ABSTRACT

BACKGROUND: The radical antegrade modular pancreatosplenectomy (RAMPS) which is a reasonable surgical approach for left-sided pancreatic cancer is emphasis on the complete resection of regional lymph nodes and tumor-free margin resection. Laparoscopic radical antegrade modular pancreatosplenectomy (LRAMPS) has been rarely performed, with only 49 cases indexed on PubMed. In this study, we present our experience of LRAMPS. METHODS: From December 2018 to February 2020, 10 patients underwent LRAMPS for pancreatic cancer at our department. The data of the patient demographics, intraoperative variables, postoperative hospital stay, morbidity, mortality, pathologic findings and follow-up were collected. RESULTS: LRAMPS was performed successfully in all the patients. The median operative time was 235 min (range 212-270 min), with an EBL of 120 ml (range 100-200 ml). Postoperative complications occurred in 5 (50.0%) patients. Three patients developed a grade B pancreatic fistula. There was no postoperative 30-day mortality and reoperation. The median postoperative hospital stay was 14 days (range 9-24 days).The median count of retrieved lymph nodes was 15 (range 13-21), and four patients (40%) had malignant-positive lymph nodes. All cases achieved a negative tangential margin and R0 resection. Median follow-up time was 11 months (range 3-14 m). Two patients developed disease recurrence (pancreatic bed recurrence and liver metastasis) 9 months, 10 months after surgery, respectively. Others survived without tumor recurrence or metastasis. CONCLUSIONS: LRAMPS is technically safe and feasible procedure in well-selected patients with pancreatic cancer in the distal pancreas. The oncologically outcomes need to be further validated based on additional large-volume studies.

ABSTRACT

Objective@#To learn the health literacy level and its influencing factors among junior high school students in Jiaxing, so as to provide basis for health promotion of adolescents. @*Methods@#The junior high school students who had been studying and living in Jiaxing for more than six months were selected by multistage cluster random sampling method. A questionnaire survey was conducted to collect general information and health literacy level (including basic knowledge and concept, healthy lifestyle and behaviors, and basic skills) of these selected students. The multivariate logistic regression model was used to analyze the influencing factors for health literacy.@*Results @#Of 1 773 questionnaires collected, 1 738 were valid, accounting for 98.03%. The level of health literacy in the junior high school students in Jiaxing was 22.84% ( 95%CI: 20.87%-24.82% ), The levels of basic knowledge and concept, healthy lifestyle and behaviors, and basic skills were 55.29% ( 95%CI: 52.95%-57.63% ), 21.75% ( 95%CI: 19.81%-23.69% ), 53.05% ( 95%CI: 50.70%-55.40%), respectively. Multivariate logistic regression analysis showed that the second grade and above ( OR: 1.609-1.835, 95%CI: 1.195-2.459 ), mother's educational level of technical secondary school/senior high school and above ( OR: 1.965-1.976, 95%CI: 1.276-3.357 ), and self-rated academic achievement of medium and above ( OR: 1.881-2.441, 95%CI: 1.359-3.335 ) were the promoting factors for health literacy level of junior high school students; self rated health status as unhealthy ( OR=0.254, 95%CI: 0.089-0.721 ) was an obstructive factor.@*Conclusions@#The health literacy level of the junior high school students in Jiaxing was 22.84%. The level of healthy lifestyle and behaviors was the lowest in three aspects. Grade, mother's educational level, self-rated academic performance and self-rated health status may have impacts on health literacy level of junior high school students.

