ABSTRACT

Understanding the paleoenvironment and phytogeographical history of the Tibetan Plateau, China relies on discovering new plant fossils. The Qaidam Basin has long been regarded as an ideal 'field laboratory' to investigate the paleoclimate and paleobiological evolution of the northern Tibetan Plateau. However, fossil angiosperms from the Qaidam Basin are rare, and our knowledge of its paleovegetation is poor. Here, we report fossil leaves and fruits of Betulaceae found from the Oligocene Shangganchaigou Formation of northwestern Qaidam Basin (Huatugou area). Comparative morphological analysis led us to assign the fruits to the Betula subgenus Betula and the leaves to Carpinus grandis. These findings, together with other reported fossil plants from the same locality, reveal a close floristic linkage between the Qaidam Basin and Europe during the Oligocene. The northern pathway of this floristic exchange may have crossed through the Qaidam Basin during the late Paleogene. This floristic linkage may have been facilitated by the continuous narrowing of the Turgai Strait and stronger westerlies, which transported moisture and provided favorable climatic conditions. Indeed, fossil plants collected from the Qaidam Basin suggest that during the Oligocene this region had warm and humid deciduous broad-leaf forest, which differs from the region's modern vegetation and indicates that the Qaidam Basin may have been a suitable region for these plants to flourish and spread during the Oligocene.

ABSTRACT

A new species of Berothidae, Jersiberothamusivum sp. nov., is described and illustrated from mid-Cretaceous (lowest Cenomanian) Myanmar amber. It is easily distinguished from other species of Berothidae by the configuration of the wing venation including: forewing with distinct areas of infuscation surrounding cross-veins and vein forks, all cross-veins simple prior to ScP-RA fusion, presence of two cross-veins ra-rp; absence of inner or outer graduate series of cross-veins; RP with three branches; and absence of ma-mp cross-veins and cua-cup cross-veins; while hind wing has cross-vein 1r-m absent. The previous diagnoses of Iceloberotha Grimaldi, 2000 and Jersiberotha Grimaldi, 2000 are quite unclear because some characters occur mosaically in both genera. In order to solve this problem and distinguish J.musivum from other species in the family, a new key to species of Berothidae from Myanmar amber has been provided and the diagnoses of Iceloberotha and Jersiberotha have been revised.

ABSTRACT

A fossil eudicot, Gansufructus saligna gen. et sp. nov., is reported from the Early Cretaceous (late Aptian-early Albian) of the Gansu Province, Northwest China, based on numerous well-preserved axes with attached leaves and infructescences. The leaves are alternate, short petiolate and linear-lanceolate with low rank pinnate to reticulate venation. The infructescences are loose panicles bearing fruits in different stages of maturity, each containing four partly free carpels borne in a whorled arrangement. Each carpel has three to five seeds borne along its ventral margin. The nature of the leaves and axes indicates a terrestrial, herbaceous habit. In general organization, Gansufructus is closely similar to the fruit-bearing axes of Sinocarpus decussatus from the Early Cretaceous Jehol Biota, as well as other more or less contemporaneous angiosperms from the Far East, which together provide evidence of diverse eudicot angiosperms of low stature colonizing areas close to environments of deposition.

ABSTRACT

Long non-coding RNAs (lncRNAs) have been reported to play important roles in Parkinson's disease (PD) pathogenesis. It was indicated that lncRNA nuclear enriched abundant transcript 1 (NEAT1) is involved in PD. However, the underlying mechanism of NEAT1 is still not fully explored. Human neuroblastoma cell line SH-SY5Y was treated with 1-Methyl-4-phenylpyridinium (MPP+) to mimic PD model in vitro. qRT-PCR was employed to detect the expression of NEAT1, IL-1ß, IL-6 and TNF-α. Starbase database, RNA pull-down assay and RNA immunoprecipitation (RIP) assay were performed to verify the relationship between NEAT1 and miR-124. MTT assay and flow cytometry assay were used to detect cell viability and apoptosis. Elisa was introduced to measure the levels of IL-1ß, IL6 and TNF-α in culture media. We found that NEAT1 expression was significantly increased in SH-SY5Y cells after MPP+ treatment in dose- and time- dependent manners. MPP+ induced the expression and secretion of IL-1ß, IL-6 and TNF-α, inhibited cell viability and induced apoptosis while these effects could be rescued by NEAT1 silencing. miR-124 was a target of NEAT1. Anti-miR-124 could reverse the effects caused by NEAT1 knockdown in MPP+ treated SH-SY5Y cells. Therefore, we speculated that NEAT1 may regulate MPP+ induced neuronal injury by targeting miR-124 in SH-SY5Y cells.

ABSTRACT

Parkinson's disease (PD) is a common neurological disorder that is detrimental to the health of older people worldwide. Long noncoding RNAs (lncRNAs) have been reported to play essential roles in the pathogenesis and therapeutics of PD. LncRNA homeobox transcript antisense intergenic RNA (HOTAIR) is expressed in PD samples; however, the exact roles of HOTAIR and its mechanism remain largely unclear. Herein, the neurotoxins 1-methyl-4-phenylpyridine (MPP+) and 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) were used to establish PD models in vitro and in vivo. The expressions of HOTAIR and microRNA-221 (miR-221) were measured by the quantitative real-time polymerase chain reaction (qRT-PCR). Cell viability and apoptosis were detected by the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) assay and western blot or flow cytometry, respectively. The interaction between HOTAIR and miR-221 was explored by luciferase activity and RNA immunoprecipitation (RIP). The tyrosine hydroxylase (TH)-positive cells in MPTP-treated-mouse midbrains were analyzed by immunohistochemistry. The HOTAIR expression was up-regulated and that of miR-221 was down-regulated in the serum of PD patients and MPP+-treated SH-SY5Y cells. Overexpression of HOTAIR inhibited cell viability and promoted apoptosis in MPP+-treated SH-SY5Y cells. However, the down-regulation of HOTAIR showed an opposite effect. Moreover, miR-221 was validated to be bound to HOTAIR, and its addition reversed the regulatory effect of HOTAIR on cell viability and apoptosis in MPP+-treated SH-SY5Y cells. Moreover, the knockdown of HOTAIR attenuated the degree of PD and cell apoptosis by regulating miR-221 in MPTP-treated mice. In conclusion, HOTAIR contributed to cell apoptosis by sponging miR-221 in PD. This study elucidates a new mechanism for understanding the pathogenesis of PD and provides a promising target for the treatment of PD.
