ABSTRACT

Objective:To explore the potential Hub genes, key miRNAs, biological processes and related signaling pathways in the pathogenesis of osteoarthritis (OA), and provide bioinformatics basis for the pathogenesis and treatment of OA.Methods:The expression profiling chip of OA synovial tissue sample from Gene Expression Omnibus (GEO) were downloaded, differentially expressed genes (DEGs) were identified, and functional enrichment analysis was performed. A protein-protein interaction network (PPI) was constructed. STRING and Cytoscape was used for module analysis, and the Hub gene was further identified, and further miRNAs mining of the Hub gene was carried out.Results:Finally, 9 Hub genes (SOCS3, BTRC, FBXO32, KLHL22, UBE3A, HUWE1, UBR4, ANAPC5, TRIM50) and 2 key miRNAs (hsa-miR-103a-3P, hsa-miR-107) related to the progression of OA were identified .They might be potential biomarkers for the pathogenesis of OA. We also found that signal transduction, the transcriptional positive regulation of RNA polymerase Ⅱ promoter, and protein serine/threoninase activity had a certain correlation with the pathogenesis of OA. In addition, our analysis results showed that cAMP signaling pathway and Rap1 signaling pathway were also involved in the progression of OA.Conclusion:The potential biological molecules, biological processes and related pathways identified in this study may guide us for the further research on the etiology and treatment of OA.

ABSTRACT

OBJECTIVE:To provide reference f or the qualit y sta ndard establishment of Amaranthus retroflexus. METHODS : Taking 7 batches of A. retroflexus medicinal materials as the research object ,the appearance properties of the medicinal materials were investigated ,and the microscopic characteristics of the medicinal powders were observed. TLC method was adopted to qualitatively identify rutin ,valine and leucine in A. retroflexus medicinal materials. According to the relevant methods of the 2015 edition of Chinese Pharmacopoeia (part Ⅳ),water content ,total ash content ,acid-insoluble ash content and water-soluble extract content were determined. HPLC method was used to determine the content of rutin in the medicinal material of A. retroflexus . The determination was performed on Agilent 5 TC-C18(2)column with mobile phase consisted of methanol- 0.3% phosphoric acid solution(40∶60,V/V),at the flow rate of 1.0 mL/min. The detection wavelength was set at 358 nm,and the column temperature was 30 ℃. The sample size was 10 μL. RESULTS:The appearance and microstructure characteristics of the medicinal materials were consistent with the existing description. The identification results of TLC meth od showed that 7 batches of medicinal materials and each reference substance (rutin,valine,leucine)showed spots of the same color at the same position. The moisture content of 7 batches of A. retroflexus medicinal materials was 7.43%-8.72%,the total ash content was 11.82%-13.78%,the acid-insoluble ash content was 0.15%-0.55%,and the water-soluble extract content was 17.27%-24.74%. The linear range of rutin was 10-200 μg/mL(R 2=1.000 0). RSDs of precision test ,stability test (24 h)and repeatability test were all less than 2.0% (n=6). The average recovery rates of rutin were 99.14%,97.98% and 98.80% in low ,medium and high concentration of samples,and RSDs were 0.97%,0.95%,0.96%(n=3). The contents of rutin in 7 batches of A. retrophylla were 0.314-1.102 mg/g. CONCLUSIONS:In this study ,character observation ,microscopic identification ,moisture content ,total ash content ,acid- insoluble ash content and water-soluble extract content of A. retroflexus are investigated ;TLC method was established for qualitative identification of leucine ,valine and rutin in A. retroflexus ,and the HPLC method was established for content determination of rutin. It provides reference for the quality standard establishment of A. retroflexus .

ABSTRACT

0bjective To explore the effect of brain health education on therapeutic adherence of hypertensive patients with subjective memory complaints based on health belief model. Methods A randomized controlled trial was designed in this study. eligible patients were selected from a community health service center in Chongqing were randomly assigned to intervention group (56 cases) and control group (60 cases). The control group received routine community health services and the intervention group received brain health education on the basis of the control group. statistical analysis was performed on the patients' dementia risk control belief, therapeutic adherence, and blood pressure control rate at baseline, after the group intervention, and after the individualized telephone intervention. Results There was a significant group× time interaction(F 交互=12.353, 22.376, P<0.05) on dementia risk control belief and therapeutic adherence (P<0.05),. The dementia risk control belief score, the therapeutic adherence and the blood pressure control rate in the intervention group was (98.74±13.62), (119.26±4.93) points and 70.0% , the control group was (85.00 ± 12.80), (106.48 ± 10.33) points and 50.0% after the intervention; the intervention group was higher than the control group (t=5.194, t=7.899, χ2=4.167, P<0.05). The subjects were rated at 4 points and above for each item of satisfaction with the project after the intervention. Conclusions Brain health education based on health belief model in hypertensive patients with subjective memory complaints, can effectively improve the dementia risk control belief and therapeutic adherence, and promote the self-management of blood pressure control.

