ABSTRACT

Dyslipidemia is a serious condition affecting an increasing number of people, and thus, preventive measures, including supplementation, are being developed. We aimed to compare the effect of linseed oil, its ethyl esters and fish oil supplementation on the serum lipid profiles of rats fed a high-fat diet. Wistar rats were divided into nine groups. Four of them were fed a high-fat diet for the whole experiment, four groups were fed a high-fat diet before the supplementation period and then the control one with supplements, and one was fed a control diet without supplements. The whole experiment lasted 12 weeks. A significant reduction in blood triglycerides, total cholesterol and the LDL fraction was noted in supplemented groups compared to the controls, especially in groups supplemented with ethyl esters of linseed oil and linseed oil compared to fish oil groups. The results were also more beneficial in groups where, in addition to supplementation, there was also a diet change from a high-fat diet to a control diet during the supplementation period. We may conclude that supplementation with omega-3 fatty acids, combined with a healthy diet, may be a good way of preventing or alleviating dyslipidemia.

ABSTRACT

Phytoremediation is a low-cost and sustainable green technology that uses plants to remove organic and inorganic pollutants from aquatic environments. The aim of this study was to investigate the phytoextraction, phytoaccumulation, and phytotransformation of three fluoroquinolones (FQs) (ciprofloxacin [CIP], enrofloxacin [ENF], and levofloxacin [LVF]) by Japanese radish (Raphanus sativus var. longipinnatus) and duckweed (Lemma minor). Determination of FQs and identification of their transformation products (TPs) were performed using ultra-high-performance liquid chromatography coupled with tandem mass spectrometry (UHPLC-MS/MS). Inter-tissue translocation of FQs in Japanese radish tissues depended on their initial concentration in the medium. CIP (IT = 14.4) and ENF (IT = 17.0) accumulated mainly in radish roots, while LVF in leaves (IT = 230.8) at an initial concentration of 10 µg g-1. CIP (2,104 ng g-1) was detected in the highest concentration, followed by ENF (426.3 ng g-1) and LVF (273.3 ng g-1) in the tissues of both plants. FQs' bioaccumulation factors were significantly higher for duckweed (1.490-18.240) than Japanese radish (0.027-0.103). The removal of FQs from water using duckweed was mainly due to their photolysis and hydrolysis than plant sorption. In the screening, analysis detected 29 FQ TPs. The biotransformation pathways of FQs are described in detail, and the factors that influence their formation are indicated.


This study has presented the efficiency of fluoroquinolone (FQ) residues phytoextraction from water by two plant species (water duckweed, Japanese radish). The use of two plant species allowed for a holistic study of the FQ phytoremediation process by determining the efficiency of extraction, tissue distribution, bioaccumulation tendency, and biotransformation. The research gap regarding FQ transformation products in the phytoremediation process and the factors determining their formation has been filled. This study indicated that duckweed can be used with great efficiency to purify water from FQ contamination.

ABSTRACT

The aim of this study was to determine the effect of supplementation with ethyl esters of linseed oil on the fatty acid profile in hair sebum, blood serum and erythrocyte membranes in healthy dogs. The material for the study included hair and blood samples of adult beagle dogs. The experiment was performed in two periods: summer and winter. Each time it lasted 16 weeks. During the first 8 weeks, the dogs received a supplement, the amount of which was determined individually so that the ratio of α-linolenic acid (ALA) to linoleic acid (LA) together in the feed and supplement was 1:1. Hair coat and blood samples were collected on days 0, 56 and 112; i.e., before the start of supplementation, after 8 weeks of supplementation and 8 weeks after supplementation was completed. The study included a determination of the fatty acid profile with a particular emphasis on polyunsaturated fatty acids (PUFAs) and the ratio of omega-6 to omega-3 in hair sebum, blood serum and erythrocyte membranes. As a result of supplementation, a significant decrease in saturated acids and an increase in unsaturated acids was observed in hair sebum both in summer and winter and especially in omega-3 fatty acids; i.e., α-linolenic (ALA) and its derivatives eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). The same relationships were observed in blood serum and in erythrocyte cell membranes in all the studied periods. Additionally, 8 weeks after the end of supplementation, the level of polyunsaturated fatty acids was still higher compared to the period before supplementation. Moreover, the supplementation resulted in a favorable decrease in the ratio of omega-6 to omega-3 acids in the tested samples, which persisted even after the withdrawal period.

