ABSTRACT

BACKGROUND: The risk factors for central venous access device-related thrombosis (CRT) in children are not fully understood. We used evidence-based medicine to find the risk factors for CRT by pooling current studies reporting risk factors of CRT, aiming to guide clinical diagnosis and treatment. METHODS: A systematic search of PubMed, Web of Science, Embase, Cochrane Library, Scopus, CNKI, Sinomed, and Wanfang databases was conducted. RevMan 5.4 was employed for data analysis. RESULTS: The review included 47 studies evaluating 262,587 children with CVAD placement. Qualitative synthesis and quantitative meta-analysis identified D-dimer, location of insertion, type of catheter, number of lumens, catheter indwelling time, and central line-associated bloodstream infection as the most critical risk factors for CRT. Primarily due to observational design, the quality of evidence was regarded as low certainty for these risk factors according to the GRADE approach. CONCLUSION: Because fewer high-quality studies are available, larger sample sizes and well-designed prospective studies are still needed to clarify the risk factors affecting CRT. In the future, developing pediatric-specific CRT risk assessment tools is important. Appropriate stratified preventive strategies for CRT according to risk assessment level will help improve clinical efficiency, avoid the occurrence of CRT, and alleviate unnecessary suffering of children. IMPACT: This is the latest systematic review of risk factors and incidence of CRT in children. A total of 47 studies involving 262,587 patients were included in our meta-analysis, according to which the pooled prevalence of CRT was 9.1%. This study identified several of the most critical risk factors affecting CRT in children, including D-dimer, insertion location, type of catheter, number of lumens, catheter indwelling time, and central line-associated bloodstream infection (CLABSI).

ABSTRACT

Delirium, a form of acute cerebral dysfunction, is a common complication of postoperative cardiac surgery in children. It is strongly associated with adverse outcomes, including prolonged hospitalization, increased mortality, and cognitive dysfunction. This study aimed to identify risk factors and incidence of delirium after cardiac surgery in children to facilitate early identification of delirium risk and provide a reference for the implementation of effective prevention and management. A systematic literature search was conducted in PubMed, Web of Science, Embase, Cochrane Library, Scopus, CNKI, Sinomed, and Wanfang for studies published in English or Chinese from the inception of each database to November 2023. The PRISMA guidelines were followed in all phases of this systematic review. The Risk of Bias Assessment for Nonrandomized Studies tool was used to assess methodological quality. A total of twelve studies were included in the analysis, with four studies classified as overall low risk of bias, seven studies as moderate risk of bias, and one study as high risk of bias. The studies reported 39 possible predictors of delirium, categorized into four broad groups: intrinsic and parent-related factors, disease-related factors, surgery and treatment-related factors, and clinical scores and laboratory parameters. By conducting qualitative synthesis and quantitative meta-analysis, we identified two definite factors, four possible factors, and 32 unclear factors related to delirium. Definite risk factors included age and mechanical ventilation duration. Possible factors included developmental delay, cyanotic heart disease, cardiopulmonary bypass time, and pain score. With only a few high-quality studies currently available, well-designed and more extensive prospective studies are still needed to investigate the risk factors affecting delirium and explore delirium prevention strategies in high-risk children.

ABSTRACT

The impact of reductive pulmonary inhalants on ultrafine particles (UFPs)-induced pulmonary oxidative stress remains a crucial consideration, yet the concentration-dependent effects of these inhalants have remained unexplored. Here we synthesized composite UFPs simulating atmospheric UFPs, primarily composed of metals and quinones. We subjected these UFPs to varying concentrations (0-7000 µM) of two reductive pulmonary inhalants, N-acetylcysteine and salbutamol, to assess their influence on oxidative potential, measured through the dithiothreitol assay (OPDTT). Simultaneously, we analysed the soluble metal content of UFPs to uncover potential relationships between oxidative potential and metal solubility. Our results unveil a dual role played by these inhalants in shaping the OPDTT of composite UFPs. Specifically, OPDTT generally increased as inhalant concentrations rose from 0 to 300 µM. However, an intriguing reversal occurred when concentrations exceeded 500 µM, resulting in a decline in OPDTT. Relative to untreated UFPs, these inhalants induced promotion and inhibition effects within concentration ranges of 100-500 and >1000 µM, respectively. While no significant correlation emerged between OPDTT and soluble metal content as inhalant concentrations ranged from 0 to 7000 µM, noteworthy positive correlations emerged at lower inhalant concentrations (e.g., N-acetylcysteine at 0-300 µM). These findings provide insights into the potential influence of reductive pulmonary inhalants on health risks associated with UFP exposure, further underscoring the need for continued research in this critical area.

