ABSTRACT

The disposal of animal remains resulting from breeding is a significant challenge that impacts the industry's growth. To address the issues with current treatment methods, such as the large space required for corpse storage, and the high energy consumption of pyrolysis. Three strains with high protease and lipase production and one strain with high keratinase production were screened. The virulence genes were evaluated, the synthesis gene clusters of degrading enzymes were mined, secondary metabolites of each strain were analyzed, and the bacterial community with both growth rate and enzyme production ability was developed. Therefore, a microbial degradation method with mild reaction conditions and rapid liquefaction of animal residues was developed. The liquid degradation of four common farm-raised animal residues (sheep, cattle, chickens, and pigs) was tested under laboratory conditions. The results showed that the liquid degradation of animal residues was achieved within 144 h, transforming the months-long anaerobic process of traditional compost fermentation process into a mere 6 days' anaerobic process. N, P, K plant nutrients accounted for 15% of the total matrix, pH value was 5.5-6.7, heavy metal content was less than 0.2 mg L-1. Designed and improved fermentation equipment, produced a 3 m³ fermentation equipment, used in chicken, pig two types of animal residues pilot test. The emissions of greenhouse gases such as CO2 in the entire degradation process were 1.6 × 104 ppm, which was 481 times less than that of composting by 7.7 × 106. This study provides a solution for the treatment of dead livestock and poultry, which has promotional and practical value.

ABSTRACT

Objective:To analyze the clinical adverse events of the first carbon ion therapy system in radiotherapy for cancer patients in China. Methods:A retrospective analysis was conducted on the clinical trial monitoring data of the carbon ion therapy system obtained by the Pharmacovigilance Center of Gansu Province. A descriptive study was conducted on the demographic characteristics, radiotherapy techniques, irradiation site and dose parameters, postoperative follow-up, and adverse event information of 46 tumor patients who received carbon ion therapy and participated in the clinical trial in Wuwei Cancer Hospital, Gansu Province from November 2018 to February 2019. Frequency and percentage were used to describe and analyze the occurrence of adverse events after carbon ion therapy for cancer patients in different groups. All subjects who received radiotherapy were grouped according to the treatment dose and fractionation method. Results:The median age of the 46 patients was 47 years old, and the male to female ratio was 30∶16. There were 15, 5, 8, 9, and 9 patients with head and neck, chest, abdomen, pelvic cavity, and limb spinal tumors, respectively. The total duration of radiotherapy was 2-4 weeks for 10-16 times. There were 246 adverse events in 45 cases, with an incidence of 98%. No severe adverse events occurred. The adverse events definitely related to carbon ion devices accounted for 19.1%, and no severe adverse events related to carbon ion devices occurred. According to the evaluation criteria of common terminology criteria for adverse events (CTCAE), the main adverse events were CTCAE grade 2 and below, with only 1 (2%) head and neck tumor patient (nasopharyngeal malignant tumor) experienced CTCAE grade 3 adverse events after treatment. In addition, 43 patients developed acute adverse reactions, with an incidence of 93%, mainly involving the skin, mucosa, eyes, ears, pharynx and esophagus, upper gastrointestinal tract, lower gastrointestinal tract (including pelvic cavity), lung, genitourinary tract, heart, central nervous system and hematology (white blood cells, platelets and neutrophils), etc. Conclusion:The adverse reactions of patients treated with the first carbon ion therapy system are mainly CTCAE grade 2 and below, and the clinical adverse events are mild and controllable.

