ABSTRACT

To explore the effect of associated bacteria on the low-temperature adaptability of pinewood nematodes (PWNs), transcriptome sequencing (RNA-seq) of PWN AH23 treated with the associated bacterial strain Bacillus cereus GD1 was carried out with reference to the whole PWN genome. Bioinformatic software was utilized to analyze the differentially expressed genes (DEGs). This study was based on the analysis of DEGs to verify the function of daf-11 by RNAi. The results showed that there were 439 DEGs between AH23 treated with GD1 and those treated with ddH2O at 10 °C. There were 207 pathways annotated in the KEGG database and 48 terms annotated in the GO database. It was found that after RNAi of daf-11, the survival rate of PWNs decreased significantly at 10 °C, and fecundity decreased significantly at 15 °C. It can be concluded that the associated bacteria GD1 can enhance the expression of genes related to PWN low-temperature adaptation and improve their adaptability to low temperatures.

ABSTRACT

We describe a catalytic strategy for direct single C(sp3)-F bond alkylation of trifluoromethylbenzimidazoles under a photoinduced thiol catalysis process. The CO2 radical anion (CO2•-) proved to be the most efficient single-electron reductant to realize such a transformation. The spin-center shift of the generated radical anion intermediate is the key step in realizing C-F bond activation under mild conditions with high efficiency.

ABSTRACT

Bisimine derivatives of salicylaldehyde with chiral diamines (salens) are privileged ligands in asymmetric organometallic catalysis, which can be used in cooperation with organocatalysts as additives. The latter can be a modifier of the metal reactivity by liganding or a true co-catalyst working in tandem or in a dual system. All scenarios encountered in the literature are reviewed and classified according to the organocatalyst. In each case, mechanistic and physical-organic chemistry considerations are discussed to better understand the gears of these complex catalytic settings.

ABSTRACT

PURPOSE: To explore the effects of traditional Chinese medicine for vaginal lavage combined with psychological intervention on the immune function and clinical efficacy in patients with cervical cancer. METHODS: Patients with cervical cancer treated in our hospital from January 2020 to May 2021 were included in this study. All patients were treated with traditional Chinese medicine for vaginal lavage combined with psychological nursing intervention. The treatment outcomes of the patients were observed, and the quality-of-life scores and depression of the patients before and after treatment were compared. Changes in T-lymphocyte subset-related indicators, changes in blood routine-related indicators, and changes in the detection level of tumor markers were compared with anxiety scores. RESULTS: After treatment, depression and anxiety were significantly reduced and the patient's quality of life significantly improved. After treatment, the patient's CD3+, CD4+, and CD4+/CD8+ proportions were dramatically higher than before treatment (P < 0.05), there was no significant difference in CD8+ proportion before and after treatment (P > 0.05), and the white blood cell (WBC), hemoglobin (Hb), platelet (PLT) of patients, and the level of tumor marker (CA125) after treatment were immensely lower than before treatment (P < 0.05). CONCLUSIONS: Treating patients with cervical cancer with traditional Chinese medicine for vaginal lavage combined with psychological nursing can effectively improve the patient's immune function, effectively reduce the level of tumor marker CA125, increase the level of T-lymphocyte subsets, and improve the bone marrow hematopoietic function.

ABSTRACT

OBJECTIVE: This study was aimed to investigate the clinical effect of Chinese herbal decoction combined with basic chemoradiotherapy and nursing intervention in the treatment of cervical cancer and the effect on serum carbohydrate antigen 125 (CA125), carcinoembryonic antigen (CEA), and tumor necrosis factor-α (TNF-α) levels. METHODS: A total of 200 cervical cancer patients in our hospital from June 2015 to November 2018 were selected and randomly divided into a study group and a control group. The control group was given chemoradiotherapy and psychological nursing treatment, and the study group was given self-made Chinese herbal decoction on the basis of the control group. The clinical efficacy and serum CEA, CA125, and TNF-α levels were assessed. RESULTS: After treatment, the total effective rate of the study group was significantly higher than that of the control group. The levels of serum CEA, CA125, and TNF-α were decreased in the two groups after treatment, and the decrease in the study group was more significant than that in the control group. After treatment, CD3+ and CD4+ levels were increased compared with those before treatment, and the increase in the study group was also more obvious than that of the control group. The level of CD8+ was decreased compared with before treatment, and the decrease in the study group was more notable than that of the control group. The two-year cumulative survival rate of the study group was markedly higher than that of the control group. The quality-of-life of patients treated for 3 months, 1 year, and 2 years was dramatically improved compared to before treatment. The incidence of adverse reactions in the study group was lower than that of the control group. CONCLUSION: The treatment of basic chemoradiotherapy and psychological nursing intervention combined with Chinese herbal decoction on cervical cancer patients can improve the clinical treatment effects, improve the patient's body immunity, reduce serum CEA, CA125, and TNF-α levels, prolong survival time, improve life quality, and reduce the incidence of adverse reactions, and it is worthy of clinical promotion.

