ABSTRACT

Blocking is often used to reduce known variability in designed experiments by collecting together homogeneous experimental units. A common modeling assumption for such experiments is that responses from units within a block are dependent. Accounting for such dependencies in both the design of the experiment and the modeling of the resulting data when the response is not normally distributed can be challenging, particularly in terms of the computation required to find an optimal design. The application of copulas and marginal modeling provides a computationally efficient approach for estimating population-average treatment effects. Motivated by an experiment from materials testing, we develop and demonstrate designs with blocks of size two using copula models. Such designs are also important in applications ranging from microarray experiments to experiments on human eyes or limbs with naturally occurring blocks of size two. We present a methodology for design selection, make comparisons to existing approaches in the literature, and assess the robustness of the designs to modeling assumptions.

ABSTRACT

Although traditionally viewed as the "powerhouse" of the cell, an accruing body of evidence in the rapidly growing field of mitochondrial biology supports additional roles of mitochondria as key participants in a multitude of cellular functions. While it has been well established that mitochondria in different tissues have distinctive ultrastructural features consistent with differential bioenergetic demands, recent and emerging technical advances in flow cytometry, imaging, and "-omics"-based bioinformatics have only just begun to explore the complex and divergent properties of mitochondria within tissues and cell types. Moreover, contemporary studies evaluating the role of mitochondria in pluripotent stem cells, cellular reprogramming, and differentiation point to a potential importance of mitochondrial subpopulations and heterogeneity in the field of stem cell biology. This review assesses the current literature regarding mitochondrial subpopulations within cell and tissue types and evaluates the current understanding of how mitochondrial diversity and heterogeneity might impact cell fate specification in pluripotent stem cells.

ABSTRACT

The objective was to compare pregnancy per AI (P/AI) with conventional (CON) or sex-sorted (SS) semen from a single sire within a fixed-time AI (FTAI) program designed for dairy heifers. Holstein heifers (n=240) were assigned to treatment (CON or SS) according to body weight and reproductive tract score. All heifers underwent FTAI by using the "Show-Me-Synch" protocol [controlled internal drug release (CIDR) insert from d 0 to 14 followed by PGF(2α) (25mg i.m.) 16d after insert removal (d 30) with GnRH (100 µg i.m.) and FTAI at 66 h after PGF(2α)]. A single professional technician performed the FTAI. Heifers were fitted with heat detection patches at PGF(2α) to characterize estrous response. Estrous response did not differ between CON (63/120; 53%) and SS (70/120; 58%) treatments. The CON heifers, however, achieved greater FTAI P/AI (82/120; 68%) compared with SS (45/120; 38%) heifers. The P/AI did not differ for CON heifers that exhibited or failed to exhibit estrus before FTAI [44/63 (70%) vs. 38/57(67%), respectively]. For SS heifers, however, those that exhibited estrus had greater P/AI compared with those that failed to exhibit estrus [32/70 (46%) vs. 13/50 (26%)]. Pregnancy per AI resulting from FTAI was greater for heifers that were inseminated with CON semen compared with those that received SS semen. The expression of estrus before FTAI did not affect P/AI when CON semen was used, whereas the P/AI with SS semen was greater for heifers detected in estrus. Further studies are required to develop strategies for using sex-sorted semen when inseminating heifers at predetermined fixed times on the basis of expression of estrus before FTAI.

