ABSTRACT

In this study, we evaluated the efficacy, expressed as a mean weight decrease of the whole echinococcal cyst mass, of novel benzimidazole salt formulations in a murine Echinococcus granulosus infection model. BALB/c mice were intraperitoneally infected with protoscoleces of E. granulosus (genotype G1). At 9 months post-infection, treatment with albendazole (ABZ), ricobendazole (RBZ) salt formulations, and RBZ enantiomer salts (R)-(+)-RBZ-Na and (S)-(-)-RBZ-Na formulations were initiated. Drugs were orally applied by gavage at 10 mg kg-1 body weight per day during 30 days. Experimental treatments with benzimidazole sodium salts resulted in a significant reduction of the weight of cysts compared to conventional ABZ treatment, except for the (S)-(-)-RBZ-Na enantiomer formulation. Scanning electron microscopy and histological inspection revealed that treatments impacted not only the structural integrity of the parasite tissue in the germinal layer, but also induced alterations in the laminated layer. Overall, these results demonstrate the improved efficacy of benzimidazole salt formulations compared to conventional ABZ treatment in experimental murine cystic echinococcosis.

ABSTRACT

OBJECTIVES: Nesfatin-1 is an antiiflammatory, antiapoptotic, and anorexigenic peptide that has many roles in cardiomyocyte metabolism and viability. Inflammation plays an important role in the pathogenesis of atherosclerosis. In this study, we aimed to evaluate the alterations in serum nesfatin-1 levels of the patients undergoing coronary artery bypass operation due to atherosclerotic coronary artery disease. MATERIALS AND METHODS: The study included 49 patients (30 men, 19 women) undergoing coronary artery bypass surgery. Serum nesfatin-1 levels were measured from venous blood samples of the patients collected before and three months after the operation. The relationship of nesfatin-1 levels with accompanying conditions was also analyzed. RESULTS: Nesfatin-1 levels at third month, postoperatively, were significantly higher than preoperative nesfatin-1 levels of the patients (41.94±13.90 pg/ml and 27.06±8.01pg/ml, respectively; p<0.001). Both preoperative and postoperative nesfatin-1 levels were negatively correlated with age (p<0.001). The postoperative increase in nesfatin-1 levels was significantly higher in diabetic patients than in nondiabetic patients (p<0.001). CONCLUSION: This study revealed that serum nesfatin-1 levels increased significantly in patients undergoing coronary artery bypass operation. Nesfatin-1 level may have a role in assessing myocardial perfusion during the follow-up of these patients (Tab. 4, Fig. 4, Ref. 25).

ABSTRACT

The objective of this study was to compare some husbandry procedures on the base of physiological stress parameters and evaluate the welfare status in sheep. Forty ewes were used as the study material. Measurements were taken during several routine husbandry procedures such as milking, shearing, weighing, loading and hoof care. Data regarding time spent for each application, as well as heart and respiratory rates were recorded during the applications. Blood samples were taken 15 min before and after each application and malondialdehyde (MDA), glutathione-peroxidase (GSH-Px), cortisol T(3) and T(4) parameters were measured. In addition, changes in the same parameters between pre- and post-application periods were evaluated. According to the results, machine milking caused less stress than hand milking. No significant difference was seen between shearing methods for hand shearer or clipper; however, both applications caused stress in animals. The results for weighing methods of animals demonstrated significant differences in cortisol, T(3) and T(4) values in favor of traditional method. Cortisol, T(3) and T(4) levels were significantly higher in manual loading compared with loading by ramp. Regarding hoof care, all the examined parameters differed in favor of modern method. On the other hand, significant differences were determined between the stress parameters regarding pre- and post-applications. All values differed for hand milking while no significant difference was observed in MDA and T(3) values in machine milking group. Parameters in weighing groups changed significantly. For loading process, GSH, cortisol, T(3) and T(4) values differed in both treatment groups. With regard to hoof care, parameters except T(4) in laying group differed significantly. An increase occurred in minute-based measurements of heart and respiratory rates parallel to physiological data. The number of the respiratory rates during the applications differed except for the shearing process. All the parameters displayed significant differences between groups in terms of heart rates. Time spent for each application also differed between groups. Time saved for milking, shearing, weighing, loading and hoof care was 3.23 min, 4.37 min, 1.71 min, 7.85 s and 1.55 min, respectively. These results appear to provide a tangible advantage of using new husbandry methods to the breeders. It was concluded that using new methods in sheep husbandry procedures provided advantages in terms of saving time and reducing labor, as well as improved conditions for welfare of animals. In addition, it facilitated the routine works and flock husbandry.

