ABSTRACT

To investigate the effects of fermented Chinese herbal medicine on growth performance, diarrhea rate, nutrient digestibility, and intestinal health of weaned piglets, and to provide the theoretical basis for applying fermented Chinese herbal medicines to weaned piglet production, a total of 162 weaned and castrated piglets at 25 days of age (Duroc × Landrace × Yorkshire, half male and half female) with an initial body weight of 7.77 ± 0.03 kg were randomly divided into the following three groups according to the principle of similar body weight: basal diet (CON) group, basal diet + 3 kg/t fermented Chinese herbal medicine (LFHM) group, and basal diet + 5 g/kg fermented Chinese herbal medicine (HFHM) group. Each group underwent six replicates and there were nine piglets in each replicate. The experiment lasted 24 days, i.e., 3 days for preliminary feeding, and 21 days for the experiment. From Day 1 of the experiment, the piglets were observed and recorded for diarrhea each day. As compared with the CON group, the results indicated: Following the addition of fermented Chinese herbal medicine, the piglets in the LFHM and HFHM groups increased final weight (FW); average daily feed intake (ADFI); average daily gain (ADG) (p < 0.01); apparent digestibility of crude protein (CP) (p < 0.05); as well as chymotrypsin, α-amylase, and lipase activities (p < 0.01). In addition, α-amylase activity in the LFHM group was higher than that in the HFHM group (p < 0.05); chymotrypsin activity in the LFHM group was lower than that in the HFHM group (p < 0.05); as compared with the CON group, the LFHM and the HFHM increased villus height (VH) and crypt depth (CD) in piglet jejunum; isovaleric acid concentration with the HFHM was higher than those with the CON and the LFHM (p < 0.05), but butyrate concentration with the HFFM was lower than those with the CON and the LFHM (p < 0.05). The high-throughput 16S rRNA sequencing of intestinal microbiota results showed that the LFHM and the HFHM affected the microbial α diversity index in weaned piglet colon (p < 0.01). In conclusion, fermented Chinese herbs can improve the growth performance of weaned piglets by promoting the secretion of intestinal digestive enzymes, changing intestinal microbial diversity, regulating the contents of intestinal short chain fatty acids (SCFAs), promoting intestinal health, and improving nutrients digestibility.

ABSTRACT

Heat stress (HS) has been revealed to damage the antioxidant system and intestinal barrier function, which greatly threatens poultry production. The present study investigated the effects of dietary ellagic acid (EA) on the antioxidant system, gut barrier function, and gut microbiota of heat-stressed broilers. Arbor Acres 14-day-old broilers numbering 360 were randomly divided into six groups, including one negative control group (NC) and five experimental groups. The broilers in the NC group were supplemented with a basal diet at a normal temperature (23 ± 2 °C). The broilers in the experimental groups were supplemented with basal diets containing EA at different doses (0, 75, 150, 300, and 600 mg/kg) at HS temperature (35 ± 2 °C). The experiment lasted for 4 weeks. Results showed that dietary EA reduced the corticosterone (CORT), LPS, and diamine oxidase (DAO) levels in the serum of heat-stressed broilers. Additionally, dietary EA improved the antioxidant enzyme activity and mRNA levels of Nrf2/HO-1 in the ileum of heat-stressed broilers. The relative abundances of Streptococcus, Ruminococcus_torques, Rothia, Neisseria, Actinomyces, and Lautropia in the cecum were significantly reduced by the EA supplementation in a dose-dependent manner. Notably, the LPS, DAO, and MDA in the serum were revealed to be positively correlated with the relative abundances of Rothia, Neisseria, Actinomyces, and Lautropia, while the GSH-px, SOD, and CAT levels in the serum were negatively correlated with the relative abundances of Ruminococcus_torques, Rothia, Neisseria, Actinomyces, Streptococcus, and Lautropia. Taken together, dietary EA improved the antioxidant capacity, intestinal barrier function, and alleviated heat-stressed injuries probably via regulating gut microbiota.

In this study, a Mg-9Gd-4Y-2Zn-0.5Zr (wt.%) alloy was subjected, after solution treatment, to hot compression deformation at different temperatures (350 °C, 400 °C and 450 °C) and different strain rates (0.001 s-1, 0.01 s-1, 0.1 s-1 and 0.5 s-1) on a Gleeble-3800 thermal simulator. The evolution of the stress-strain curves under different conditions was compared. The changes in microstructure caused by the different deformation parameters and the change law of the long-period stacking-ordered (LPSO) phase during compression were observed and analyzed by optical microscope (OM) and scanning electron microscope (SEM). The results show that with the increase in the deformation temperature and the decrease in the strain rate, the degree of dynamic recrystallization (DRX) gradually increased, and the morphology of the phase also changed through, for example, twist fracture. The continuous dynamic recrystallization (CDRX) and discontinuous dynamic recrystallization (DDRX) mechanisms activated during the thermal deformation process can effectively refine the grains and weaken the texture in the alloy.

