ABSTRACT

Catalytic methods which control multiple stereogenic centers simultaneously are highly desirable in modern organic synthesis and chemical manufacturing. Herein, we report a regio-, enantio-, and diastereoselective NiH-catalyzed hydroalkylation process which proceeds with simultaneous control of vicinal stereocenters originating from two readily accessible partners, prochiral internal alkenes (enamides or enecarbamates) and racemic alkyl electrophiles (α-bromoamides or Katritzky salts). This reaction produces high-value ß-aminoamides and their derivatives under mild conditions and with precise selectivity. Preliminary studies of the mechanism indicate that the reaction involves an enantioselective syn-hydronickelation to generate an enantiomerically enriched alkylnickel(II) species. Subsequent enantioconvergent alkylation with a racemic alkyl electrophile generates the desired product as a single stereoisomer.

ABSTRACT

The global COVID-19 pandemic is having a significant impact on the development of many aspects all over the world. As an important part of public services, rail transit requires effective response countermeasures to control the spread of COVID-19. Considering the current development of the epidemic situation, this article discusses the characteristics of COVID-19 transmission and identifies vulnerable areas to target in order to prevent and control the spread of the epidemic in the rail transit system. Countermeasures adopted to prevent the spread of COVID-19 are analyzed in terms of external and internal categories, which were classified into six groups: passenger service, case care, information, staff, equipment and operation management. An evaluation architecture was also constructed, which was established from the perspective of effectiveness, economic efficiency, acceptability, privacy and so on. The effect of implementing the measures was evaluated by a social survey, and their advantages and shortcomings were analyzed, which can be used to guide future epidemic prevention and control for rail transit systems around the world. It is important to formulate a reasonable work schedule according to local conditions, providing a reference for rapid response to future public health emergencies of international concern.

ABSTRACT

PURPOSE: Temporal lobe necrosis (TLN), a late complication of nasopharyngeal carcinoma (NPC) after concurrent chemoradiotherapy (CCRT), causes permanent neurologic deficits. We aimed to investigate the risk factors for the development of CCRT-induced TLN in locally advanced NPC patients. METHODS AND MATERIALS: The incidence of CCRT-induced TLN was assessed in consecutive patients with NPC initially staged with T3-4N0-3M0 receiving curative intensity modulated radiation therapy (IMRT) and cisplatin-based chemotherapy with long-term follow-up. The TLN risk was evaluated with radiation dose-volume histograms (a dosimetric risk indicator of organ injury) and the dynamics of blood circulating neutrophil-to-lymphocyte ratios (a clinical indicator of systemic inflammation) by linear and logistic regression models. High-throughput unbiased T cell receptor-beta (TCRbeta) sequencing was performed to correlate the different TCRbeta repertoires of NPC-infiltrating lymphocytes (a biological factor of the immune microenvironment) with TLN incidence. RESULTS: In the era of modern IMRT-based CCRT, radiation doses of up to 74 Gy achieved local control rates of more than 90% in both T3 and T4 diseases but still induced a remarkably higher incidence of TLN in the T4 patients (30.14%) compared with the rare incidence of TLN observed in the T3 patients (2.78%) (P < .0001). We found that in the T4 NPC patients, univariate and multivariate analyses showed the radiation tolerance dose-volume effect was not an absolutely independent factor influencing TLN occurrence. However, increased TLN risk was observed in association with higher pre-CCRT baseline and post-CCRT neutrophil-to-lymphocyte ratios. There was also a link between intratumoral TCRbeta repertoire subtypes and TLN incidence. Combining the inherent TCRbeta genomic susceptibility with the clinical variable neutrophil-to-lymphocyte ratio better predicted the risk of TLN for T4 NPC patients after CCRT. CONCLUSIONS: The associations of tumor-infiltrating lymphocyte repertoires and blood circulating neutrophil-to-lymphocyte ratios with TLN occurrence in T4 NPC patients suggest that the immune and inflammatory milieus play roles in the late brain damage caused by CCRT. Modulated or provoked by CCRT locally and systemically, the reciprocal interactions of neutrophils and lymphocytes in the intracranial NPC-associated immune microenvironment could be a key driver of chronic TLN pathogenesis.