ABSTRACT

Extubation failure (EF) is a significant concern in mechanically ventilated newborns, and predicting its occurrence is an ongoing area of research. To investigate the predictors of EF in newborns undergoing planned extubation, we conducted a systematic review and meta-analysis. A systematic literature search was conducted in PubMed, Web of Science, Embase, and Cochrane Library for studies published in English from the inception of each database to March 2023. The PRISMA guidelines were followed in all phases of this systematic review. The Risk of Bias Assessment for Nonrandomized Studies tool was used to assess methodological quality. Thirty-four studies were included, 10 of which were overall low risk of bias, 15 of moderate risk of bias, and 9 of high risk of bias. The studies reported 43 possible predictors in six broad categories (intrinsic factors; maternal factors; diseases and adverse conditions of the newborn; treatment of the newborn; characteristics before and after extubation; and clinical scores and composite indicators). Through a qualitative synthesis of 43 predictors and a quantitative meta-analysis of 19 factors, we identified five definite factors, eight possible factors, and 22 unclear factors related to EF. Definite factors included gestational age, sepsis, pre-extubation pH, pre-extubation FiO2, and respiratory severity score. Possible factors included age at extubation, anemia, inotropic use, mean airway pressure, pre-extubation PCO2, mechanical ventilation duration, Apgar score, and spontaneous breathing trial. With only a few high-quality studies currently available, well-designed and more extensive prospective studies investigating the predictors affecting EF are still needed. In the future, it will be important to explore the possibility of combining multiple predictors or assessment tools to enhance the accuracy of predicting extubation outcomes in clinical practice.

ABSTRACT

BACKGROUND: Although breastfeeding is strongly recommended, the breastfeeding rate of preterm infants in China remains significantly low. In addition to the global structural challenges to breastfeeding and the physiological immaturity of preterm infants, Chinese mothers of preterm infants face unique challenges of maternal-infant separation after birth. Moreover, little is known about Chinese mothers' specific needs in coping with the difficulties posed by these challenges. This study utilized the Behaviour Change Wheel to investigate the breastfeeding needs of Chinese preterm mothers that may facilitate its practice in the future. METHOD: A qualitative descriptive design was implemented in Wuhan in 2022. Based on purposeful sampling, 13 preterm mothers were recruited from a NICU in a Grade III Class A hospital in Wuhan, China. Face-to-face semi-structured interviews were conducted to collect data using the interview guide developed by the Theoretical Domains Framework. Theoretical Thematic Analysis was used to review the data in 6 steps to identify themes. RESULTS: Five major themes emerged: (1) capability: ability to interpret infants' cues and identify problems, and need for breastfeeding knowledge and skills training; (2) physical opportunity: cleanliness and quietness in household environment, private lactation spaces and breastfeeding tools in workplaces and hospitals; (3) social opportunity: family support, peer support, and authoritative support from healthcare providers; (4) reflective motivation: information on health impacts of breastfeeding; (5) automatic motivation: maternal-infant bonding, free of aversive stimulus. CONCLUSION: Preterm mothers' needs to enable breastfeeding were diverse, including increasing their capability, physical and social opportunities, and reflective and automatic motivation. People, resources and environments associated with these needs should be engaged together to stablish a conducive structural environment for breastfeeding. The policy change for "zero separation" and implementation of kangaroo care should also be implemented in Chinese neonatal intensive care units. Future studies are needed to design effective interventions according to mothers' specific needs.