ABSTRACT

This study was conducted to evaluate the effects of tea powder in diets on the growth performance, meat quality, muscular amino acid, fatty acid profile, and serum biochemical indices of pigs. A total of 120 local Chinese pigs (Tibetan × Bama miniature pigs) were randomly assigned to five treatment groups, each with six pens and four pigs per pen. During a 60-day experiment, these groups of pigs were fed the normal diet and the diet supplemented with 1%, 2%, 4%, and 6% tea powder, respectively. The results showed that the supplementation of tea powder did not affect the growth performance of pigs. However, the dietary tea powder inclusion decreased (p < 0.05) the average fat thickness, total fat, and abdominal fat, and increased (p < 0.05) the total muscle as well. In addition, the dietary 2% tea powder decreased (p < 0.05) the muscle lightness (L*) and yellowness (b*). Compared with the control group, the dietary supplementation with 1%, 2%, and 4% tea powder raised (p < 0.05) the total amino acids (TAA) and essential amino acids (EAA), and dietary 4% and 6% tea powder increased (p < 0.05) the C20:5n3 in the muscle tissue. Furthermore, the serum lipid metabolism-related biochemical indices and mRNA expression levels were improved with the addition of tea powder. These results indicated that dietary tea powder might improve the carcass traits and meat quality of the Chinese native finishing pigs, but it does not affect their growth performance. Tea powder could be fully developed and reasonably applied as a dietary supplement.

ABSTRACT

Guanidinoacetic acid is the direct precursor of creatine and its phosphorylated derivative phosphocreatine in the body. It is a safe nutritional supplement that can be used to promote muscle growth and development. Improving the growth performance of livestock and poultry and meat quality is the eternal goal of the animal husbandry, and it is also the common demand of today's society and consumers. A large number of experimental studies have shown that guanidinoacetic acid could improve the growth performance of animals, promote muscle development and improve the health of animals. However, the mechanism of how it affects muscle development needs to be further elucidated. This article discusses the physical and chemical properties of guanidinoacetic acid and its synthesis pathway, explores its mechanism of how it promotes muscle development and growth, and also classifies and summarizes the impact of its application in animal husbandry, providing a scientific basis for this application. In addition, this article also proposes future directions for the development of this substance.

ABSTRACT

The objective of the study is to evaluate and compare the effects of betaine or glycine on carcass trait, meat quality and lipid metabolism of finishing Huan Jiang mini-pigs. Betaine called trimethylglycine is a methyl derivative of glycine, but few researches were conducted to compare the impact of dietary betaine and glycine on pigs. One hundred and forty-four Huan Jiang mini-pigs (body weight = 10.55 ± 0.15 kg; 70 d) were randomly divided to 3 treatment groups (basal diet, glycine or betaine). Results indicated that dietary betaine increased the average daily gain (ADG) and final weight (P < 0.05). Dietary glycine or betaine markedly reduced average backfat thickness (P < 0.05) and heightened lean percentage (P < 0.01) compared to the control group. Moreover, in comparison with the control group, betaine significantly improved the redness (a∗) and tenderness (shear force) of the longissimus dorsi (LD) muscle (P < 0.05), whereas glycine only raised the value of a∗ of the LD muscle (P < 0.05). These results showed that diet supplemented with 0.25% betaine and equimolar amounts of glycine could regulate cascass trait and meat quality of finishing Huan Jiang mini-pigs, and the effect of betaine was superior to that of glycine.

ABSTRACT

This work is to study the microstructure and texture evolution of AZ31 alloy prepared by cyclic expansion extrusion with an asymmetrical extrusion cavity (CEE-AEC) at different deformation temperatures. The result shows AZ31 alloy undergoes continuous dynamic recrystallization (CDRX) and discontinuous dynamic recrystallization (DDRX) during CEE-AEC processing. At the initial stage of deformation, AZ31 alloys exhibit similar bimodal microstructure of coarse deformed grains surrounded by fine DRXed grains. As the passes increase, the cumulative strain increases, and the coarse grains of all samples are almost replaced by fine equiaxed grains. The average grain sizes and the basal texture intensities of the deformed samples increase as the deformation temperature increases. In addition, due to the existence of an asymmetrical cavity, as the passes increase, the basal textures of all samples are deflected with maximum intensities increase, and even an unusual bimodal texture is formed, resulting in a soft orientation that is easy to basal slip.

