ABSTRACT

Two new C21 steroidal glycosides, paniculatumosides H and I, together with four known ones were isolated from the roots of Cynanchum paniculatum (Bge.) Kitag. Their structures were identified by spectroscopic methods including extensive 1D and 2D NMR techniques. All compounds were subjected to detect the anti-tobacco mosaic virus (TMV) activities and their cytotoxities against three human tumor cell lines (SMMC-7721, MDA-MB-231 and A549). The results showed that compounds 1 and 5 exhibited potent protective activities against TMV, while 2, 4 and 6 had moderate effects on the SMMC-7721 cancer cells viability.

ABSTRACT

The phytochemical study of Euphorbia resinifera afforded 18 structurally diverse diterpenoids, including 14 new ingol-type diterpenoids, euphorblins A-N (1-14), a new rhamnofolane diterpenoid, euphorblin O (15), and three known analogues (16-18). The structures of these compounds were deduced using 2D NMR spectroscopy and NOE experiments. The structure of compound 1 was confirmed by single-crystal X-ray crystallography. The abilities of the compounds to enhance lysosomal biosynthesis were evaluated through LysoTracker Red staining. Among the 10 active compounds, compounds 2, 4, and 18 showed remarkable immunofluorescence strength, and their LysoTracker staining intensities were 155.9%, 143.5%, and 140.7%, respectively, greater than that of the control. A series of lysosomal genes were also found to be upregulated by these compounds, which further confirms their ability to induce lysosome biosynthesis and suggests that these diterpenoids have potential as lead compounds for the development of drugs for the treatment of lysosome-related diseases.

ABSTRACT

Aim To observe whether asiatic acid ( AA) can inhibit lipopolysaccharide ( LPS )-induced inflammatory response in VSMCs , and explore its mechanism of action .Methods The VSMCs isolated from aorta of SD rats were primarily cultured . The effect of AA on the cell viability of VSMCs was meas-ured by MTT assay .The protein and mRNA expression of IL-6, MCP-1, and TNF-α, were measured by ELISA assay and real-time PCR, respectively.The protein and mRNA of TLR4 and PPAR-γwere meas-ured by Western blot and real-time PCR, respectively . Results AA exhibited no effect on cellular viability between the concentration from 0 to 30 μmol · L-1 . After treating VSMCs with LPS (500μg· L-1 ) for 6h or 24 h, the protein and mRNA expression of IL-6, MCP-1, TNF-α, and TLR4 significantly increased ( P<0.05 );and on the contrary , the activity of PPAR-γwas significantly reduced ( P<0.05 ) .Treatment with AA (10, 20, 30 μmol· L-1 ) could concentration-de-pendently inhibit LPS-induced protein and mRNA ex-pression of IL-6, MCP-1, TNF-α.AA could also re-duce LPS-induced protein and mRNA expression of TLR4, and pretreatment of the cells with TLR4-siRNA could reduce LPS-induced inflammation . Moreover , treatment with AA could up-regulate the mRNA and protein expression of PPAR-γin VSMCs; however , GW9662 , a PPAR-γantagonist , partially attenuated AA' s anti-inflammatory effect .Conclusion AA can significantly inhibit LPS-induced mRNA and protein expression of IL-6, MCP-1, TNF-α, in VSMCs, which is partially dependent on suppressing TLR 4 and up-regulating PPAR-γ.

ABSTRACT

We have determined the complete mitochondrial genome of Lista haraldusalis Walker, 1859. The mitochondrial genome of L. haraldusalis is 15,213 bp in size with 81.5% A+T content. It consists of 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA), 2 ribosomal RNA (rRNA) genes and an A+T-rich region. All PCGs start with ATN codon, except for COI, which begins with TTAG. Eleven PCGs stop with typical stop codon TAA and TAG. But ND6 and COII use incomplete stop codon T. All of the 22 tRNAs have the typical cloverleaf structure except for tRNASer(AGN) lacking of the dihydrouridine (DHU) stem. The conserved motif ATACTA, poly-T stretch, ATAGA, ATTTA and microsatellite-like (AT)11 were found in the intergenic region between tRNASer(UCN) and ND1 and the A+T-rich region of L. haraldusalis, respectively.