ABSTRACT

BACKGROUND: Curd architecture is one of the most important characters determining the curd morphology of cauliflower. However, the genetic mechanism dissection of this complex trait at molecular level is lacking. Genes/QTLs responsible for the morphological differences between present-day loose-curd and compact-curd cauliflower haven't been well revealed. RESULTS: Herein, by using a common compact-curd parent and two loose-curd parents, we developed two double haploid (DH) populations including 122 and 79 lines, respectively. For each population, we decomposed the curd architecture concept into four parameters (basal diameter, stalk length, stalk angle and curd solidity), and collected corresponding phenotypic data for each parameter across two environments. The Kosambi function and composite interval mapping algorithm were conducted to construct the linkage map and analyze the QTLs associated with curd architecture parameters. A total of 20 QTLs were detected with the minimum likelihood of odd (LOD) values ranging from 2.61 to 8.38 and the percentage of the phenotypic variance explained by each QTL (PVE) varying between 7.69 and 25.10%. Of these, two QTLs controlling stalk length (qSL.C6-1, qSL.C6-2) and two QTLs controlling curd solidity (qCS.C6-1 and qCS.C6-2) were steadily expressed in both environments. Further, qSL.C6-1, qSL.C6-2, qCS.C6-1 and qCS.C6-4 fell into the same chromosomal region of the reference genome, indicating that these loci are involved in pleiotropic effects or are tightly linked. CONCLUSION: The current study identified a series of QTLs associated with curd architecture parameters, which might contribute essentially to the formation of present-day loose-curd cauliflower that is widely cultivated in China. These results may pave the way for intensive deciphering the molecular mechanisms of curd development and for marker-assisted selection of curd morphology in cauliflower breeding.

ABSTRACT

BACKGROUND: MADS-box genes play important roles in vegetative growth and reproductive development and are essential for the correct development of plants (particularly inflorescences, flowers, and fruits). However, this gene family has not been identified nor their functions analyzed in Brassica oleracea. RESULTS: In this study, we performed a whole-genome survey of the complete set of MADS-box genes in B. oleracea. In total, 91 MADS-box transcription factors (TFs) were identified and categorized as type I (Mα, Mß, Mγ) and type II (MIKCC, MIKC*) groups according to the phylogeny and gene structure analysis. Among these genes, 59 were randomly distributed on 9 chromosomes, while the other 23 were assigned to 19 scaffolds and 9 genes from NCBI had no location information. Both RNA-sequencing and quantitative real-time-PCR analysis suggested that MIKC genes had more active and complex expression patterns than M type genes and most type II genes showed high flowering-related expression profiles. Additional quantitative real-time-PCR analysis of pedicel and four flower whorls revealed that the structure of the B.oleracea MIKC genes was conserved, but their homologues showed variable expression patterns compared to those in Arabidopsis thaliana. CONCLUSION: This paper gives a detailed overview of the BolMADS genes and their expression patterns. The results obtained in this study provide useful information for understanding the molecular regulation of flower development and further functional characterization of MADS-box genes in B. oleracea.

ABSTRACT

A new analytical method was developed to detect neomycin in complex biological samples using molecularly imprinted polymer to construct an optical sensor. Fluorescent neomycin-imprinted polymers (fMIPs) containing both imprinted cavity and boronate affinity site were synthesized on the surface of silica-modified quantum dots. The fMIPs exhibited high selectivity to neomycin by having two binding sites for the target analyte. Neomycin analogues (competing for imprinted cavity) and D-glucose (competing for the boronate affinity site) did not affect the selectivity of the fMIPs. When combined with a fluorescent microplate reader, the obtained fMIP sensor displayed a linear concentration-dependent fluorescence quenching in response to neomycin in the range of 2-1000 µg/L, with a limit of detection as 0.16 µg/L. The fMIP sensor was able to detect trace neomycin in biological samples accurately after simple sample pre-treatment. The sensitivity of the fMIP sensor was higher than HPLC equipped with a fluorescence detector. The fMIP sensor containing the doubly selective binding sites provides a selective, sensitive, accurate, and high through-put approach for neomycin monitoring.

