ABSTRACT

Six previously unreported solanidane steroidal alkaloids, namely lyrasolanosides A-F, were isolated from Solanum lyratum. In addition, five known steroidal alkaloids were also identified. The structures of these compounds were determined through the use of NMR, HRESIMS,UV, IR and ECD analysis. To assess their bioactivities, the cytotoxic effects of the six previously unreported compounds were evaluated on A549 cells. The results revealed that lyrasolanoside B (2) exhibited the highest potency among them. Lyrasolanoside B (2) exhibited significant inhibition of cell migration, invasion, and adhesion dramatically. Mechanistically, it was found to suppress the activity of JAK2/STAT3 signaling pathway by downregulating the expression of phosphorylated JAK2/STAT3 in an exosome-dependent manner. In addition, lyrasolanoside B (2) was found to significantly upregulate the expression of E-cadherin and downregulate the expression of N-cadherin and vimentin. These findings indicate that lyrasolanoside B (2) inhibits the metastasis of A549 cells by suppressing exosome-mediated EMT. These findings suggest that lyrasolanoside B (2) may inhibit the metastasis of lung cancer by regulating A549-derived exosomes.

ABSTRACT

Background: The growing population of middle-aged and older empty nesters is characterized by poorer health, and social participation (SP) has been shown to improve this situation. However, few studies have investigated specific performance and gender differences between SP and health. The present study aims to address these issues. Methods: A total of 1207 middle-aged and older empty nesters over 45 years old were selected from the China Health and Retirement Longitudinal Study (CHARLS, 2011-2018). Random-effects analyses were used to explore the association between changes in SP (diversity, frequency, type) and changes in health status. Health status include physical health, mental health, self-reported health (SRH). Results: Female middle-aged and olderly empty nesters have significantly poorer health and participate in SP more frequently. The higher the diversity of SP, the better the health of middle-aged and olderly empty nesters, while higher frequency is beneficial to SRH. Female's participation in sports and Internet had better mental health and SRH, and mahjong helped female's mental health. Clubs are helpful for male's SRH. Conclusions: This study reveals the specifics of the association between SP and health status of middle-aged and older empty nesters. Therefore, all aspects of SP and gender differences should be taken into account when predicting and improving the health status. Help the government to better formulate policies to better cope with the increasing empty nest phenomenon and build a harmonious and stable society.

ABSTRACT

Both oxygen vacancies and surface hydroxyls play a crucial role in catalysis. Yet, their relationship is not often explored. Herein, we prepare two series of TiO2 (rutile and P25) with increasing oxygen deficiency and Ti3+ concentration by pulsed laser defect engineering in liquid (PUDEL), and selectively quantify the acidic and basic surface OH by fluoride substitution. As indicated by EPR spectroscopy, the laser-generated Ti3+ exist near the surface of rutile, but appear to be deeper in the bulk for P25. Fluoride substitution shows that extra acidic bridging OH are selectively created on rutile, while the surface OH density remains constant for P25. These observations suggest near-surface Ti3+ are highly related to surface bridging OH, presumably the former increasing the electron density of the bridging oxygen to form more of the latter. We anticipate that fluoride substitution will enable better characterization of surface OH and its correlation with defects in metal oxides.

ABSTRACT

Two previously undescribed steroidal alkaloids, compounds 1-2, along with two known ones(3-4), were isolated from the 80% ethanol extract of ripe berries of Solanum nigrum by chromatographic methods, including silica gel, ODS, and HPLC. Based on spectroscopic and chemical evidence, including IR, NMR, and HR-ESI-MS data, the structures of the isolated compounds were identified as 12ß,27-dihydroxy solasodine-3-O-ß-D-glucopyranoside(1), 27-hydroxy solasodine-3-O-ß-D-glucopyranosyl-(1→4)-α-L-rhamnopyranosyl-(1→2)-[α-L-rhamnopyranosyl-(1→4)]-ß-D-glucopyranoside(2), solalyraine A(3), and 12ß,27-dihydroxy solasodine(4). Compounds 1-2 were tested for their potential effects against the proliferation of A549 cells, which revealed that compounds 1-2 had weak cytotoxic activity.

