ABSTRACT

ObjectiveTo explore the optimal formula of Maxing Shigantang in regulating epidermal growth factor receptor(EGFR)expression and alleviating airway injury in asthmatic rats and to reveal the underlying mechanism. MethodSD male rats were randomly divided into normal group, model group, dexamethasone group (5×10-4 g·kg-1) and Maxing Shigantang 1∶0.5, 1∶1, 1∶2 groups (group A, B, C, 10 g·kg-1), with 8 rats in each group. The other groups except the normal group received nebulization of 2% acetylcholine chloride and 0.4% histamine phosphate for the modeling of asthma. One hour before modeling, the normal group and the model group were given the same amount of normal saline, and the other groups were given the same amount of corresponding drugs, once a day for 7 days. On the 7th day, the model was established and the incubation period of asthma was recorded. The rats were then immediately anesthetized, and arterial blood and tracheal tissue were collected. Enzyme-linked immunosorbent assay (ELISA) was employed to detect the levels of interleukin-2 (IL-2), interleukin-4 (IL-4), and tumor necrosis factor-α (TNF-α) in serum. Pathological sections were prepared for the observation of the pathological changes of tracheal tissues and the ultrastructure of epithelial cells in each group. Terminal-deoxynucleotidyl transferase-mediated nick-end labeling (TUNEL) was adopted to detect epithelial cell apoptosis, and in situ hybridization and Western blot were employed to determine the mRNA and protein levels of epidermal growth factor receptor (EGFR), respectively. ResultCompared with the model group, groups A, B and C prolonged the incubation period of asthma (P<0.05,P<0.01). Compared with the control group, the model group showed declined IL-2 level (P<0.01), risen IL-4 and TNF-α levels (P<0.05,P<0.01), increased airway pathology score, collagen volume fraction, and airway epithelial cell apoptosis index (P<0.01), and up-regulated mRNA and protein levels of EGFR in trachea tissue (P<0.01). Compared with the model group, group A showed increased IL-2 level (P<0.05) and declined IL-4 (P<0.05,P<0.01) level, and group B showed declined IL-4 level (P<0.05). The level of TNF-α in groups A, B, and C declined compared with that in the model group (P<0.01). Maxing Shigantang repaired the tracheal tissue to different degrees (P<0.05). Among the three groups, group A inhibited tracheal fibrosis (P<0.05) and had the most significant effect of repairing the ultrastructural changes of airway epithelial cells. Groups A, B and C all inhibited the apoptosis of airway epithelial cells (P<0.05). All the three groups inhibited the up-regulation of EGFR mRNA level (P<0.05,P<0.01), and groups B and C inhibited the up-regulation of EGFR protein level (P<0.05,P<0.01). ConclusionMaxing Shigantang can inhibit the abnormal changes of airway epithelial structure, alleviate airway injury, and can down-regulate the expression of EGFR in the tracheal tissue of asthma model rats. In this study, the optimal compatibility of Maxing Shigantang to repair airway epithelial injury in asthmatic rats was group A, with the Ephedrae Herba-Armeniacae Semen Amarum-Glycyrrhizae Radix et Rhizoma-Gypsum Fibrosum ratio of 1∶0.5∶4∶1.

ABSTRACT

The innovation and patent layout of traditional Chinese medicine compounds reflects the innovation level of the traditio-nal Chinese medicine industry to a certain extent. Lianhua Qingwen Formula was taken as an example to analyze the innovation and patent layout of traditional Chinese medicine compounds. The study first proposed an innovative technology system for traditional Chinese medicine compounds, and then analyzed the advantages and disadvantages of Lianhua Qingwen Formula in innovation and patent layout based on 56 patents and other relevant patents. The analysis results showed that Lianhua Qingwen Formula had the following characte-ristics in terms of patent layout. In terms of innovation technical route, the patented technical route of Lianhua Qingwen Formula was mainly based on the composition and preparation of granules as the first choice, followed by corresponding process improvements, new uses, and detection methods as the main improvement routes. In terms of the corresponding patent layout, the basic patent protection scope of Lianhua Qingwen Formula completely covered marketed drugs, and then took new functions as the main layout strategy for subsequent patent applications. With the advancement of modern technical means, the preparation process and testing methods have been optimized continuously. At the same time, the international patent layout was given the priority in domestic patent application. Based on the above characteristics, the study gave suggestions for follow-up innovation and patent work for Lianhua Qingwen Formula, so as to provide enlightenment for other traditional Chinese medicine companies in exploring original innovation of traditional Chinese medicine compounds, improving innovative methods, and enhancing the ability of patent layout of innovative achievements.

ABSTRACT

BACKGROUND: This study aims to examine the effect of supportive psychological intervention (SPI) on psychological disorders (PD) in clinical medicine students (CMS) with English Learning Difficulties (ELD). METHODS: We will perform a comprehensive literature search from the following databases: Cochrane Library, MEDLINE, EMBASE, Allied and Complementary Medicine Database, Chinese Biomedical Literature Database, and China National Knowledge Infrastructure. All databases will be performed from their inception to the present without language limitation by 2 independent reviewers. We will also look for grey literature, such as conference proceedings, dissertations or theses. Newcastle-Ottawa Scale will be used to assess study quality, and RevMan 5.3 software will be applied to carry out statistical analysis. RESULTS: This study will summarize the most recent evidence to assess the effect of SPI on PD in CMS with ELD. CONCLUSION: This study may provide helpful evidence of SPI on PD in CMS with ELD. ABSTRACT