ABSTRACT

Objective: To explore the effect of the family-centered empowerment model (FECM) on reducing anxiety, improving care ability, and readiness for hospital discharge of main caregivers of preterm infants. Methods: The primary caregivers of preterm infants who were admitted to the Neonatal intensive care Unit (NICU) of our center from September 2021 to April 2022 were selected as the research objects. According to the wishes of the primary caregivers of preterm infants, they were divided into group A (FECM group) and group B (non-FECM group). The intervention effects were evaluated with the Anxiety Screening Scale (GAD-7), the Readiness for Hospital Discharge Scale-Parent Version (RHDS-Parent Form), and the Primary Caregivers of Premature Infants Assessment of Care Ability Questionnaire. Results: Before the intervention, there was no statistically significant difference in the general information, anxiety screening, the scores of each dimension, and total score of the comprehensive ability of the main caregivers, and the score of caregiver preparedness between the two groups (P > 0.05). After the intervention, there were statistically significant differences in the anxiety screening, the total score and total score of each dimension of the care ability, and the score of caregiver preparedness between the two groups (P < 0.05). Conclusions: FECM can effectively reduce the anxiety of primary caregivers of premature infants and improve their readiness for hospital discharge and care ability. To improve the quality of life of premature infants by implementing personalized training, care guidance, and peer support.

BACKGROUND: Muscle strength and depression have been identified as independent risk factors for mortality in the elderly population. This study aimed to quantify the association between handgrip strength (HGS) and depression in community-dwelling older adults. METHODS: Research data were obtained from the China Health and Retirement Longitudinal Study (CHARLS). Depression was assessed using the Center for Epidemiologic Studies Depression Scale (CESD) with a cut-off score of 20 or more. HGS was assessed with a dynamometer. Binary logistic regression and multiple linear regression models were used to test the association between HGS and depression. RESULTS: The sample consisted of 7,036 CHARLS participants, with an average age of 68.9 ± 7.2. After adjusting for gender, age, marriage, body mass index, comorbidities, smoking, drinking, and sleep time, compared with participants in the lowest quartile of HGS, those in the second to the fourth quartile group had a 0.84- (95% CI 0.72 to 0.98), 0.70- (95% CI 0.58 to 0.84) and 0.46- (95% CI 0.35 to 0.61) fold risk of depression, respectively. CONCLUSIONS: There were a negative association between HGS and depression in community-dwelling older adults. It is critical to assess muscle strength in community older adults through accessible and valid objective measures to enhance depression screening.

ABSTRACT

Health risk posed by ultrafine particles (UFPs) is potentially increased by reducing substances present in lung fluid, although knowledge of the underlying mechanisms is insufficient. Here, UFPs mainly consisting of metals and quinones were prepared. The reducing substances examined included lung endogenous and exogenous reductants. UFPs were extracted in simulated lung fluid containing reductants. Extracts were used to analyze metrics relevant to health effects, including the bioaccessible metal concentration (MeBA) and oxidative potential (OPDTT). The MeBA of Mn (974.5-9896.9 µg L-1) was higher than those of Cu (155.0-599.6 µg L-1) and Fe (79.9-500.9 µg L-1). Correspondingly, UFPs containing Mn had higher OPDTT (2.07-12.0 pmol min-1 µg-1) than those containing Cu (2.03-7.11 pmol min-1 µg-1) and Fe (1.63-5.34 pmol min-1 µg-1). Endogenous and exogenous reductants can increase MeBA and OPDTT, and the increments were generally higher for composite than pure UFPs. Positive correlations between OPDTT and MeBA of UFPs in the presence of most reductants emphasized the importance of the bioaccessible metal fraction in UFPs for inducing oxidative stress by reactive oxygen species (ROS)-generating reactions between quinones, metals, and lung reductants. Present findings provide novel insight into the toxicity and health risks of UFPs.

