ABSTRACT

Background and Aim: Raw goat milk is a highly nutritious dairy product and a suitable medium for the growth of antimicrobial-resistant Staphylococcus aureus, the leading cause of subclinical mastitis. This study aimed to investigate the resistance status of S. aureus which isolated from goat milk associated with subclinical mastitis cases in Siliragung Subdistrict, Banyuwangi District, East Java, Indonesia. Materials and Methods: The S. aureus isolates were recovered from 258 raw goat milk samples from seven different dairy goat farms. Preliminary screening of subclinical mastitis using the California mastitis test, then samples with score +3 and +4 were taken for further isolation and identification, followed by a biochemical test to determine the S. aureus. Moreover, the bacteria susceptibility test against several antimicrobials was done using the disk diffusion method. Results: Based on our findings, a total of 66 (25.58%) raw goat milk samples were tested positive for S. aureus, of which 36.36% were identified as multidrug-resistant. Moreover, S. aureus were also identified as resistant to penicillin (81.82%), ampicillin (65.15%), erythromycin (50.52%), and gentamicin (36.09%). Conclusion: The prevalence of S. aureus isolated from raw goat milk associated with subclinical mastitis in Siliragung Subdistrict, Banyuwangi District, Indonesia, was recorded at 25.58%. Moreover, 36.36% of S. aureus isolates were categorized as resistant to three or more classes of antibiotics. The biosafety and biosecurity procedures during the milking process should be strengthened in dairy goat farms to prevent the transmission of antimicrobial resistance among animals, humans, and environments.

Background and Aim: Exotic pet snakes are more susceptible to infection, especially parasitic helminths than wild-caught. There is no comprehensive report on the prevalence of acanthocephalan parasite infection in Indonesian snakes. Therefore, this study aimed to estimate the prevalence rate and to identify the acanthocephalan infection in wild-caught Asian vine snake (Ahaetulla prasina) from the Mojokerto District, East Java, Indonesia. Materials and Methods: A total of 60 snakes were collected from the local sellers in the Mojokerto District, East Java, Indonesia. Then, snakes were euthanized and necropsied to observe various predilections of acanthocephalan larval stage (cystacanth). Morphological identification of the cystacanth was conducted using the carmine staining method and microscopic examination. Results: Acanthocephalan infection was recorded with a high prevalence rate of 80.06%. A total of 696 cystacanths were examined from the muscle, subcutaneous tissues, and visceral with 32.90, 16.37, and 50.71% intensity rates, respectively. Conclusion: Acanthocephalan prevalence rate was recorded at 80.06% in this study. Constant disease monitoring is necessary, considering wild-caught Asian vine snakes were susceptible host and lack of data regarding parasitological surveys. Therefore, further studies are needed in new areas and various species of wild-caught snakes in Indonesia, because of the potential of parasitic helminth transmission between snake and other reptiles.

Background and Aim: Surra is caused by Trypanosoma evansi. The detection method using conventional parasitological tests has not always shown positive results in blood parasite detection, although the livestock has presented with clinical signs. Therefore, a fast and accurate diagnosis is necessary to prevent the disease predominately in field isolates. This study aimed to investigate the sensitivity of molecular detection method using two different specific primers, namely, Internal Transcribed Spacer 1 (ITS-1) and Trypanosoma brucei repeat 1/2 (TBR-1/2) against T. evansi field isolates from Banten Province, Indonesia. Materials and Methods: The isolates of T. evansi used in this study were collected from Banten Province and cultured and preserved by the National Research Center for Veterinary Science, Indonesia. Eighteen experimental rats were divided into three equal groups, which were categorized as control, 1 × 101, and 1 × 104 infective doses. The isolates were injected into all experimental albino rats intraperitoneally. All samples were tested using conventional blood smear, card agglutination test (CATT), and polymerase chain reaction (PCR) method. Results: The results of the CATT examination in all treatments showed negative results. However, PCR results showed that two different primers, namely, ITS-1 and TBR-1/2 had been successfully detected T. evansi from infected experimental rats, proven by positive PCR band appeared in 480 base pairs (bp) and 164 bp, respectively. Conclusion: Based on the molecular diagnostic test using PCR method, TBR-1/2 primer is more sensitive to detect T. evansi compared to ITS-1 primer. The present finding provides preliminary data for studying the efficiency of different primers if practically applied as a standard diagnostic test for trypanosomiasis, especially in Indonesian livestock.

