ABSTRACT

BACKGROUND: The association of Sjögren's syndrome (SS) and lymphoma is similar. Mucosa-associated lymphoid tissue (MALT) or extranodal marginal zone B-cell lymphoma was the most common lymphomatous histology in SS patients. MALT in SS patients is frequently located in the parotid gland, while MALT lymphoma of the skin with SS is an exceedingly rare entity that needs to be recognized. CASE SUMMARY: A 60-year-old woman presented with a 3-year history of progressive dry mouth associated with a 1-year history of enlarging cutaneous nodules. Physical examination revealed two hard subcutaneous nodules on her right lower leg. The results of Schirmer's test were positive, despite the absence of dry eyes. Labial salivary gland biopsy revealed lymphocytic infiltration and chronic inflammation with a focus score of 2. The patient was diagnosed with SS. She underwent resection of one cutaneous nodule, and histopathological analysis identified the nodule as MALT lymphoma. Her dry mouth symptoms improved, and the nodules decreased after 6 mo of treatment with hydroxychloroquine sulfate and chemotherapy (thalidomide, cyclophosphamide, and dexamethasone). CONCLUSION: Lymphoma is a severe complication of SS, shown by the reported unique case of cutaneous MALT lymphoma with SS.

ABSTRACT

Although hepatic metabolism of hexabromocyclododecanes (HBCDs) played critical roles in the selective bioaccumulation of HBCDs in humans, the hepatic metabolism patterns of its enantiomers remained ambiguous. Aiming to elucidate the mechanism on hepatic metabolism of hexabromocyclododecanes (HBCDs) enantiomers, the enantiomers ((+)-α-HBCD, (-)-α-HBCD, (+)-γ-HBCD, and (-)-γ-HBCD), the diastereoisomers (α-, ß-, and γ-HBCDs) and the mixed of α- and γ-HBCDs were incubated with human HepG2 cell under different exposure levels in the present study. The clearance percentages ranked as γ-HBCD enantiomers >ß-HBCD enantiomers >α-HBCD enantiomers at the same exposure levels. The clearance percentages of (+)- and (-)-α-HBCDs increased when cells were exposed to racemic α-HBCD and the mixture of racemic α- and γ-HBCDs (p < 0.05). (-)-γ-HBCD was more resistant to human hepatic metabolism than (+)-γ-HBCD, leading to the enantiomer fractions (EFs) of γ-HBCD lower than 0.50. (-)-α-HBCD was slightly more metabolized when independently exposed to α-HBCD, while (+)-α-HBCD was more preferentially metabolized after exposure to α- and γ-HBCD mixtures. Hydroxylation and debromination HBCD metabolites were identified. In addition, the different EFs of HBCDs in cells and mediums suggested the selective transfer of chiral HBCDs and HBCD metabolites through the cell membrane. This study provided new insight into the enantiomer-selective metabolism of HBCDs.

ABSTRACT

HYPOTHESIS: Finding an efficient dispersant for obtaining a good dispersion of 5-nm detonation nanodiamond (DND) is always a challenge. Two newly designed diblock copolymers, both poly(ammonium methacrylate)-block-poly(2-phenoxyethyl acrylate) (PMA-b-PBEA) but with different molar ratios of PMA to PBEA, were proposed to be efficient dispersants in stabilizing the concentrated aqueous suspensions of DND. EXPERIMENTS: The dispersion efficiency of dispersants for DND in aqueous suspensions was studied by the measurements of particle size, sedimentation property, and rheological behavior. The interactions between the added dispersants and DND were identified by the zeta potential and adsorption analyses. Calculations based on Derjaguin-Landau-Verwey-Overbeek (DLVO) theory were conducted for clarifying the dominant parameters relating to the dispersion efficiency of dispersants. FINDINGS: Compared with the commercially popular dispersant ammonium polyacrylate, these two diblock dispersants exhibited superior efficiency in the stabilization of DND suspensions. Using the diblock copolymers as dispersants, good dispersion stability in a DND suspension with an extremely high solid content of 30 wt% was achieved. According to experimental analyses and based on DLVO calculations, a low number of accompanied counter-ions, high adsorption capability, and thick PMA-b-PBEA adsorption layer are the main reasons for the extremely high dispersion efficiency of the two new dispersants.

ABSTRACT

Tofacitinib, a small molecule JAK inhibitor, has been widely used to reduce inflammation and inhibit progression of bone destruction in rheumatoid arthritis. STAT3, a downstream signaling molecule of JAK, plays a key role in the activation of signaling in response to inflammatory cytokines. Thus, targeting STAT3 may be an inspiring strategy for treating osteoclast-related diseases such as rheumatoid arthritis. In this study, we first investigated the effects of Stattic, a STAT3 inhibitor, on receptor activator of NF-κB ligand (RANKL)-mediated osteoclastogenesis. Stattic inhibited osteoclast differentiation and bone resorption in RANKL-induced RAW264.7 cells in a dose-dependent manner. Stattic also suppressed RANKL-induced upregulation of osteoclast-related genes tartrate-resistant acid phosphatase, matrix metalloproteinase 9, cathepsin K, RANK, tumor necrosis factor receptor-associated factor 6, and osteoclast-associated receptor in RAW264.7 cells. Moreover, Stattic exhibited an inhibitory effect on cell proliferation and cell cycle progression at higher dosages. At the molecular level, Stattic inhibited RANKL-induced activation of STAT3 and NF-κB pathways, without significantly affecting MAPK signaling. In addition, Stattic inhibited RANKL-induced expression of osteoclast-related transcription factors c-Fos and NFATc1. Importantly, Stattic also prevented bone loss caused by ovariectomy. Together, our data confirm that Stattic restricts osteoclastogenesis and bone loss by disturbing RANKL-induced STAT3 and NF-κB signaling. Thus, Stattic represents a novel type of osteoclast inhibitor that could be useful for conditions such as osteoporosis and rheumatoid arthritis.

ABSTRACT

A newly designed dispersant for water-based suspensions, ammonium poly(methacrylate)-block-poly(2-phenoxyethyl acrylate) (PMA-b-PBEA), is proposed in this study. According to the results of rheological analysis, the dispersion efficiency of this new dispersant is superior to that of the commercially available ammonium polyacrylate (PAA-NH4). The diblock structure of PMA-b-PBEA, which simultaneously contains a low-polar anchoring head group and a water-dissociable stabilizing moiety, is the main cause for its extremely high efficiency for powder dispersion. The unique structure not only results in effective adsorption approximately double that of PAA-NH4, but also produces a low number of counter-ions that compress the electrical double layer and ruin powder stabilization. Based on Derjaquin-Landau-Verwey-Overbeek calculations, the large adsorbance of PMA-b-PBEA gives the powder, titania (TiO2) in this study, a high steric stabilization energy. In addition, PMA-b-PBEA provides TiO2 with a remarkably high electrostatic energy because it generates fewer counter-ions. This energy provides excellent dispersity of powder in the suspensions with a high solid content of 60wt% without showing any rheological hysteresis.

ABSTRACT

A new strategy to synthesize 4,9- and 5,10-dialkylated α-aNDTs as well as 4,9- and 5,10-dialkylated ß-aNDTs is described. Four isomeric precursors with different dithienyl-ene-diyne arrangements undergo base-induced double 6π-cyclization to construct the central naphthalene cores, leading to the formation of the regiospecific products. These 2,7-distannylated dialkylated aNDT-based monomers can be used for Stille cross-coupling to produce promising conjugated materials for various optoelectronic applications.