ABSTRACT

Background: The improvement in survival of preterm infants is accompanied by an increase in neonatal intensive care unit (NICU) admissions. Prolonged length of stay in the NICU (LOS-NICU) increases the incidence of neonatal complications and even mortality and places a significant economic burden on families and strain on healthcare systems. This review aims to identify risk factors influencing LOS-NICU of newborns and to provide a basis for interventions to shorten LOS-NICU and avoid prolonged LOS-NICU. Methods: A systematic literature search was conducted in PubMed, Web of Science, Embase, and Cochrane library for studies that were published in English from January 1994 to October 2022. The PRISMA guidelines were followed in all phases of this systematic review. The Quality in Prognostic Studies (QUIPS) tool was used to assess methodological quality. Results: Twenty-three studies were included, 5 of which were of high quality and 18 of moderate quality, with no low-quality literature. The studies reported 58 possible risk factors in six broad categories (inherent factors; antenatal treatment and maternal factors; diseases and adverse conditions of the newborn; treatment of the newborn; clinical scores and laboratory indicators; organizational factors). Conclusions: We identified several of the most critical risk factors affecting LOS-NICU, including birth weight, gestational age, sepsis, necrotizing enterocolitis, bronchopulmonary dysplasia, and retinopathy of prematurity. As only a few high-quality studies are available at present, well-designed and more extensive prospective studies investigating the risk factors affecting LOS-NICU are still needed in the future.

ABSTRACT

The incidence of infections caused by the H7N9 subtype of the influenza virus has expanded rapidly in China in recent decades, generating massive economic loss and posing a significant threat to public health. In the absence of specialized antiviral treatments or long-term effective preventative vaccinations, it is critical to constantly enhance vaccines and create effective antiviral drugs to prevent the recurrence of pandemics. In the present study, a transmembrane-substituted (TM) virus-like particle (VLP)-based vaccine was created by replacing the transmembrane region of hemagglutinin (HA) protein with the transmembrane region of the H3 HA protein and then used to immunize BALB/c mice. Sera and T cells were collected from the immunized mice to evaluate the passive immune effects. Our results showed that naïve mice achieved 80-100% protection against homologous and heterologous H7N9 influenza strains after receiving passive serum immunization; the protective effect of the TM VLPs was more evident than that of the wild-type HA VLPs. In contrast, mice immunized with passive T cells achieved only 20 to 80% protection against homologous or heterologous strains. Our findings significantly contribute to understanding the control of the H7N9 virus and the development of a vaccine.

ABSTRACT

Background: Since the 2019 novel coronavirus (COVID-19) pneumonia outbreak in late 2019, an endless stream of research has emerged surrounding physical activity. This study analyzes the 50 most influential articles on COVID-19 and physical activity over the past 2 years to describe the research landscape and hotspots from bibliometric citation analysis. Methods: The top-50 cited articles were extracted from the Web of Science Core Collection database, and bibliometric citation analysis was performed by Excel 2019 and VOSviewer software. Results: The top-50 articles were cited 160.48 ± 106.90 (range: 70-587). Most of the articles were from the United States (14), followed by Italy (11) and England (9). The International Journal of Environmental Research and Public Health (n = 10) is the journal with the top-50 cited articles. The collaboration between authors was mainly among three teams, including Smith L, Musumeci G, and Napoli C. The hotspot of research around COVID-19 and physical activity focused on lifestyle change (sedentary behavior, sitting time), mental health (depressive, anxiety, loneliness), the credibility of physical activity assessment tools (reliability, validity), and physical activity of different populations (gender, youth, children). Conclusions: Based on a bibliometric analysis of high-impact articles on COVID-19 and physical activity highlights physical activity as an essential lifestyle change and developments and hotspots in this field. These data will provide insights for future researchers regarding the direction of physical activity research in the COVID-19 pandemic.

