ABSTRACT

Liposarcoma (LPS) is one of the most common subtypes of sarcoma with a high recurrence rate. CENPF is a regulator of cell cycle, differential expression of which has been shown to be related with various cancers. However, the prognostic value of CENPF in LPS has not been deciphered yet. Using data from TCGA and GEO datasets, the expression difference of CENPF and its effects on the prognosis or immune infiltration of LPS patients were analyzed. As results show, CENPF was significantly upregulated in LPS compared to normal tissues. Survival curves illustrated that high CENPF expression was significantly associated with adverse prognosis. Univariate and multivariate analysis suggested that CENPF expression could be an independent risk factor for LPS. CENPF was closely related to chromosome segregation, microtubule binding and cell cycle. Immune infiltration analysis elucidated a negative correlation between CENPF expression and immune score. In conclusion, CENPF not only could be considered as a potential prognostic biomarker but also a potential malignant indicator of immune infiltration-related survival for LPS. The elevated expression of CENPF reveals an unfavorable prognostic outcome and worse immune score. Thus, therapeutically targeting CENPF combined with immunotherapy might be an attractive strategy for the treatment of LPS.

ABSTRACT

Carbapenemase-resistant Klebsiella pneumoniae (CR-KP) has become one of the nosocomial infections that seriously threaten the lives of patients, greatly increasing the burden on patients. In order to explore the resistance mechanism of clinically isolated CR-KP to carbapenems and perform multilocus sequence typing (MLST), to study the clinical characteristics of patients with different ST types of infection, we collected 74 CR-KP strains clinically isolated from the main 6 hospitals in Zhejiang province from January 2018 to July 2020. The sensitivity of the tested strains to 23 antibacterial drugs was determined by the microbroth dilution method, and PCR was applied. Gene amplification technology and DNA sequencing methods were used to detect the carbapenemase gene of the tested strains. Through the MLST of the tested strains, the clonal correlation and molecular epidemiological characteristics of the tested strains were explored, and the characteristics of CR-KP resistance, resistance mechanisms, and clinical characteristics of bacterial infections under different MLST types were analyzed at the same time. The results showed that 74 carbapenem-resistant Klebsiella pneumoniae strains showed high resistance to 21 commonly used antibacterial drugs, and all carbapenemase phenotypic screening tests were positive. MLST typing showed that 74 CR-KP strains had 17 ST typings, and ST11 was the dominant type (54.05%). The study also found that these ST11 strains are more likely to be resistant to carbapenem antibiotics. Most of them produce KPC carbapenemase, and a few are IMP, VIM, and NDM. Univariate analysis suggested that the proportion of patients in the ST11 group receiving treatment in ICU, the use rate of mechanical ventilation, and the proportion of drainage tube indwelling were higher than those in the non-ST11 group, and the survival rate of the ST11 group was lower than that of the non-ST11 group. Clinical data suggested that the same hospital was dominated by the same clonal epidemic in the same period. In view of the analysis of clinical data suggesting that patients who have received ICU treatment, mechanical ventilation, and drainage tube indwelling are prone to the risk of CR-KP strain (especially ST11) infection and low survival rate, such patients should arouse extensive clinical attention.

ABSTRACT

BACKGROUND: The recent advancement in massively parallel sequencing technologies has empowered liquid biopsies, in particular circulating tumor DNA (ctDNA) analysis, to be the new paradigm in personalized cancer management. Plasma ctDNA detection overcomes the current limitations in tumor tissue procurement and serves as a convenient and non-invasive method to capture tumor heterogeneity and genetic evolution along patients' cancer journey. In breast cancer, the current clinical application of ctDNA includes real-time monitoring of tumor response, detection of drug-resistant clones, assessing dynamic variations in tumor mutational landscape, identifying actionable mutations, detecting minimal residual disease and screening of early tumor. PURPOSE: This review aims to summarize the current clinical evidence of ctDNA application in the management of breast cancer.