ABSTRACT

The pharmacological effects of the presence of a sugar moiety, 1,2,3-triazole ring and silyl groups in the structure of biologically active compounds have been extensively studied in drug design and medicinal chemistry. These components can be useful tools to tailoring the bioavailability of target molecules. Herein we present the study on the impact of the sugar substituent structure and triisopropylsilyl group presence on the anticancer activity of mucochloric acid (MCA) derivatives containing the furan-2(5H)-one or 2H-pyrrol-2-one core. The obtained results clearly indicated that tested compounds caused a significant decrease in cell viability of HCT116 and MCF-7 cell lines. MCF-7 cells indicate serious resistance toward investigated compounds in comparison with HCT116 cell line, it suggests that estrogen-dependent breast cancer cells are significantly less sensitive to the tested derivatives. Depending on the structure of the sugar, the type and site of connection with the furanone or 2H-pyrrol-2-one derivative and the presence of the silyl group, the selectivity of the compound towards cancer cells can be controlled. The obtained results may have an impact on the design of new furanone-based anticancer compounds.

ABSTRACT

The haloalkane dehalogenase LinB is a well-known enzyme that contains buried active site and is used for many modelling studies. Using classical molecular dynamics simulations of enzymes and substrates, we searched for transient binding sites on the surface of the LinB protein by calculating maps of enzyme-ligand interactions that were then transformed into sparse matrices. All residues considered as functionally important for enzyme performance (e.g., tunnel entrances) were excluded from the analysis to concentrate rather on non-obvious surface residues. From a set of 130 surface residues, twenty-six were proposed as a promising improvement of enzyme performance. Eventually, based on rational selection and filtering out the potentially unstable mutants, a small library of ten mutants was proposed to validate the possibility of fine-tuning the LinB protein. Nearly half of the predicted mutant structures showed improved activity towards the selected substrates, which demonstrates that the proposed approach could be applied to identify non-obvious yet beneficial mutations for enzyme performance especially when obvious locations have already been explored.

ABSTRACT

The study aimed to examine an effect of the preparation based on nanosilver suspension on mineral carrier on poultry litter microbiological profile. The study was conducted on Ross 308 broiler chickens. Three groups were formed, 84 birds in each. Preparation used in the study was composed of aqueous nanosilver suspension sprayed on mineral sorbent. Birds were maintained on straw-sawdust litter; the groups were differentiated due to preparation application (C - control without preparation, I - preparation applied once at the beginning, II - preparation added each week). Pooled litter samples were collected from the top layer of the litter (six samplings) in order to determine mesophilic bacteria count. Additionally, on the last day of the experiment litter samples were collected from three points (by drinker, feeder, pen corner) to analyze the total number of microorganisms, Salmonella spp., Escherichia coli, Enterococci, and molds. In the case of mesophilic bacteria count, the highest decrease was noted for group II. Total number of microorganisms determined in various points of the pen did not give clear relationship; in some cases even an increase was found. Salmonella spp. decreased as a result of preparation addition; the highest decrease was noted for samples collected by feeders. The results for Escherichia coli are not unequivocal. However, a decrease was found in the case of drinkers and feeders compared to control, especially in group II. An addition of preparation caused a decrease in Enterococci, especially for samples collected by feeders in group II. Similar tendency was found for molds. The study demonstrated that the preparation exhibits bactericidal properties. However, its effect varies depending on microorganism kind and sample collection point.

