ABSTRACT

Objective:To explore the molecular mechanism of Wendan Decoction in the treatment of Hypertension by the network pharmacolog. Methods:The potential active components and targets of Wendan Decoction were screened in TCMSP and TCMIP database. The TTD, genecards and OMIM databases were used to collect hypertension targets. Then the protein interaction was analyzed by using String database. The software of Cytoscape v3.7.1 was used to construct the network diagram of Wendan Decoction in the treatment of Hypertension and using the software of ClueGO to analyze GeneOntology and KEGG pathway of the potential targets. Finally, we used molecular docking technology for the appropriate of molecules and targets. Results:A total of 141 potential active components and 108 potential therapeutic targets were selected from Wendan Decoction. And 20 of core targets selected by MCC in CytoHubba. The GeneOntology analysis contained 164 GO terms, 133 of biological processes, 24 of molecular functions, and 7 of cell components. Seven key pathways were selected by enrichment analysis of KEGG pathway. Molecular docking results showed good docking between key components and targets. Conclusion:Wendan Decoction can treat hypertension through multiple ways and targets. This study predicts the possible mechanism of Wendan Decoction and provides data for its clinical application.

ABSTRACT

BACKGROUND: Osteoarthritis (OA) pertains to a chronic disease of degenerative joints distinguished by articular cartilage destruction, subchondral bone remodeling, osteophyte formation, and inflammatory changes. Chondrocyte apoptosis is inextricably linked to cartilage degeneration. SRY-related high-mobility-group-box 9 (SOX9) is a well-acknowledged transcription factor in the chondrogenesis. Nevertheless, the detailed function of miR-138-5p/SOX9 in OA remains to be fully clarified. MATERIALS AND METHODS: qRT-PCR was performed to measure the expressions of miR-138-5p and SOX9 mRNA in OA and normal cartilage tissues and cells. Human chondrocyte cell lines, CHON-001 and ATDC5, were treated with different doses of interleukin-1ß (IL-1ß) to simulate the inflammatory response environment of OA. miR-138-5p mimics, miR-138-5p inhibitors, and SOX9 small interfering RNA (siRNA) were constructed and transfected into CHON-001 and ATDC5 cells. CCK-8 was conducted to determine the cell viability and transwell assay was used to monitor the migration of cells. Western blot was carried out to detect the expressions of apoptosis- related factors. Enzyme-linked immunosorbent assay (ELISA) was adopted to measure the contents of inflammatory factors. TargetScan predicted SOX9 was a target gene of miR-138-5p, which was then verified by luciferase assay. RESULTS: miR-138-5p expression was down-regulated in OA and regulated SOX9 expression. The downregulation of miR-138-5p facilitated the proliferation and migration of CHON-001 and ATDC5 cells, while impeded their apoptosis and inflammatory response. Besides, down-regulated SOX9 can counteract the promoting effect of down-regulated miR-138-5p on the proliferation and migration of chondrocytes. CONCLUSION: miR-138-5p can arrest the proliferation and migration of CHON-001 and ATDC5 via restraining SOX9, and facilitate the apoptosis and inflammation. This study revealed the protective effect of down-regulated miR-138-5p on the inflammatory injury of chondrocytes caused by IL-1ß.

ABSTRACT

Objective:To observe the effects of bama nectar in kidney-yang deficienct mice and on the mating ability and sperm counts of normal mice. Methods:The weight, body temperature, locomotor activity, cold-resistant,genital organ coefficient and lipid peroxidation of the hydrocortisone-induced kidney-yang deficient mice were detected. The genital organ ccoefficient, immune organ co-efficient and erectile function of the ovariectomized mice were detected. The mating ability and the sperm counts of normal mice were measured. Results:Compared with the model group, bama nectar obviously increased the weight, body temperature and locomotor ac-tivity of the hydrocortisone-induced kidney-yang deficient mice, and improved the locomotor activity, the organ coefficient of seminal vesicle, prostate gland, preputial gland and epididymis. Furthermore, bama nectar significantly increased the SOD activity in serum and reduced the level of MDA in serum when compared with the model group (P<0.05 or P<0.01). Compared with the model group, bama nectar could increase the organ coefficient of foreskin and thymus gland in the ovariectomized mice, and shorten the erec-tile latency (P<0. 05). Compared with the normal group, bama nectar improved the mating ability and the sperm counts of the male mice (P<0. 05 or P<0. 01). Conclusion:Bama nectar has beneficial effects on invigorating kidney and strengthening yang.

ABSTRACT

Objective To develop user-friendly information education-communication (IEC)materials and guidelines of malaria control for various target groups. Methods The participatory methods were used to make sure participation of target groups at each stages of IEC material development. Results A package consisted of interactive video, 2 posters for primary groups, sto ry booklet for students, manual for activities of "buddy system" for teachers, flipcharts for health staff at grass-root levels, guidelines on communication skill and how to use the first 5kinds of materials, was developed. Conclusion A set of user-friendly IEC materials and guidelines of malaria control has successfully been developed with sufficient participation of target groups.

ABSTRACT

Objectives To learn the bednet use and influential factors among Wa and Lahu ethnics, and further to explore feasible ways of promoting the use of bednets and insecticide-treated nets (ITNs). Methods Questionnaire surveys and statistical analysis with EpiInfo 2000 software package were carried out. Results Only did 2.8% [95% confidence intervals(95%CI):1.2%-5.4%] of Wa and 4.8 % (95%CI:2.8%-8.1%) of Lahu ethnic members use bednets frequently or sometimes. Bednets were not available for 67.7% (95% CI:62.0%-73.1%) of Wa and 59.7% (95% CI:53.1%- 66.1%) of Lahu ethnic peoples. Most people who had bednets did not often use them for anti-mosquitoes. Results of multivariate logistic regression confirmed significant correlation between bednet use and knowledge of malaria prevention. Conclusion The coverage and use rates are very low in the two ethnical minorities. Health education, communication and social marketing are the strategies for promoting the usage of bednet and ITNs.