ABSTRACT

Isochorismate synthase (ICS) is a key enzyme for the synthesis of salicylic acid (SA) in plants. SA plays an important role in the response of plants to abiotic stress. In this study, transgenic barley was constructed to evaluate the function of ICS under salt stress. ICSOE lines showed obvious salt stress tolerance, this results from the increased outward Na+ flux and inward K+ flux in roots, thereby maintaining a lower cytosolic Na+/K+ ratio under salt stress. Overexprssion of ICS also improved Na+ sequestration in shoots under salt stress. In addition, ICSOE lines displayed less accumulation of reactive oxygen species and oxidative damage, accompanied by higher activity of antioxidant enzymes. The improved Na+/K+ ratio, Na+ sequestration, and antioxidative competence play an important role in the enhanced salt tolerance of ICSOE lines. These findings help to elucidate the abiotic stress resistance of the ICS pathway in barley.

ABSTRACT

The first example of a ruthenium-catalyzed C-H bond alkylation via six-membered ruthenacycles is presented. This is disclosed for the C-H bond alkylation of biologically relevant cyclic amides with maleimide derivatives. The cyclic tertiary amide core acted as a directing group (DG) enabling formation of six-membered cycloruthenated species responsible for the control of the regio- and site selectivity of the reaction as well as the excellent functional group tolerance. Unexpectedly, cyclic amides were found to be better DGs than pyridine-containing ones or cyclic imides for this type of C-H bond functionalization.

ABSTRACT

Biologically relevant N-arylisoindolinones efficiently underwent arylation reactions under ruthenium catalysis via C-H bond functionalization. The reactions exclusively led to monoarylated products, and only ortho selectivity was observed in the aromatic ring connected to the nitrogen atom. Interestingly, no C-H bond functionalization was observed in the other benzene ring in the ortho position with respect to the carbonyl group. This ruthenium-catalyzed reaction displayed a high functional group tolerance, and it employed readily available and benchmark stable boronic acid and potassium aryltrifluoroborate derivatives as coupling partners. An appealing late-stage functionalization of indoprofen applying this methodology is showcased.

ABSTRACT

Site- and regio-selective aromatic C-H bond benzoxylations were found to take place using biologically appealing N-arylisoindolinones under ruthenium(ii) catalysis in the presence of (hetero)aromatic carboxylic acid derivatives as coupling partners. Besides the presence of two potential C(sp2)-H sites available for functionalization in the substrates, exclusive ortho selectivity was achieved in the phenyl ring attached to the nitrogen atom. Notably, the reactions occurred in a selective manner as only mono-functionalized products were formed and they tolerated a large number of functional chemical groups. The ability of the cyclic tertiary amide within the isoindolinone skeleton to act as a weak directing group in order to accommodate six-membered ring ruthenacycle intermediates appears to be the key to reach such high levels of selectivity. In contrast, the more sterically demanding cyclic imides were unreactive under identical reaction conditions.

ABSTRACT

This study aimed to investigate the expression of arterial blood gas and acid-base balance in patients with pregnancy-induced hypertension syndrome and their influence on newborns. A total of 348 patients with pregnancy-induced hypertension syndrome (research group) admitted and treated in the First People's Hospital of Jining from March 2013 to March 2016 were retrospectively analyzed, and another 156 cases of healthy pregnant women who were physically examined were selected as the control group. Blood gas analysis was performed on pregnant women, and pH value, partial pressure of oxygen (PO2), partial pressure of carbon dioxide (PCO2), bicarbonate radical (HCO3-) and base excess (BE) were included as test indexes. Apgar score was made for newborns for neonatal asphyxia. The evaluation was performed at 1, 5, and 10 min after birth to analyze the correlation between the maternal arterial blood pH and the neonatal Apgar score. The pH value, and PO2 and HCO3- levels in the research group were lower than those in the control group (p<0.05). PCO2 and BE levels in the research group were higher than those in the control group (p<0.05). The incidence of neonatal mild asphyxia (Apgar score, 4-6 points) and neonatal severe asphyxia (Apgar score, <4 points) in the research group were higher than that in the control group (p<0.05). The Apgar score of newborns was positively correlated with the pH value of the arterial blood of pregnant women. Blood gas analysis in patients with pregnancy-induced hypertension syndrome is useful to a certain extent to evaluate the condition of the patient. The combination of the pH value of the patient and the Apgar score of the newborns may help improve the judgement rate of neonatal asphyxia so as to improve the quality of childbirth by early monitoring and intervention of the newborns.