Dose-dependent effects of chronic Pb exposure-induced injuries, especially on the trace elements and gut microbiota in mice, have not been explored. In the present study, we investigated these aspects using C57BL/6 mouse models that were exposed to Pb via drinking water with Pb concentrations of 0.1, 0.5, and 1.0 g/L for 8 weeks. The results showed that with the increase in chronic Pb exposure dose, the Pb levels in the blood and tissues, Zn levels in the kidney and brain were elevated, and the levels of bone Zn, kidney Fe, brain Mg, Ca, and Fe, renal catalase activity, and glutathione levels, as well as the expression of colonic zonula occludens-1 and occludin, decreased with a strong linear correlation. Moreover, the relative abundance of Marvinbryantia and Ruminococcus 1 increased, while that of Lactobacillus and Roseburia decreased linearly with the Pb exposure dose. PICRUSt analysis revealed that chronic Pb exposure had a greater impact on the metabolism of macronutrients, trace elements, and neurodegenerative injury. These findings suggest that chronic Pb exposure disrupts trace element levels in tissues, especially in the brain, and induces gut dysbiosis in a dose-dependent manner, which is different from the dose-effect of acute Pb toxicity.

ABSTRACT

Human pegivirus (HPgV-1), previously known as GB virus C (GBV-C) or hepatitis G virus (HGV), is a single-stranded positive RNA virus belonging to the genus Pegivirus of the Flaviviridae family. It is transmitted by percutaneous injuries (PIs), contaminated blood and/or blood products, sexual contact, and vertical mother-to-child transmission. It is widely prevalent in general population, especially in high-risk groups. HPgV-1 viremia is typically cleared within the first 1-2 years of infection in most healthy individuals, but may persist for longer periods of time in immunocompromised individuals and/or those co-infected by other viruses. A large body of evidences indicate that HPgV-1 persistent infection has a beneficial clinical effect on many infectious diseases, such as acquired immunodeficiency syndrome (AIDS) and hepatitis C. The beneficial effects seem to be related to a significant reduction of immune activation, and/or the inhabitation of co-infected viruses (e.g. HIV-1). HPgV-1 has a broad cellular tropism for lymphoid and myeloid cells, and preferentially replicates in bone marrow and spleen without cytopathic effect, implying a therapeutic potential. The paper aims to summarize the natural history, prevalence and distribution characteristics, and pathogenesis of HPgV-1, and discuss its association with other human viral diseases, and potential use in therapy as a biovaccine or viral vector.

ABSTRACT

Health risk of F in soil is of special concern due to the continuously elevated concentration of F in soil. However, there is still a dearth of risk assessments of F in soil based on in-vitro bioaccessibility posed by multiple exposure routes. Herein, the oral, inhalation, and dermal bioaccessibility of F in soil was firstly obtained by adapting and combining in-vitro methods, which then was introduced to remedy an information gap of a comprehensive risk of F in soil posed by a multi-exposure pathway. Combined in-vitro tests indicate the oral, inhalation, and dermal bioaccessibility of F was 13.15 ± 2.63%, 16.55 ± 2.63%, and 1.27 ± 0.73%, respectively. Plasma yielded a detoxic potential for the absorbed F after digesting in small intestine, while effects of enzymes, sweat, and food on the oral bioaccessibility of F were insignificant. Different with metals, the major dissolving phase of F was the interstitial fluid in the deep lung instead of in the alveolar macrophages intracellular environment. A potentially major release of F in the exocrine sweat was noted than in the apocrine sweat. Risk assessments based on the daily exposure incorporated with the in-vitro bioaccessibility suggested that compared with inhalation and dermal contact, oral ingestion was the main exposure route of F in soil to human. Present findings provide insights into the bioaccessibility and health risk of F in soil by multiple exposure routes, which are crucial for the risk control of F contamination in soil.