ABSTRACT

OBJECTIVE: Delayed thrombolytic therapy with recombinant tissue plasminogen activator (tPA) may exacerbate blood-brain barrier (BBB) breakdown after ischemic stroke and lead to catastrophic hemorrhagic transformation (HT). Rosiglitazone(RSG), a widely used antidiabetic drug that activates peroxisome proliferator-activated receptor-γ (PPAR-γ), has been shown to protect against cerebral ischemia through promoting poststroke microglial polarization toward the beneficial anti-inflammatory phenotype. However, whether RSG can alleviate HT after delayed tPA treatment remains unknown. In this study, we sort to examine the role of RSG on tPA-induced HT after stroke. METHODS AND RESULTS: We used the murine suture middle cerebral artery occlusion (MCAO) models of stroke followed by delayed administration of tPA (10 mg/kg, 2 hours after suture occlusion) to investigate the therapeutic potential of RSG against tPA-induced HT. When RSG(6 mg/kg) was intraperitoneally administered 1 hour before MCAO in tPA-treated MCAO mice, HT in the ischemic territory was significantly attenuated 1 day after stroke. In the tPA-treated MCAO mice, we found RSG significantly mitigated BBB disruption and hemorrhage development compared to tPA-alone-treated stroke mice. Using flow cytometry and immunostaining, we confirmed that the expression of CD206 was significantly upregulated while the expression of iNOS was down-regulated in microglia of the RSG-treated mice. We further found that the expression of Arg-1 was also upregulated in those tPA and RSG-treated stroke mice and the protection against tPA-induced HT and BBB disruption in these mice were abolished in the presence of PPAR-γ antagonist GW9662 (4 mg/kg, 1 hour before dMCAO through intraperitoneal injection). CONCLUSIONS: RSG treatment protects against BBB damage and ameliorates HT in delayed tPA-treated stroke mice by activating PPAR-γ and favoring microglial polarization toward anti-inflammatory phenotype.

ABSTRACT

In this work, three Cu metal-organic framework samples with tunable rhombic, squama, and trucated bipyramid morphologies have been synthesized at 0, 25, and 60 °C, respectively, and further employed as precursors to initially prepare Cu@C composites by the calcination-thermolysis procedure. Then Cu@C composites have been etched with HCl and subsequently activated with KOH to obtain activated porous carbon (APC-0, -25, and -60). Interestingly, APC-25 presents a loose multilevel morphology of cabbage and possesses the largest specific surface area (1880.4 m2 g-1) and pore volume (0.81 cm3 g-1) among these APC materials. Consequently, APC-25 also exhibits the highest specific capacitance of 196 F g-1 at 0.5 A g-1, and the corresponding symmetric supercapacitor cell (SSC) achieves a remarkable energy density of 11.8 Wh kg-1 at a power density of 350 W kg-1. Furthermore, APC-25 shows excellent cycling stability, and the loss of capacitance is only 7.7% even after 10000 cycles at 1 A g-1. Significantly, five light-emitting diodes can be lit by six SSCs, which proves that APC-25 can be used in energy storage devices.

ABSTRACT

Interface engineering has been regarded as an effective and noninvasive means to optimize the performance of perovskite solar cells (PSCs). Here, doping engineering of a ZnO electron transport layer (ETL) and CsPbI3/ZnO interface engineering via introduction of an interfacial layer are employed to improve the performances of CsPbI3-based PSCs. The results show that when introducing a TiO2 buffer layer while increasing the ZnO layer doping concentration, the open-circuit voltage, power conversion efficiency, and fill factor of the CsPbI3-based PSCs can be improved to 1.31 V, 21.06%, and 74.07%, respectively, which are superior to those of PSCs only modified by the TiO2 buffer layer or high-concentration doping of ZnO layer. On the one hand, the buffer layer relieves the band bending and structural disorder of CsPbI3. On the other hand, the increased doping concentration of the ZnO layer improves the conductivity of the TiO2/ZnO bilayer ETL because of the strong interaction between the TiO2 and ZnO layers. However, such phenomena are not observed for those of a PCBM/ZnO bilayer ETL because of the weak interlayer interaction of the PCBM/ZnO interface. These results provide a comprehensive understanding of the CsPbI3/ZnO interface and suggest a guideline to design high-performance PSCs.

