ABSTRACT

To conduct an anti-tumor research by using targeted drug-loaded cell-derived microparticles to target the tumor microenvironment and enhance NK cell killing function. In this experiment, we obtained HepG2 tumor cell-derived microparticles by physical extrusion, high speed centrifugation and filtration, modified the hepatocellular carcinoma targeting peptide SP94 on the surface of microparticles and encapsulated the TGF-ß inhibitor SB505124. Finally we validated and analyzed whether the new drug delivery system can target to tumor site and enhance the anti-tumor function of NK cells. This type of novel targeted cell-derived microparticles drug delivery system will provide a novel idea for tumor immunotherapy.

ABSTRACT

Hepatocellular carcinoma (HCC) is a common malignant tumor with poor prognosis and high mortality. In this study, we demonstrated a novel vaccine targeting HCC and tumor neovascular endothelial cells by fusing recombinant MHCC97H cells expressing porcine α-1,3-galactose epitopes (αGal) and endorphin extracellular domains (END) with dendritic cells (DCs) from healthy volunteers. END+/Gal+-MHCC97H/DC fusion cells induced cytotoxic T lymphocytes (CTLs) and secretion of interferon-gamma (IFN-γ). CTLs targeted cells expressing αGal and END and tumor angiogenesis. The fused cell vaccine can effectively inhibit tumor growth and prolong the survival time of human hepatoma mice, indicating the high clinical potential of this new cell based vaccine.

ABSTRACT

Bacillus spp. are probiotics and can secrete a variety of natural antimicrobiol active substances, of which lipopeptides are an important class. Up to now, about 90 lipopeptides have been identified, and most of them are cyclic lipopeptides. surfactin, iturin, fengycin, bacillomycin and polymyxins are widely studied, and the first three have huge potential for application due to their properties of surfactants and anti-fungal, anti-bacterial, anti-viral, anti-tumor and anti-inflammatory functions. In this paper, the research progress in the structure, function, synthesis regulation, separation, purification and production of surfactin, iturin and fengycin was reviewed. Synthetic biology is a vital means to increase the yield of lipopeptides, and in the future, lipopeptides can be used in crop cultivation, animal farming, food, medicine and petroleum industries as well as environmental protection. Future research should be strengthened on the discovery of new lipopeptides, synthesis of high-activity lipopeptides, economical production of lipopeptides on a large scale and their safety evaluation.

ABSTRACT

This paper proposed a quarantine-susceptible-exposed-infectious-resistant (QSEIR) model which considers the unprecedented strict quarantine measures in almost the whole of China to resist the epidemic. We estimated model parameters from published information with the statistical method and stochastic simulation, we found the parameters that achieved the best simulation test result. The next stage involved quantitative predictions of future epidemic developments based on different containment strategies with the QSEIR model, focused on the sensitivity of the outcomes to different parameter choices in mainland China. The main results are as follows. If the strict quarantine measures are being retained, the peak value of confirmed cases would be in the range of [52438, 64090] and the peak date would be expected in the range February 7 to February 19, 2020. During March18-30, 2020, the epidemic would be controlled. The end date would be in the period from August 20 to September 1, 2020. With 80% probability, our prediction on the peak date was 4 days ahead of the real date, the prediction error of the peak value is 0.43%, both estimates are much closer to the observed values compared with published studies. The sensitive analysis indicated that the quarantine measures (or with vaccination) are the most effective containment strategy to control the epidemic, followed by measures to increase the cured rate (like finding special medicine). The long-term simulation result and sensitive analysis in mainland China showed that the QSEIR model is stable and can be empirically validated. It is suggested that the QSEIR model can be applied to predict the development trend of the epidemic in other regions or countries in the world. In mainland China, the quarantine measures cant be relaxed before the end of March 2020. China can fully resume production with appropriate anti-epidemic measures beginning in early April 2020. The results of this study also implied that other countries now facing the epidemic outbreaks should act more decisively and take in time quarantine measures though it may have negative short-term public and economic consequences.