ABSTRACT

Objective@#To explore the level of the subjective memory complaints and its influencing factors among the elderly people in the community.@*Methods@#The Memory Satisfaction of the Muti-factorial Memory Questionnaire was to evaluate the subjective memory complaints of the community elders, the objective memory function and general cognitive function were evaluated by the Auditory Verbal Learning Test (AVLT) and Mini-mental State Examination (MMSE) respectively, the depression was evaluate by the Geriatric Depression Scale (GDS-1), and the general population was collected by the general information questionnaire.@*Results@#The incidence of subjective memory complaints in the community was 76%, multiple regression analysis showed that GDS score (β=-0.522, P<0.001) , gender (β=0.137,P<0.05) and AVLT delayed recall score (β=0.136,P<0.05) were the main complaints of subjective memory in the elderly influencing factors.@*Conclusion@#Depression and objective memory are the main influencing factors of elderly subjective memory complaints, suggesting that when developing a primary preventive strategy, consideration should be given to cognitive screening of elderly people who with subjective memory complains and inclusion of prevention and management as a risk population for dementia, with a focus on providing health services and family visits.

ABSTRACT

Objective To study the memory disorder perception characteristics of subjective memo-ry complaints(SMCs)and to analyze the influencing factors. Methods 302 subjects conforming to the in-clusion criteria were selected.All the subjects completed the illness perception questionnaire-memory of Chi-nese version(IPQ-M). Results (1)There was no statistical difference between the score of social compari-son(2.95±0.78)and the middle value 3.The differences between the score of the remaining dimensions of memory impairment perception(timeline acute/chronic(3.57 ± 0.89),consequences(2.24 ± 0.77),timeline stability/decline(3.71±0.88),personal control(blame)(3.61±0.92),personal control(helplessness)(3.31± 0.73),emotional representation(2.29 ± 0.70),treatment control(2.78 ± 0.67),illness coherence(3.14 ± 0.64))and the middle value 3 were statistically significant(all P<0.05).(2)Gender,education level,living condition,marital status,economic condition,family history of dementia and knowledge education of dementia were the influencing factors of memory perception of SMCs. Conclusion The memory complaining people are less aware of the related knowledge of memory decline,and there are more errors and negative percep-tions.The factors that affect the perception of memory disorders are complex.

ABSTRACT

Objective To investigate the characteristics of everyday memory of adults with subjective memory complaints (SMC),and probe into the correlation between everyday memory function and life quality.Methods Through convenience sampling,150 adults with SMC were face-to-face interviewed by an investigator,with demographic characteristics collected,and the cognition function,everyday memory function and life quality were also assessed during the interview.Results Among the 150 adults with SMC,the impairment rate of everyday memory function reached 96%,and the median score of RBMT-II was 16.00 (interquartile range 6.00),indicating a low level of everyday memory function,with the Name andDelayed recall Story items having lower scores.All participants were divided into two groups according to cognitive status,with 67 (44.70%) adults having normal cognitive function and 83 (55.30%) impaired cognitive function.After comparison,significant differences were found in the total score of RBMT-II and scores ofFaces recognition andDelayed recall Story between the two groups (P<0.05).In all dimensions of SF-36,the median score of general health dimension for the 150 adults was 55.00 (inter-quartile range 31.25).The Spearman rank correlation analysis showed that the scores of several dimensions in RBMT-II were positively correlated with the scores of several dimensions in SF-36 (P<0.05).Conclusion Everyday memory function of people with SMC was positively CorTelated with their life quality.

