ABSTRACT

Ulvan is a complex sulfated polysaccharide extracted from Ulva, and ulvan lyases can degrade ulvan through a ß-elimination mechanism to obtain oligosaccharides. In this study, a new ulvan lyase, EPL15085, which belongs to the polysaccharide lyase (PL) 28 family from Tamlana fucoidanivorans CW2-9, was characterized in detail. The optimal pH and salinity are 9.0 and 0.4 M NaCl, respectively. The Km and Vmax of recombinant EPL15085 toward ulvan are 0.80 mg·mL-1 and 11.22 µmol·min -1 mg-1·mL-1, respectively. Unexpectedly, it is very resistant to high temperatures. After treatment at 100 °C, EPL15085 maintained its ability to degrade ulvan. Molecular dynamics simulation analysis and site-directed mutagenesis analysis indicated that the strong rigidity of the disulfide bond between Cys74-Cys102 in the N-terminus is related to its thermostability. In addition, oligosaccharides with disaccharides and tetrasaccharides were the end products of EPL15085. Based on molecular docking and site-directed mutagenesis analysis, Tyr177 and Leu134 are considered to be the crucial residues for enzyme activity. In conclusion, our study identified a new PL28 family of ulvan lyases, EPL15085, with excellent heat resistance that can expand the database of ulvan lyases and provide the possibility to make full use of ulvan.

ABSTRACT

Metamaterials have become a powerful technique in interdisciplinary research, especially in the field of designing terahertz devices. In this paper, two pairs of different structural units of aluminum-polymer composite metamaterials (APCM) for tunable selectivity are designed. One is designed to achieve high-contrast near-field imaging of linear polarized waves, the other is designed to achieve high-contrast near-field imaging of circularly polarized waves, which means the structural units have very large circular dichroisms. After theoretical design and simulation optimization, it can be found that the contrast of near-field imaging can be effectively controlled by using vanadium oxide (VO2) to fill the open gap of the structure. When the conductivity of VO2 is 200 S/m, both the reflection difference under linear polarization excitation and the reflection difference under the excitation of the circularly polarized wave are at the maximum. The former has a modulation depth of 0.8, and the latter has a modulation depth of 0.55. This work shows excellent tunable selective absorption ability, which will promote the application of metamaterials in terahertz absorber, such as biomedical, non-destructive testing, security inspection, wireless communication and so on.

ABSTRACT

Photothermal materials based on organic small molecules have the characteristics of structural diversity and easy modification for solar-driven water evaporation and power generation technology. However, there still exist limitations, such as the utilization of solar energy and photostability. Therefore, it is the focus of current research to design organic photothermal materials with excellent photothermal stability, strong solar absorption capacity, and high photothermal conversion efficiency. Herein, photothermal conversion materials based on tetrapyridylporphyrin (TPyP) is studied, which possesses polypyrrole macrocyclic framework (18π electrons), which makes it exhibit strong absorption in the 300-800 nm region with high photothermal conversion. The interfacial-heating evaporation system based on polyurethane (PU) foam loaded with TPyP was prepared, whose solar-to-vapor conversion efficiency and vapor evaporation rate of PU + TPyP foam solar energy reached 56% and 0.81 kg m-2 h-1, respectively. In addition, TPyP-loaded solar evaporator equipped with abundant microchannels for water flow are integrated with thermoelectric devices, thus achieving an evaporation rate and voltage as high as 0.69 kg m-2 h-1 and 60 mV under 1 kW m-2 solar irradiation, respectively. The successful application of TPyP in water evaporation and power generation effectively addresses the difficulties faced in the process of using organic small molecule photothermal materials to solve the energy crisis.

