient (D*), perfusion fraction (f), distributed diffusion coefficient (DDC), water molecular diffusion heterogeneity index (α), mean kurtosis (MK), and mean diffusivity (MD) were calculated and compared between low-risk and non-low-risk groups. STATISTICAL TESTS: Individual sample t test, analysis of variance, and logistic regression. A P-value <0.05 was considered statistically significant. RESULTS: The α, ADC, D, DDC, and MD were significantly higher and the f, MK, and MTRasym (3.5 ppm) were significantly lower in the low-risk group than in the non-low-risk group. The difference in D* between the two groups was not significant (P = 0.289). MTRasym (3.5 ppm), D, and MK were independent predictors of risk stratification. The combination of these three parameters was better able to identify low- and non-low-risk groups than each individual parameter. DATA CONCLUSION: The IVIM, DKI, and APTWI parameters have potential as imaging markers for risk stratification in early-stage EC. LEVEL OF EVIDENCE: 2 TECHNICAL EFFICACY: Stage 3.

OBJECTIVES: To investigate whether amide proton transfer-weighted imaging (APTWI) and diffusion kurtosis imaging (DKI) can be used to evaluate endometrial carcinoma (EC) in terms of clinical type, histological grade, subtype, and Ki-67 index. METHODS: Eighty-eight patients with EC underwent pelvic DKI and APTWI. The non-Gaussian diffusion coefficient (Dapp), apparent kurtosis coefficient (Kapp), and magnetization transfer ratio asymmetry (MTRasym (3.5 ppm)) were calculated and compared based on the clinical type (type I, II), histological grade (high- and low-grade), and subtype (endometrioid adenocarcinoma (EA) and non-EA). Correlation coefficients were calculated for each parameter with histological grades and the Ki-67 index. RESULTS: The MTRasym (3.5 ppm) and Kapp values were higher in the type II group and high-grade group than in the type I and low-grade groups, respectively, while the Dapp values were lower in the type I and low-grade groups, respectively (all p < 0.05). The Kapp value was higher in the EA group than in the non-EA group (p = 0.022). The Kapp value was the only independent predictor for the histological grade of EA and the clinical type of EC. The AUC (DKI) was higher than the AUC (APTWI) in the identification of type I and II EC and high- and low-grade EA (Z = 2.042, 2.013, p = 0.041, 0.044), while in the identification of EA and non-EA, only the difference in Kapp was statistically significant. Moreover, the Kapp and MTRasym (3.5 ppm) values and Dapp values correlated positively and negatively, respectively, with histological grade (r = 0.759, 0.555, 0.624, and 0.462, all p < 0.05) and Ki-67 index (r = -0.704, -0.507, all p < 0.05). CONCLUSION: Both DKI- and APTWI-related parameters have potential as imaging markers in estimating the histological features of EC, while DKI shows better performance than APTWI in this study. KEY POINTS: • DKI and APTWI can be used to preliminarily evaluate the histological characteristics of endometrial carcinoma (EC). • The Kapp was the only independent predictor for the histological grade of EA and the clinical type of EC. • The Kapp, MTRasym (3.5 ppm), and Dapp correlated positively and negatively, respectively, with histological grade and Ki-67 index.

OBJECTIVES: To compare the value of amide proton transfer-weighted imaging (APTWI) and diffusion kurtosis imaging (DKI) in differentiating benign and malignant breast lesions and analyze the correlations between the derived parameters and prognostic factors of breast cancer. METHODS: One hundred thirty-five women underwent breast APTWI and DKI. The magnetization transfer ratio asymmetry (MTRasym (3.5 ppm)), apparent kurtosis coefficient (Kapp), and non-Gaussian diffusion coefficient (Dapp) were calculated according to the histological subtype, grade, and prognostic factors (Ki-67, estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor-2 (HER-2), lymph node metastasis, and maximum lesion diameter). The differences, efficacy, and correlation between the parameters were determined. RESULTS: The Kapp value was higher and the Dapp and MTRasym (3.5 ppm) values were lower in the malignant group than in the benign group (all p < 0.001; AUC (Kapp) = 0.913, AUC (Dapp) = 0.910, and AUC (MTRasym (3.5 ppm)) = 0.796). The differences in the AUC between Kapp and MTRasym (3.5 ppm) and between Dapp and MTRasym (3.5 ppm) were significant (p = 0.023, 0.046). Kapp was moderately correlated with the pathological grade (|r| = 0.724) and mildly correlated with Ki-67 and HER-2 expression (|r| = 0.454, 0.333). Dapp was moderately correlated with the pathological grade (|r| = 0.648) and mildly correlated with Ki-67 expression (|r| = 0.400). MTRasym (3.5 ppm) was only mildly correlated with the pathological grade (|r| = 0.468). CONCLUSION: DKI is superior to APTWI in differentiating between benign and malignant breast lesions. Each parameter is correlated with some prognostic factors to a certain extent. KEY POINTS: • DKI and APTWI provide valuable information regarding lesion characterization. • Kapp, Dapp, and MTRasym (3.5 ppm) are valid parameters for the characterization of tissue microstructure. • DKI is superior to APTWI in the study of breast cancer.

