ABSTRACT

Benzodiazepines(BDZs)are used in clinics for anxiolysis,anticonvulsants,sedative hypnosis,and muscle relaxation.They have high consumptions worldwide because of their easy availability and potential addiction.They are often used for suicide or criminal practices such as abduction and drug-facilitated sexual assault.The pharmacological effects of using small doses of BDZs and their detections from complex biological matrices are challenging.Efficient pretreatment methods followed by accurate and sensitive detections are necessary.Herein,pretreatment methods for the extraction,enrichment,and preconcentration of BDZs as well as the strategies for their screening,identification,and quantitation developed in the past five years have been reviewed.Moreover,recent advances in various methods are summarized.Characteristics and advantages of each method are encompassed.Future directions of the pretreatment and detection methods for BDZs are also reviewed.

ABSTRACT

Nucleotides, which are important low-molecular-weight compounds present in organisms, are precursors of nucleic acids and participate in various regulatory and metabolic functions. Sensitive and valid methods for monitoring and determining nucleotides and nucleosides in different samples are urgently required. Due to the presence of numerous endogenous interferences in complex matrices and the high polarity of the molecules of the phosphate moiety, the determination of nucleotide content is challenging. This review summarizes the pretreatment and analysis methods of nucleotides in different samples. Advanced pretreatment methods, including different microextraction methods, solid-phase extraction based on novel materials, QuEChERS, are clearly displayed, and continuous progress which has been made in LC, LC-MS/MS and capillary electrophoresis methods are discussed. Moreover, the strengths and weaknesses of different methods are discussed and compared. Highlight:Advanced pretreatment and detection methods of nucleotides were critically reviewed.Microextraction technology was one of the trends of nucleotides pretreatment in the future.Applications of novel materials and supercritical fluid were highlighted.The evolution and advance of HRMS analyzers were in detailed.

ABSTRACT

Carbohydrates in foods and other matrices plays vital roles in their diverse biological functions. Carbohydrates serve not only as functional substances but also as structural materials, such as components of membranes, and participate in cellular recognition. The fact that carbohydrates are indispensable has contributed to the need for pretreatment and analytical methods to be developed for their characterization. The aim of this review is to provide a comprehensive overview of carbohydrate pretreatment and determination methods in various matrices. The pretreatment methods include simple and more developed approaches (e.g., solid phase extraction, supercritical fluid extraction, and different microextraction methods, among others). The analytical methods include those by liquid chromatography (including high-performance anion-exchange chromatography), capillary electrophoresis, gas chromatography and supercritical fluid chromatography, and others. Different pretreatment methods and determination approaches are updated, compared, and discussed. Moreover, we discuss and compare the strengths and weaknesses of different methods and suggest their future prospects.

ABSTRACT

Coumarins are widely used due to their wide range of biological activities, but the long-term or excessive use of coumarin flavors can pose serious health hazards. Therefore, sensitive and specific methods for the quantification of these compounds in different matrices have been developed. In this review, an updated overview of the latest trends in sample preparation techniques and methods used to detect coumarins from March 2012 to April 2019 is provided. This study reviews different analytical methods (such as liquid chromatography coupled with different detectors, electrochemical sensors, capillary electrophoresis, etc.) and different pretreatment methods (such as liquid-liquid extraction, solid-phase extraction, dispersive liquid-liquid microextraction, etc.). Different methods for the pretreatment and determination of coumarins in plant, food, environmental, pharmaceutical and biological samples are summarized, discussed and compared.HighlightsProgress in pretreatment and analytical methods of coumarins are summarized.Fundamentals, instrumentation and applications of purification and quantification are summarized and compared.Optimization of experimental conditions are discussed.Newly emerged eco-friendly methods are introduced.

ABSTRACT

This study was designed to explore the interventional mechanism involving "multi-components, multi-targets and multi-pathways" of Gu-Chang-Zhi-Xie pills (GCZX) for treatment of irritable bowel syndrome (IBS) using pharmacological network technology. Firstly, 96 active ingredients from GCZX pills were screened by ADME parameter filtration and chemical space principal component analysis, and the targets of anti-IBS function were predicted using PharmMapper online database. Secondly, AutoDock Vina was used to validate the docking between the active ingredients and predicted disease targets, and to establish the corresponding relationship between "pharmacodynamic molecules and target proteins". Finally, the target elements were mapped into the KEGG biological pathway by CluoGO plug-in, which further elucidates the potential relationship between the key targets and the mechanism of action of Gu-Chang-Zhi-Xie pills for treatment of IBS. The results showed that most of the top 11 key pharmacodynamic molecules were isoquinoline alkaloids, which mainly acted on inflammatory or pain targets, with different degrees of anti-inflammatory and analgesic effects. A total of 39 key targets were identified, including TPH1, TNF-α, IL-6, IFN-γ, MAO-A and IL-10. These targets were mapped to 29 KEGG pathways, of which the P-value of 5-HT signaling pathway was the smallest. Therefore, the pharmacodynamic molecules mainly act on 6 core targets and may play a major role in the regulation of 5-HT signal synthesis or transport pathway. This study sets an example for drug development and mechanistic investigation using innovative technology.

ABSTRACT

The molecular delivery vectors used in gene therapy need provide the features of safety,stability, efficiency and capacity. The current studies on the structure and action mechanism of pRNA, a packaging RNA of phage?29, showed that pRNA with multiple binding sites can through cell membrane easily and escort exogenous molecules to target cell, without inducing immune reaction. As an ideal nano-scale gene therapy vehicle, pRNA presents a promising application in delivering multiple therapeutic components to detect and treat human diseases.
