ABSTRACT

Objective: To investigate the effects of the active ingredients combined therapy on inflammatory factors interleukin 1 beta (IL-1β) and neuropeptide Y (NPY) based onpharmacodynamics in rats. Methods: The animal model was built by transient middle cerebral artery occlusion (MCAO). The method for evaluating the concentrations of the FA-Pr-Al components in rat plasma was established by using HPLC and the expression levels of IL-1βand NPY were determined by ELISA. A new mathematics method of the trend of percentage rate of change (PRC) was used to assess the correlation between pharmacokinetics (PK) and pharmacodynamics (PD). Results: FA-Pr-Al in combination reduced neurological deficits, decreased infarct volume and inhibited the expression levels of IL-1βand NPY (all P<0.05)compared with the model group. FA, Pr and Al all displayed two compartment open models in rats. Clockwise hysteresis loops were obtained by time-concentration-effect curves. IL-1βand NPY level changes in the plasma followed an opposite trend to the plasma concentration tendency after Cmax was reached. Astragaloside’s PRC value was significantly higher than those of FA and puerarin between 120 to 180 min. Conclusions: The pharmacokinetics of FA-Pr-Al in combination were closely related its pharmacodynamics in treating ischemia/reperfusion injury, and the components of FA-Pr-Al may have a synergistic pharmacological effect. Astragaloside may play a more pronounced role in regulating IL-1毬and NPY levels comparedwith puerarin or FA.

ABSTRACT

This study was undertaken to determine the effect of total flavones from Elsholtzia blanda (Benth.) Benth. (TFEB), a traditional Chinese medicine, on myocardial ischemia induced by coronary occlusion in Beagle dogs. Infarct size of left ventricle, serum activity of creatine kinase-MB (CK-MB) and malondialdehyde (MDA), hemorrheologic variables and homodynamic parameters including mean arterial pressure (MAP), coronary blood flow (CBF), coronary vascular resistance (CVR), end-diastolic pressure of left ventricle (LVEDP), rate of rise and decline of left ventricular pressure (+/-dp/dtmax) were measured in this study. Administration with TFEB produced a dose-dependent reduction in infarct size. TFEB 100 mg/kg exerted notable inhibition in the elevation of serum CK-MB and MDA activity. TFEB significantly reduced MAP and CVR. The decrease in CBF also tended to be smaller in treated dogs. TFEB improved the recovery of myocardial function by depressing the degree of reduction in +/-dp/dtmax and LVEDP. TFEB also showed a capacity to resist ischemic damage through lowering blood viscosity. The results indicated that TFEB kept heart from ischemic damage due to coronary occlusion in Beagle dogs.

ABSTRACT

Objective To investigate the regulatory effects of icariin (ICA) combined with the Panax notoginseng saponins (PNS) on immunological function in mice and provide some experimental evidences for the combination mechanism improving the spatial learning and memory abilities of Alzheimer's disease (AD) animal model. Methods Based on serum pharmacological method, the ICR mice were individually ig administrated with ICA+PNS [(40+320), (80+640), and (160+ 1 280 ) mg/kg] or ICA (80 mg/kg), and PNS (640 mg/kg) only for 7 d. Drug-containing serum was prepared and effects on spleen lymphocyte proliferation of Bable/c mice induced by concanavalin-A (ConA) or lipopolysaccharide (LPS) and on interleukin-2 (IL-2) secretion were observed in vitro. Meanwhile, the immunological organ indexes of treated mice were evaluated. Results Drug-containing serum of ICA+PNS [(80+640) and (160+ 1 280 ) mg/kg] could improve the spleen lymphocyte proliferation induced by ConA (P0.05). Drug-containing serum of ICA+PNS [(80+640) and (160+ 1 280) mg/kg] could improve IL-2 production (P0.05), respectively. Conclusion ICA combined with PNS could improve immunological function selectively and promote T cell function in mice.