Objective@#To investigate the relationship between the expression of mucin 1 and collagen Ⅳ and the clinical characteristics in invasive breast cancer by using the quantum dots immunofluorescence imaging technique.@*Methods@#The expressions of mucin 1 and collagen Ⅳ were detected simultaneously by quantum dots immunofluorescence imaging technique in 94 cases of breast cancer from July 2007 to July 2010 in Affiliated Hospital of Guilin Medical University. The correlation of mucin 1 and collagen Ⅳ quantitative parameters with clinicopathological features and the prognosis were also analyzed.@*Results@#The positive rates of mucin 1 in human breast cancer tissues marked by quantum dots immunofluorescence imaging technique and immunohistochemistry were 73.4 % (69/94) and 69.1 % (65/94), the positive rates of collagen Ⅳ were 53.2 % (50/94) and 47.9 % (45/94), and the differences were not statistically significant (both P > 0.05) Quantum dots immunofluorescence imaging technique was consistent with conventional immunohistochemistry (IHC) in detecting the expressions of mucin 1 and collagen Ⅳ in human breast cancer tissues (κ = 0.763, P = 0.000; κ=0.759, P = 0.000). The expression of mucin 1 was negatively correlated with collagen Ⅳ (r = -0.883, P < 0.01). The expressions of mucin 1 and collagen Ⅳ were respectively associated with tumor size (F = 3.683, P = 0.029; F = 4.922, P = 0.009), histological grading (F = 3.529, P = 0.033; F = 3.912, P = 0.023), lymph node metastasis (t = -4.868, P = 0.000; t = 3.868, P = 0.000), pathological stage (t = -8.337, P = 0.000; t = 5.962, P = 0.000) and 5-year disease free survival rate (both P = 0.000), and the differences were statistically significant (all P < 0.05).@*Conclusion@#The co-detection of mucin 1 and collagen Ⅳ by using quantum dots immunofluorescence imaging technique provides direct evidence determining the biologic behaviors of breast cancer and evaluating the prognosis.

ABSTRACT

Objective To investigate the prevalence of iron deficiency(ID)and iron deficiency anemia (IDA) in pregnant women in urban areas of China. Methods The study was a national cross-sectional survey conducted from September 19th, 2016 to November 20th, 2016. According to the classification of the National Bureau of Statistics, all survey sites were set up in 6 regions of the country. Pregnant women were continuously selected using multistage stratified sampling. A total of 12 403 pregnant women were collected and examined for serum ferritin and hemoglobin levels. Results The median serum ferritin level during pregnancy was 20.60 μg/L(11.78-36.98 μg/L), the hemoglobin level was(118±12)g/L. With the progress of pregnancy, the levels of serum ferritin and hemoglobin decreased gradually. The median serum ferritin levels in the first, second trimester and third trimester were 54.30 μg/L(34.48-94.01 μg/L), 28.60 μg/L(16.40-50.52 μg/L), and 16.70 μg/L(10.20-27.00 μg/L)respectively(P<0.01). The mean hemoglobin levels were(127 ± 10)g/L,(119 ± 11)g/L and(117 ± 11)g/L respectively(P<0.01). The prevalence of ID in urban pregnant women was 48.16%(5 973/12 403), and IDA prevalence was 13.87% (1 720/12 403). The prevalence of IDA in the first, second trimester and third trimester were 1.96% (20/1 019), 8.40%(293/3 487)and 17.82%(1 407/7 897), respectively(P<0.01). The prevalence of standardized ID and IDA were significantly different in various regions of China(P<0.01). The standardized prevalence of ID were relatively higher in East China and Northeast China, 57.37% and 53.41% respectively, while it was the lowest in Southwest China, 30.51%. The standardized prevalence of IDA in South Central, Northwest, and East China were relatively high, 21.30%, 16.97% and 17.53% respectively, and the standardized prevalence of IDA in Southwest China was the lowest, 5.44%,the differents in various regions were significant(all P<0.01). Conclusion The current phenomenon of ID and IDA in pregnant women is still very common,and nutrition and health care during pregnancy should be strengthened.