ABSTRACT

Objective To introduce the illness perception questionnaire-memory (IPQ-M) and translate it into Chinese and to evaluate its reliability and validity.Methods The Chinese version of IPQ-M was translated from the original,back translated and adjusted for culture adaptation.A total of 152 community-dwelling people with subjective memory complaints (SMCs),who were aged 50 years or above,were investigated with the Chinese version of IPQ-M.40 participants among them were re-evaluated 2 weeks later.Results Exploratory factor analysis got 9 factors which could explain 70.757% of the total variance.The factor loading of each item on the corresponding factors was higher than 0.4.The correlation between Chinese version of IPQ-M and GDS were-0.208 to 0.763 (P<0.01).The Cronbach's α coefficient was 0.872,the testretest reliability was 0.894,and the Spearman-Brown split coefficient was 0.890.Conclusion The Chinese version of IPQ-M has good reliability and validity,and it can be used in Chinese elderly people with SMCs.

ABSTRACT

Objective To evaluate the effect of dexmedetomidine on lipid peroxidation during lung ischemia-reperfusion (I/R) in rats.Methods Fifty-four pathogen-free healthy male Wistar rats,weighing 250-350 g,were randomly divided into 3 groups (n=18 each) using a random number table:sham operation group (S group),I/R group,and dexmedetomidine group (Dex group).In group Dex,dexmedetomidine 5 μg/kg was injected via the caudal vein once a day for 2 consecutive days.The equal volume of normal saline was given in S and I/R groups.At 30 min after administration on 2nd day,lung I/R was produced by clamping the left hilum of lung for 45 min followed by 120 min of reperfusion in I/R and Dex groups.At 45 min of ischemia,and 60 and 120 min of reperfusion,6 rats selected from each group were sacrificed,and the left lungs were removed for examination of the pathological changes and for determination of wet/dry lung weight ratio (W/D ratio),malondialdehyde (MDA) content and superoxide dismutase (SOD) activity.Results Compared with S group,the SOD activity was significantly decreased,and the MDA content and W/D ratio were significantly increased at 60 and 120 min of reperfusion in I/R and Dex groups (P<0.05).Compared with I/R group,the SOD activity was significantly increased,and the MDA content and W/D ratio were significantly decreased at 60 and 120 min of reperfusion in Dex group (P<0.05).The pathological changes of lungs were significantly attenuated in Dex group as compared with I/R group.Conclusion Dexmedetomidine mitigates lung I/R injury through inhibiting lipid peroxidation in rats.

ABSTRACT

Objective To evaluate the effect of lappaconitine on renal ischemia-reperfusion (I/R) injury in mice.Methods Thirty-six male Wistar rats,weighing 180-220 g,were randomly divided into 3 groups (n=12 each) using a random number table:sham operation group (group S),group I/R and lappaconitine group (group LA).Renal I/R was induced by occlusion of the left renal pedicle for 45 min with atraumatic microclips followed by reperfusion,and the right kidney was removed after atraumatic microclips were released.At 30 min before reperfusion,lappaconitine 4 mg/kg was injected intraperitoneally in group LA,and normal saline 2 ml was given in S and I/R groups.In group S,the left renal pedicle was only isolated.At 5 and 24 h of reperfusion,blood samples were taken from the inferior vena cava for determination of serum creatinine (Cr) and blood urea nitrogen (BUN) concentrations,and kidney specimens were obtained for histopathologic examination (with light microscope) and for determination of the expression of cyclooxygenase-2 (COX-2) and matrix metalloproteinase-2 (MMP-2) in renal tissues (by immunohistochemistry).Results Compared with group S,the serum Cr and BUN concentrations were significantly increased,and the expression of COX-2 and MMP-2 in renal tissues was up-regulated at 5 and 24 h of reperfusion in I/R and LA groups.Compared with group I/R,the serum Cr and BUN concentrations were significantly decreased,the expression of COX-2 and MMP-2 in renal tissues was down-regulated at 5 and 24 h of reperfusion and histopathologic changes were reduced in group LA.Conclusion Lappaconitine can attenuate renal I/R injury through inhibiting the expression of COX-2 and MMP-2 in rats.