ABSTRACT

Resistin-like molecule-ß (RELM-ß), focal adhesion kinase (FAK), and survivin may be involved in the proliferation of cultured human pulmonary artery smooth muscle cells (HPAMSCs), which is involved in pulmonary hypertension. HPAMSCs were treated with human recombinant RELM-ß (rhRELM-ß). siRNAs against FAK and survivin were transfected into cultured HPASMCs. Expression of FAK and survivin were examined by RT-PCR and western blot. Immunofluorescence was used to localize FAK. Flow cytometry was used to examine cell cycle distribution and cell death. Compared to the control group, all rhRELM-ß-treated groups demonstrated significant increases in the expression of FAK and survivin (P<0.05). rhRELM-ß significantly increased the proportion of HPASMCs in the S phase and decreased the proportion in G0/G1. FAK siRNA down-regulated survivin expression while survivin siRNA did not affect FAK expression. FAK siRNA effectively inhibited FAK and survivin expression in RELM-ß-treated HPASMCs and partially suppressed cell proliferation. RELM-ß promoted HPASMC proliferation and upregulated FAK and survivin expression. In conclusion, results suggested that FAK is upstream of survivin in the signaling pathway mediating cell proliferation. FAK seems to be important in RELM-ß-induced HPASMC proliferation, partially by upregulating survivin expression.

ABSTRACT

BACKGROUND: Vibroarthrographic (VAG) signals are used as useful indicators of knee osteoarthritis (OA) status. The objective was to build a template database of knee crepitus sounds. Internships can practice in the template database to shorten the time of training for diagnosis of OA. METHODS: A knee sound signal was obtained using an innovative stethoscope device with a goniometer. Each knee sound signal was recorded with a Kellgren-Lawrence (KL) grade. The sound signal was segmented according to the goniometer data. The signal was Fourier transformed on the correlated frequency segment. An inverse Fourier transform was performed to obtain the time-domain signal. Haar wavelet transform was then done. The median and mean of the wavelet coefficients were chosen to inverse transform the synthesized signal in each KL category. The quality of the synthesized signal was assessed by a clinician. RESULTS: The sample signals were evaluated using different algorithms (median and mean). The accuracy rate of the median coefficient algorithm (93 %) was better than the mean coefficient algorithm (88 %) for cross-validation by a clinician using synthesis of VAG. CONCLUSIONS: The artificial signal we synthesized has the potential to build a learning system for medical students, internships and para-medical personnel for the diagnosis of OA. Therefore, our method provides a feasible way to evaluate crepitus sounds that may assist in the diagnosis of knee OA.

ABSTRACT

This paper investigates the reverberation time estimation methods which employ backward integration of adaptively identified room impulse responses (RIRs). Two kinds of conditions are considered; the first is the "ideal condition" where the anechoic and reverberant signals are both known a priori so that the RIRs can be identified using system identification methods. The second is that only the reverberant speech signal is available, and blind identification of the RIRs via dereverberation is employed for reverberation time estimation. Results show that under the "ideal condition," the average relative errors in 7 octave bands are less than 2% for white noise and 15% for speech, respectively, when both the anechoic and reverberant signals are available. In contrast, under the second condition, the average relative errors of the blindly identified RIR-based reverberation time estimation are around 20%-30% except the 63 Hz octave band. The fluctuation of reverberation times estimated under the second condition is more severe than that under the ideal condition and the relative error for low frequency octave bands is larger than that for high octave bands under both conditions.

ABSTRACT

To develop the logistic and the probit models to analyse electromyographic (EMG) equivalent uniform voltage- (EUV-) response for the tenderness of tennis elbow. In total, 78 hands from 39 subjects were enrolled. In this study, surface EMG (sEMG) signal is obtained by an innovative device with electrodes over forearm region. The analytical endpoint was defined as Visual Analog Score (VAS) 3+ tenderness of tennis elbow. The logistic and the probit diseased probability (DP) models were established for the VAS score and EMG absolute voltage-time histograms (AVTH). TV50 is the threshold equivalent uniform voltage predicting a 50% risk of disease. Twenty-one out of 78 samples (27%) developed VAS 3+ tenderness of tennis elbow reported by the subject and confirmed by the physician. The fitted DP parameters were TV50 = 153.0 mV (CI: 136.3-169.7 mV), γ 50 = 0.84 (CI: 0.78-0.90) and TV50 = 155.6 mV (CI: 138.9-172.4 mV), m = 0.54 (CI: 0.49-0.59) for logistic and probit models, respectively. When the EUV ≥ 153 mV, the DP of the patient is greater than 50% and vice versa. The logistic and the probit models are valuable tools to predict the DP of VAS 3+ tenderness of tennis elbow.