ABSTRACT

Objective To analyze the imaging features and follow-up changes of high-resolution CT(HRCT) in Goodpasture syndrome.Methods HRCT imaging features and follow-up findings of 15 cases Goodpasture syndrome confirmed by clinical were analyzed retrospectrively.The imaging features included extent,forms and follow-up changes.Results The lung lesion of Goodpasture syndrome involved two lobes(n=1), three lobes(n=2), four lobes(n=5) and five lobes (n=7).Upper lobe of the right lung was the most common involved region.Centered on the hilum of lung consolidations confused ground glass opacity (GGO) were showed in 7 cases, GGO distribution of pulmonary leaflets in 5 cases.On follow-up observation, lobar or segmental consolidation could change into GGO,GGO could disappear in short times.Conclusion Multiple lobar or segmental consolidations confused GGO without the lung bottom and periphery involvement is the imaging characteristics of Goodpasture syndrome patients with anemia and hemoptysis.HRCT is a helpful method for the diagnosis and following up of Goodpasture syndrome.

ABSTRACT

OBJECTIVE To investigate the tinnitus' prevalence in Healthy check-up population in Dalian City. METHODS 1748 cases (male 806, female 942) were chosen from the Physical Examination Center of our hospital from February 1st, 2015 to April 1st, 2015, the tinnitus questionnaire was performed. For the cases with the tinnitus symptoms, the characteristics of tinnitus were further inquired, including the course of tinnitus, seizure frequency, the duration of tinnitus, pulsatile or persistent, unilateral or bilateral. RESULTS The total prevalence of tinnitus is 32.4%(566/1748), the prevalence of persistent tinnitus is 18.3%(320/1748), and the transient tinnitus is 14.1%(246/1748); The prevalence of tinnitus in female are 36.4%(343/942), 20.0%(188/942) and 16.5%(155/942) in turn, which are higher than that of the male 27.7%(223/806), P<0.01; 16.4%(132/806), P =0.054, and 11.3%(91/806), P<0.01. Both the total and persistent tinnitus prevalence increase with age(P<0.01), but the transient tinnitus doesn't increase with age. The prevalence of objective and subjective tinnitus are 1.3%(6.9% out of total prevalence of tinnitus), and 17.0%(93.1% out of total prevalence of tinnitus), respectively. The rate of bilateral tinnitus is 78.1% out of total tinnitus, and rate of unilateral tinnitus is 21.9%(right 11.6%, left 10.3%). In addition, the rates of acute tinnitus, subacute tinnitus, and chronic tinnitus are 4.4%, 3.8%, and 91.9% out of total, respectively. Otherwise, the rates of sleep disorders and hearing loss complaining accounted for 31.9% and 14.1% respectively. CONCLUSION The prevalence of tinnitus is high in healthy check-up population of Dalian, the total prevalence of tinnitus is 32.4%, mainly bilateral,chronic and subjective.

ABSTRACT

Objective To observe the preventive effects of puerarinon development of deep vein thrombosis (DVT) in elderly women with osteoporosis after hip replacement. Methods 100 elderly women with osteoporosis who scheduled for femoral head arthrolastybetween January 2012 and January 2014 in Cangzhou Central Hospital were selected and then randomly divided into a study group (n=50) and a control group (n=50). The control group received routine anti?inflammatory and anticoagulant therapy ,while the study group received intravenous drip of puerarinof 400 mg mixed with 5%glucose liquid 500 ml daily for 15 days as a treatment course. After the treatment, DVT incidence rate ,change of early diagnostic indexes and hemorheology were compared with their baselines. Results DVT incidence rate was significantly lowerin the study group than in the control group (8.0%vs. 24.0%, P<0.05). Ascompare with the control group,thromboxane A2/prostacyclin,plasma homocysteine and hemorheology?were significantly reduced (P < 0.05). Conclusions Puerarin has a preventive effect for deep vein thrombosis in elderly women with osteoporosis after hip replacement.