ABSTRACT

ObjectiveTo explore the effect of guided learning in the eye optical applied optics teaching.Methods30 students of Grade 2013 were chosen as experimental group which was given guided learning teaching, and other 30 students of Grade 2013 were chosen as control group which was given used traditional teaching. In the guided learning teaching, self-study and practice students were re-quired to master the key and found the difficulties in the course firstly, answering questions after the group discussion. Then teachers focused on parsing the teaching materials. By the repeated communication and practice students mastered knowledge finally. The control group used the traditional teaching method which was based on the teacher's lecture. Through comparing the theory examination scores, practical as-sessment grades and questionnaire of the students self-assessment, teaching effect based on guided learn-ing was evaluated. Normal distribution measurement data was showed by standard deviation, T test was used in two independent sample measurement data; The count data was showed by rate, using Chi-squared test whichα=0.05.ResultsIn experiment group,the theoretical examination average score was(78.35±7.14) points, while in control group, the average score was(69.15±8.88)points. There were significance differ-ences between the two groups(t=-4.339,P=0.000). The experimental group practical assessment grades and the self-assessment score were superior to control group, and the difference has statistical sig-nificance (P<0.05).ConclusionGuided learning teaching can guide the students to obtain and master relevant knowledge independently, fully arouse their initiative of learning and improve their ability to analyze and solve problems, which can help reach a good teaching effect.

ABSTRACT

Objective To observe the effects of curcumin on hepatic oxidant stress in water arsenic-exposed rats and to study its mechanism,which can offer references for curcumin used in antioxidant therapy of arsenic poisoning.Methods Thirty-two SD rats were divided into 4 groups according to body weight by random number table,half male and half female.Including control group (lavaged 135 days with deionized water),arsenic poisoning group (lavaged 45 days with deionized water after lavaging 90 days with 10 mg/kg sodium arsenite),pure curcumin group (lavaged 135 days with 1 000 mg/kg curcumin solution) and curcumin treatment group (lavaged 45 days with 1 000 mg/kg curcumin solution after lavaging 90 days with 10 mg/kg sodium arsenite),8 rats in each group.The arsenic contents of urine (urine creatinine corrected) and liver were detected by hydride generation inductively coupled plasma optical emission spectrometer (HG-ICP-OES);the activity of Cu/Zn-superoxide dismutase (SOD1) and catalase (CAT),the contents of malondialdehyde (MDA) in serum and liver homogenate by colorimetric method;the protein expression of liver antioxidant enzyme (SOD 1 and CAT) was assayed by Western blotting.Results The arsenic contents of urine and liver in arsenic poisoning group [(5.83 ± 0.29)μg/g Cr,(15.76 ± 1.65)μg/g] and the arsenic contents of urine in curcumin treatment group [(1.07 ± 0.14)μg/g Cr] were obviously higher than those of control group [(0.40 ± 0.14)μg/g Cr,(4.56 ± 1.05)μg/g,all P < 0.05];compared to arsenic poisoning group,the arsenic contents of urine and liver in curcumin treatment group [(1.07 ± 0.14)μg/g Cr,(5.42 ± 1.76)μg/g] were obviously lower (all P < 0.05).The contents of serum and liver SOD1,CAT and MDA in control group respectively were (102.46 ± 5.03),(29.33 ± 8.13)U/ml,(3.11 ± 0.49)μ mol/L and (204.05 ± 18.33),(126.26 ± 13.19)U/mg prot,(1.62 ± 0.42) μmol/g prot.Compared to the control,the activity of serum and liver SOD1 and CAT in arsenic poisoning group [(60.97 ± 7.94),(13.56 ± 5.14)U/ml and (133.66 ± 11.51),(74.01 ± 13.30)U/mg prot] were lower,the contents of MDA [(7.26 ± 0.54)μmol/L and (2.61 ± 0.52)μmol/g prot] were higher (all P < 0.05).Compared to arsenic poisoning group,the activity of serum and liver SOD1 and CAT in curcumin treatment group [(87.39 ± 9.38),(20.45 ± 6.49) U/ml and (178.27 ± 9.32),(93.70 ± 20.35)U/mg prot] were higher,the contents of MDA [(4.34 ± 0.79)μmol/L and (1.92 ± 0.18)μmol/g prot] were lower (all P < 0.05).The protein expressions of SOD1 and CAT in control group respectively were 0.64 ± 0.32 and 0.72 ± 0.31.Compared to the control group,the protein expressions of SOD1 and CAT in pure curcumin group (1.03 ± 0.23,1.02 ± 0.20) were significantly higher (all P < 0.05) and in arsenic poisoning group (0.34 ± 0.12,0.39 ± 0.11) were lower (all P < 0.05);Compared with the arsenic poisoning group,the protein expressions of SOD1 and CAT in curcumin treatment group (0.58 ± 0.09,0.68 ± 0.29) were significantly higher (all P < 0.05).The arsenic content of urine in rats were positively related with arsenic content of liver and the content of MDA [correlation coefficient (r) =0.952,0.732,all P < 0.05],but negativity related with the activity of SOD1 and CAT in liver (r =-0.874,-0.679,all P < 0.05);the activity of SOD1 and CAT and the content of MDA in serum and liver were positively related (r =0.796,0.484,0.607,all P < 0.05),the activity and protein expression of SOD1 and CAT in liver were positively related (r =0.748,0.424,all P < 0.05).Conclusion The curcumin may improve the activity of hepatic antioxidant enzyme in water arsenic-exposed rats and effectively decrease lipid poroxidation damage caused by arsenic via promoting the excretion of arsenic and the protein expression of hepatic antioxidant enzyme.

