ABSTRACT

Crystal facet engineering is an effective strategy for precisely regulating the orientations and electrochemical properties of metal oxides. However, the contribution of each crystal facet to pseudocapacitance is still puzzling, which is a bottleneck that restricts the specific capacitance of metal oxides. Herein, α-MnO2 nanorods with different exposed facets were synthesized through a hydrothermal route and applied to pseudocapacitors. XRD and TEM results verified that the exposure ratio of active crystal facets was significantly increased with the assistance of the structure-directing agents. XPS analysis showed that there was more adsorbed oxygen and Mn3+ on the active crystal facets, which can provide strong kinetics for the electrochemical reaction. Consequently, the α-MnO2 nanorods with {110} and {310} facets exhibited much higher pseudocapacitances of 120.0 F g-1 and 133.0 F g-1 than their α-MnO2-200 counterparts (67.5 F g-1). The theoretical calculations proved that the {310} and {110} facets have stronger adsorption capacity and lower diffusion barriers for sodium ions, which is responsible for the enhanced pseudocapacitance of MnO2. This study provides a strategy to enhance the electrochemical performance of metal oxide, based on facet engineering.

ABSTRACT

Adulteration of higher priced milks with cheaper ones to obtain extra profit can adversely affect consumer health and the market. In this study, pure buffalo milk (BM), goat milk (GM), camel milk (CM), and their mixtures with 5-50% (vol/vol) cow milk or water were used. Mid-infrared spectroscopy (MIRS) combined with modern statistical machine learning was used for the discrimination and quantification of cow milk or water adulteration in BM, GM, and CM. Compared to partial least squares (PLS), modern statistical machine learning-especially support vector machines (SVM), projection pursuit regression (PPR), and Bayesian regularized neural networks (BRNN)-exhibited superior performance for the detection of adulteration. The best prediction models for the different predictive traits are as follows: The binary classification models developed by SVM resulted in differentiation of CM-cow milk, and GM/CM-water mixtures. PLS resulted in differentiation of BM/GM-cow milk and BM-water mixtures. All of the above models have 100% classification accuracy. SVM was used to develop multi-classification models for identifying the high and low proportions of cow milk in BM, GM, and CM, as well as the high and low proportions of water adulteration in BM and GM, with correct classification rates of 94%, 100%, 100%, 99%, and 100%, respectively. In addition, a PLS-based model was developed for identifying the high and low proportions of water adulteration in CM, with correct classification rates of 100%. A regression model for quantifying cow milk in BM was developed using PCA + BRNN, with RMSEV = 5.42%, and RV2 = 0.88. A regression model for quantifying water adulteration in BM was developed using PCA + PPR, with RMSEV = 1.70%, and RV2 = 0.99. Modern statistical machine learning improved the accuracy of MIRS in predicting BM, GM, and CM adulteration more effectively than PLS.

ABSTRACT

Semiconductors exhibit great potential as a surface enhanced Raman scattering (SERS) substrate due to their low cost, good stability and biocompatibility. However, the extensive application of semiconductors has been restricted by their intrinsically low SERS sensitivity. It is urgently required to design uniform metal oxide substrates with enhanced charge transfer and SERS activity. Herein, three facet-defined ({101̄0}, {0001} and {101̄1}) ZnO films were synthesized via an electrodeposition procedure with the assistance of KCl or ethylenediamine. According to the results, the ZnO films with {0001} and {101̄1} exposed facets exhibit appreciable SERS enhancement factors (EFs) of 1.6 × 104 and 2.8 × 104 for 4-nitrobenzenethiol (4-NBT), as well as a relatively low limit of detection (LOD) down to 1 × 10-6 M and 5 × 10-7 M, respectively. Simultaneously, the electrodeposited ZnO films deliver good repeatability and SERS stability, with relative standard deviation (RSD) less than 6% and 85.2% of their original activity retained after 40 days. Theoretical calculations verified that the {0001} and {101̄1} facets can transfer more electrons from ZnO to the molecules on account of their low facet-related electronic work functions, thus generating the noticeable improvement of SERS activity. The current study provides theoretical and technical support for the crystal facet engineering and property improvement of semiconductors.