ABSTRACT

Objective To observe the effects of recombinant human interleukin 11(rhIL-11) on proliferation, migration and invasion of A549 cells, and the mechanism thereof. Methods Final concentrations of 0, 10, 20, 50 and 100μg/L rhIL-11 were added into pulmonary adenocarcinoma A549 cells. The cell proliferation was detected by MTT. The wound-healing, transwell migration assay were used to validate the capability of the migration and invasion of A549 cells. Matrix metallopro-teinases (MMP)-2 and MMP-9 protein expressions were revealed by Western blot assay. Results The proliferation of A549 cells was not significantly changed by rhIL-11. The cell capability to migrate and invade was significantly increased 24 h af-ter treatment with rhIL-11 (P<0.05). The expression levels of MMP-2 and MMP-9 were significantly un-regulated, and which were increased with the increased concentrations of rhIL-11 (P<0.05). Conclusion rhIL-11 can promote the migra-tion and invasion of A549 cells, and the up-regulation of MMP-2 and MMP-9 expression might be one of the mechanisms.

ABSTRACT

Objective:To implore the effect of continuous positive airway pressure (CPAP) therapy on dizziness patients and its mechanism. Method:Seventy-five dizziness patients were enrolled. All patients accepted polysomnographic test, dizziness handicap inventory, Pitsburgh sleep quality index and Epworth sleepiness scale before and after CPAP therapy. Patients were divided into two groups according to a polysomnographic test: OSAHS group (AHI≥15) and without OSAHS group (AHI<15); divided into three groups according to dizziness handicap inventory score: mild dizziness group (0<DHI≤30), moderate dizziness group (30<DHI≤60), severe dizziness group (60<DHI≤100); divided into three groups according to therapy time: 0-<3 months group, 3-<6 months group, 6-12 months group; divided into four groups according to age: youngth group (age<45), middle age group (45-<60), younger old group (60-<75) and old group (≥75). Result:①There was a significant difference in all dizziness patients before and after CPAP therapy according DHI (P<0.01). ②The effectiveness of CPAP therapy between OSAHS group and without OSAHS group has no difference (P>0.05), but CPAP therapy could ameliorate sleep disorder of the OSAHS group. ③Different levels of dizziness had no impact on improving DHI during CPAP therapy. However, the effect on sleep improvement decreased when patients had heavier dizziness. ④Treating by CPAP after three months could significantly improve patients dizziness (P<0.01). ⑤The dizziness of the patients in younger old group and old group have significant improvement after CPAP therapy (P<0.01). However, only younger old group has better sleep. Conclusion:CPAP therapy could dramatically improve dizziness in patients with sleep disorders.

ABSTRACT

OBJECTIVE:To investigate the effects of puerarin on bone mineral density around the femoral prosthesis of el-derly women after osteoporotic fracture artificial hip joint replacement. METHODS:99 elderly women after osteoporotic fracture artificial hip joint replacement were divided into control group(49 cases)and test group(50 cases)according to random number table. Control group received conventional treatment:calcium carbonate and vitamin D3+alendronate sodium+salmon calcitonin;test group was additionally given Puerarin injection 200-400 mg dissolved in Glucose injection 500 ml intravenously,qd,on the basis of control group. A treatment course lasted for 20 d,and both groups received 2 courses of treatment. The hip joint function score and bone mineral density around the femoral prosthesis of 2 groups were observed and compared after surgery,and the oc-currence of ADR was also observed. RESULTS:3 and 2 patients withdrew from control group and test group,respectively. 18 months after surgery,the patients with hip joint function score ranged 70-79 in test group was significantly less than in control group;the rate of excellent hip joint function score in test group was significantly higher than in control group,with statistical significance (P0.05);bone mineral density in R6-R7 range was signifi-cantly higher than control group(89.58% vs. 69.57%),with statistical significance(P<0.05). The prosthesis loosening was not found in both groups,and ADR was also not found as fever,erythra,nausea,vomiting,headache,dizziness,etc. CONCLU-SIONS:For the use of puerarin in elderly women after osteoporotic fracture artificial hip joint replacement,puerarin can increase the periprosthetic femur bone mineral density with good safety.