ABSTRACT

Objective To evaluate the effect of sinomenine on apoptosis in renal tubular epithelial cells of rats subjected to renal ischemia-reperfusion (I/R), and the relationship with C-Jun N-terminal kinase (JNK) signaling pathway.Methods Fifty-four male Wistar rats, aged 6-8 weeks, weighing 180-220 g, were randomly divided into 3 groups (n =18 each) using a random number table: sham operation group (group S), I/R group and sinomenine group (group SIN).Renal ischemia was induced by occlusion of the left renal pedicle for 45 min followed by reperfusion, and the right kidney was removed immediately after onset of reperfusion in anesthetized rats in I/R and SIN groups.In group SIN, sinomenine 60 mg/kg was injected intraperitoneally at 30 min before reperfusion, while the equal volume of normal saline was given instead of sinomenine at the same time point in S and I/R groups.Six animals in each group were selected at 0.5, 6 and 24 h of reperfusion, blood samples were collected by cardiac puncture for determination of serum creatinine (Cr) and blood urea nitrogen (BUN) concentrations.Immediately after blood sampling, the left kidney was removed for examination of pathological changes in renal tissues (with light microscopes) and for determination of phosphorylated JNK (p-JNK) and caspase-3 expression (by immune-histochemistry) and apoptosis in renal tubular epithelial cells (by TUNEL).The apoptotic rate was calculated.Results Compared with group S, the serum Cr and BUN concentrations were significantly increased, the expression of p-JNK and caspase-3 was up-regulated, and the apoptotic rate was increased in I/R and SIN groups.Compared with group I/R, the serum Cr and BUN concentrations were significantly decreased, the expression of p-JNK and caspase-3 was down-regulated, and the apoptotic rate was decreased in group SIN.The microscopic examination showed that the pathological changes of kidney were significantly attenuated in group SIN compared with group I/R.Conclusion The mechanism by which sinomenine attenuates renal I/R injury is related to inhibited activation of p-JNK signaling pathway and reduced apoptosis in renal tubular epithelial cells of rats.

ABSTRACT

Objective To investigate the effect of sinomenine on the levels of nuclear factor kappa B (NF-κB) and inducible nitric oxide synthase (iNOS) during renal ischemia-reperfusion (I/R) in rats.Methods Seventy-two healthy male Wistar rats, aged 6-9 weeks, weighing 180-220 g, were randomly assigned into 4 groups (n =18 each) using a random number table: control group (group C), sinomenine group (group SIN), group I/R, and sinomenine+I/R group (group SIN+I/R).The animals were anesthetized with 10% chloral hydrate 350 mg/kg.The left renal pedicles were clamped with atraumatic microclips for 45 min followed by reperfusion, and the right kidney was removed immediately after onset of reperfusion to establish the model of renal I/R injury.Sinomenine 60 mg/kg was injected intraperitoneally at 30 min before reperfusion in group SIN +I/R, and at the corresponding time point in group SIN.At 6, 8 and 12 h of reperfusion, the blood samples were drawn by cardiac puncture for measurement of serum creatinine (Cr) and blood urea nitrogen (BUN) concentrations.The left renal specimens were obtained for examination of pathological changes (with light microscopes) and for determination of the rate of NF-κB-positive cells and iNOS expression in renal tissues (by immunohistochemistry).Results Compared with group C, the concentrations of serum Cr and BUN, and rate of NF-κB-positive cells were significantly increased, and the expression of iNOS was up-regulated in I/R and SIN+I/R groups, and no significant change was found in the parameters mentioned above in group SIN.Compared with group I/R, the concentrations of serum Cr and BUN, and rate of NF-κB-positive cells were significantly decreased, and the expression of iNOS was down-regulated in group SIN+I/R.The microscopic examination showed that the pathological changes of kidney were significantly attenuated in group SIN+I/R compared with group I/R.Conclusion The mechanism by which sinomenine attenuates renal I/R injury is related to inhibited activity of NF-κB and down-regulated expression of iNOS in rats.