ABSTRACT

BACKGROUND: To investigate the antimicrobial profiles and genomic characteristics of MDR-Citrobacter spp. strains isolated from Fennec fox imported from Sudan to China. METHODS: Four Citrobacter spp. strains were isolated from stool samples. Individual fresh stool samples were collected and subsequently diluted in phosphate buffered saline as described previously. The diluted fecal samples were plated on MacConkey agar supplemented with 1 mg/l cefotaxime and incubated for 20 h at 37 °C. Matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF-MS) was used for identification. Antimicrobial susceptibility testing was performed using the broth microdilution method. Whole-genome sequencing was performed on an Illumina Novaseq-6000 platform. Acquired antimicrobial resistance genes and plasmid replicons were detected using ResFinder 4.1 and PlasmidFinder 1.3, respectively. Comparative genomic analysis of 277 Citrobacter genomes was also performed. RESULTS: Isolate FF141 was identified as Citrobacter cronae while isolate FF371, isolate FF414, and isolate FF423 were identified as Citrobacter braakii. Of these, three C. braakii isolates were further confirmed to be extended-spectrum ß-lactamases (ESBL)-producer. All isolates are all multidrug resistance (MDR) with resistance to multiple antimicrobials. Plasmid of pKPC-CAV1321 belong to incompatibility (Inc) group. Comparative genomics analysis of Citrobacter isolates generated a large core-genome. Genetic diversity was observed in our bacterial collection, which clustered into five main clades. Human, environmental and animal Citrobacter isolates were distributed into five clusters. CONCLUSIONS: To our knowledge, this is the first investigation of MDR-Citrobacter from Fennec Fox. Our phenotypic and genomic data further underscore the threat of increased ESBL prevalence in wildlife and emphasize that increased effort should be committed to monitoring the potentially rapid dissemination of ESBL-producers with one health perspective.

ABSTRACT

The emergence of carbapenem resistance (CR) caused by hydrolytic enzymes called carbapenemases has become a major concern worldwide. So far, CR genes have been widely detected in various bacteria. However, there is no report of CR gene harboring Comamonas thiooxydans. We first isolated a strain of an IMP-8-producing C. thiooxydans from a patient with urinary tract infection in China. Species identity was determined using MALDI-TOF MS analysis and carbapenemase-encoding genes were detected using PCR. The complete genomic sequence of C. thiooxydans was identified using Illumina Novaseq and Oxford Nanopore PromethION. Antimicrobial susceptibility analysis indicated that the C. thiooxydans strain ZDHYF418 was susceptible to imipenem, intermediate to meropenem, and was resistant to aztreonam, fluoroquinolones, and aminoglycosides. The bla IMP- 8 gene was chromosomally located, and was part of a Tn402-like class 1 integron characterized by the following structure: DDE-type integrase/transposase/recombinase-tniB-tniQ-recombinase family protein-aac(6')-Ib-cr-bla IMP- 8-intI1. Phylogenetic analysis demonstrated that the closest relative of ZDHYF418 is C. thiooxydans QYY (accession number: CP053920.1). We detected 330 SNP differences between ZDHYF418 and C. thiooxydans QYY. Strain QYY was isolated from activated sludge in Jilin province, China in 2015. In summary, we isolated a strain of C. thiooxydans that is able to produce IMP-8 and a novel bla OXA . This is the first time that a CR gene has been identified in C. thiooxydans. The occurrence of the strain needs to be closely monitored.

ABSTRACT

To investigate the oral microflora of patients with oesophageal squamous cell carcinoma (ESCC), saliva samples were collected from 20 patients with ESCC and 21 healthy controls. The V3-V4 region of 16S rDNA was amplified and sequenced by the Illumina MiSeq high-throughput sequencing platform. The final sequences were used for OTU analysis. Alpha and beta diversity analysis showed that the bacterial diversity and richness of the ESCC group were lower than those of the control group, while the variability of the ESCC group was higher than that of the control group. According to the Metastats difference analysis and LEfSe analysis, the high risk of ESCC may be related to Actinomyces and Atopobium, while the healthy control group is closely related to Fusobacterium and Porphyromonas (the analysis was performed at the genus level). The establishment of the relationship between oral microbiota and risk of ESCC may lead to significant advances in understanding the aetiology of cancer and may open a new research paradigm for cancer prevention.