ABSTRACT

The aim of this study was to measure fat thickness, area and depth of the longissimus dorsi muscle using ultrasonography, to estimate carcass composition in live Akkaraman lambs. Fat thickness, area and depth of the longissimus dorsi muscle between the 12th and 13th ribs were measured in vivo and on the carcass after slaughter, using real time ultrasound in 40 Akkaraman lambs. To estimate the carcass composition, one-half of a carcass was dissected into muscle, fat and bone after slaughter. Overall, correlation coefficients between ultrasound and carcass longissimus dorsi muscle area, depth and fat thickness were 0.82, 0.60 and 0.77, respectively. Estimates of carcass composition for Akkaraman lambs based on LW explained 78%, 82%, 74%, 52%, 75%, 36% and 72% of the variations for muscle, total carcass fat, subcutaneous fat, inter-muscular fat, non-carcass fat, tail fat and bone, respectively. The introduction of UFT, ULMA and ULMD as independent variables in addition to LW in the multiple linear regression equations further improved the variations for total muscle (80%), carcass fat (84%) and bone weight (76%) whereas no improvement was observed for subcutaneous, intermuscular, non-carcass and tail fat. The results showed that in vivo ultrasound fat thickness and measurement of area and depth of the longissimus dorsi muscle in association with live weight could be used to estimate muscle, total body fat and bone weight in Akkaraman lambs.

ABSTRACT

The bicuspid aortic valve is a common congenital cardiac anomaly, having an incidence in the general population of 0.9% to 2.0% and a frequency of 54% in all patients aged >15 years with valvular aortic stenosis. In most cases it remains undetected until infective endocarditis or calcification supervenes. The bicuspid aortic valve may function normally throughout life, may develop progressive calcification and stenosis or may develop regurgitation with or without infection. The association of the bicuspid aortic valve with dissection of the aorta is also common. The recognition of the bicuspid valve in patients with aortic valve disease remains an important challenge to the clinician, whereas preoperative knowledge of valve morphology would be helpful in planning the surgery. Antibiotic prophylaxis is also recommended in such patients, since these valves are likely to become the most important intrinsic cardiac predisposition for infective endocarditis with the virtual disappearance of rheumatic fever in developed countries.

ABSTRACT

The fluid that resides within cranial and spinal cavities, cerebrospinal fluid (CSF), moves in a pulsatile fashion to and from the cranial cavity. This motion can be measured hy magnetic resonance imaging (MRI) and may he of clinical importance in the diagnosis of several brain and spinal cord disorders such as hydrocephalus, Chiari malformation, and syringomyelia. In the present work, a geometric and hydrodynamic characterization of an anatomically relevant spinal canal model is presented. We found that inertial effects dominate the flow field under normal physiological flow rates. Along the length of the spinal canal, hydraulic diameter was found to vary significantly from 5 to 15 mm. The instantaneous Reynolds number at peak flow rate ranged from 150 to 450, and the Womersle number ranged from 5 to 17. Pulsatile flow calculations are presented for an idealized geometric representation of the spinal cavity. A linearized Navier-Stokes model of the pulsatile CSF flow was constructed based on MRI flow rate measurements taken on a healthy volunteer. The numerical model was employed to investigate effects of cross-sectional geometry and spinal cord motion on unsteady velocity, shear stress, and pressure gradientfields. The velocity field was shown to be blunt, due to the inertial character of the flow, with velocity peaks located near the boundaries of the spinal canal rather than at the midpoint between boundaries. The pressure gradient waveform was found to be almost exclusively dependent on the flow waveform and cross-sectional area. Characterization of the CSF dynamics in normal and diseased states may be important in understanding the pathophysiology of CSF related disorders. Flow models coupled with MRI flow measurements mnay become a noninvasive tool to explain the abnormal dynamics of CSF in related brain disorders as well as to determine concentration and local distribution of drugs delivered into the CSF space.
