ABSTRACT

This experiment was carried out to study the feeding value of ground date pits (DP) (Phoenix dactylifera L.) with and without enzyme supplementation on laying hens' performance. Apparent metabolizable energy value of DP was determined by the total collection method using 10 adult Leghorn cockerels. After that, a total number of 144 Lohmann 50-week-old LSL-Lite hens were randomly allocated into six groups consisting of four replicates of six birds, based on a 3 × 2 factorial arrangement of the treatments. Six iso-caloric and iso-nitrogenous experimental diets including I-corn-soya bean meal-based control, II-corn-soya bean meal oil-based control and III- corn-soya bean meal-based diet (180 g/kg DP) were formulated. Each of the diets was supplemented with two levels of an enzyme (0.0 and 0.07 g/kg Natozim Plus). The experiment lasted 10 weeks after 7 days of adaptation. The results revealed that there was no significant difference in feed intake, feed conversion ratio, egg production, egg mass, eggshell weight, eggshell thickness and Haugh unit among the treatments. Dietary inclusion of DP significantly decreased body weight gain (BWG), egg weight and yolk colour score. On the other hand, corn-soya bean meal-based control diet with vegetable oil significantly increased egg weight and BWG of birds in comparison with other treatments. The serum biochemical metabolites were not affected by DP and enzyme supplementation. Thus, DP can be used as alternative feedstuff in laying hen diets, up to 18% of the diet with little effect on the performance of hens, including egg weight, and also, it had an adverse effect on yolk colour. Eventually, in terms of performance, the results failed to demonstrate any positive effect of Natozim Plus on either the controls or 18% DP diets.

ABSTRACT

An experiment was conducted to determine dietary energy (ME) and CP requirements of quail chicks using response surface methodology. A total of 40 floor pens of 20 birds each were assigned to 9 diets of central composite design (CCD) containing 5 levels of ME (2,809 to 3,091 kcal/kg) and CP (19 to 24.8% of diet) from 7 to 14 d of age. The experimental results of CCD were fitted with quadratic response models, and ridge analysis was used to compute the optimal response for BW gain (BWG) and feed conversion ratio (FCR). Regression analysis showed that the linear effect of independent variables was significant on bird responses. The quadratic and cross-product effects did not have significant effects on performance. Dietary levels of CP linearly affected BWG and FCR, but the effect of dietary ME was not significant. The ridge maximum analysis on BWG and minimum analysis on FCR models revealed that the maximum BWG may be achieved with 2,950 kcal of ME/kg and 25% CP; and minimum FCR may be obtained with 2,878 kcal of ME/kg and 24.4% CP. The results of this study showed that response surface analysis with the CCD platform was successfully used to optimize dietary requirements of Japanese quail and this methodology could be used for other nutrients.

ABSTRACT

A double-stranded calf thymus DNA (dsDNA) was physisorbed onto a polypyrrole (PPy) nanofiber film that had been electrochemically deposited onto a Pt electrode. The surface morphology of the polymeric film was characterized using scanning electron microscopy (SEM). The electrochemical characteristics of the PPy film and the DNA deposited onto the PPy modified electrode were investigated by cyclic voltammetry (CV), differential pulse voltammetry (DPV), and electrochemical impedance spectroscopy (EIS). Then the interaction of DNA with salicylic acid (SA) and acetylsalicylic acid (ASA), or aspirin, was studied on the electrode surface with DPV. An increase in the DPV current was observed due to the oxidation of guanine, which decreased with the increasing concentrations of the ligands. The interactions of SA and ASA with the DNA follow the saturation isotherm behavior. The binding constants of these interactions were 1.15×10(4)M for SA and 7.46×10(5)M for ASA. The numbers of binding sites of SA and ASA on DNA were approximately 0.8 and 0.6, respectively. The linear dynamic ranges of the sensors were 0.1-2µM (r(2)=0.996) and 0.05-1mM (r(2)=0.996) with limits of detection of 8.62×10(-1) and 5.24×10(-6)µM for SA and ASA, respectively.