ABSTRACT

In this study, we used 10 healthy sheep, which gave birth to healthy twins. Stool samples were collected from mothers and their offspring 3 times during the study (0, 28 and 56 day postpartum). Milk samples were taken from the mothers at the same time. RT PCR analysis of faeces and milk was performed in order to assess the level of bacteria from the Firmicutes and Bacteroidetes phyla including the family Lactobacillaceae (phylum Firmicutes). The composition of mother's milk was also analyzed and their BCS. The data were compiled statistically. The obtained results showed that the level of the studied groups of bacteria may change due to the change of diet. Additionally, there were significant differences between lambs and mothers in the levels of the studied groups of bacteria. Analysis also shown that in the digestive system of mothers was a smaller disproportion in the level of the studied bacterial phyla than in lambs. The results also indicated the occurrence of differences in the bacterial composition at the individual level, both in ewes and their offspring. Additionally, in the conducted experiment, there were differences in the level of Firmicutes and Bacteroidetes groups depending on the sex.

ABSTRACT

The pharmacological effects of carbon to silicon bioisosteric replacements have been widely explored in drug design and medicinal chemistry. Here, we present a systematic investigation of the impact of different silyl groups on the anticancer activity of mucobromic acid (MBA) bearing furan-2(5H)-one core. We describe a comprehensive characterization of obtained compounds with respect to their anticancer potency and selectivity towards cancer cells. All four novel compounds exert stronger antiproliferative activity than MBA. Moreover, 3b induce apoptosis in colon cancer cell lines. A detailed investigation of the mechanism of action revealed that 3b activity stems from the down-regulation of survivin and the activation of caspase-3. Furthermore, compound 3b attenuates the clonogenic potential of HCT-116 cells. Interestingly, we also found that depending on the type of the silyl group, compound selectivity towards cancer cells could be precisely controlled. Collectively, we demonstrated the utility of silyl groups for adjusting both the potency and selectivity of silicon-containing compounds. These data reveal a link between the types of silyl group and compound potency, which could have bearings for the design of novel silicon-based anticancer drugs.

ABSTRACT

For decades, skin has been assigned the main role of an insulator of the inside of the body from the external environment, but it also plays a role in maintaining homeostasis. In this study, the level of selected bacterial phyla (Firmicutes, Bacteroidetes, Proteobacteria and Actinobacteria) was assessed in three sheep breeds (Swiniarka sheep, Uhruska sheep and BCP line (synthetic sheep breed; n = 6) and in two breeds of goats (Boer, Saenian; n = 6) living in the same environment and fed on the same feed, where the aim was to identify differences in terms of race, species and individual differences. Significant differences were found in Firmicute, Actinobacteria and Proteobacteria phyla (p ≤ 0.05). Statistically significant and positive correlations were demonstrated between Actinobacteria and Proteobacteria, Proteobacteria and Bacteroidetes or Firmicutes and Bacteroidetes. The obtained results suggest that the species and racial differences in the level of the studied bacterial phyla may also result from the physicochemical differences of the skin surface, as they could exacerbate the variations in humidity, temperature, composition of antimicrobial peptides (AMP) and lipid content. In addition, individual differences were observed, which indicate a similar effect of an individual on the microbiological composition of its organism.

Fatty acids from the omega-3 family are an important element of both human and animal diets. Their activity involves a range of functions for the functioning of a whole organism, and their presence in animal diets can be considered as a means for animal origin product enrichment for human benefit or as compounds profitable for an animal's health status. The aim of this preliminary study was to compare the effect of supplements rich in omega-3 fatty acids (linseed oil, linseed oil ethyl esters, and fish oil) in rat feed on the fatty acid profile of their intramuscular and perimuscular fat. The results demonstrated beneficial changes in fatty acid profiles (a decrease in saturated acids, an increase in unsaturated ones, i.e., omega-3 acids share) of examined tissues in the case of all supplements however, particular attention should be paid to linseed oil ethyl esters, which significantly increased the content of all omega-3 acids. Supplementation of animal diet with linseed oil ethyl esters may be beneficial for both animals, as omega-3 fatty acids exhibit profitable properties related to an animal's health status and productivity, and humans who consume such enriched products.