ABSTRACT

PURPOSE: To study the correlations of the recurrence of gastric cancer in patients after radical surgery with serum gastrointestinal hormones, serum anti-Helicobacter pylori (anti-HP) immunoglobulin G (IgG) antibody and vascular endothelial growth factors (VEGFs). METHODS: According to whether gastric cancer recurred within five years after surgery, the patients were divided into recurrence group (RE group, gastric cancer recurred within five years after surgery, n=78) and non-recurrence group (NR group, gastric cancer did not recur within five years after surgery, n=69). Differences in lymph node metastasis, gastrointestinal hormones, VEGFs, anti-HP IgG antibody and the tumor-node-metastasis (TNM) stage between RE group and NR group were detected and compared, so as to analyze the correlations of these factors with the recurrence of gastric cancer in patients after radical surgery. RESULTS: The levels of gastrin (GAS) and motilin (MTL) after meals in RE group and NR group were slightly higher than those before meals. The levels of GAS and MTL in RE group were higher than those in NR group in the two periods (p<0.05). Besides, compared with NR group, RE group had lower pepsinogen (PG) I, PG II and PG I/II ratio (PGR), but a higher positive value of anti-HP IgG antibody (p<0.05) and higher levels of VEGF-A, VEGF-C and VEGF-D (p<0.05). Moreover, there were markedly more cases of gastric cancer in stage III, remarkably few cases of gastric cancer in stage I and obviously more cases of lymph node metastasis in RE group than those in NR group (p<0.05). Multivariate analysis showed that gastrointestinal hormones, lymph node metastasis, VEGFs, the TNM stage of gastric cancer and anti-HP IgG antibody were all risk factors for the recurrence of gastric cancer after radical surgery (p<0.05). CONCLUSIONS: The recurrence of gastric cancer in patients after radical surgery is related to the TNM stage of gastric cancer, gastrointestinal hormones, VEGFs, lymph node metastasis, anti-HP IgG antibody and other factors.

ABSTRACT

Objective To quantitatively assess anterior chamber angle of primary angle-closure glaucoma (PACG) and primary angle-closure (PAC) patients after laser pheripheral iridotomy (LPI) by spectral-domain anterior segment optical coherence tomography (SD-AS-OCT),and investigate which parameters should be used in these patients and the clinical significance of these parameters in screening and follow-ups of glaucoma. Methods Fifty-five eyes of 40 patients diagnosed with PACG remission (11 eyes,8 patients),PAC (20 eyes,20 patients) or PAC suspect (24 eyes,12 patients) were examined. Each eye was examined by SD-AS-OCT at upper, lower, nasal, and temporal sides.Anterior chamber angle (ACA),trabecular iris angle (TIA),and trabecular iris space area (TISA500/750) were measured or calculated. All results were analyzed. Results The points from the inside to the corneal scleral spur 500 μm and 750 pm were selected,AOD,TIA,ACA,TISA at upper, lower, nasal, and temporal sides were measured, which were included into 500 series and 750 series. The mean values of TIA,ACA and AOD in the 500 series and 750 series were the largest in the temporal side and the lowest in the nasal side. The mean values of TISA measurement was the largest in the lower side and the smallest at the upper side.Pearson correlation analysis showed that TIA500 and ACA500 (r =0.799,P =0.00),TIA750 and ACA750 (r =0.799,P =0.00),TIA500 and TISA500 (r=0.919,P =0.00),TIA750 and TISA750 (r =0.920,P =0.00) had positive correlation. With TIA500 as independent variable x and TISA500 as dependent variable y,the regression equation was obtained:y =-0.300 + 0.005x (F =1195,P =0.00),which showed TISA500 and TIA500 were linearly positive correlated. With TIA750 as independent variable x,and TISA750 as dependent variable y,the regression equation was obtained:y =-0.440 + 0.009x (F =854,P =0.00),which showed TISA750 and TIA750 were linearly positive correlated. The coefficient of variation of the 750 series parameters were less than the 500 Series in the four directions. Conclusion SD-AS-OCT is an efficient tool for efficiency evaluation and follow-up during the course of PACG and PAC after LPI.For narrow angles,as the parameters of 750 series parameters have less variability,so 750 series are more recommended in clinical application.

