ABSTRACT

We have built a Fizeau fiber interferometer to investigate the internal cylindrical defects in an aluminum plate based on laser ultrasonic techniques. The ultrasound is excited in the plate by a Q-switched Nd:YAG laser. When the ultrasonic waves interact with the internal defects, the transmitted amplitudes of longitudinal and shear waves are different. The experimental results show that the difference in transmission amplitudes can be attributed to the high frequency damping of internal cylinders. When the scanning point is close to the internal defect, the longitudinal waves attenuate significantly in the whole defect area, and their amplitude is always smaller than that of shear waves. By comparing the transmitted amplitudes of longitudinal and shear waves at different scanning points, we can achieve a C scan image of the sample to realize the visual inspection of internal defects. Our system exhibits outstanding performance in detecting internal cylinders, which could be used not only in evaluating structure cracks but also in exploring ultrasonic transmission characteristics.

ABSTRACT

Studies on elevational gradients in biodiversity have accumulated in recent decades. However, few studies have compared the elevational patterns of diversity between the different slopes of a single mountain. We investigated the elevational distribution of rodent diversity (alpha and beta diversity) and its underlying mechanisms along the southern and northern slopes of Mt. Taibai, the highest mountain in the Qinling Mountains, China. The species richness of rodents on the two slopes showed distinct distribution patterns, with a monotonically decreasing pattern found along the southern slope and a hump-shaped elevational pattern evident along the northern slope. Multi-model inference suggested that temperature was an important explanatory factor for the richness pattern along the southern slope, and the mid-domain effect (MDE) was important in explaining the richness pattern along the northern slope. The two slopes also greatly differed in the elevational patterns of species turnover, with the southern slope demonstrating a U-shaped curve and the northern slope possessing a roughly hump-shaped pattern. Our results suggest that even within the same mountain, organisms inhabiting different slopes may possess distinct diversity patterns, and the underlying mechanisms may also differ. The potential role of the factors associated with slope aspect in shaping diversity, therefore, cannot be ignored.

ABSTRACT

A hydroponics experiment was aimed at identifying the lead (Pb) tolerance and phytoremediation potential of Moso bamboo (Phyllostachys pubescens) seedlings grown under different Pb treatments. Experimental results indicated that at the highest Pb concentration (400 µmol/L), the growth of bamboo seedlings was inhibited and Pb concentrations in leaves, stems, and roots reached the maximum of 148.8, 482.2, and 4282.8 mg/kg, respectively. Scanning electron microscopy revealed that the excessive Pb caused decreased stomatal opening, formation of abundant inclusions in roots, and just a few inclusions in stems. The ultrastructural analysis using transmission electron microscopy revealed that the addition of excessive Pb caused abnormally shaped chloroplasts, disappearance of endoplasmic reticulum, shrinkage of nucleus and nucleolus, and loss of thylakoid membranes. Although ultrastructural analysis revealed some internal damage, even the plants exposed to 400 µmol/L Pb survived and no visual Pb toxicity symptoms such as necrosis and chlorosis were observed in these plants. Even at the highest Pb treatment, no significant difference was observed for the dry weight of stem compared with controls. It is suggested that use of Moso bamboo as an experimental material provides a new perspective for remediation of heavy metal contaminated soil owing to its high metal tolerance and greater biomass.

ABSTRACT

BACKGROUND: The etiology of Hirschsprung's disease associated enterocolitis (HAEC) is unknown. Previous investigations have suggested that several factors such as dilation of proximal bowel, changes in colonic mucosal defence, and overgrowth of toxigenic bacteria may be related with it. This study was to quantify bifidobacteria and lactobacilli in the feces of Hirschsprung's disease (HD) patients with or without enterocolitis and those of normal children. METHODS: Fresh stool specimens were collected at the first three days of the admission from 30 HD patients (aged 2 weeks to 2 years) and 15 healthy age-matched non-HD patients in the morning once a day for at least three days. All of them have not been given probiotics or antibiotics at least 7 days before stool collection. Hematoxylin-eosin and acetylcholinesterase histochemical staining on rectal biopsies of patients with HD confirmed the diagnosis of HD in all 30 patients. The 30 HD patients were divided into two groups based on the clinical history of enterocolitis: the HAEC group (n=10) and HD group (n=20). Fecal bifidobacteria and lactobacilli were consecutively quantified by SYBR Green I-based real-time PCR assay. Data were analyzed using SAS v. 12.6 for Windows. All tests were two-tailed, and P values <0.05 were considered statistically significant. RESULTS: The mean levels of bifidobacteria were 7.35+/-0.59, 8.16+/-1.17, and 8.35+/-0.74 in the HAEC, HD and control groups, respectively. The bifidobacteria colonization levels were lower in the HAEC group than in the HD and control groups (P<0.05, P<0.001 respectively). The mean level of lactobacilli in the HAEC (5.51+/-0.65) and HD groups (5.87+/-0.78) was significantly lower than that in the control group (6.39+/-0.56) (P<0.05). But there was no difference in log numbers of lactobacilli between HAEC and HD groups (P>0.05). CONCLUSIONS: The scarcity of bifidobacteria and lactobacilli in HAEC patients may result in a decrease in epithelial barrier function and be a predisposing factor in the development of HAEC. This decline suggests that treatment with probiotics or prebiotics may be beneficial in these individuals. Further research will focus on whether probiotics can decrease the incidence of HAEC.