ABSTRACT

The aim of this study was to investigate the occurrence and genomic characteristics of extended-spectrum-ß-lactamase-producing Escherichia coli (ESBL-EC) in fennec fox imported from Sudan to China. We screened 88 fecal samples from fennec fox for ESBL-EC, using cefotaxime- and meropenem-supplemented selective medium. Antimicrobial susceptibility testing was performed by the agar dilution method except for colistin and tigecycline; for colistin and tigecycline, testing was conducted by the broth microdilution method. ESBL-EC bacteria were sequenced, and their genomes were characterized. Plasmid conjugation, S1 nuclease pulsed-field gel electrophoresis (PFGE), and Southern blotting were performed for a MCR-1-producing isolate. The genetic environment of mcr-1 and ESBL genes was also investigated. A total of 29 ESBL-EC bacteria were isolated from 88 fennec fox (32.9%), while no carbapenemase producers were found. The most prevalent genotypes were the blaCTX-M-55 and blaCTX-M-14 genes, followed by blaCTX-M-15 and blaCTX-M-64 We detected nine sequence types among 29 ESBL-EC. Furthermore, the mcr-1 gene was detected in isolate EcFF273. Conjugation analysis confirmed that the mcr-1 gene was transferable. S1 PFGE, Southern blotting, and whole-genome sequencing revealed that mcr-1 and blaCTX-M-64 were both located on a 65-kb IncI2 plasmid. This study reports for the first time the occurrence of ESBL-EC in fennec fox. The high prevalence of ESBL producers and the occurrence of MCR-1 producer in fennec fox imported into China from Sudan are unexpected. In addition, it clearly demonstrated that commensal E. coli strains can be reservoirs of blaCTX-M and mcr-1, potentially contributing to the dissemination and transfer of such genes to pathogenic bacteria among fennec fox. Our results support the implication of fennec fox as a biological vector for ESBL-producing members of the Enterobacteriaceae family.IMPORTANCE The extended-spectrum-ß-lactamase (ESBL)-producing members of the Enterobacteriaceae family are a global concern for both animal and human health. There is some information indicating a high prevalence of ESBL producers in food animals. Moreover, there have been an increasing number of reports on ESBL-producing strains resistant to the last-resort antibiotic colistin with the global dissemination of the plasmid-mediated mcr-1 gene, which is believed to have originated in animal breeding. However, little is known regarding the burden of ESBL-producing Enterobacteriaceae on wild animals. No data were available on the prevalence of antimicrobial resistance (AMR) among wild animals imported into China. This is the first study to investigate the microbiological and genomics surveillance investigation of ESBL colonization among fennec fox (Vulpes zerda) imported from Sudan to China, and we uncovered a high prevalence of ESBL-EC. Furthermore, the underlying mechanism of colistin resistance in an isolate that harbored mcr-1 was also investigated. Results of characterization and analysis of 29 ESBL-producing E. coli may have important implications on our understanding of the transmission dynamics of these bacteria. We emphasize the importance of improved multisectoral surveillance for colistin-resistant E. coli in this region.

ABSTRACT

The relationship between acupuncture and stress is discussed from three aspects, including is it possible for acupuncture stimulation to be a stressor, whether acupuncture will start stress reaction, and whether acupuncture effects contain some stress factors. It is believed that correct acupuncture manipulation will not cause stress response, however, under some circumstances, such as inaccurate manipulation, improper treatment or patients who are very nervous but do not receive effective intervention, acupuncture is likely to cause stress response. Acupuncture-induced stress response is totally different from acupuncture anti-stress. The possible stress factors in acupuncture effect are explored, which can provide a new angle for the research on action mechanism of acupuncture. From the view of stress to review acupuncture treatment, there are three enlightenments: emphasizing on communication between doctors and patients, avoiding over-pursuit of deqi and focusing on analysis of the body constitution.

ABSTRACT

OBJECTIVE: To explore the action mechanism of electroacupuncture (EA) at "Shenmen" (HT 7) and "Sanyinjiao" (SP 6) on insomnia. METHODS: Thirty SD rats were randomly divided into a blank group, a model group and an EA group, 10 cases in each group. The insomnia model was made by immobilization method in the model group and EA group. After model establishment, rats in the blank group and model group were treated with fixation and no treatment was given. Rats in the EA group were treated with EA at "Shenmen" (HT 7) and "San-yinjiao" (SP 6) for 15 min, once a day for 4 days. After treatment, the level of daytime and nighttime activity, open-arm entry percentage and open-arm time percentage of elevated plus-maze test were measured; the content of noradrenaline (NE), dopamine (DA) and epinephrine (EPI) in plasma and NE, DA in thalamus and brainstem were detected by using euzymelinked immunosorbent assay method. RESULTS: Compared with the blank group, the daytime activity was increased and nighttime activity was reduced in the model group (both P<0. 05); the open-arm entry percentage and open-arm time percentage of elevated plus-maze test were both reduced in the model group (both P<0. 05); the contents of NE, DA, EPI in the plasma and NE, DA in thalamus and brainstem were increased in the model group (all P<0. 05). Compared with the model group, the daytime activity was reduced and nighttime activity was increased in the EA group (both P<0. 05); the open-arm entry percentage and open-arm time percentage of elevated plus-maze test were both increased in the EA group (both P<0. 05); the contents of NE, DA, EPI in the plasma and NE, DA in thalamus and brainstem were reduced in the EA group (all P< 0. 05). CONCLUSION: Electroacupuncture at "Shenmen" (HT 7) and "Sanyinjiao" (SP 6) can restrain the over-activity of the sympathetic-adrenal medullary system to treat the insomnia.