ABSTRACT

OBJECTIVE To investigate the effect of clopidogrel(Clog),a platelet aggregation inhibitor,on the development of colitis-associated colon cancer(CAC)and its possible mechanism. METHODS To establish a CAC model, male BALB/c mice were treated with single azoxymethane(AOM) 10 mg · kg-1 by ip. One week later, the mice drank 2.5% dextran sulfate sodium(DSS)for one week and water for two weeks, which lasted three cycles. From the first day mice received 2.5%DSS water, Clog 12.5,25.0 and 50.0 mg · kg-1 was ig administered once a day. Body mass, clinical symptoms, the number of colon tumor and tumor size in colon tissue were recorded. Hyperplasia of tumors was analyzed by HE staining. In the early inflammatory phase of the CAC model, the length of colons was measured, histological structure and epithelium cell proliferation of colon tissues were evaluated by HE staining and Ki67 staining, respectively. In the tumorigenesis and progression phase of the CAC model, epithe?lium cell proliferation of colon tissues was evaluated by Ki67 staining. The mRNA expression of tumor necrosis factor-α(TNF-α)was detected by real-time quantitative PCR. The expression of chemokine(C-X-C motif)ligand 2(CXCL2)and its receptor 2(CXCR2)in colon tissues was detected by PCR and immu?nohistochemistry. RESULTS Compared with model group, clinical symptoms of mice in Clog 12.5 mg · kg-1 group were alleviated, the size of colon tumors was decreased(P<0.05),and hyperplasia of tumors was reduced(P<0.05). During the inflammatory phase, the clinical symptoms of mice in Clog 12.5 mg·kg-1 group were significantly alleviated(P<0.05),the decrease of body mass was reduced(P<0.01),the colon shrinkage was ameliorated(P<0.01),the inflammatory injury and epithelium cell proliferation in colon tissues were reduced(P<0.05). During the tumorigenesis and progression phase, epithelium cell prolif?eration in colon tissues in Clog 12.5 mg·kg-1 group was reduced(P<0.01),and the mRNA and protein expression of TNF-α,CXCL2 and CXCR2 of colon tissues was decreased(P<0.05). CONCLUSION Clog can alleviate inflammation during the CAC early inflammatory phase and inhibit the formation of CAC. The antitumor effect of Clog may be related to the decrease in expression of CXCL2 and CXCR2.

ABSTRACT

Objective To explore the feasibility of adding a flexible linker between two-pore-domain potassium channel TREK-1 (TWIK related K + channel 1)monomers to construct a tandem-linked dimer. Methods PCR was used to add a flexible linker between the two TREK-1 monomers. The cRNA obtained from in vitro transcription using the above vector was injected into Xenopus oocytes. After 24 -48 h,currents were recorded from these oocytes using a two-electrode voltage clamp. The effects of extracellular Ba2 + and pH on TdTREK-1 were observed and compared with those of native dimeric TREK-1.Results The tandem-linked dimeric TdTREK-1 was highly expressed in Xenopus oocytes. The currents through these channels were inhibited by extracellular Ba2 +and acidification.Furthermore, the responsiveness of the concatenated dimers to these extracellular stimuli was similar to that of native dimers. Conclusion Adding a flexible linker between the two monomers to construct the tandem-linked dimer does not affect the expression and gating properties of TREK-1, suggesting that the method be feasible.Such a method will allow the manipulation of a single subunit, which will help basis study the structure and function of TREK-1.

ABSTRACT

Objective The alm of this study was to determine the solubility and permeability of daldzin and daldzein and the interaction of these two components. Methods With the method inChinese Pharmacopoeia and in situ single-pass intestinal perfusion model we tested the solubility and permeability of daldzin, daldzein and their interaction. Results In pH 7.4 K-R buffer the solubility of daldzin was 6 times than daldzein and both the solubility of these two components were enhanced when they were determined together. In small intestine of rat, the permeability of daldzein was 3 times than daldzin. Daldzin could enhance the permeability of daldzein but the daldzein manifested an opposite trend. Conclusion When compared to daldzin, daldzein owned a lower solubility but a better permeability. When used together, both the solubility and permeability of daldzein would be enhanced. The solubility of daldzin could be enhanced slightly but its permeability would be reduced.

