ABSTRACT

Objective • To investigate the role of Rho-associated coiled coil protein kinase 1 (ROCK1) in amyloid β-protein (Aβ) induced damage of rat hippocampal neurons. Methods • The rat primary neurons were treated with Aβ40 oligopeptides to establish a neurotoxicity model. Western blotting was used to detect the protein expression of ROCK1. Its activity was detected by the kit. Confocal laser scanning was used to observe the calcium signal in neurons, and apoptosis of neurons was detected by TUNEL assay. Y-27632, an inhibitor of ROCK1, was added into the culture medium in order to observe its effect on Aβ40. Results • Aβ40 (10 μmol/L) could significantly induce calcium overload, increase ROCK1 expression and activity, and promote apoptosis in primary neurons. Furthermore, ROCK1 inhibitor could decrease all the effect induced by Aβ40. Conclusion • ROCK1 is involved in both Aβ- induced neuronal calcium overload and neurotoxicity, and ROCK1 inhibitor can antagonize the toxic effects of Aβ.

ABSTRACT

AIM To investigate the effects of Tongmai Decoction on PLT,PAgR,D-D and TEG in elderly patients with intertrochanteric fracture in perioperative period.METHODS One hundred and fifteen recruited patient subjects were randomly divided into two groups.Fifty-seven cases in the control group were treated with Low Molecular Weight Heparins Calcium Injection,and 58 cases in the treatment group were intervened with both Low Molecular Weight Heparins Calcium Injection and Tongmai Decoction.The preoperative and postoperative dynamic changes of PLT,PAgR,D-D and TEG (R value,K value,alpha angle,MA),and adverse drug reactions in the two groups were detected 1,3,7 d prior to,and after the treatments,respectively.RESULTS No statistically significant difference between the two groups' PLT,PAgR and D-D contents were observed before the operation (P > 0.05).The levels of PLT,PAgR and D-D in both groups increased one day after the operation with similar change degree (P > 0.05).On the 3rd and 7th days after the surgery,the PLT,PAgR and D-D contents in the two groups began to drop,and the decline in the treatment group was more significant (P < 0.05).No significantly different TEG (R value,K value,alpha angle,MA) between the two groups was noticed before the operation (P > 0.05).Patients of both groups displayed their R value,K value decrease,and alpha Angle,MA increase (P > 0.05) one day after the operation with similar change degree.Their R value,K value increased,and alpha angle,MA decreased,and as well as the statistically significant change between the two groups were discovered on the 3rd and 7th postoperative days (P < 0.05).CONCLUSION Tongmai Decoction can effectively reduce blood hypercoagulability and prevent deep venous thrombosis in terms of PLT,PAgR,D-D and TEG (R value,K value,alpha angle,MA) improvement in elderly patients with intertrochanteric fracture in perioperative period.

ABSTRACT

Objective To investigate the epidemioiogical pattern of Borna disease virus (BDV) among different canine breeds in Ili, China, and to analyze its potential phylogeny. Methods BDV p24 RNA fragments were detected from peripheral blood mononuclear cells (PBMCs) of canine by modified nested RT-PCR (nRT-PCR). Possible false positives were excluded by determination of both BDV p40 RNA fragments and PMD19 plasmid standards. Analysis were performed on genetic sequence, homologous comparison, amino acid sequence and phylogeny after p24 positive products were validated. Results BDV p24 RNA fragments were found only in Kazakh Tobet (a shepherd dog) in 8 breeds of 150 cases and their overall positive rate was 11.0% (10/91). Compared with the strain of He/80 from horse and that of S6 from sheep in Germany, the homologous similarities of Kazakh Tobet was 99.2% and 95.7%, and that of amino acid as 100% and 89.3%, respectively. The kinship of Kazakh Tobet was close to He/80 and next to S6. Conclusion There was potential natural BDV infection in Kazakh Tobet in Ili, and its endemic strain was concerned with He/ 80 infecting Ili horse and S6 of German Merino sheep introduced into the region from Germany.
