ABSTRACT

Gastrointestinal (GI) parasites may impose detrimental consequences on wildlife populations due to their capacity to cause mortality and reduce fitness. Additionally, wild animals play an important role in the transmission of zoonoses. Despite this importance, information on GI parasites of tropical wild mammals is critically lacking. The present study aimed to document GI parasites of six wild-dwelling large mammal taxa in Sri Lanka: Asian elephant (Elephas maximus), Sloth bear (Melursus ursinus), civet (Paradoxurus sp.), Leopard (Panthera pardus), Grey langur (Semnopithecus priam) and buffalo (Bubalus sp). Fresh faecal samples (n = 56) collected from the Wasgomuwa National Park, Sri Lanka were subjected to coprological examination using faecal smears, and the brine floatation technique followed by microscopic identification; quantitative data were accrued using the formol-ether method. The survey revealed a high prevalence of GI parasites, where 86% (48/56) of faecal samples screened positive for parasitic infections. Faecal samples of the civet, buffalo and Leopard recorded 100% prevalence, while the lowest (40%) was recorded for the Grey langur. Eight types of GI parasites were documented: protozoan cysts, platyhelminth ova (three types of digenean and a single cyclophillidean type), nematode ova (strongyle, strongyloid, ascarid, and trichuroid types) and rhabditiform larvae. The buffaloes and civets had a comparatively high number and diversity of GI parasites (buffalo: 7 types, H' = 1.02; civet: 6 types, H' = 1.52), whilst only a single type (digenean) was detected in the Grey langur. Likewise, parasite loads were also highly variable; highest in the bear (486 per g faeces) and lowest in the monkey (10 per g faeces). The outcome of this survey is important on two accounts; i) to fill the knowledge gap on GI parasites of tropical wild mammals, and ii) the revelation of many first-time parasite-host records for some of the threatened wild-dwelling large mammals in Sri Lanka.

ABSTRACT

Chromium discharged in industrial effluents frequently occurs as an environmental pollutant, but the lethal and sub-lethal effects the heavy metal might cause in animals exposed to it have been insufficiently investigated. Selecting the amphibian Duttaphrynus melanostictus, we carried out laboratory tests to investigate the effects of short and long term exposure to hexavalent chromium (Cr(VI)) in both tadpoles and adult toads. The concentrations used were 0.002, 0.02, 0.2, 1.0 and 2.0mg/L, the first three corresponding to field levels. In vitro exposures were also carried out using toad erythrocytes and Cr(VI) concentrations of 0.0015, 0.003, 0.015, 0.03, 0.15mg/L. Mortality, growth retardation, developmental delays and structural aberrations were noted in the metal-treated tadpoles, with increasing incidence corresponding to increase in Cr(VI) level and duration of exposure. Many of the sub-lethal effects were evident with long term exposure to environmentally relevant levels of the toxicant. Changes in selected blood parameters and erythrocyte morphometry were also detected in Cr(VI) exposed toads, indicating anaemic and leucopenic conditions. In the genotoxicity study, DNA damage indicated by comet assay and increased micronuclei frequency, occurred at the low Cr(VI) concentrations tested. The multiple deleterious effects of exposure to chromium signal the need for monitoring and controlling the discharge of chromium to the environment. The dose-dependency and genotoxic effects observed in this widely distributed Asian toad indicates its suitability for monitoring heavy metal pollution in aquatic systems.

ABSTRACT

Continuous subchronic exposure experiments were conducted to assess the effects of diazinon, an organophosphate pesticide, on the survival, growth and activity of larvae of the Asian common toad Bufo melanostictus. Two larval stages, the gill stage (Gosner stages 21 and 22) and gill-atrophy stage (Gosner stages 24 and 25), were continuously exposed to 4 microg/L, 400 mirog/L, and 10 mg/L of commercial-grade diazinon for 30 d. Treatments and untreated controls were maintained in triplicate with water changed and pesticide concentrations renewed every 3 d. Observations showed that subchronic exposure to 400 microg/L and 10 mg/L diazinon caused a significant dose-dependent increase in mortality compared to the control, regardless of the age at which larvae were exposed. One hundred percent mortality was observed in larvae exposed to 10 mg/L. No clear age-related sensitivity was evident in this study. The lethal concentrations at which 50% of the tadpoles (LC50) died during 30 d of continuous exposure were 6 and 7.5 mg/L for gill stage and gill-atrophy stage larvae, respectively. Diazinon impaired larval growth and activity. Tail abnormalities were apparent in larvae exposed to 400 microg/L and 10 mg/L of diazinon. This investigation provides the first empirical evidence of the negative effects of diazinon on the survival, growth and activity of B. melanostictus. The high degree of diazinon toxicity in this study highlights the need to consider important nontarget groups when recommending safe levels of pesticide application.