ABSTRACT

Eight undescribed, along with five known steroidal alkaloids were isolated from Solanum nigrum L., a plant used in traditional Chinese medicine. Their structures were elucidated by NMR, HR-ESI-MS, and IR spectroscopy. Two compounds displayed an unusual structure in steroidal alkaloids with an open E-ring and without an F-ring present. To evaluate their bioactivities, nine compounds were selected to intervene five human cancer cell lines including H1299, HepG2, HeLa, HCT116, and MCF7 respectively. All compounds exhibited inhibitory effects for the five cell lines, revealing potential anti-tumor activities from Solanum nigrum.

ABSTRACT

Fatal road traffic crashes are often related to multifarious risk factors, among which driving under the influence of drugs (DUID) has been reported as a significantly contributing cause. The first worry about the side-effect of influencing driving drugs is central nervous system adverse reaction, and anti-hypersensitivity drugs are a class of drugs with such side effects meanwhile has been widely used for common allergic diseases thus posing a great challenge to road safety and demanding a rapid and efficient method to detect. In this work, a method based on magnetic graphene oxide dispersive solid phase extraction (MGO-D-SPE) combined with ion mobility spectrometry (IMS) was firstly introduced for simultaneous determination of ephedrine, pseudoephedrine, diphenhydramine, promethazine and terfenadine in saliva and urine matrices. The prepared MGO was characterized by Fourier transform infrared (FT-IR) spectroscopy and thermo gravimetric analysis (TGA). Various parameters affecting extraction efficiency as well as instrumental acquisition sensitivity were studied and optimized. Under the optimum experimental conditions, the method was fully validated and the results demonstrated that the proposed method exhibited some advantages, including a good linearity covering large concentration ranges of 51.0-3040 ng ml-1 for five anti-hypersensitivity drugs, and good accuracy was also obtained with high precision (CV% < 5.0 %). LODs and LOQs were 10.2-50.4 ng·mL-1 and 30.6-101.3 ng·mL-1, respectively. Consequently, the MGO-D-SPE-IMS methodology succeeded in building a hitherto unexplored tool for quantifying anti-hypersensitivity drugs in saliva and urine matrices of interest in DUID research field.

ABSTRACT

In this study, seven previously undescribed steroidal glycoalkaloids, compounds 1-7, were isolated from Solanum lyratum, along with two known ones (8 and 9). Comprehensive spectroscopy techniques were used to determine their structures. Although 1-8 only showed a weak inhibitory effect on the proliferation of the tumor-derived vascular endothelial cells, however, in a former study we found both total steroidal glycoalkaloids from Solanum lyratum (TSGS) and 9 significantly inhibited tumor angiogenesis and its mechanism was linked to its ability to interfere with cell membrane lipid rafts. Lipid rafts are closely related to the functions of tumor-derived exosomes, a vital factor in cancer progression. Thus, we investigated the impacts of TSGS and 9 on the functions of A549-derived exosomes. Our results indicated that A549-derived exosomes can significantly enhance the angiogenesis abilities of human umbilical vein endothelial cells, whereas the intervention of TSGS or 9 significantly inhibited this activity of A549-derived exosomes. These findings suggest that TSGS and 9 exert anti-tumor angiogenesis by inhibiting the pro-angiogenic activity of A549-derived exosomes.

Potential genotoxic impurities (PGIs) are a series of compounds that could potentially damage DNA. Therefore, a sensitive method is needed for detection and quantification. The present work described and validated a method for the quantification of one PGI (namely 1,4-benzodioxane-6-aldehyde) in Eliglustat tartrate (EGT) active pharmaceutical ingredient (API) substances using dispersive liquid-liquid microextraction (DLLME) as sample preparation to remove matrix effect and detected by HPLC-UV. Parameters influencing the microextraction efficiency were systematically investigated. The combined application of DLLME and HPLC-UV provided the sensitivity of the method. The achieved limit of detection (LOD) and the limit of quantification (LOQ) were adequate for the specific purpose and found to be 1.29 µg g-1 and 2.58 µg g-1, respectively. This simple and effective methodology offers a key advantage in the ease of removing matrix effect and improves sensitivity obviously. In addition, no costly instrumentation and skilled personnel are needed when using this method, which is available and can be successfully implemented in routine factory drug quality control analysis.