ABSTRACT

Stress exerts profound effects on cognitive and emotional functions. As a regulator of response to stress, the corticotropin releasing factor (CRF) during stress impacts organism function through hypothalamic-pituitary-adrenal axis. Recent research has shown that dendritic branches were abnormal and dendritic spine was lost under stress. In contrast, stressed mice with CRF receptor 1 (CRFR1) deficiency or CRFR1 antagonist exhibited normal dendritic morphology of hippocampal neurons .The structure and related factors expression of hippocampal neurons were affected by CRFR1 as a G protein-coupled receptor(GPCR), but the molecular mechanism about CRF-induced direct injury effect on hippocampal neurons has been poorly understood. The paper reviews the research progress on the contribution of stress activated CRF-CRF receptor signaling to the structure and related factors of hippocampal neurons.

ABSTRACT

Antitumor effects of erythromycin and the related mechanism were investigated in the present study. Neuroblastoma cells (SH-SY5Y) were exposed to erythromycin at different concentrations for different durations. Cell proliferation was measured by cell counting, and cell viability was examined by MTT assay. Cell cycle phase distribution and the cytosolic calcium level were detected by flow cytometry. Mitochondrial membrane potential was measured by the JC-1 probe staining and fluorescent microscopy. The expression of an oncogene (c-Myc) and a tumor suppressor [p21 (WAF1/Cip1)] proteins was analyzed by using Western blotting. Erythromycin could inhibit the proliferation of SH-SY5Y cells in a concentration- and time-dependent manner. The cell cycle was arrested at S phase. Mitochondrial membrane potential collapsed and the cytosolic calcium was overloaded in SH-SY5Y cells when treated with erythromycin. The expression of c-Myc protein was down-regulated, while that of p21 (WAF1/Cip1) protein was up-regulated. It was concluded that erythromycin could restrain the proliferation of SH-SY5Y cells. The antitumor mechanism of erythromycin might involve regulating the expression of c-Myc and p21 (WAF1/Cip1) proteins.

ABSTRACT

Objective To investigate whether there is a thinning of the retinal nerve fiber layer (RNFL) in the eyes of diabetic patients and to analyze the relationship between RNFL thickness and several diabetic risk factors. Methods Forty-two type 2 diabetes mellitus patients aged 47-70 years were enrolled in this study. All the subjects underwent ophthalmologic examinations and glaucoma diagnosis (GDx) nerve fiber analyzer scanning with two different modes: variable cornea compensation (VCC) and enhanced cornea compensation(ECC).The GDx parameters included the temporal-superior-nasal-inferior-temporal average (TSNITave), superior and inferior averages, the TSNIT standard deviation and the nerve fiber indicator (NFI). The receiver operating characteristic (ROC) was applied to study the diagnostic ability of indices. The association between risk factors and the NFI was studied. Results The area under the curve for the ROC of the NFI was the largest for both the ECC and VCC modes. There was a significant relationship between age and RNFL thinning. However, there was no significant association between diabetic duration and fasting blood glucose with RNFL thinning. The influence of age on NFI was greater in diabetic patients than in control subjects. Conclusions Age has an important influence on the NFI of diabetic patients. Diabetic patients with normal ocular fundus may have RNFL thinning.

ABSTRACT

The name of SR proteins is derived from their typical RS domain that is rich in serine (Ser, S) and arginine (Arg, R). They are conserved in evolution. Up to now, 10 members of the SR protein family have been identified in humans. SR proteins contain one or two RNA binding motifs aside from the RS domain, and also possess special biochemical and immunological features. As to the functions of SR proteins, they facilitate the recruitment of the components of splicesome via protein-protein interaction to prompt the assembly of early splicesome; while in alternative splicing, tissue-specifically expressed SR protein along with the relative ratio of SR protein and heterogeneous nuclear ribonucleoprotein (hnRNP) is composed of two main regulative mechanisms to alternative splicing.