ABSTRACT

Undifferentiated granulosa cells from prehierarchal (6- to 8-mm-diameter) hen follicles express very low to undetectable levels of LH receptor (LH-R) mRNA, P450 cholesterol side chain cleavage (P450scc) enzyme activity, and steroidogenic acute regulatory (StAR) protein, and produce negligible progesterone, in vitro, following an acute (3-h) challenge with either FSH or LH. It has previously been established that culturing such cells with FSH for 18-20 h induces LH-R, P450scc, and StAR expression, which enables the initiation of progesterone production. The present studies were conducted to characterize the ability of activin and transforming growth factor (TGF) beta, both alone and in combination with FSH, to promote hen granulosa cell differentiation, in vitro. A 20-h culture of prehierarchal follicle granulosa cells with activin A or transforming growth factor beta (TGFbeta)1 increased LH-R mRNA levels compared with control cultured cells. Activin A and TGFbeta1 also promoted FSH-receptor (FSH-R) mRNA expression when combined with FSH treatment. Neither activin A nor TGFbeta1 alone stimulated progesterone production after 20 h culture. However, preculture with either factor for 20 h (to induce gonadotropin receptor mRNA expression) followed by a 3-h challenge with FSH or LH potentiated StAR expression and progesterone production compared with cells challenged with gonadotropin in the absence of activin A or TGFbeta1 preculture. Significantly, activation of the mitogen-activated protein (MAP) kinase pathway with transforming growth factor alpha (TGFalpha) (monitored by Erk phosphorylation) blocked TGFbeta1-induced LH-R expression, and this effect was associated with the inhibition of Smad2 phosphorylation. We conclude that a primary differentiation-inducing action of activin A and TGFbeta1 on hen granulosa cells from prehierarchal follicles is directed toward LH-R expression. Enhanced LH-R levels subsequently sensitize granulosa cells to LH, which in turn promotes StAR plus P450scc expression and subsequently an increase in P4 production. Significantly, the finding that TGFbeta signaling is negatively regulated by MAP kinase signaling is proposed to represent a mechanism that prevents premature differentiation of granulosa cells.

ABSTRACT

Highly resolved aerosol size distributions measured from high-altitude aircraft can be used to describe the effect of the 1991 eruption of Mount Pinatubo on the stratospheric aerosol. In some air masses, aerosol mass mixing ratios increased by factors exceeding 100 and aerosol surface area concentrations increased by factors of 30 or more. Increases in aerosol surface area concentration were accompanied by increases in chlorine monoxide at mid-latitudes when confounding factors were controlled. This observation supports the assertion that reactions occurring on the aerosol can increase the fraction of stratospheric chlorine that occurs in ozone-destroying forms.

ABSTRACT

Halite particles about 2 micrometers in size were collected by a quartz crystal microbalance cascade impactor from the El Chichón eruption cloud in the lower stratosphere during April and May 1982. These particles are probably derived from the erupted chloride-rich, alkalic magma. Enrichments of hydrogen chloride and increases in optical depolarization in the eruption cloud observed by lidar measurements may reflect the influence of the halite particles. There is evidence that the halite particles reacted with sulfuric acid after about 1 month, releasing gaseous hydrogen chloride, which can influence the catalytic destruction of ozone in the stratosphere.

ABSTRACT

The size distributions of fine particles measured at tropospheric altitudes in the periphery of the eruption plume formed during the 17 April 1979 eruption of Soufriere Volcano and in the low-level effluents on 15 May 1979 were found to be bimodal, having peak concentrations at geometric mean diameters of 1.1 and 0.23 micrometers. Scanning electron microscopy and energy-dispersive x-ray analysis of the samples revealed an abundance of aluminum and silicon and traces of sodium, magnesium, chlorine, potassium, calcium, and iron in the large-particle mode. The submicrometer-sized particles were covered with liquid containing sulfur, assumed to be in the form of liquid sulfuric acid.

ABSTRACT

Measurements of mass concentration and size distribution of aerosols from eruptions of Mount St. Helens as well as morphological and elemental analyses were obtained between 7 April and 7 August 1980. In situ measurements were made in early phreatic and later, minor phreatomagmatic eruption clouds near the vent of the volcano and in plumes injected into the stratosphere from the major eruptions of 18 and 25 May. The phreatic aerosol was characterized by an essentially monomodal size distribution dominated by silicate particles larger than 10 micrometers in diameter. The phreatomagmatic eruption cloud was multimodal; the large size mode consisted of silicate particles and the small size modes were made up of mixtures of sulfuric acid and silicate particles. The stratospheric aerosol from the main eruption exhibited a characteristic narrow single mode with particles less than 1 micrometer in diameter and nearly all of the mass made up of sulfuric acid droplets.