ABSTRACT

In response to the dangerous behavior of pedestrians roaming freely on unsupervised train tracks, the real-time detection of pedestrians is urgently required to ensure the safety of trains and people. Aiming to improve the low accuracy of railway pedestrian detection, the high missed-detection rate of target pedestrians, and the poor retention of non-redundant boxes, YOLOv5 is adopted as the baseline to improve the effectiveness of pedestrian detection. First of all, L1 regularization is deployed before the BN layer, and the layers with smaller influence factors are removed through sparse training to achieve the effect of model pruning. In the next moment, the context extraction module is applied to the feature extraction network, and the input features are fully extracted using receptive fields of different sizes. In addition, both the context attention module CxAM and the content attention module CnAM are added to the FPN part to correct the target position deviation in the process of feature extraction so that the accuracy of detection can be improved. What is more, DIoU_NMS is employed to replace NMS as the prediction frame screening algorithm to improve the problem of detection target loss in the case of high target coincidence. Experimental results show that compared with YOLOv5, the AP of our YOLOv5-AC model for pedestrians is 95.14%, the recall is 94.22%, and the counting frame rate is 63.1 FPS. Among them, AP and recall increased by 3.78% and 3.92%, respectively, while the detection speed increased by 57.8%. The experimental results verify that our YOLOv5-AC is an effective and accurate method for pedestrian detection in railways.

ABSTRACT

Background: Endometrial cancer (EC) is a common tumor of the genital tract that affects the female reproductive system but with only limited treatment options. We aimed to discover new prognostic biomarkers for EC. Methods: We used mRNA-seq data to detect differentially expressed genes (DEGs) between EC and control tissues. Detailed clinicopathological information was collected, and changes in the mRNA and protein levels of hub DEGs were analyzed in EC. Copy number variation (CNV) was also evaluated for its association with the pathogenesis of EC. Gene set enrichment analysis (GSEA) was conducted to enrich significant pathways driven by the hub genes. Cox regression analysis was used to select variables to create a nomogram. The nomogram was calibrated by applying the concordance index (C-index), and net benefits of the nomogram at different threshold probabilities were quantified using decision curve analysis (DCA). Results: Differential expression analysis identified 24 DEGs as potential risk factors for EC. Survival analysis revealed that TPX2 expression was related to worsening overall survival in patients with advanced EC. A high CNV was associated with the overexpression of TPX2; this suggested that modifications in the cell-cycle pathway might be crucial in the advancement of EC. Moreover, an individualized nomogram was developed for TPX2 incorporating clinical factors; this was also evaluated for its ability to predict EC. Calibration and DCA analyses confirmed the robustness and clinical usefulness of the nomogram. Conclusion: We offer novel insights into the pathogenesis and molecular mechanisms of EC. The overexpression of TPX2 was related to a poorer prognosis and could serve as a biomarker for predicting prognostic outcomes in EC patients.

ABSTRACT

Long non-coding RNAs are a kind of endogenous ncRNAs with a length of more than 200 bp. Accumulating evidence suggests that long non-coding RNAs function as pivotal regulators in tumorigenesis and progression. However, their biological roles in breast cancer remain largely unknown. Here, we found that IGF2 antisense RNA (IGF2-AS) was significantly decreased in breast cancer tissues, cell lines, and plasma. Patients with low IGF2-AS were more likely to develop larger tumor size and later clinical stage. Overexpression of IGF2-AS evidently inhibited the proliferation and induced apoptosis of MCF-7 and T47D cells in vitro, as well as retarded tumor growth in vivo. Further investigation revealed that IGF2-AS inhibited the expression of its sense-cognate gene IGF2 in an epigenetic DNMT1-dependent manner, resulting in the inactivation of downstream oncogenic PI3K/AKT/mTOR signaling pathway. Enforced expression of IGF2 could significantly block the tumor inhibitory effect of IGF2-AS. Importantly, we found that IGF2-AS could be used as an effective biomarker for breast cancer diagnosis and prognosis. Taken together, our study indicates that IGF2-AS is a tumor suppressor in breast cancer, restoration of IGF2-AS may be a promising treatment for this fatal disease.

ABSTRACT

This paper presents a novel skeleton-based method for deforming meshes (using an approximate skeleton, rather than a precise medial axis). The significant difference from previous skeleton-based methods is that the latter use the skeleton to control movement of vertices whereas we use it to control the simplices defining the model. By doing so, errors that occur near joints in other methods can be spread over the whole mesh, using an optimization process, resulting in smooth transitions near joints of the skeleton. By controlling simplices, our method has the advantage that no vertex weights need to be defined on the bones, which is a tedious requirement in previous skeleton-based methods. Our method can also easily be extended to control deformation by moving a few chosen line segments or vertices embedded in the object, rather than a skeleton.