ABSTRACT

By using the principle of stereovision, 3D digital image correlation (3D-DIC) can determine the 3D morphology and deformation of a target and has been widely used in experimental mechanics as a noncontact 3D measurement technique. To eliminate the limitations of the conventional 3D-DIC system, this study proposes a calibration-free single-lens 3D-DIC system based on a bilateral telecentric lens and a bi-prism. The performance of the proposed system is verified by tests of rigid-body translation along the out-of-plane direction. As a comparison, the same rigid-body translations are measured using a single-entocentric-lens 3D-DIC system. The results show that the measurement accuracy of the proposed system is higher than that of the entocentric-lens-based one. As an application, the proposed system is used to measure the thermal linear expansion of a ceramic plate at elevated temperatures. The reasonable measurement results verify its applicability in deformation measurements, even in high-temperature environments.

ABSTRACT

A reverberation time (RT) estimation method is presented which consists of three steps, the anechoic speech is first recovered by maximizing the skewness of the linear prediction residual of the reverberant speech, then room impulse response (RIR) is identified using the recovered anechoic and reverberant speech, finally RIR is truncated to compensate the estimation errors and RT is estimated using the Schroeder's method. Simulations show that the proposed method successfully estimates RT less than 1.4 s and is insensitive to the speech content such as the number of long pauses and sharp offsets.

To reveal the genetic structures and relationships of the four ethnic populations from the maternal inheritance and explore the origins and migrations of nationalities, the genetic polymorphism of mtDNA in Dong, Gelao, Tujia, and Yi populations from Guizhou was studied by direct sequencing of hypervariable segment I(HVS I) and PCR-RFLP of coding region. Thirty-seven (sub-) haplogroups were identified in the classification tree of mtDNA haplogroups. Haplogroup distributions and principal component (PC) analysis showed that the Dong has high frequencies of south-prevalent haplogroups, which indicates that it is a typically southern population. The Yi harbors high frequencies of the south-prevalent and northern-prevalent haplogroups, which demonstrates that it inherits the maternal characteristics from both southern and northern populations. The Yi and Gelao cluster together, the reason for which might be that their ancestries frequently underwent gene exchanges and mixtures.

ABSTRACT

OBJECTIVE: Through maternal inheritance, to explore the genetic structures and relationships of Dong, Gelao, Tujia and Yi ethnic population in Guizhou of China. METHODS: The mtDNA D-loop hypervariable segment I (HVS I ) in 108 samples of four ethnic populations were sequenced. Then, the nucleotide diversity was estimated and a phylogenetic tree was constructed by Neighbor-Joining method. RESULTS: In the detected 497 bp fragments, 86 polymorphic sites were found, and 82 different haplotypes were identified. The phylogenetic tree of four ethnic populations showed: Yi, Tujia and Gelao clustered more closely than Dong did. CONCLUSION: Yi and Tujia population are very closely related, the reason may be that they either originate from a common ancestry or frequently undergo the gene exchanges and admixtures. The genetic relationship between Tujia and Gelao population is nearer, perhaps because they have settled in the adjacent regions. Dong and Yi population show the farthest genetic relationship, this is probably due to their different historical origins and geographic segregation.

OBJECTIVE: To obtain Maackia amurensis callus and investigate some factors influencing total isoflavonoid production in callus. METHOD: Different media were used for callus induction from cotyledonary and hypocotyl explants. Total isoflavonoid content was determined by UV spectrophotometer. RESULT: Medium types and hormone combinations influenced both callus formation and their developmental states. MS medium supplemented with NAA/BA or containing 2,4-D was suitable for callus induction from cotyledonary explants. Among the basal media tested, MS, N6 and B5 were suitable for cotyledonary callus formation and SH for hypocotyl callus formation. The formed callus could synthesize isoflavones. Media containing NAA were suitable for isoflavonoid production in cotyledonary callus and media containing 2,4-D/BA were suitable for hypocotyl callus, but 2,4-D/KT at high concentration inhibited the isoflavonoid accumulation in hypocotyl and coytledonary calli. According to the developmentally morphological characters, the formed callus could be classified into four types. II-type callus with yellow in color and friability in texture showed the highest accumulation of isoflavones. CONCLUSION: The influences of medium type and hormone combinations on Maackia amurensis callus formation are reflected in both developmentally morphological characters and isoflavonoid accumulation of calli. Yellow, friable callus induced from cotyledonary and hypocotyl explants in MS or N6 medium supplemented with 2,4-D and BA exhibit the optimum growth and isoflavonoid production.