Here, we investigated the roles and mechanisms of flavonoids from mulberry leaves (FML) on lipid metabolism in high fat diet (HFD)-fed mice. ICR mice were fed either a control diet (Con) or HFD with or without FML (240 mg/kg/day) by oral gavage for six weeks. FML administration improved lipid accumulation, alleviated liver steatosis and the whitening of brown adipose tissue, and improved gut microbiota composition in HFD-fed mice. Microbiota transplantation from FML-treated mice alleviated HFD-induced lipid metabolic disorders. Moreover, FML administration restored the production of acetic acid in HFD-fed mice. Correlation analysis identified a significant correlation between the relative abundances of Bacteroidetes and the production of acetic acid, and between the production of acetic acid and the weight of selected adipose tissues. Overall, our results demonstrated that in HFD-fed mice, the lipid metabolism improvement induced by FML administration might be mediated by gut microbiota, especially Bacteroidetes-triggered acetic acid production.

ABSTRACT

Tea trees have a long history of cultivation and utilization. People in many countries have the habit of drinking tea and choosing green tea, oolong tea, or black tea according to different regions and personal tastes. Tea polyphenols are a general term for polyphenol compounds in tea, and has been shown to have good effects on antioxidant, anti-inflammatory, cancer prevention and regulation of lipid metabolism. Tea polyphenols have been widely used as antioxidants in disease treatment and animal husbandry, but their specific mechanism of action needs to be further clarified and revealed. This review focuses on the definition, classification, antioxidant activity and the regulation of signaling pathways of tea polyphenols. This paper also aims to examine the application of tea polyphenols in human and animal health, providing a scientific basis for this application in addition to proposing future directions for the development of this resource.

ABSTRACT

OBJECTIVES: The aim of this study was to explore the effects of ß-hydroxy-ß-methylbutyrate (HMB) on intestinal function of lipopolysaccharide (LPS)-challenged piglets. METHODS: Forty weaned piglets were used in a 2 × 2 factorial design. The major factors were challenge (saline or LPS) and diet (basal diet or 0.6% HMB-Ca diet). After 15 d of treatment with LPS or HMB, blood and intestine samples were obtained. RESULTS: The results showed that in LPS-injected pigs, HMB supplementation significantly increased jejunal villus height and ileal villus height-to-crypt depth ratio and decreased ileal crypt depth (P < 0.05). HMB also improved intestinal function indicated by elevated activities of intestinal mucosal disaccharidase and tricarboxylic acid cycle key enzymes. Furthermore, HMB significantly downregulated mRNA expression of Sirt1 in jejunum and mRNA expression of AMPKα1 and Sirt1 in ileum (P < 0.05), with a concurrent decrease of AMPKα phosphorylation in jejunum and ileum. Microbiota analysis indicated that HMB supplementation significantly increased α-diversity and affected relative abundances of Romboutsia and Sarcina at the genus level, accompanied by increased concentrations of all short-chain fatty acids except propionate in the terminate ileum of LPS-injected piglets. CONCLUSION: Dietary HMB supplementation could improve intestinal integrity, function, microbiota communities, and short-chain fatty acid concentrations in LPS-challenged piglets, suggesting its potential usage as a feed additive in weaned piglets to alleviate intestinal dysfunction triggered by immune stress.

ABSTRACT

This study was conducted to investigate the effects of dietary amylose/amylopectin (AM/AP) ratio and amylase on growth performance, apparent digestibility of energy and starch, serum biochemical index, and digestive enzymes. The experiment used a 4 × 3 factor design, and 960 one-day-old Arbor Acres (AA) broilers were randomly divided into 12 groups fed diets containing different AM/AP ratio of 0.11, 0.23, 0.35 and 0.47 and combined with 0, 3,000 and 6,000 U/kg amylase. Results showed that 0.23-0.35 AM/AP ratio increased growth performance, while dietary addition of 6,000 U/kg amylase significantly reduced average daily weight gain in broilers. The energy digestibility was significantly reduced along with the increase of dietary AM/AP ratio and in the 6,000 U/Kg amylase-supplemented groups. The digestibility of starch also decreased significantly with the increase of dietary AM/AP ratio, but high dose (6,000 U/Kg) of amylase increased. High AM/AP diet reduced serum insulin concentration, which was increased in amylase-supplemented groups. Furthermore, exogenous amylase increased amylase activity in the jejunal chyme. In conclusion, dietary 0.23-0.35 AM/AP ratio was suggested to maintain a higher growth performance in broilers and high AM/AP ratio diets reduced energy and starch digestibility and serum insulin concentration, which was reversed by dietary amylase.