The study aimed to investigate the effect of supplementation of fish oil after the process of low-temperature crystallization (LTC-FO) enriched with long-chain polyunsaturated fatty acids (LC-PUFAs) on cow milk parameters. The experiment was carried out on 24 Polish Holstein Friesian cows. For 4 weeks, experimental (EXP) group animals (n = 12) were fed LTC-FO (1% of dry matter). Milk was collected two times: on days 14 and 30. LTC-FO supplementation decreased milk fat yield and concentration (p < 0.01). Higher levels of polyunsaturated fatty acids (PUFAs), including these with beneficial biological properties, i.e., eicosapentaenoic (EPA), docosahexaenoic (DHA), docosapentaenoic (DPA), CLA, alpha-linolenic acid (ALA), and TVA (p < 0.01), and lower levels of SFAs, especially short- (p < 0.01) and medium-chain ones (p < 0.05, p < 0.01), were found in the EXP group. The addition of LTC-FO reduced the value of atherogenic and thrombogenic indices as well as SFA/UFA and n-6/n-3 ratios and increased the content of n-3 PUFA and functional fatty acids (p < 0.01). The addition of LTC-FO also increased the delta-9 desaturase index for CLA/TVA and decreased it for pairs C14:1/C14:0 and C16:1/C16:0 (p < 0.05, p < 0.01).

The aim of the study was to examine the effect of dietary supplementation of isomerized poppy seed oil (IPO) enriched with conjugated dienes of linoleic acid (CLA) on cow and sheep milk parameters (fat content, fatty acid profile, Δ9-desaturase index, and atherogenic index). The process of poppy seed oil alkaline isomerization caused the formation of CLA isomers with cis-9,trans-11, trans-10,cis-12, and cis-11,trans-13 configurations in the amounts of 31.2%, 27.6%, and 4.1% of total fatty acids (FAs), respectively. Animal experiments were conducted on 16 Polish Holstein Friesian cows (control (CTRL) and experimental (EXP), n = 8/group) and 20 East Friesian Sheep (CTRL and EXP, n = 10/group). For four weeks, animals from EXP groups received the addition of IPO in the amount of 1% of dry matter. Milk was collected three times: on days 7, 14, and 30. Diet supplementation with IPO decrease milk fat content (p < 0.01). Milk fat from EXP groups had higher levels of polyunsaturated fatty acids, including FAs with beneficial biological properties, that is, CLA and TVA (p < 0.01), and lower levels of saturated fatty acids, particularly short- (p < 0.01) and medium-chain FAs (p < 0.05). The addition of IPO led to a decrease in the atherogenic index.

ABSTRACT

This review aims to explain the influence and characterization of the microbiome in the ruminant digestive system by presenting the knowledge collected so far. The knowledge presented in this work is focused on the main factors affecting the microbiome and the main dependencies that have been found in it so far. The microbiome in the rumen is the first to come into contact with the biomass of the forage and its main purpose is to decompose into smaller particles or compounds. With the gradual increase in knowledge about the microbiome, there is a chance to manipulate it so that the animal continues to live in a symbiotic relationship with it, while reducing greenhouse gas emissions to the environment as well as increasing feed efficiency. Therefore, understanding the influence of the ruminant microbiome is the main step to achieve such results. However, learning the relationship between microorganisms is only at an early stage, because research focuses mainly on taxonomy. Future research should focus on interactions in the ecosystem which is the microbiome, on explaining individual functions and on influence of environmental factors.

ABSTRACT

The most important nutrients affecting wool production include sulfur amino acids, such as methionine, and minerals, such as zinc, and their deficiency may cause wool production decrease. The aim of this study was to evaluate an effect of zinc and methionine chelate addition on the qualitative characteristics of Merino ewes wool and zinc content in wool fibers and on their surface. Histological evaluation of fibers was also performed. The study included 22 Polish Merino ewes divided into two groups: control and experimental (0.4 g chelates daily per head). The experiment lasted for 4 months (3.5 months of pregnancy and 2 weeks of early lactation). The length, thickness and breaking force of wool fibers were higher in the experimental group compared to the control group. Higher content of zinc in wool of sheep from the experimental group was also demonstrated. The histological structure of fibers from both groups did not differ, as the content of zinc on their surface. It can be concluded on the basis of the obtained results that the addition of chelates favorably influenced the thickness compensation of wool fibers and amount of wool obtained from sheep during pregnancy and early lactation, as well as mechanical features of wool.