The effects of sewage irrigation on the growth of rice seedlings and soil environment under wheat straw returning were examined with a pot experiment. Root morphology, root activity, tiller number, plant height, dry matter accumulation of rice seedling, soil ferrous ions content, organic acid content and enzyme activity were measured. The results showed that sewage irrigation significantly increased the number of tillers and root activity at 41 days after transplanting under no N fertilizer application. Under the same N input level, sewage irrigation combined with N fertilizer promoted the growth of rice seedlings and root, and increased the root length, root surface area, root volume, root activity, tiller number and dry matter accumulation. Sewage irrigation significantly reduced the contents of soil ferrous ions and organic acid, while significantly increased the activities of soil urease and catalase. These results indicated that the combination of sewage irrigation and N fertilizer could effectively reduce the negative effect of straw returning on rice seedling and thus enhance soil fertility and quality.

ABSTRACT

Six isomorphous metal-organic frameworks (MOFs) with a 3D window-beam architecture have been synthesized from solvothermal reactions, and are named Zn, Cd, Ni, Co, Mn and Cu-MOF, respectively. The series of MOFs was utilized as precursors to synthesize MOF-derived carbon with different morphologies. Zn and Cd-MOFs lead to the derivation of porous carbons (PCs), which exhibit remarkable BET specific surface areas. For derivates of Ni, Co and Mn-MOFs, graphitized carbons (GCs) show some carbon graphitization, but their BET specific surface areas are relatively small. C-Cu has the smallest BET specific surface area, and there is no carbon graphitization. Therefore, the metal ion of the parent MOF exerts a crucial effect on the preparation of MOF-derived carbon, such as the pore-forming effect of Zn and Cd species, and catalytic graphitization of Ni, Co, and Mn species. The capacitances of MOF-derived carbon follow the sequence of PCs>GCs>C-Cu, which reveals that the specific surface area plays a dominant role in the capacitive performance of electrical double layer capacitors (EDLCs), and that the graphitization could improve the capacitance. Significantly, PC-Zn exhibits the best specific capacitance (138 F g-1 at 0.5 Ag-1 ), and excellent life cycle, which can be applied as an electrode material in supercapacitors.

ABSTRACT

Building a nutrient channel between eutrophic water and agricultural fields could reduce nutrient input into fields and alleviate eutrophication by returning nitrogen. In order to determine the feasibility of returning nitrogen by biochar loading, a rhizobox experiment was conducted with two nitrogen applied methods, namely SN (applied nitrogen by nitrogen fertilizer solution) and BN (applied nitrogen by nitrogen-loaded biochar). The results showed that BN, in comparison with SN, decreased the biomass and nitrogen uptake of the aboveground paddy by 16% and 14%, respectively, increased biomass root-shoot ratios by 25%-27%, and reduced nitrogen recovery use efficiency. Two nitrogen application methods affected the length and volume of paddy adventitious roots. Paddy underground biomass and nitrogen uptake were positively correlated with soil ammonium content, whereas paddy aboveground nitrogen uptake was negatively correlated with root tips. It was suggested that the paddy biomass and nitrogen uptake would be influenced when nitrogen was applied solely by nitrogen-loaded biochar. However, no affinity and no significance in nitrogen use efficiency were found for plant uptake between chemical nitrogen and biochar-loaded nitrogen. Additionally, biochar promoted soil mineral nitrogen content for further plant uptake. Therefore, biochar could be used as the carrier for returning nitrogen from waterbodies to fields. The replacement rate of chemical nitrogen fertilizer is the key to influencing plant growth and needs future study.