ABSTRACT

We report on cyclic imides as weak directing groups for selective monohydroxylation reactions using ruthenium catalysis. Whereas acyclic amides are known to promote the hydroxylation of the C(sp2)-H bond enabling five-membered ring ruthenacycle intermediates, the cyclic imides studied herein enabled the hydroxylation of the C(sp2)-H bond via larger six-membered ruthenacycle intermediates. Furthermore, monohydroxylated products were exclusively obtained (even in the presence of overstoichiometric amounts of reagents), which was rationalized by the difficulty to accommodate coplanar intermediates once the first hydroxyl group was introduced into the substrate. The same reactivity was observed in the presence of palladium catalysts.

BACKGROUND: The aim of this study was to investigate the relationship of serum iron and ferritin concentrations in early pregnancy with gestational diabetes mellitus (GDM) complicated in pregnant metaphase through detecting the serum iron and ferritin concentrations in early pregnancy of normal pregnant women, so as to provide new ideas for the early detection and prevention of GDM. METHODS: Spontaneously pregnant women with single fetus receiving prenatal routine examination in our hospital from December 2014 to October 2016 were selected. They were in good health before with normal fasting blood-glucose in early pregnancy without anemia during pregnancy and any medication history. The serum iron and ferritin concentrations were measured at 12 gestational weeks, the fasting blood-glucose was detected at 12 weeks of pregnancy, and 75g oral glucose tolerance test was performed at 24-26 weeks. According to the results of oral glucose tolerance test, the pregnant women were divided into GDM group (N.=52) and normal control group (N.=310). The detection results of pregnant women in the two groups were statistically analyzed. RESULTS: The serum ferritin and iron levels at 12 gestational weeks in GDM group were higher than those in normal pregnancy group (P<0.05). The cut-off values of serum ferritin and iron at 12 gestational weeks in the prediction of GDM were 67.8 µg/L and 52.9 mmol/L. CONCLUSIONS: The high concentrations of serum iron and ferritin in early pregnancy have a certain correlation with the incidence of GDM, and the early detection of serum iron and ferritin levels can improve the detection rate of GDM in pregnant metaphase.

Ovarian carcinoma is the most lethal of the gynecologic malignancies worldwide. Increasing evidence suggests dysfunction of microRNAs (miRNAs) plays an important role in human cancers. The function of miR-222 was detected in ovarian carcinoma to verify the regulation of phosphatase and tensin homolog (PTEN) by miR-222. miR-222 expression in ovarian carcinoma tissues and cell lines were examined using RT-qPCR. Transwell assay was used to detect miR-222 effects on ovarian carcinoma cell migration and invasion. Western blot analysis and luciferase assays were performed to validate PTEN as miR-222 targets. miR-222 expression was upregulated in ovarian carcinoma tissues and three cell lines (A2780, SKOV-3 and OVCAR-3). Ectopic overexpression of miR-222 in ovarian carcinoma cells was sufficient to promote invasion and migration. PTEN acted as a direct target of miR-222. Overexpression of PTEN inhibited human ovarian carcinoma cell migration and invasion. In summary, our findings suggest that miR-222 plays an important role in promoting ovarian carcinoma cell invasion and migration and miR-222/PTEN may be a novel therapeutic target of miRNA-mediated promotion of cell invasion and migration in ovarian carcinoma.

ABSTRACT

A new visible-light-induced trifluoromethylation of isonitrile-substituted methylenecyclopropanes is developed. A range of substituted 6-(trifluoromethyl)-7,8-dihydrobenzo[k]phenanthridine derivatives are readily furnished by this newly developed tandem reaction with moderate to good yields. This reaction allows the direct formation of two six-membered rings and three new C-C bonds, including the C-CF3 bond, under visible light irradiation.

ABSTRACT

A novel intramolecular oxycyanation of methylenecyclopropanes is reported that proceeds through oxidative cleavage of the N-CN bond and subsequent palladium transfer from N to O of the amide group. A range of substituted benzo[d][1,3]oxazines with a cyano group are readily furnished by this newly developed oxycyanation reaction. Tris(4-trifluoromethylphenyl)phosphine as a ligand has been found to be crucial to effectively promote the transformation with high chemo- and regioselectivity. Moreover, the reaction outcome can be significantly affected by the electronic effect of the acyl group attached to the nitrogen atom of methylenecyclopropanes. When R(3) is a chloromethyl group, the pyrrolo[2,3-b]quinoline derivative is obtained by thermal-induced [3+2] cycloaddition of methylenecyclopropane to the methanediimine intermediate.

ABSTRACT

The chemoselective [4+2] vs. [2+2] cycloaddition between allenoates and dithioesters can be controlled by switching the nucleophilic amine catalyst. The two modes of cyclizations represent the first example of controllable and chemoselective annulations between allenoates and dienophiles catalyzed by amine. These cyclizations are useful in offering a divergent synthesis of sulfur-containing heterocycles. On the basis of this investigation, it can be realized that dithioesters with a vicinal electron-withdrawing group can react not only like a Michael acceptor but also as a ketone or imine.