ABSTRACT

Objective To analyze the influence of hepatosteatosis on pancreatic β-cell function in type 2 diabetes mellitus (T2DM).Methods A total of 213 subjects with T2DM from Metabolic Disease Hospital,Tianjin Medical University from January 2013 to December 2013 were included in the study.Non-alcoholic fatty liver disease (NAFLD) was diagnosed with abdominal ultrasonography.Patients were divided into two groups:T2DM with NAFLD and T2DM without NAFLD.ALT,AST,γ-glutamyltransferase,serum lipid,glycosylated hemoglobin A1 c (HbA1c),fructosamine,fasting glucose,insulin and 2 hours plasma glucose,insulin after 75g glucoseload were detected.The insulin resistance and β-cell function were assessed by HOMA-IR and HOMA-β.Results Among the 213 T2DM subjects,51% (108 cases) were with NAFLD.The HOMA-IR [4.76 (2.83,7.21) vs 2.79 (1.76,4.37),P < 0.05] and HOMA-β [49.18 (37.78,85.09) vs 29.50 (18.09,45.54),P < 0.05] were significantly higher in T2DM with NAFLD than those in T2DM alone.Within subjects with T2DM and NAFLD,the HOMA-IR [6.28 (2.87,8.17) vs 2.95 (2.07,3.66) P < 0.05] and HOMA-β [59.18 (37.78,85.09) vs 30.59 (28.56,34.49),P < 0.05] levels were higher in subjects with normal liver function than those with abnormal liver function.Conclusions T2DM patients with NAFLD have severer insulin resistance than those without NAFLD.The β-cell function of those patients was compensatory increased,which was decreased in subjects with abnormal liver function.

ABSTRACT

In an attempt to produce effective and lower price gaseous Hg(0) adsorbents, two methods of HCl and KMnO4/heat treatment were used respectively for the surface modification of liginite semicoke from inner Mongolia. The different effects of modification process on the surface physical and chemical properties were analyzed. The characteristics and stability of mercury vapor adsorption over two kinds of modified semicoke were investigated. The results indicated that modification process caused lower micropore quantity and volume capacity of semicoke; the C-Cl functional groups, C=O bond and delocalized electron π on the surface of Cl-SC, the amorphous higher valency Mn (x+) , and O=C-OH functional groups on the surface of Mn-H-SC were the active sites for oxidation and adsorption of gaseous Hg(0). Modification process led to higher mercury removal efficiency of semicoke at 140°C and reduced the stability of adsorbed mercury of semicoke in simulated water circumstance simultaneously.

Objective To construct recobinant pmRNA IRES-hKDR and lay a primary foundation for further study of gene therapy for tumors.Methods pmRNA IRES-hKDR was obtained from pDC520 hkDR and pmRNA IRES-Luc which was reserved by our laboratory through molecular biology technology and was transfected into Hepall-6 via lipsome after identification of PCR,restriction enzyme digesting and DNA ELISA and Western blot.Results The recombiant plasmid had pmRNA IRES-hKD gene which was expressed in Hepall-6//G418 in vitro and the expressed protein was secreted out of the cells could response with specific hKDR antibody,which was identified by Western blot.Conclusion An eukaryotic expression recombiant plasmid pmRNA IRES-hKDR can be constructed successfully which provides the possibility of further researches on its anti-tumor effect.