ABSTRACT

The aim of this study was to investigate the effects of dietary ß-hydroxy-ß-methylbutyrate (HMB) on lipopolysaccharide (LPS)-induced muscle atrophy and to investigate the mechanisms involved. Sixty pigs (21 ± 2 days old, 5.86 ± 0.18 kg body weight) were used in a 2 × 3 factorial design and the main factors included diet (0, 0.60%, or 1.20% HMB) and immunological challenge (LPS or saline). After 15 d of treatment with LPS and/or HMB, growth performance, blood parameters, and muscle protein degradation rate were measured. The results showed that in LPS-injected pigs, 0.60% HMB supplementation increased the average daily gain and average daily feed intake and decreased the feed : gain ratio (P < 0.05), with a concurrent increase of lean percentage. Moreover, 0.60% HMB supplementation decreased the serum concentrations of blood urea nitrogen, IL-1ß, and TNF-α and the rate of protein degradation as well as cell apoptosis in selected muscles (P < 0.05). In addition, dietary HMB supplementation (0.60%) regulated the expression of genes involved in mitochondrial biogenesis and increased the phosphorylation of Akt and Forkhead Box O3a (FoxO3a) in selected muscles, accompanied by decreased protein expression of muscle RING finger 1 and muscle atrophy F-box. These results indicate that HMB may exert protective effects against LPS-induced muscle atrophy by normalizing the Akt/FoxO3a axis that regulates ubiquitin proteolysis and by improving mitochondrial biogenesis.

ABSTRACT

The effects of repetitive upsetting extrusion under different starting temperatures on the microstructure and texture of GWZK124 alloy were investigated. The results clearly showed that the particles and second phases induced dynamic recrystallization (DRX), which can be explained by the particle-stimulated nucleation (PSN) mechanism. It was shown that grain refinement during repetitive upsetting extrusion (RUE) is dominated by a complicated combination of continuous dynamic recrystallization and discontinuous dynamic recrystallization. The RUEed alloys under different starting temperatures exhibited a bimodal microstructure comprising fine DRXed grains with weak texture and coarse deformed grains with strong texture. The DRXed grains could weaken the texture. As the RUE starting temperature decreased, the average grain size increased and the volume fraction of DRXed grains decreased.

Mg-Gd-Y-Zn-Zr Mg alloys show excellent performance in high-end manufacturing due to its strength, hardness and corrosion resistance. However, the hot deformation and dynamic recrystallization (DRX) behaviors of Mg-13.5Gd-3.2Y-2.3Zn-0.5Zr were not studied. For this article, hot compression behavior of homogenized high rare-earth (RE) content Mg-13.5Gd-3.2Y-2.3Zn-0.5Zr (wt%) alloy was investigated by using the Gleeble-3500D thermo-simulation test machine under the temperature of 350⁻500 °C and the strain rate of 0.001⁻1 s-1. It was found that the high flow stress corresponded to the low temperature and high strain rate, which showed DRX steady state curve during the hot compression. The hot deformation average activation was 263.17 kJ/mol, which was obtained by the analysis of the hyperbolic constitutive equation and the Zener-Hollomon parameter. From observation of the microstructure, it was found that kink deformation of long period stacking ordered (LPSO) phase was one of the important coordination mechanisms of hot deformation at low temperature. The processing map with the strain of 0.5 was established under the basis of dynamic material model (DMM); it described two high power dissipation domains: one appearing in the temperature range of 370⁻440 °C and the strain rate range of 0.001⁻0.006 s-1, the other appearing in the temperature range of 465⁻500 °C and strain rate range of 0.001⁻0.05 s-1, in which dynamic recrystallization (DRX) mainly ocurred. The highest degree of DRX was 18% from the observation of the metallographic.

Repetitive upsetting extrusion (RUE) was applied to the as-homogenized Mg-12.0Gd-4.5Y-2Zn-0.4Zr (wt %) alloy at 773 K. The microstructure evolution of the alloy during RUE was investigated. The results indicated that almost all Mg5(Gd,Y,Zn) phases and fine-lamellar long-period stacking-ordered (LPSO) phases were dissolved into the matrix after homogenization treatment at 793 K for 16 h. After one RUE pass, dynamic recrystallization (DRX) occurred. During subsequent RUE passes (from one to three passes), average volume fractions of DRXed grains increased from 43.9% to 65.8%, and that of fine-lamellar and block-shaped LPSO phases gradually decreased. All samples exhibited a typical bimodal microstructure consisting of some initial grains containing fine-lamellar LPSO phases, but consisting mostly of fine-DRXed grains with a mean grain size of 6 µm. Because of an increase in the accumulated strains, the coarse grains were substituted with fine-DRXed grains, the grains were gradually refined, and the microstructure distribution became more homogeneous.