Objective To evaluate the effects of sinomenine on the expression of hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) during renal ischemia-reperfusion (I/ R) in rats.Methods Fifty-four male Wistar rats, weighing 180-220 g, were randomly divided into 3 groups (n=18 each) using a random number table: sham operation group (group S) , group I/R and sinomenine group (group SIN).Renal I/R was induced by clamping the left renal pedicle for 45 min followed by reperfusion in I/R and SIN groups.In group S, the bilateral renal pedicels were only exposed.Sinomenine 60 mg/kg was intraperitoneally injected at 30 min before reperfusion in group SIN, while the equal volume of normal saline was given in group S and group I/R.At 6, 12 and 24 h (T1-3) of reperfusion, 6 rats from each group were chosen, and blood samples were drawn from the hearts for determination of the serum creatinine (Cr) and blood urea nitrogen (BUN) concentrations.The animals were then sacrificed, and the left kidney was removed and embedded into a paraffin block for determination of the expression of HIF-1α and VEGF in the renal tissues (by immuno-histochemistry).Results Compared with group S, the serum Cr and BUN concentrations were significantly increased at T1-3 , and the expression of HIF-1α was significantly up-regulated at T2,3 in group I/R and group SIN, and the expression of VEGF was significantly upregulated at T2,3in group I/R, and at T1-3 in group SIN.Compared with group I/R, the serum Cr concentration at T1-3 and serum BUN concentration at T2,3 were significantly decreased and the expression of HIF-1α at T2,3and VEGF at T1-3was significantly up-regulated in group SIN.Conclusion The mechanism by which sinomenine attenuates renal I/R injury is related to up-regulation of the expression of HIF-1α and VEGF in the renal tissues of rats.

ABSTRACT

Objective To investigate the effect of continued nursing education manual on surgical nursing education.Methods A continued nursing education manual was designed and applied to surgical nursing education.The effects were assessed by comparing the nursing training results before and after intervention by the manual in terms of nursing theory test,nursing techniques test.Results After the application of manual,the training coverage rate increased from 94.59%to 100.00%(P<0.05),the pass rate of theory test increased from 87.57%to 94.21%(P<0.05),and the pass rate of nursing skills test increased from 90.27%to 96.14%(P<0.05). Conclusion The continued nursing education manual is effective in improving the educational effect of surgical nursing.

ABSTRACT

Objective To explore the cognitive status of community-dwelling older adults with subjective memory complaints (SMC).Methods A total of 262 older adults with SMC were included based on convenience sampling.Face-to-face interview was conducted for demographic information collection and cognitive assessment.Statistical analyses Were undertaken using SPSS version 19.0.Results The participants with SMC had a low level of general cognitive function,with a mean score of 22.18±4.21 on the MoCA; the two cognitive domains of abstraction' and deIayed memory had the lowest scores,with an average score of 0.85±0.82 and 2.91± 1.50 respectively,and the average scoring rate lower than 60.00%.About 54.20% of the older adults with SMC presented with cognitive impairment.The scores ofabstraction were low in the participants with and without cognitive impairment.In addition,the scores of visuospatial/executive function, language and delayed memory were low in the participants with cognitive impairment.The cognitive impairment of persons with SMC may be associated with age and marital status.The people over the age of 70 years and widowed ones are more likely to suffer from cognitive dysfunction.Conclusion The older adults with SMC have a low level of general cognitive function,and cognitive imPairment detection rate is higher.Advanced age and widowhood are important factors affecting their cognitive function.