ABSTRACT

?AlM: To study the correlation between monocular mild myopic juvenile myopia degree and dominant eye. ? METHODS: Totally 158 patients with juvenile monocularly mild myopia in our hospital from December 2012 to December 2013 were retrospectively analyzed, and cylindrical mirror astigmatism was used for spherical equivalent conversion. On the basis of the myopic degree, they were divided into three groups, 30 cases in group A (-0. 25~-0. 75D), 92 cases in B group (-1. 0~-2. 0D), 36 cases in group C (-2. 25 ~ -3. 0D). The card hole method was selected to measure dominant eye for subjectsat nearly 33cm and 5m. After glasses correction of ametropia, the far and near dominant eyes were received measurement again. ?RESULTS:The monocular mild myopia of dominant eye was compared with the non-dominant eye adjustment function, and there was no significant difference ( P>0. 05 ) . The dominant eye, non - dominant eye mean diopter and other correlations of eyes in the three groups were compared, and there was no significant difference (P>0. 05). The dominant eye of three groups at 5m was compared, and there was significant difference (P ?CONCLUSlON: The formation of myopic anisometropia is related to the degree of clearly seeing objects, although glasses correction can improve the clarity and visual, but affect the choice of the dominant eye, because the myopic anisometropia appears most early in the dominant eye, therefore optician correction stage, which should be taken into consideration, and avoid the severity of the adolescent myopia.

ABSTRACT

OBJECTIVE: To study the effect of protein phosphatase 2A (PP2A) in the negative regulation of progesterone on the proliferation of mouse endometrial epithelial cells (EECs). METHODS: Mouse EECs were isolated and cultured in vitro, which were divided into four groups when they grown to confluence: control group (P4) was treated with 1 micromol/L progesterone only, group A, B and C were treated respectively with 1 micromol/L progesterone and different concentrations of okadaic acid (5 nmol/L, 10 nmol/L and 20 nmol/L). After 24 h, the numbers of cells in different phases of the cell cycle were counted with flow cytometry. RESULTS: The effect of OA on mouse EECs was concentration-dependent. Compared with control group of P4, the change of cell cycle procession in group A was not obvious. Lower proportion of cells in G1 and G2/M phase and higher proportion of cells in S phase in group B, higher proportion of cells in G1 and S phase and lower proportion of cells in G2/M phase in group C were observed. CONCLUSION: Adequate dose OA inhibiting PP2A could release the inhibitory effect of progesterone on proliferation of mouse EECs obviously, this suggested that PP2A was involved in the inhibitory effect of progesterone on proliferation of EECs by influencing the process of cell cycle.

ABSTRACT

It is hard and costly to test resolution directly, because the focal length of testing equipment could be nearly ten meters. Solving resolution by diffraction wavefront aberration indirectly is an effective solution to this problem. A normalization model of solving resolution using fitting coefficients of Zernike polynomials was established based on the spectral imaging theory of Fourier optics. The relationship between resolution and wavefront aberration of diffraction gratings was illustrated by this model. Finally, a new method of testing resolution using fitting coefficients of Zernike polynomials was proposed. According to this method, the resolution of a grating is tested by ZYGO interferometer indirectly. Compared with direct method, results indicate that the error of indirect method is less than 4.22%, and this method could be an effective way to avoid the difficulty of direct method to solve resolution. Meanwhile, this method can be used in ZYGO interferometer to solve resolution by wavefront testing easily.

ABSTRACT

A new-style system that automatically measures the diffraction efficiency of plane reflection grating was designed. The continuous illuminant was adopted for illumination, the duplex grating spectrograph structure was applied, and the linear array NMOS was the receiving component. Wielding relevant principle of the grating spectrograph, theoretical analysis principle was carried out for the testing system. Integrating the aberration theory of geometrical optics, the image quality of this optics system was analyzed. Analysis indicated that the systematic device structure is compact, and electronics system is simplified. The system does not have the problem about wavelength sweep synchronization of the two grating spectrographs, and its wavelength repeatability is very good. So the precision is easy to guarantee. Compared with the former automated scheme, the production cost is reduced, moreover it is easy to operate, and the working efficiency is enhanced. The study showed that this automatic measurement instrument system features a spectral range of 190-1 100 nm and resolution is less than 3 nm, which entirely satisfies the design request. It is an economical and feasible plan.