Continuous fluorine (F) accumulation in soil by anthropogenic activities leads to variously global environmental and health issues. Herein, 300 farmland soil samples were collected from different anthropogenic activity dominated districts for studying the distribution and related health risk of F in soils. Co-existing metal concentrations in soil samples were also analysed to evaluate the relationship between the distribution of F and metals in soil. The median value of the total F concentration of 488 mg kg-1 in the present samples was higher than the median background F concentration in topsoil in Sichuan province of China (261 mg kg-1). Concentration of water-soluble F (1.33-26.2 mg kg-1) was two or three orders of magnitude less than that of total F in soil. Levels of total and water-soluble F in soils collected from the district with longer contamination history were higher than that from other districts with shorter contamination period, indicating a historical contribution of anthropogenic activities to F accumulation in soil. Notable positive correlation between the total F and vanadium (V) concentration in soil can be partly linked to the usually negative charged form or a common source of F and V in soil (e.g. coal combustion). Compared with inhalation and dermal contact, present human exposure of F in soil was mainly caused by oral ingestion, and the health risks posed by F in soil for both children and adults were acceptable. However, considering the higher potential risk for children than adults, the accumulation of F in soil induced by anthropogenic activities should not be neglect.

Background: Cholangiocarcinoma (CCA) is primary liver cancer originating from the biliary tract. The epidemiology of CCA is diverse across the globe. There are no reliably effective options for systemic therapy and CCA outcomes are poor. Herein, we examined the association between overall survival and clinical characteristics of CCA patients in our region. Methods: We included 62 CCA cases diagnosed between 2015 and 2019. Demographics, clinical history, therapeutic procedures, and concomitant diseases were abstracted. Patient survival was obtained from a household registration system. Results: The cohort was 69% male and 31% female, with 26 (42%) iCCA, 27 (44%) pCCA, and 9 (15%) dCCA. No age differences were observed between the three subtypes. Bile duct and metabolic disorders were the major concomitant diseases and showed varying associations with CCA subgroups. Serum triglycerides (TG) were higher in pCCA and dCCA than iCCA patients (p < 0.05), and TG and total cholesterol (TC) were highest among pCCA patients with cholelithiasis. Liver function appeared significant difference between iCCA, pCCA and dCCA subtypes (p < 0.01), and also in the subgroups without cholelithiasis (p < 0.01). The obstructive jaundice in pCCA patients was associated with survival time after surgery, and the presence of cholelithiasis was also another influential factor. Conclusion: We found that pCCA was more frequently associated with metabolic disorders compared to iCCA and dCCA. Postoperative survival was also associated with the degree of jaundice in pCCA compared to iCCA or dCCA. And biliary drainage is an important predictor of outcome of pCCA.

Fluorine (F) is widely dispersed in the environment and frequently used in industry and agriculture with a high migration ability. Thus, it is essential to understand the leaching characteristic of F in soil from industry and agriculture sources. Several sources of F pollutants in soil, including fertilizers, pesticides, phosphogypsum, and atmospheric deposition, were selected to investigate leaching characteristics of F in soil by leaching experiments. The addition of phosphate fertilizer and compound fertilizer (N:P:K = 20:10:15) enhanced the leachability of F in soil and the proportion of F leached out from soil treated by these fertilizers were 0.25% and 0.24%, respectively. However, unanticipated lower leachability of F appeared in compound fertilizer (N:P:K = 17:17:17), nitrogen fertilizer, dipterex, fluoroglycofen, fluopimomide, simulative dry deposition (YF3), and phosphogypsum loaded soils compared with additive-absent treatment. Although phosphogysum had a high F concentration, minimum proportion of F released (0.18%) was observed in phosphogypsum-coverd soil. The amounts of F leaching-out from surface soils (0-25 cm) treated with nitrogen fertilizer decreased 1.03 kg ha-1 comparing with blank control. Soil with phosphate fertilizer leached 5.47 kg F ha-1 a year, having the highest environment risk to deeper soil and groundwater. However, phosphogypsum and dry deposition of airbone F chemical had few effects on F leaching in soil. F-containing materials from agricultural process may leach more F from surface soils than industrial sources.