Background and Aim: In Indonesia, Madura cattle are native breeds that are expected to contribute to the improvement of regional meat self-sufficiency. Eimeria spp. are protozoans that are commonly found in ruminants. This study aimed to identify the occurrence and diversity of Eimeria spp. in Madura cattle. Materials and Methods: In this study, fresh fecal samples were collected from 100 cattle in Kamal Subdistrict, Bangkalan District, Madura Island, Indonesia. Morphological detection was performed using a light microscope, and molecular identification was performed using a polymerase chain reaction. DNA amplification was conducted using various species-specific primers for Eimeria bovis, Eimeria zuernii, Eimeria auburnensis, Eimeria alabamensis, Eimeria ellipsoidalis, and Eimeria cylindrica. Results: The results obtained 21% (21/100) of Eimeria spp. based on morphological detection. A total of 15 positive samples with 500-25,000/mL oocysts were selected for DNA extraction and amplification, resulting in 12 positive samples. Four Eimeria spp. were obtained based on molecular identification: E. bovis, E. zuernii, E. auburnensis, and E. cylindrica. Conclusion: Four species of Eimeria namely E. bovis, E. zuernii, E. auburnensis, and E. cylindrica were identified from fecal sample of Madura cattle using PCR method in this study. Further comprehensive studies are required to investigate the pathogenicity of Eimeria spp. in Madura cattle. Therefore, improved and integrated management practices should be strengthened by local governments to prevent pathogenic diseases and increase national livestock productivity in Indonesia.

BACKGROUND AND AIM: The high prevalence of sparganosis has been reported globally, especially in Asian countries where the majority of individuals consume raw meat from wild-caught reptiles. In Indonesia, similar cases regarding the high prevalence of sparganosis were recorded from wild reptiles such as snakes that utilized for culinary purposes, whereas, there are no data regarding other species such as water monitor lizard (Varanus salvator), which also provided as daily culinary with a high number of trades. Therefore, this study aims to investigate the prevalence of sparganosis in water monitor lizard (V. salvator), which is frequently utilized as culinary products in East Java Province, Indonesia. MATERIALS AND METHODS: A total of 313 living wild-caught and captive-bred Asian water monitor lizards were collected from the reptile markets and breeders. All samples were euthanized and observed for the presence of plerocercoid. Identification of the plerocercoid as larval infective stage was made using carmine staining method. RESULTS: The total prevalence of sparganosis was recorded at 69.64%. A total of 393 plerocercoids were collected in which divided 280 (71.24%) infecting muscles and 113 (28.75%) located in subcutaneous tissues. CONCLUSION: To the best of our knowledge, this study not only recorded as the first evidence but also confirms the role of monitor lizards as sparganosis transmitters in Asia and reveals additional routes of sparganosis transmission in Indonesian reptiles. Local conservation laws should be strengthened to effectively control or ban wildlife trade in traditional markets. Moreover, public awareness regarding sparganosis as a neglected zoonotic disease should be applied to prevent disease transmission in Indonesia.

ABSTRACT

AIM: Spirometra parasites cause sparganosis, a zoonotic disease, especially in reptiles and humans. This study aimed to report on the prevalence and effects of Spirometra parasites infection on public health and provide a scientific foundation for its prevention. MATERIALS AND METHODS: A total of 378 living Indonesian wild-caught and captive-bred Bronzeback snakes (Dendrelaphis pictus) were selected. The snakes were euthanized using ethyl ether anesthesia before checking for Spirometra parasites. The numbers of Spirometra located in the muscle tissue, subcutaneous tissue, and coelom (including the viscera) were each counted to investigate the distribution of Spirometra inside the snake body cavity. RESULTS: The total prevalence in the sample was 50.85%. The prevalence values in wild-caught and captive-bred snakes were 70.7% and 48.7%, respectively. More than half (56.6%) of the Spirometra parasites were located in the muscular tissue, while 29.5% were in the subcutaneous tissue and 13.8% were in the coelomic cavity. CONCLUSION: Wild-caught Indonesian Bronzeback snakes, which are sold as food in markets, and captive-bred snakes, which are collected as exotic pets in Indonesia, have similar opportunities to transmit the Spirometra parasite and cause global health problems due to their high prevalence.