ABSTRACT

Increasing evidence suggests that the mesolimbic reward system plays critical roles in the regulation of depression and nociception; however, its circuitry and cellular mechanisms remain unclear. In this study, we investigated the output-specific regulatory roles of dopaminergic (DA) neurons within the ventral tegmental area (VTA) in depressive-like and nociceptive behaviors in mice subjected to unpredictable chronic mild stress (CMS), using the projection-specific electrophysiological recording, pharmacological manipulation, behavioral test, and molecular biology technologies. We demonstrated that CMS decreased the firing activity in VTA projecting to medial prefrontal cortex (VTA → mPFC), but not in VTA to nucleus accumbens (VTA → NAc), DA neurons. However, both VTA → mPFC and VTA → NAc DA neurons showed increased firing activity in response to morphine perfusion in CMS mice. Behavioral results showed that intra-VTA microinjection of morphine (25.5 ng/0.15 µL) relieved depressive-like behaviors, intriguingly, accompanied by a thermal hyperalgesia. Furthermore, the relief of depressive-like behaviors induced by intra-VTA injection of morphine in CMS mice could be prevented by blocking brain-derived neurotrophic factor (BDNF) signaling and mimicked by the administration of exogenous BDNF in mPFC rather than in NAc shell. Nociceptive responses induced by the activation of VTA DA neurons with morphine in CMS mice could be prevented by blocking BDNF signaling or mimicked by administration of exogenous BDNF in NAc shell, but not in mPFC. These results reveal projection-specific regulatory mechanisms of depression and nociception in the mesolimbic reward circuitry and provide new insights into the neural circuits involved in the processing of depressive and nociceptive information.

ABSTRACT

In this work, five new cadmium metal-organic frameworks (Cd-MOFs 1-5) have been synthesized from solvothermal reactions of Cd(NO3 )2 ⋅4 H2 O with isophthalic acid and 1,4-bis(imidazol-1-yl)-benzene under different solvent systems of CH3 OH, C2 H5 OH, (CH3 )2 CHOH, DMF, and N-methyl-2-pyrrolidone (NMP), respectively. Cd-MOF 1 shows a 3D diamondoid framework with 1D rhombic and hexagonal channels, and the porosity is 12.9 %. Cd-MOF 2 exhibits a 2D (4,4) layer with a 1D parallelogram channel and porosity of 23.6 %. Cd-MOF 3 has an 8-connected dense network with the Schäfli symbol of [424 ⋅64 ] based on the Cd6 cluster. Cd-MOFs 4-5 are isomorphous, and display an absolutely double-bridging 2D (4,4) layer with 1D tetragonal channels and porosities of 29.2 and 28.2 %, which are occupied by DMF and NMP molecules, respectively. Followed by the calcination-thermolysis procedure, Cd-MOFs 1-5 are employed as precursors to prepare MOF-derived porous carbon materials (labeled as PC-me, PC-eth, PC-ipr, PC-dmf and PC-nmp), which have the BET specific surface area of 23, 51, 10, 122, and 96 m2 g-1 , respectively. The results demonstrate that the specific surface area of PCs is tuned by the porosity of Cd-MOFs, where the later is highly dependent on the solvent. Thereby, the specific surface area of PCs could be adjusted by the solvent used in the synthese of MOF precusors. Significantly, PCs have been further activated by KOH to obtain activated carbon materials (APCs), which possess even higher specific surface area and larger porosity. After a series of characterization and electrochemical investigations, the APC-dmf electrode exhibits the best porous properties and largest specific capacitances (153 F g-1 at 5 mV s-1 and 156 F g-1 at 0.5 Ag-1 ). Meanwhile, the APC-dmf electrode shows excellent cycling stability (ca. 84.2 % after 5000 cycles at 1 Ag-1 ), which can be applied as a suitable electrode material for supercapacitors.

Herein three novel cobalt metal-organic frameworks (Co-MOFs) with similar ingredients, [Co(bib)(o-bdc)]∞ (1), [Co2(bib)2(m-bdc)2]∞ (2), and {[Co(bib)(p-bdc)(H2O)](H2O)0.5}∞ (3), have been synthesized from the reaction of cobalt nitrate with 1,4-bis(imidazol-1-yl)benzene (bib) and structure-related aromatic acids (1,2-benzenedicarboxylic acid = o-bdc, 1,3-benzenedicarboxylic acid = m-bdc, and 1,4-benzenedicarboxylic acid = p-bdc) by the solvothermal method. It is aimed to perform systematic research on the relationship among the conformation of benzoate acid, lattice dimension of Co-MOF, and pore diameter of MOF-derived carbon composite. Through the precursor strategy, Co-MOFs 1-3 have been utilized to synthesize porous cobalt@carbon nanotube composites (Co@CNTs). After the in situ gas-sulfurization, secondary composites CoS2@CNTs were successfully obtained, which kept similar morphologies of corresponding Co@CNTs without destroying previous highly dispersed structures. Co-MOFs and two series of composites (Co@CNTs and CoS2@CNTs) have been well characterized. Topology and Brunauer-Emmett-Teller analyses elucidate that the bdc2- ion could control the pore diameters of MOF-derived carbon composites by adjusting the lattice dimension of Co-MOFs. The systematic studies on electrochemical properties demonstrate that (p)-CoS2@CNT possesses hierarchical morphology, moderate specific surface area, proper pore diameter distribution, and high graphitization, which lead to remarkable specific capacitances (839 F g-1 at 5 mV s-1 and 825 F g-1 at 0.5 A g-1) in 2 M potassium hydroxide solution. In addition, the (p)-CoS2@CNT electrode exhibits good electrochemical stability and still retains 82.9% of initial specific capacitance at the current density of 1 A g-1 after 5000 cycles.

