ABSTRACT

Metabolism-associated genes (MAGs) are important regulators of tumor progression and can affect a variety of physiological processes. In this study, we focused on the relationship between MAGs and Ovarian cancer (OC) prognosis. METHOD: Metabolism-related genes were extracted from the Cancer Genome Atlas (TCGA) database. Through univariate COX and lasso regression models, a dynamic risk model based on MAGs was established. Compared with other clinical factors, demonstrated the ability of the model to predict the prognosis of patients with OC. The clinical samples were used to verify the expression of these MAGs. RESULTS: A metabolism-associated gene signature was constructed by LASSO Cox regression analysis in OC, which was composed of 3-MAGs (PTGIS, AOC3, and IDO1). The signature was used to classify the OC patients into high-risk and low-risk groups. The overall survival of the low-risk group was significantly better than that of the high-risk group. The analysis of the therapeutic effect of bevacizumab showed that bevacizumab was not conducive to improving the prognosis of the low-risk group. CONCLUSIONS: We constructed a prognostic model of MAGs in OC, which can be used to predict the prognosis of OC patients and may have a good guiding significance in the individualized treatment of patients.

ABSTRACT

Objective To investigate the effect of clinical nursing pathway on patients with cerebral infarction. Methods Two hundred patients with cerebral infarction were randomized by random digit table into the clinical pathway group and the conventional nursing group.The former was managed with the clinical nursing pathway and the latter received conventional nursing. The neurologic impairment and the activities of daily life were assessed respectively by national institute of health stroke scale(NIHSS) and Barthel index(BI).The two groups were compared in terms of hospital stay,medical expense,patient’s satisfaction and curative effect. Results Compared to the conventional nursing group,the clinical pathway group was shorter at hospital stay,lower at lower hospitalization cost,higher at patient’s satisfaction and higher in the score by Barthel index,compared to the conventional nursing group(all P<0.05). Conclusion The application of clinical nursing pathway in patients with cerebral infarction can improve the activities of daily living and the quality of care,shorten length of stay,reduce medical cost and increase patients’satisfaction drgree.

ABSTRACT

Objective An effective reproducible protocol for complete plant regeneration via somatic embryogenesis has beendeveloped for Herpetospermum pedunculosum,an endangered Tibetan medicinal herb.Methods The cotyledonexplants used in this study were excised from seedlings germinated in vitro.Callus was induced from cotyledonexplants on Murashige and Skoog's medium,supplemented with 2,4-dichlorophenoxyacetic acid(2,4-D,0.1-1.0mg/L)alone or in combination with 6-benzylaminopurine(BA,0.5,1.0,and 2.0 mg/L).Results The calli showeddifferentiation of globular embryos after three weeks of incubation on MS medium supplemented with variouscombinations of BA and NAA.Sixty-two percent of the embryogenic calli produced somatic embryos in MS basalmedium supplemented with BA(1.0 mg/L)+NAA(2.0 mg/L).The addition of KN(0.5 mg/L)to MS mediumcontaining both BA and NAA(2.0 mg/L each)significantly increased the frequency of somatic embryogenesis.Themaximum percentage of embryogenic calli formation was 83%,and globular embryos formed and germinatedsuccessfully in this medium.Then,transferring the regenerated plants from this medium to hormone-free MSmedium will further enhanced the development of the plants,and the healthy plantlets are formed successfullywithin four weeks.The plantlets were transferred to soil to acclimatize under greenhouse conditions and 75%survived.Conclusion Somatic embryogenesis protocol as reported here can play a key role in the propagation andconservation of this endangered species.

ABSTRACT

OBJECTIVE: The purpose of this study was to determine the cut-offs of serum biomarkers of liver fibrosis in HBsAg-positive patients by receiver operating characteristic (ROC) curves, and to determine the validity of these markers in parallel and serial interpretation. METHODS: This study included 444 HBsAg-positive patients who had liver biopsy performed between January 1, 2003 and March 31, 2006. Some routine and clinical chemistry tests were run, and the stage of liver fibrosis was measured. The cut-offs of those markers were identified by using ROC curves. Sensitivity, specificity, predictive values (both positive and negative), and the Youden's index of these markers were calculated in parallel and serial interpretation. RESULTS: The 444 patients were divided into a training group (322 patients) and a validation group (122 patients). All markers except gender, HB and TP were found to be statistically significant factors associated with significant fibrosis in the training group. The AUROC of GGT, APRI, AGE-PLT and AST in the training group were 0.772, 0.769, 0.748 and 0.700, respectively, and their sensitivities in the validation group were 65.00%, 70.00%, 75.00% and 60.00%, respectively. In parallel interpretation, the highest sensitivity was 85.00% at AGE-PLT union or logical sum AST, and in serial interpretation, the best specificity was 96.34% at GGT intersection AGE-PLT. CONCLUSION: A few routine and serum biomarkers can be used to effectively assess most patients with HBsAg-positive CHB with and without significant liver fibrosis in parallel and serial interpretation.
