ABSTRACT

Human DAB2 interaction protein (DAB2IP) is a member of Ras-GTPase activating protein family and functions as a tumor suppressor, implying it could serve as a prognostic biomarker in cancers. Here we generated a mouse monoclonal antibody, 2A4, directed against human DAB2IP. This antibody was identified as IgG1 and specifically recognizes DAB2IP in both its native and denatured forms. It will serve as a useful and versatile tool for further mechanistic study and development of the potential prognostic significance of DAB2IP.

ABSTRACT

OBJECTIVE: To discuss the changes in Wnt pathway inhibiting factors in esophageal precancerosis lesions induced by methyl benzyl nitrosamine (MBNA) and the effect of Gexia Zhuyu decoction. METHOD: Wistar rats were subcutaneously injected with MBNA (3.5 mg x kg(-1) for twice per week to establish the model. Since the 1st day after the model establishment, they were orally administered with Gexia Zhuyu decoction (16, 8 mg x kg(-1)). At the 10th week, esophageal tissues were collected to observe the pathological changes of esophageal mucosa, detect SFRP1, sFRP4, Axin1, Axin2 and GSK-3ß mRNA levels.by fluorescent quantitation PCR analysis and ß-catenin protein level by Western blotting. RESULT: Being induced by MBNA, rats in the model group showed slight atypical hyperplasia in the histopathological examination. Compared with the normal group, Gexia Zhuyu decoction dose high and low groups showed no significant pathomorphological and histological changes. The model group showed lower gene transcription levels of esophageal tissues sFRP1, sFRP4, Axin1 and Axin2 (P < 0.05 or P < 0.01) and higher ß-catenin protein expression level (P < 0.01) than the normal control group. The Gexia Zhuyu decoction low dose group showed higher gene transcription levels of esophageal tissues sFRP1, sFRP4, Axin1 and Axin2 (P < 0.05 or P < 0.01) and lower ß-catenin protein expression level (P < 0.01) than the normal control group. CONCLUSION: Up-regulated ß-catenin protein level and down-regulated Wnt pathway could enhance Wnt pathway activity of MBNA-induced esophageal precancerous lesions. Gexia Zhuyu decoction could down-regulate the ß-catenin protein level and up-regulate the transcription level of Wnt pathway inhibiting factors, but could not block MBNA-induced esophageal precancerosis lesions.

ABSTRACT

Nitrification-based microbial fuel cell (N-MFC) is a novel inorganic microbial fuel cell based on nitrification in the anode compartment. So far, little information is available on the substrates and pathway of N-MFC. The results of this study indicated that apart from the primary nitrification substrate (ammonium), the intermediates (hydroxylamine and nitrite) could also serve as anodic fuel to generate current, and the end product nitrate showed an inhibitory effect on electricity generation. Based on the research, a pathway of electricity generation was proposed for N-MFC: ammonium was oxidized first to nitrite by ammonia-oxidizing bacteria (AOB), then the nitrite in anolyte and the potassium permanganate in catholyte constituted a chemical cell to generate current. In other words, the electricity generation in N-MFC was not only supported by microbial reaction as we expected, but both biological and electrochemical reactions contributed.

ABSTRACT

Effect of substrate concentration on substrate degradation and electricity generation in anodic denitrification microbial fuel cell (AD-MFC) was investigated over a broad range of substrate concentrations. Substrate degradation rates and power generation could be promoted with increasing substrate concentration in a certain range, but both of them would be inhibited at high substrate concentrations. Maximum denitrification rate of 1.26 ± 0.01 kg NO(-)-N/m(3)d and maximum output voltage of 1016.75 ± 4.74 mV could be achieved when initial NO3(-)-N concentration was 1999.95 ± 2.86 mg/L. Based on Han-Levenspiel model, kinetics of substrate degradation and power generation in the AD-MFC were established. According to the kinetic model, the half-saturation coefficient and the critical inhibitory concentration for nitrate were more than 200 and 4300 mg/L, respectively. The results demonstrated that AD-MFC was capable of treating nitrate-containing wastewater and generating electricity simultaneously, and tolerant to high strength nitrate-containing wastewater.

ABSTRACT

Two-chamber Microbial Fuel Cells (MFC) using graphite rods as electrodes were operated for simultaneous anaerobic sulfide and nitrate removal coupled with electricity generation. The MFC showed good ability to remove substrates. When the influent sulfide and nitrate concentrations were 780 mg/L and 135.49 mg/L, respectively, the removal percentages of sulfide and nitrate were higher than 90% and the main end products were nitrogen and sulfate. The MFC also showed good ability to generate electricity, and the voltage went up with the rise of influent substrate concentrations. When the external resistance was 1000 Ω, its highest steady voltage was 71 mV. Based on the linear relationship between the electrons released by substrates and accepted by electrode, it was concluded that the electricity generation was coupled with the substrate conversion in the MFC.

ABSTRACT

BACKGROUND: To investigate the clinical value of serum total MMP-9 detection in lung malignancies. METHODS: Serum total MMP-9 levels were detected in 63 patients with lung malignancies by sandwich ELISA with monoclonal antibody against human total MMP-9. RESULTS: Serum total MMP-9 was (92.2± 74.39) µg/L in lung malignancy group, which was significantly higher than that of healthy control [(9.5± 6.74) µg/L](P < 0.05). In lung cancer group, there were significant differences in serum total MMP-9 level between patients with and without lymph node metastasis, and between progressive disease+death group and complete response+partial response group. Significantly negative correlation was observed between serum CA242 and serum total MMP-9 levels (P=0.021). CONCLUSIONS: Detection of serum total MMP-9 may be helpful to predict metastasis and treatment response of lung cancer.