ABSTRACT

The objectives were (i) to conduct an overview of systematic reviews to summarize evidence from and evaluate the methodological quality of systematic reviews assessing the impact of ambient temperature on morbidity and mortality; and (ii) to reanalyse meta-analyses of cold-induced cardiovascular morbidity in different age groups. The registration number is PROSPERO-CRD42016047179. PubMed, Embase, the Cochrane Library, Web of Science, the Cumulative Index to Nursing and Allied Health Literature (CINAHL), and Global Health were systematically searched to identify systematic reviews. Two reviewers independently selected studies for inclusion, extracted data, and assessed quality. The Assessment of Multiple Systematic Reviews (AMSTAR) checklist was used to assess the methodological quality of included systematic reviews. Estimates of morbidity and mortality risk in association with heat exposure, cold exposure, heatwaves, cold spells and diurnal temperature ranges (DTRs) were the primary outcomes. Twenty-eight systematic reviews were included in the overview of systematic reviews. (i) The median (interquartile range) AMSTAR scores were 7 (1.75) for quantitative reviews and 3.5 (1.75) for qualitative reviews. (ii) Heat exposure was identified to be associated with increased risk of cardiovascular, cerebrovascular and respiratory mortality, but was not found to have an impact on cardiovascular or cerebrovascular morbidity. (iii) Reanalysis of the meta-analyses indicated that cold-induced cardiovascular morbidity increased in youth and middle-age (RR=1.009, 95% CI: 1.004-1.015) as well as the elderly (RR=1.013, 95% CI: 1.007-1.018). (iv) The definitions of temperature exposure adopted by different studies included various temperature indicators and thresholds. In conclusion, heat exposure seemed to have an adverse effect on mortality and cold-induced cardiovascular morbidity increased in the elderly. Developing definitions of temperature exposure at the regional level may contribute to more accurate evaluations of the health effects of temperature.

Cd-MOF 1 shows a 2D (4,4) lattice with parallel interpenetration, whereas 2 displays an interesting 3D interpenetrating dia network, 3 exhibits an unusual 3D interpenetrating dmp network, and 4 presents a 3D self-catenated pillar-layered framework with a Schäfli symbol of [43 ⋅63 ]2 ⋅[46 ⋅616 ⋅86 ]. The structural diversity indicates that the backbone of the bis(imidazole) ligand (including the terminal group and spacer) plays a crucial role in the assembly of mixed-ligand frameworks. By using the pore-forming effect of cadmium vapor, for the first time we have utilized these Cd-MOFs as precursors to further prepare porous carbon materials (PCs) in a calcination-thermolysis procedure. These PCs show different porous features that correspond to the topological structures of Cd-MOFs. Significantly, it was found that the specific surface area and capacitance of PCs are tuned by the Cd/C ratio of the MOF. Furthermore, the as-synthesized PCs were processed with KOH to obtain activated porous carbon materials (APCs) with higher specific surface area and porosity, which greatly promoted the energy-storage capacity. After full characterization, we found that APC-bib displays the largest specific surface area (1290 m2 g-1 ) and total pore volume (1.37 cm3 g-1 ) of this series of carbon materials. Consequently, APC-bib demonstrates the highest specific capacitance of 164 F g-1 at a current density of 0.5 A g-1 , and also excellent retention of capacitance (≈89.4 % after 5000 cycles at 1 A g-1 ). Therefore, APC-bib has great potential as the electrode material in a supercapacitor.