Objective To investigate the immune protection of dendritic cells(DCs) harboring kinase domain-containing receptor(KDR) fusion gene on mice carrying B16 melanoma.Methods The bone marrow precursor-derived dendritic cells(BMDCs) were induced from bone marrow progenitors of mice by GM-CSF.The KDR fusion gene mRNA was transfected into DCs in vitro.Mice were immunized with the same amount of DCs at 7-day interval and then each mouse was injected with 5?10~5 B16 cells.The mice without tumors were injected with B16 cells again 20 days later.Mice were randomly divided into 4 groups: group A(n=7),group B(n=8),group C(n=8) and group D(n=7).The mice were immunized with DCs one time in group A,2 times in group B,3 times in group C and as controls in group D.Results After one week,all mice in group D had tumors with average survival 15 days.All mice in group A,B and C had no tumors after 20 days later.After the second injection of B16 cells,2 mice in group A and 2 mice in group B had tumors.The mice in group C had no tumor.The average survival periods were calculated from first injection of B16 cells to the study end.The average survival period of group A was 50 days and that of group B was 72 days.Conclusions The dendritic cells vaccine harboring KDR fusion gene has immune protection against melanoma in mice.

ABSTRACT

Objective Clinical reports on perilymphatic fistulae(PLF) of the round window membrane(RWM) have shown different degrees of hearing loss. For the unknown mechanisms of hearing loss, the morphological changes of cochlea with perilymphatic fistulae(PLF) was studied.Methods Among 10 guinea pigs, each ear on one side was punctured and the ear of another side was used as control. 4 hours after operation, the morphological changes of cochlea were observed.Results It was found that 2 ears with endolymphatic hydrops, 1 ear with strica vascularis stripped, venous hyperemia and hemorrhage in cochlea, 7 ears with no changes.Conclusion The study suggestes that the normal and the abnormal morphological changes of cochlea can all be seen after the rupture of RWM.

Objective To explore the significance of RPR te st and Tp -IgM-Capture -ELISA in fol-lowing-up of early syphilis.Methods Sixty-one cases of primary syphilis with less than one-year course and 77cases of secondary syphilis were enr olled into the study.The patients we re followed up by RPR test and Tp -IgM-Capture -ELISAat intervals of three months for two years after treatment .Results By the end of 3,6,9,12,15,18,21and 24months,RPRtest became negative in 6.2%,31.5%,61.5%,83.8%,90.7%,92.3%,94.6%,and95.3%of patients,respectively.By the end of 3,6,9,12and 15months,Tp -IgM-Capture -ELISA became negative in 25.4%,56.5%,86.2%,97.1%and 100.0%of patients,respectiv ely.In primary syphilis the RPR test became nonreactive in 8.2%(3months after treatment ),31.1%(6months),57.4%(9months),75.4%(12months),83.6%(15months),85.2%(18months),and 100.0%(21months)of patients,and Tp -IgM-Cap-ture -ELISA in 45.9%(3months),85.2%(6months),98.4%(9months),100.0%(12months)of patients.In secondary syphilis RPRtest became n onreactive in 2.6%(3months after treatment ),28.6%(6months),58.4%(9months),81.8%(12months),84.4%(15months),88.3%(18months),90.9%(21months)and 92.2%(24months)of patients,and Tp -IgM-Capture -ELISAin 9.1%(3months),33.8%(6months),71.4%(9months),94.8%(12months)and 100.0%(15months)of patients.Tp -IgM-Capture -ELISA was found to be negative earlier than that of RPR test in all ca ses.No sero-resistant was shown in T p -IgM-Capture -ELISA.Conclusions Tp -IgM turns to negative in 12months after treatment for nearly all patie nts with early syphilis.Tp -IgM-Cap-ture -ELISA may be served as a useful t ool to follow up early syphilis patie nts after treatment.[