ABSTRACT

BACKGROUND: Amide proton transfer-weighted imaging (APTWI) and intravoxel incoherent motion imaging (IVIM) are valuable MRI techniques applied to cancer. PURPOSE: To compare APTWI and IVIM in the diagnosis of benign and malignant breast lesions and to evaluate the correlations between different parameters (MTRasym [3.5 ppm], D, D*, and f) and prognostic factors for breast cancer. STUDY TYPE: Retrospective. POPULATION: In all, 123 breast lesions were studied before treatment, including 58 benign lesions and 65 malignant lesions. FIELD STRENGTH/SEQUENCE: Conventional MRI (T1 WI, T2 WI, and diffusion-weighted imaging [DWI]), APTWI, and IVIM MRI at 3T. ASSESSMENT: The magnetization transfer ratio asymmetry at 3.5 ppm (MTRasym [3.5 ppm]), diffusion coefficient (D), pseudo diffusion coefficient (D*), and perfusion fraction (f) values were compared between the benign and malignant groups and between groups with different expression levels of prognostic factors. STATISTICAL TESTS: Individual sample t-test, χ2 test, Spearman correlation, logistic regression, and the Delong test. RESULTS: The D and MTRasym (3.5 ppm) values of the malignant group were lower than those of the benign group; however, D* and f values were higher than those of the benign group (all P < 0.05). The areas under the curve (AUCs) of D, MTRasym (3.5 ppm), D*, and f were 0.809, 0.778, 0.670, and 0.766, respectively; however, only the difference between AUC (D) and AUC (D*) was significant (Z = 2.374, P < 0.05). The D value showed a low correlation with the pathological grade and Ki-67 expression (| r | = 0.294, 0.367); the f value showed a low correlation with estrogen receptor (ER) expression (| r | = 0.382); and the MTRasym (3.5 ppm) value showed a low correlation with pathological grade (| r | = 0.371). DATA CONCLUSION: This analysis revealed that both IVIM and APTWI could be used for the differential diagnosis of benign and malignant breast lesions, and APTWI-derived MTRasym (3.5 ppm), IVIM-derived D, D*, and f values showed correlations with some prognostic factors for breast cancer. ABSTRACT

OBJECTIVES: To analyze the value of amide proton transfer-weighted imaging (APTWI) and diffusion kurtosis imaging (DKI) in differentiating cervical cancer (CC) pathological type, grade, and stage. METHODS: One hundred and twelve women underwent pelvic APTWI and DKI. The magnetization transfer ratio asymmetry (MTRasym, 3.5 ppm), apparent kurtosis coefficient (Kapp), and non-Gaussian diffusion coefficient (Dapp) were calculated by histological subtype, grade, and stage. The differences, efficacy, and correlation between parameters were determined. RESULTS: The MTRasym(3.5 ppm) and Dapp values of the adenocarcinoma (CA) group were higher than those of the cervical squamous carcinoma (CSC) group, while the Kapp values were lower than those of the CSC group. The MTRasym(3.5 ppm) and Kapp values of the high-grade group were higher than those of the low-grade group, while the Dapp values were lower than those of the low-grade group. The Dapp values of the advanced-stage group were lower than those of the early-stage group, while the Kapp values were greater than those of the early-stage group. The Kapp showed the highest efficacy in differentiating CSC and CA, high- and low-grade CC, and advanced- and early-stage CC. In the CSC and CA groups, both the Kapp and Dapp were highly correlated with pathological grade, and the MTRasym(3.5 ppm) was weakly correlated with pathological grade. The Kapp, Dapp, and MTRasym(3.5 ppm) were all weakly correlated with pathological stage. CONCLUSION: Both DKI and APTWI can be used in preliminary evaluations of CC, but DKI has advantages in the identification of pathological type, grade, and stage. KEY POINTS: • PTWI and DKI provide new information regarding cervical cancer. • MTRasym(3.5 ppm), Dapp, and Kapp are valid parameters to characterize tissue microstructure. • DKI is superior to APTWI in the study of cervical cancer.

