ABSTRACT

The pathogenesis-related protein-1 (PR1) gene is important for plants to respond to various biotic and abiotic stresses. Unlike those in model plants, PR1 genes in wheat have not been systematically studied. Herein, we identified 86 potential TaPR1 wheat genes using bioinformatics tools and RNA sequencing. Kyoto Encyclopedia of Genes and Genomes analysis indicated that the TaPR1 genes were involved in the salicylic acid signalling pathway, MAPK signalling pathway, and phenylalanine metabolism in response to Pst-CYR34 infection. Ten of the TaPR1 genes were structurally characterized and validated by RT‒PCR. One particular gene, TaPR1-7, was found to be associated with resistance to Puccinia striiformis f. sp. tritici (Pst) in a biparental wheat population. Virus-induced gene silencing showed that TaPR1-7 is important for Pst resistance in wheat. This study provides the first comprehensive study on wheat PR1 genes, improving our overall understanding of these genes in plant defenses, particularly against stripe rust.

ABSTRACT

Objective To explore the role of next-generation sequencing(NGS)technology in the assisted diagnosis of RA-Sopathies.Methods Peripheral blood was extracted from 1 child patient with suspected Noonan syndrome and her parents,and the gene mutations were detected by adopting the aCGH and NGS.The results were verified by Sanger sequencing.Results The NGS results revealed that the heterozygous mutation of c.1406G>A existed in BRAF gene,and the results of Sanger sequencing in this child case was consistent with the NGS results.The Sanger sequencing results in her parents showed the locus was G/G wild type. Conclusion This child case was diagnosed as CFC.NGS plays a good auxiliary role in the differentiation diagnosis of RASopathies.

ABSTRACT

Origin of Chinese Characters (Shuowen Jiezi) records a large number of drug names, providing rich materials for the study of the history of pharmacy. But in the aftertime circulating process, errors were inevitable, making the ancient scholars disagree on the relationship between some drug names and their referents. This paper would give a textual research on medication names about chu, huo, huo, and hua. Relevant content was analyzed by using Shuowen Jiezi, Er Ya,Yu Pian and previous annotations of these exegesis books, as well as herbal literature. The relationship between the names and referents, as well as their lexical meanings were clarified by comparing textual content in specialized book about Chinese exegetics and previous herbal literature. Their medicinal efficacy was explained, and the value of herbalism in these exegesis books was explored.

ABSTRACT

Many medicine names were recorded in Shuowen Jiezi. The later scholars interpreted these names, but they had different standpoints on the relationship between the name and nature of some medicines. In this study, the medicine names of diao, ji, li, tiao, and di with greater different standpoints were verified by textual research. Shuowen Jiezi, Erya, Guangya and previous annotations of these exegesis books were combined with herbal literature to summarize and analyze the relevant content. The herbal content in exegesis books were compared with the records in herbal literature to hackle the relationship between name and nature of medicines, clarify the lexical meaning and illustrate the efficacy of the medicines. Meanwhile, the value of herbalism in these exegesis books was explored.

ABSTRACT

AIM OF THIS STUDY: Chinese herbal pill Yi Shen Juan Bi (YJB), which is a compound herbal drug, has traditionally been used as an anti-rheumatic drug in Chinese local clinics. Thus, we investigated the therapeutic effect of the YJB in rheumatoid arthritis (RA), using Freund's complete induced adjuvant arthritis (AA) in rat model. METHODS: For that purpose, macrophage derived cytokine tumor necrosis factor alpha (TNF-α) and interleukin-1 (IL-1) were measured in ex vivo by enzyme linked immunosorbent assay (ELISA). We also assayed the effect of YJB on peritoneal macrophage derived nitric oxide (NO) in ex vivo by Griess reaction. Prostaglandin E (PGE) in metapedes was assayed by ultraviolet spectrophotometer method. Further synovial Bax level was examined by Western blot analysis. RESULTS: YJB significantly decreased the production of peritoneal macrophages derived TNF-α, IL-1 and NO. YJB also significantly decreased prostaglandin E (PGE) and upregulated the Bax expression in AA rat's synovium. CONCLUSION: YJB is a potential anti-rheumatic agent targeting the inflammatory and immunomodulatory response of macrophages while down regulating the PGE and up-regulating the pro-apoptotic Bax expression. Such characteristics of YJB on AA may be advantageous to the treatment of clinical rheumatoid arthritis.

ABSTRACT

Objective To investigate the expression and clinical significance of peripheral blood CD4~+, CD25~+ and CD4~+CD25~+ T subpopulations in patients with systemic lupus erythematosis.Methods The per- centage and fluorescence intensities of peripheral blood CD4~+,CD25~+ and CD4~+CD25~+ subpopulations from 34 SLE and 18 normal controls were measured with flow cytometry assay,then the correlation with clincal data was analyzed.The CD25~+ cells were defined as the CD25~(high) cells if their fluorescence intensity was higher than 10. Results The percentage of CD4~+CD25~+,CD4~+CD25~(high) T lymphocytes in active SLE patients[(4.80?1.21)% and (0.25?0.10)%]was lower than that in normal controls[(8.92?3.21)% and(0.44?0.22)% and non-active SLE patients(11.28?2.09)% and(0.59?0.34)%](P<0.05).However,as for the CD25~+ cells in the CD4~+ T cells,there was no difference between SLE patients and normal control group.Peripheral blood CD4~+CD25~+,CD4~+CD25~(high) cells in SLE were reversely correlated with SLEDAI(r=-0.74,P=0.004 and r=-0.614,P=0.026),but not with others such as complements,ANA titers etc.Peripheral blood CD4~+ and CD25~+ lymphocytes in active SLE pa- tients were also lower than those in normal controls[(23?7)vs(34?7)and(7.4?1.8)vs(13.9?3.4),P<0.05]. CD25 fluorescence intensities were higher in the SLE patients those in the normal controls,but CD4 fluores- cence intensities were not.Conclusion CD4~+CD25~+ may play a role in the pathogenesis of SLE.

ABSTRACT

Objective To summarize the manifestation and treatment of acquired hemophilia A in pa- tients with systemic lupus erythematosus(SLE).Method A case was investigated retrospectively and the lit- erature was reviewed.Results A 25-year-old woman with a 5 year history of SLE was admitted to hospital due to abdominal pain.She was diagnosed with acquired factorⅧinhibitor deficiency based on a prolonged activated partial-thromboplastin time(APTT,135.3 s),reduced factorⅧactivity(0.9%)and factorⅧin- hibitor(26.1 BU/ml).Sonography and magnetic nuclear resonance of the abdomen confirmed the presence of a retro-uterine hematoma.The patient was initially treated with intravenous pulse and oral corticosteroids,factorⅧplasma concentrated and intravenous immunoglobulin.Clinical and biological improvement was promptly obtained.Conclusions Attention should be paid to the association between SLE and acquired hemophilia A. Combination therapy may be recommended as initial therapy for the management of acquired hemophilia A in patients with SLE.But no standardized treatment can be recommended at present.