Currently, increasing importance has been attached to heavy metal pollution of water environment. In order to help geologist and environmentalist to obtain on-site experimental data, there is an urgent need to develop portable analysis techniques of water quality, which can be used to give an timely and accurate assessment of water quality of the contaminated area with low cost and to monitor water environment conveniently in daily preventing and controlling work of water pollution. Based on adsorption with Purolite S930 chelating resin at pH 4 and thin-film sample preparation technique, a rapid and simple on-site-oriented analysis method of heavy metal ions, namely Mn, Fe, Ni, Cu, Zn and Pb in aqueous solutions, was developed utilizing a handheld energy-dispersive X-ray fluorescence in this paper. The correlation coefficients of the calibration curves were greater than 0.992 for all six metal ions, the lower limits of detection were between 5.8 and 18.6 µg·L-1. Precision tests carried out on multi-element mixed solutions showed that the relative standard deviations (RSD, n=10) were better than 15%. The method has been used on-site to analysis 21 water samples collected from different river or well sites near a mining area northeast China. Out of the five underground water samples one was found contaminated by Cu and Zn (Class Ⅲ), one by Zn and Fe (Class Ⅱ), and one by Fe(in between Class Ⅱ to Ⅲ). Surface water samples showed better qualities. But two out of the sixteen were found to be contaminated by Mn heavily (Class Ⅳ). Laboratory ICPMS was used to analyze the same samples. The results for Cu, Ni and Pb were coincident with the on-site data in general. But for Zn and Mn with higher concentrations, on-site data were lower than that by ICPMS. The reason for that might be the inclusion of the fine suspending particles in the samples by ICPMS. We conclude that the on-site data were effective. With the described method, an overall data acquisition time, including sampling, processing and measuring, can be within 12 hours for a batch of 10 samples. The pre-concentration device and the XRF instrument are both small, light, portable and can operate without external power supply. So, the method is suitable for on-site water sample analysis, especially in remote areas.

ABSTRACT

Fluorescence enhancement reaction of flavoxate hydrochloride (FX) in strong alkali solution was studied, the mechanism of the reaction was investigated, and a novel fluorimetric method for analysis of FX in drug sample was established. FX has no intrinsic fluorescence, but it can slowly produce fluorescence in strong alkali solution. Heating can promote the fluorescence enhancement reaction. In 3D fluorescence spectra of the decomposition product of FX, two fluorescence peaks, located respectively at excitation wavelengths λex/ emission wavelength λem =223/410 nm, and 302/410 nm, were observed. Using quinine sulfate as a reference, fluorescence quantum yield of the decomposition product was measured to be 0.50. The structural characteriza- tion and spectral analysis of the decomposition product reveal that ester bond hydrolysis reaction of FX is firstly occurred during heating process, forming 3-methylflavone-8-carboxylic acid (MFA), then a cleavage reaction of the γ-pyrone ring of MFA occurred, producing α, β-unsaturated ketone. This product includes adjacent hydroxyl benzoic acid group in its molecule, which can form intramolecular hydrogen bond under alkaline condition, so that increase the conjugate degree and enhance the rigidity of the molecule, and thereby cause fluorescence enhancement. Based on this fluorescence enhancement reaction, a fluorimetric method was proposed for the determination of FX. A linear calibration curve covered the concentration range 0.020 3-0.487 µg · mL. The regression equation was I(F) = 23.9 + 5357.3 c, with correlation coefficient r = 0.999 7 (n = 8), detection limit D = 1.1 ng · mL(-1). The method was applied to the analysis of FX tablets, with a spiked recovery rate of 100.2%. The reliability of the method was verified by a UV-spectrophotometric method.

ABSTRACT

<p><b>OBJECTIVE</b>To assess the effects of seawater immersion of open abdominal wound and drowning on plasma electrolyte levels in dogs.</p><p><b>METHODS</b>Twenty-one dogs with open abdominal wounds were randomly divided into 3 groups, namely the control group (n=7), seawater immersion group (n=7) and seawater immersion and drowning group (n=7). The dogs in the control group were subjected to simple open abdominal wound, and those in the latter two groups were immersed into seawater after the abdominal injury. The changes in the plasma electrolytes and osmotic pressure were observed after the treatments. RESULTS AMD CONCLUSION: Seawater immersion of the open abdominal wound results in hypematremia, hyperkalemia and hypertonic dehydration, and the effect is proportional to the length of immersion time, but seawater airway perfusion does not cause obvious changes of the plasma electrolytes and osmotic pressure in dogs.</p>

ABSTRACT

A method was developed for the determination of As, Sb, Bi, Cd and Sn in degrees C, iodides of As, Sb, Bi, Cd and Sn were formed and sublimated from the sample matrix and then extracted by diluted HCl. The prepared solution was determined by ICP-AES. The sample treatment procedure, such as the ratio of NH4 I to sample, and sublimation and extraction conditions, was studied. The instrument determination parameters, including ICP power, Ar gas flow rate, spectral wavelengths, background subtraction, spectral interferences correction and internal standard, were selected and optimized. The established procedure is rapid and simple, the detection limits for the five elements are in the range of 0.01-0.35 microg x g(-1), and the analytical results for soil and sediment reference materials were in good agreement with the reference values.

ABSTRACT

An alkaline sample digestion procedure using fusion with lithium metaborate (LiBO2) after dry ashing was developed. Silicon, aluminium and other, a total of 10 elements in different kinds of biological samples, were measured by ICP-AES. The detection limit of this method was 0.15 microg x mL(-1) for Si and 0.03 microg x mL(-1) for Al. The accuracy and precision of the method were demonstrated by analyzing certified reference materials poplar leaves (GBW07604). The precision was less than 3% and the accuracy was -0.03% for Si and -3% for Al. This method was applied to the certification of a series of ten biological reference materials. The results were satisfied.