ABSTRACT

Brassica juncea L. is an attractive species in PTMs contaminated soil remediation ascribing to its high tolerance under stress and great accumulation capacity of metals. To identify the potential Cd/Zn accumulators from numerous different Chinese mustard cultivars for practical phytoremediation is a promising strategy in China. In present work, a pot experiment involving elevated Cd/Zn concentrations was performed among 21 cultivars. Regarding physiological and biochemical indicators under Cd/Zn stress, principal component analysis and cluster analysis were used for cultivar tolerance evaluation and classification. Results showed that BJ (Bao Jie, var. involutus) cultivar was distinguished as a potential phytoremediation candidate comparing with other cultivars. Moreover, BJ accumulated the maximum Cd content of 63.85 and 77.29 mg kg-1 DW in shoots and roots, respectively, and the maximum Zn uptake by BJ were 6693 and 4777 mg kg-1 DW in shoots and roots, respectively. Accordingly, BJ had the highest Cd/Zn tolerance, remarkable accumulation and translocation capacity (accumulation factor (AF) > 1 for Cd and Zn; translocation factor (TF) > 0.8 for Cd and TF > 1 for Zn). In addition, the superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activities of the mustard increased initially under low Cd/Zn stress as compared to the control and then declined dramatically with the increasing metals exposure concentration. Therefore, the antioxidant enzymes may play a protective role against reactive oxygen species (ROS) under low Cd/Zn stress, whereas the defense system might be collapsed under relatively high Cd/Zn stress. Furthermore, the enhanced Cd/Zn exposure led to an increase in malondialdehyde (MDA) content in the mustard cultivars, indicating that Cd/Zn had induced more severe oxidative stress and higher degree of lipid peroxidation had occurred. The present investigation results indicated that BJ (Bao Jie, var. involutus), as a native cultivar, can be further applied in the field trials of phytoremediation practices in contaminated soil.

ABSTRACT

PURPOSE: To explore the possibility of using amide proton transfer-weighted imaging (APTWI) for the identification and diagnosis of cervical squamous carcinoma (CSC), cervical adenocarcinoma (CA) and different levels of CSC. MATERIALS AND METHODS: Seventy-six patients with newly diagnosed uterine cervical cancer (UCC) were studied prior to treatment, including 20 with poorly differentiated (Grade 3) CSC, 23 with moderately differentiated (Grade 2) CSC, 17 with well-differentiated (Grade 1) CSC, and 16 with CA (13 with poorly differentiated (Grade 3) CA and 3 with moderately differentiated (Grade 2) CA). Differences in the magnetization transfer ratio at 3.5 ppm (MTRasym (3.5 ppm)) were identified between CSC and CA and between high-level (Grade 3) CSC and low-level (Grade 2 and Grade 1) CSC, as well as among all three grades of CSC differentiation. Receiver operating characteristic (ROC) curve analysis was used to evaluate the diagnostic thresholds and performance of the parameters. Spearman correlation analysis was used to examine the correlation between the MTRasym (3.5 ppm) and histological grade. RESULTS: The MTRasym (3.5 ppm) in CA was higher than that in CSC (P = 0.001). The MTRasym (3.5 ppm) in high-level CSC was higher than that in low-level CSC (P = 0.001). The MTRasym (3.5 ppm) was positively correlated with the grade of CSC differentiation (r = 0.498, P = 0.001). The MTRasym (3.5 ppm) in Grade 3 CSC was higher than that in Grade 2 and Grade 1 CSC (P = 0.02/0.01). No significant difference in the MTRasym (3.5 ppm) was found between Grade 2 CSC and Grade 1 CSC (P = 0.173). The area under the ROC curve (AUC) for the MTRasym (3.5 ppm) in distinguishing CSC and CA was 0.779, with a cut-off, sensitivity, and specificity of 2.97%, 60.0% and 82.5%, respectively. The AUC for distinguishing high-/low-level CSC was 0.756, with a cut-off, sensitivity, and specificity of 3.29%, 68.8% and 83.3%, respectively. CONCLUSION: APTWI may be a useful technique for the identification and diagnosis of CSC, CA and different levels of CSC, which may have an important impact on clinical strategies for treating patients with UCC.

ABSTRACT

PURPOSE: This study evaluated and compared the performances of diffusion kurtosis imaging (DKI) and diffusion-weighted imaging (DWI) for diagnosing and histologically grading endometrial cancer. MATERIALS AND METHODS: In this retrospective study, DKI and DWI data for 61 patients with endometrial cancer and 30 patients with a normal endometrium were analyzed, and the mean kurtosis (MK), mean diffusion coefficient (MD) and apparent diffusion coefficient (ADC) values for the endometrial cancer tissue and normal endometrial tissue were acquired. The parameters for the normal endometrium group (G0) and the endometrial cancer groups (G1, G2 and G3) were compared and analyzed. The receiver operating characteristic (ROC) curve was used to evaluate each parameter's diagnostic accuracy and threshold. Spearman correlation analysis was used to analyze the correlations between all parameters and histological grades. RESULTS: The MK values for the G0, G1, G2 and G3 groups increased gradually, while the MD and ADC values decreased gradually. Except for the differences in the ADC values between G0 and G1, the differences among the groups were statistically significant (P < 0.05). The MK values had the highest diagnostic accuracy in differentiating G0 and (G1 + G2 + G3), G0 and G1, G1 and G2, and G2 and G3 (AUC = 0.93, 0.76, 0.91, 0.91, P < 0.05). MK was maximally correlated with histological grade, followed by MD and ADC (MK > MD > ADC; r = - 0.85, + 0.82, + 0.76, P < 0.01). CONCLUSION: Both DKI and DWI can be used to evaluate the diagnosis and histological grading of endometrial cancer. Compared with DWI, the DKI model is a more complete mathematical model with more sensitive parameters, which can more effectively evaluate the pathological and physiological characteristics of endometrial cancer.