ABSTRACT

Objective To investigate the genotype and antifungal susceptibility of AspergiIlus species isolated from clinical settings and environmental sources. Methods Random amplification of polymorphic DNA (RAPD) analysis was performed to profile the genotypes of 48 Aspergillus fumigatus strains, 59 Aspergillus flavus strains and 30 Aspergillus niger strains isolated from clinical settings and environmental sources. NCCLS M38-A protocol was carried out for antifungal susceptibility testing of these Aspergillus isolates. Results As RAPD analysis showed, the 48 Aspergillus fumigatus strains were classified into 8 genotypes, 59 Aspergillus flavus strains into 12 genotypes, and 30 Aspergillus niger strains into 5 genotypes. A significant difference was observed in the susceptibility to terbinafine among the 8 genotypes of Aspergillus fumigatus (χ2 = 33.092, P <0.01 ) as well as in that to amphotericin B among the 5 genotypes of Aspergillus niger (χ2 = 15.185, P< 0.05).No statistical difference was found in the susceptibility to amphoteriein B, itraconazole, fluconazole or flucytosine among the 8 genotypes of Aspergillus fumigatus or in that to terbinafine, itraconazole, fluconazole or flucytosine among the 5 genotypes of Aspergillus niger. Conclusion There is some difference in the susceptibility to some antifungal agents between different genotypes of Aspergillus fumigatus and Aspergillus niger.

ABSTRACT

Objective To observe the changes of SOD and MDA after the rabbits were exposed to acute hypoxia when cured by oxygen therapy.Methods New Zealand rabbits underwent hypoxia imitating the 4500-meter sea level in altitude chamber(group E,n=6),then some received oxygen therapy,the volume ratio of oxygen-nitrogen mixture was 25%,30% and 35% respectively for 10,5 and 3 hours as group B,C and D.Another 5 rabbits received no treatment as normal group.The rabbits samples were collected at the time points of 1,3,5,10 and 24 hours after the oxygen therapy started to detect serum biochemical indicators.Results Comparing with normal control group,serum SOD activity in simple hypoxia group noticeably decreased and the content of MDA noticeably increased;in three oxygen therapy groups,serum SOD activity in 25% oxygen therapy group was highest and the content of MDA was lowest.Serum SOD activity in 25% oxygen therapy group had ascendant tendency and the content of MDA had descendant tendency.Conclusions Low concentration oxygen therapy can prevent acute hypoxia-induced injury in rabbits,besides increase oxygen partial pressure,oxygen therapy may improve superoxide dismutase activity.

ABSTRACT

OBJECTIVE To supervise the environmental fungal load and species distribution in transplantation department and intensive care unit of the Southwest Hospital,and to analyze the relationship with season,temperature,humidity,ventilation and personnel activities.METHODS Data from Dec 2005 to Jan 2006 were collected from liver transplantation department(LTD),cerebral surgery intensive care unit(CSICU) and central intensive care unit(CICU).Air,surfaces and tap water were sampled twice a month at each department.RESULTS The air fungal load was 123.63 CFU/m3,139.90 CFU/m3,7 CFU/m3 and 217.71 CFU/m3 at LTD,CSICU,CICU and outdoor,respectively.The five most prevalent fungi collected from air and surface were Penicillium spp,Cladosporium spp,Alternaria spp,Aspergillus spp and Saccharomyces spp in turn.The five most prevalent fungi collected from water were Saccharomyces spp,Candida spp,Aspergillus spp,Penicillium spp and Rhodotorula spp in turn.The fungal load in LTD was positively correlated with the average temperature and the average humidity;the fungal load in CSICU was correlated with the average temperature and the average humidity,but the correlation between air fungal load and personnel activities wasn′t observed.CONCLUSIONS It demonstrated the fungi are found in the environment of the hospital including air,surface and water.The air fungal load varies throughout the year.The crest-time is May to June and September to October.Air fungal load is lower in winter and higher in summer and autumn.The correlation between air fungal load and temperature and humidity is observed.

ABSTRACT

The study was to investigate thrombolysis in vivo with ultrasound, and to discuss effects of thrombolysis with ultrasound on the structure of erythrocyte and its safety threshold, under different ultrasound intensity and exposure time. The structure of erythrocyte in thrombus was evaluated under light microscope. The relationship between the structure of erythrocyte in thrombus and ultrasound intensity and exposure time was obtained. The results showed that ultrasound eliminated the thrombus. According to the change of the structure of erythrocyte in thrombus and ultrasound intensity and exposure time, the effects of thrombolysis with ultrasound could be divided into three kinds of areas: the A, B, C area. The area A was the safe area, the area B was the relatively safe area, and the area C was the irreversible damage area. The study suggested that ultrasound intensity and exposure time had significant impact on the structure of erythrocyte. Too much ultrasound intensity or too long exposure time could cause erythrocytes irreversible damaged. It could accelerate thrombolysis and shorten the exposure time that the ultrasound intensity was little bit increased. This study of effects of thrombolysis with ultrasound on the structure of erythrocyte and its safety threshold were important for practical applications.