ABSTRACT

The aim of this study was to determine selected characteristics of hair originating from adult females of three primate species: lar gibbon, crested black macaque and common chimpanzee. The research material consisted of 300 hairs collected from the top of the head. The analysis included the length, diameter, strength, breaking load, elongation and cross-section area of the hairs, and the hair index. Additionally, the scale number per 1 mm of hair length was determined using scanning electron microscope images. Lar gibbon hairs were the thinnest and the longest, while common chimpanzee hairs were the thickest and the shortest, and concurrently the least resistant. Hairs from various primate species differ in terms of their morphological structure and histological features and thus can be used for differentiation and identification of species.

BACKGROUND: The substituted 1,8-Naphthalimides (1H-benzo[de]isoquinoline-1,3(2H)- diones) are known as DNA intercalators stabilizing DNA-Topoisomerase II complexes. This interaction disrupts the cleavage-relegation equilibrium of Topo II, resulting in formation of broken strands of DNA. OBJECTIVE: To investigate the influence of type of substituents and substitution positions in 1,8- naphthalimde skeleton on the inhibition of Topoisomerase II activity. METHODS: The starting 1,8-naphthalimide were prepared from acenaphthene by introduction of appropriate substituents followed by condensation with ω-hydroxylakylamines of different chain length. The substituents were introduced to 1,8-naphthalimide molecule by nucleophilic substitution of leaving groups like nitro or bromo present in 4 or 4,5- positions using the ω- hydroxylalkylamines. The bioactivity of obtained compounds was examined in model cell lines. RESULTS: Antiproliferative activity of selected compounds against HCT 116 human colon cancer cells, human non-small cell lung cells A549 and non-tumorigenic BEAS-2B human bronchial epithelium cells was examined. Several of investigated compounds exhibit a significant activity (IC50 µM to 7 µM) against model cancer cell lines. It was demonstrated that upon treatment with concentration of 200 µM, all derivatives display Topo II inhibitory activity, which may be compared with activity of Amonafide. CONCLUSION: The replacement of the nitro groups in the chromophore slightly reduces its anticancer activities, whereas the presence of both nitro group and ω-hydroxylalkylamine chain resulted in seriously increased anticancer activity. Obtained compounds showed Topo II inhibitory activity, moreover, influence of the substitution pattern on the ability to inhibit Topo II activity and cancer cells proliferation was observed.

ABSTRACT

A series of 5-alkoxy derivatives of 3,4-dichloro-5-hydroxyfuran-2-(5H)-one (mucochloric acid, MCA) were obtained and subsequently subjected to modification in the C-4 position of 2(5H)-furanone ring. The cytotoxicity of newly synthesized compounds was evaluated in MTT assay against non-small cell lung cancer (A549) and healthy lung epithelial cell line (BEAS-2B). The derivatives containing a branched alkoxy substituent in the C-5 position demonstrated the highest anticancer properties, whereas modification of compounds in the C-4 position of 2(5H)-furanone ring only slightly improve their antiproliferative properties. Compounds 12 and 15 exhibited the best selectivity towards A549 cells and were also evaluated in a panel of cancer cell lines of different origin. Further investigation revealed that treatment of A549 cell line with compounds 12 and 15 led to G2 phase cell cycle arrest and induction of caspase-independent cell death. Moreover, compound 12 was found to act synergistically with erlotinib.

ABSTRACT

A series of UDP-sugar analogues was synthesized and their preliminary biological activity was evaluated. Glycoconjugates of uridine 1 and 2 were synthesized by condensation of uridine-5'-carboxylic acid and 1-amino sugars derivatives of d-glucose and d-galactose, glycoconjugates 3 and 4 were synthesized by azide-alkyne 1,3-dipolar cycloaddition (CuAAC) of 1-azido sugars and propargylamide derivatives of uridine while glycoconjugates 5 and 6 were synthesized by CuAAC of propargyl ß-O-glycosides and 5'-azido uridine. Evaluation of inhibitory activity of compounds 1-6 against commercially available ß-1,4-galactosyltransferase I (ß4GalT) show that compound 5 inhibited the enzyme in µmolar range.