ABSTRACT

BACKGROUND: AEB071, an orally available PKC inhibitor, prevents organ rejection after transplantation in rodents and man. Furthermore, pro-inflammatory cytokines and inflammatory processes are important mediators of transplanted organ rejection. We therefore examined whether single or combination therapies of AEB071 and/or tacrolimus affect cytokine profiles in a rat cardiac allograft model. METHODS: AEB071 (60 mg/kg twice a day) and tacrolimus (0.6 or 1.2 mg/kg once a day) were orally administered daily after cardiac transplantation. Interferon (IFN)-gamma, interleukin (IL)-1beta, IL-2, IL-4, IL-6, IL-10, and tumor necrosis factor (TNF)-alpha levels in serum were subsequently measured 5 days after cardiac transplantation using a multiplex protein assay system. RESULTS: All cytokine levels were significantly depressed in cardiac transplanted rats treated with AEB071, whereas tacrolimus only reduced IFN-gamma, IL-2, IL-4, IL-6, and IL-10 levels. When administered in combination, AEB071 and low- or high-dose tacrolimus had additive effects on IFN-gamma, IL-4, IL-6, and TNF-alpha. CONCLUSIONS: These results suggest that AEB071 inhibits T cell activation by blocking the production of proinflammatory cytokines, and that tacrolimus combined with AEB071 can effectively regulate inflammatory cytokines in the transplantation setting.

ABSTRACT

A study was carried out to study the accumulative levels and dynamic changes of organochlorine pesticides in human milk in Beijing from 1982 to 1998, and the effect of prohibition and restriction on the use of DDT and BHC (666) in China in 1983. The milk samples were collected from women during the lactation period in four districts of Beijing from 1982 to 1998, in 1991, 1993 and 1998, respectively. The contents of DDT and BHC in human milk were determined according to the standard method of biology-monitoring of PR China. The average fat content of human milk was about 3% from 1982-1998 in Beijing. The average concentration of pp-DDT and op-DDT in 1985 was less than 0.10 mg/kg and remained so until 1998. The average concentration of pp'-DDT decreased from 1.80 mg/kg in 1982 to 0.24 mg/kg in 1998 and that of pp-DDE descended from 5.87 mg/kg to 1.72 mg/kg and the total concentration of DDT dropped from 7.71 mg/kg to 2.04 mg/kg. Since prohibition of organochlorine pesticides 1983, the average concentration of beta-BHC and total BHC fell respectively from 9.66 mg/kg and 10.1 mg/kg in 1983 to 1.18 mg/kg in 1998. However, the accumulative concentration of DDT and BHC was much higher than that of some foreign countries. Therefore, it was important and necessary to monitor organochlorine compounds in human milk continuously.

ABSTRACT

OBJECTIVE: To investigate the basic contents of organochlorine pesticides (DDT, BHC and HCB) in human milk of normal residents in Beijing urban areas. METHODS: Through continuous survey on the trend of organochlorine contents in human milk since DDT and BHC pesticides were forbidden in use in 1983, and to evaluate the effects of this program, the required lying-in women residents in urban districts for at least 5 years were enrolled in this study. All these lying-in women and also their family members were not exposed to or engaged in producing or selling organochlorine pesticides. In this study, the women chosen were giving birth to a child just after one week to 4 months. The detection method used is the national standard biological detection method. RESULTS: In 1982, the total amount of DDT and BHC contents in human milk were 6.45 mg/kg and 6.97 mg/kg, respectively, which were found the highest level in the word. The contents of organochlorine were descending apparently since the DDT and BHC pesticides were forbidden by Chinese government in 1983. According to the standards of FAO/WHO, the average intake of total DDT and beta-BHC in the infants born in 1998 was 11 and 6 microg/kg, respectively. Both of them were less than the allowable daily intake amount regulated by WHO and the Ministry of Health of Japan. CONCLUSION: The study might make clear that the basic levels of organochlorine pesticides (DDT, BHC and HCB) in human milk of normal residents and the attenuation trend of 2 pesticides cumulative levels in human body, through estimating the intake quantity of organochlorine by infants, showing that the DDT and BHC daily intake from human milk absorbed by infants, who were born after 1998, were at a safe level. It was indicated that the measure for forbidding use of DDT and BHC pesticides is contributory.
