ABSTRACT

Automatic and rapid whole-body 3D shape measurement has attracted extensive attention in recent years and been widely used in many fields. Rapid 3D reconstruction, automatic 3D registration, and dedicated system layout are critical factors to enable an excellent 3D measurement system. In this paper, we present an automatic and rapid whole- body 3D measurement system that is based on multinode 3D sensors using speckle projection. A rapid algorithm for searching homologous point pairs is suggested, which takes advantage of the optimized projective rectification and simplified subpixel matching techniques, leading to an improved time efficiency of 3D reconstruction. Meanwhile, a low-cost automatic system with a flexible setup and an improved calibration strategy are proposed, where system parameters of each node sensor can be simultaneously estimated with the assistance of a cubic and a planar target. Furthermore, an automatic range data registration strategy by employing two aided cameras is investigated. Experiment results show that the presented approach can realize automatic whole-body 3D shape measurement with high efficiency and accuracy.

ABSTRACT

Objective To observe the effect of Modified Dachengqi Decoction on recovery of gastrointestinal function after closure of protective ileostomy of rectal cancer. Methods Seventy pa- tients with rectal cancer, scheduled to receive protective ileostomy were enrolled and randomized into treatment group (35 cases) and control group (35 cases). The treatment group was subjected to ante-grade enema with Modified Dachengqi Decoction (500 mL) once a day before ileostomy, the therapeutic course was 2 weeks ,and the control group received ileostomy alone. The oral feeding time, anal exhaust time, ileus rate and length of hospital stay were observed. Results Compared with the control group, the oral feeding time(h,22 ±6 vs.41 ±8), anal exhaust time(h,27 ±5 vs.48 ±8), ileus rate(2. 85% vs. 14. 29%) and length of hospital stay ( d,5. 5 ± 1. 0 vs.7. 6 ± 1. 2) were decreased in the treatment group (P <0. 01 , P <0. 05).Conclusion Modified Dachengqi Decoction could promote the recovery of gastroin- testinal function after surgery in patients with rectal cancer.

ABSTRACT

The development and application of single nucleotide polymorphisms (SNPs) is in its infancy for pepper. Here, a set of 15,000 SNPs were chosen from the resequencing data to develop an array for pepper with 12,720 loci being ultimately synthesized. Of these, 8,199 (~64.46%) SNPs were found to be scorable and covered ~81.18% of the whole genome. With this array, a high-density interspecific genetic map with 5,569 SNPs was constructed using 297 F2 individuals, and genetic diversity of a panel of 399 pepper elite/landrace lines was successfully characterized. Based on the genetic map, one major QTL, named Up12.1, was detected for the fruit orientation trait. A total of 65 protein-coding genes were predicted within this QTL region based on the current annotation of the Zunla-1 genome. In summary, the thousands of well-validated SNP markers, high-density genetic map and genetic diversity information will be useful for molecular genetics and innovative breeding in pepper. Furthermore, the mapping results lay foundation for isolating the genes underlying variation in fruit orientation of Capsicum.

ABSTRACT

OBJECTIVE: To detect and analyze the characteristic miRNAs profile of anal fistula and explore their possible target genes and potential clinical significance. METHODS: The anal mucosa close to the hemorrhoids were collected from three patients undergoing fistulectomy and hemorrhoidectomy (fistula group) as well as three patients receiving only hemorroidectomy(hemorrhoids group), matching with fistula group in age, gender and body weight. miRNA microarray was used to compare the expression of 1 285 human miRNAs of the anal mucosa between two groups. Cluster analysis was adopted to analyze the accumulation of the differentially expressed miRNAs(P<0.05, fold≥2.0 or ≤0.5) and their target genes were predicted with 10 softwares such as DIANAmT, miRanda, miRDB, miRWalk etc. Comprehensive scoring was performed to identify genes with highest predictive score. Gene ontology (GO) concentration technique was used to analyze the target gene-associated biological process. Immunohistochemistry was used to examine protein expression of genes with the highest score. RESULTS: Among 1285 miRNAs in fistula group, 13 miRNAs were differentially expressed with those in hemorrhoid group, including 2 of up-regulation and 11 of down-regulation. Paired t test showed that in fistula group, miRNA-3609 up-regulation was 5.98 folds(P=0.0231) and miR-181a-2-3p down-regulation was 0.13 folds(P=0.0067) compared to those in hemorrhoid group, which had the greatest differential expression. Cluster analysis suggested that up-regulated miR-3609 and miR-6086 had similar change trend in both groups. Among 11 down-regulated miRNAs, miR-125bp-1-3p and miR-548q had similar expression and other 9 miRNAs had similar expression as well, including miR-1185-1-3p, miR-532-3p, miR-1233-5p, miR-769-5p, miR-149-5p, miR-99b-3p, miR-141-3p, miR-138-5p, and miR-181a-2-3p. Target gene prediction analysis of above 13 genes showed that 7 miRNAs(53.8%) were eligible to predict their potential target genes, yielding totally 104 possible target genes. The rest of 6 miRNAs(46.2%) failed to predict any target gene. The highest score in prediction of target gene was chitinase 1(ChIT1) and its corresponding differential miRNA was miR-769-5p(r=-0.94286, P=0.0167). Gene ontology analysis showed that the most associated biological process related with these 104 target genes was keratinization, immune response and signal transduction. Immunohistochemistry revealed ChiT1 expression of anal mucosa in fistula group was significantly higher compared to hemorrhoid group(P<0.01). CONCLUSIONS: There is a characteristic miRNAs profile in anal fistula patients, which may play a role in the occurrence and development of anal fistula.

ABSTRACT

OBJECTIVE: To optimize the freeze-dried powder preparation technology of recombinate hirudin-2 (rHV2) nanoparticle which has bio-adhesive characteristic for nasal delivery, also to investigate its stability and permeability through nasal membrane in vitro. METHODS: Taking the appearance, rediffusion of nanoparticle and rHV2 encapsulation efficiency as the evaluation indexes. Cryoprotector, the preparative technique and the effect of illumination and high temperature factors on its stability for rHV2 freeze-dried powder were investigated. Using Fraze diffusion cell technique, the permeability of rHV2 across rabbit nasal mucous membrane in chitosan solution, chitosan nanoparticle, and nanoparticle frozen-dried powder were compared with that in normal saline solution. RESULTS: The optimized preparation of rHV2 nanoparticle freeze-dried powder was as follows: 5% trehalose and glucose (1:1) was used as cryoprotector, nanoparticle solution was freezed for 24 h in vacuum frozen-dryer after being pre-freezed for 24 h. The content of rHV2 in the freeze-dried powder was 1.1 ug/mg. Illumination had little effect on the appearance, rediffusion and encapsulation efficiency of the rHV2 freeze-dried powder. High temperature could obviously influence the appearance of nanoparticle freeze-dried powder. The permeability coefficient (P) of nanoparticle was 5 times more than that in chictonson solution. It was indicated that chitosan nanoparticle has effect on increasing the permeability of rHV2. CONCLUSION: The freeze-dried powder of chitosan nanoparticle can be a good nasal preparation of rHV2.