ABSTRACT

Hepatocellular carcinoma (HCC) is one of the most common cause of malignancy-related deaths. Recently, RING-finger protein 5 (RNF5), an E3 ubiquitin ligase, was revealed to be associated with the development of several human cancers. However, the clinical implication and functional role of RNF5 in HCC are poorly understood. We analysed RNF5 expression in HCC samples and observed that both the mRNA and protein levels of RNF5 were significantly increased in HCC tissues. RNF5 upregulation was markedly associated with larger tumour size, more satellite foci, and higher alpha fetoprotein (AFP) level, indicating poor prognosis in patients with HCC. Knockdown and overexpression experiments demonstrated that RNF5 promoted the proliferation, migration, and invasion of HCC cells in vitro. Moreover, RNF5 facilitated HCC growth in vivo. Our findings indicated that RNF5 was an oncogene of HCC progression and could be used as a novel prognostic biomarker and therapeutic target for patients with HCC.

ABSTRACT

The metal tolerance mechanism of plants is of great importance to explore the plant-based clean-up of environmental substrata contaminated by heavy metals. Indoor experiment of tobacco (Nicotiana tabacum L.) seedlings growing hydroponically in nutrient solution containing 0, 0.1, 0.5, 2.0, and 4.0 mg L-1 V was conducted. The results indicated that plant overall growth performance was significantly affected at ≥ 2.0 mg L-1 V. Oxidative stress degree as indicated by foliar O2-· and H2O2 content intensified markedly at ≥ 0.5 mg L-1 V treatments. In response, the plant activated its enzyme and non-enzyme protecting mechanism to cope with oxidative stress inflicted by vanadium. The activities of antioxidant enzymes, including SOD, POD, CAT, APX, and the concentration of non-enzyme antioxidants, e.g., AsA and GSH were all conspicuously (p < 0.5 or p < 0.1) enhanced at ≥ 0.5 mg L-1 V treatments. Vanadium accumulated in leaves, stems, and roots increased with increasing vanadium level. The majority of the absorbed vanadium retained in plant root, and minor portions were transferred to aerial parts. Vanadium concentration in plant tissues ordered as root ˃ stem ˃ leaf. Translocation factors (TF) in vanadium-treated tobaccos (TF « 1) were significantly lower than that of control (TF ˃ 1). In conclusion, although vanadium at ≥ 2.0 mg L-1 inhibited plant growth, tobacco exhibited a relatively good vanadium tolerance through self-adaptive regulation and has the potential as a phytostabilizer in decontaminating the environment contaminated by vanadium.

ABSTRACT

Lead (Pb) toxicity has been widely studied, but its dose-dependent toxic effects on the gut remain unclear, therefore, the aim of this study was to evaluate the effects of different doses of Pb exposure on the gut microbiota and gut barrier in vitro and in vivo. The HT-29 cell model was used to determine the Pb-induced effects on cell viability, reactive oxygen species (ROS), and tight junction proteins (TJPs) in vitro, and C57BL/6 mice models exposed to 0, 20, 100, 500, and 1000 mg/kg Pb were used to investigate the Pb-induced dose-dependent effects on the gut microbiota, TJP expression, and colon histopathology. Our results showed that the exposure of HT-29 cells to 8 mM Pb decreased cell viability by 50%, elevated ROS levels by 200%, and suppressed the expression of the TJPs, zonula occludens-1 (ZO-1) and occludin by 23% and 35%, respectively. Consistently, Pb-exposed mice showed significant increases in colon tissue damage and inflammation and reductions in ZO-1 mRNA levels in a dose-dependent manner. The occludin mRNA levels decreased in the 500 and 1000 mg/kg groups. At the genus level, the relative abundance of Coprococcus and Oscillospira decreased and that of Lactobacillus increased in linear manner with the Pb exposure dose. PICRUSt analysis based on 16S rRNA sequencing revealed Pb dose-dependent alterations in metabolism through the gut microbiota. These findings suggest that Pb exposure can not only disrupt the barrier by generating oxidative stress, but can also induce gut dysbiosis, colon tissue damage, and gut inflammation in a dose-dependent manner.
