ABSTRACT

To prepare a lipid nanoparticle (LNP)-based subunit vaccine of Mycobacterium tuberculosis (Mtb) antigen EsxV and study its immunological characteristics, the LNP containing EsxV and c-di-AMP (EsxV: C: L) was prepared by thin film dispersion method, and its encapsulation rate, LNP morphology, particle size, surface charge and polyphase dispersion index were measured. BALB/c mice were immunized with EsxV: C: L by nasal drops. The levels of serum and mucosal antibodies, transcription and secretion of cytokines in lung and spleen, and the proportion of T cell subsets were detected after immunization. EsxV: C: L LNPs were obtained with uniform size and they were spherical and negatively charged. Compared with EsxV: C immunization, EsxV: C: L mucosal inoculation induced increased sIgA level in respiratory tract mucosa. Levels of IL-2 secreted from spleen and ratios of memory T cells and tissue-resident T cells in mice were also elevated. In conclusion, EsxV: C: L could induce stronger mucosal immunity and memory T cell immune responses, which may provide better protection against Mtb infection.

ABSTRACT

Chemotherapy resistance of cancer mainly has innate resistance and acquired drug resistance, which is mainly caused by the gene mutation induced by chemotherapy drugs. Alternative splicing of CD44 has been shown to be associated with the occurrence and development of tumors especially the multidrug resistance, but the underlying mechanism of CD44 in drug resistance remains unclear. Previous studies have found CD44 variants could significantly improve tumor resistance, therefore, we hypothesized the CD44v16, a CD44 variant found in our lab, might confer drug resistance in K562 cell. Through lentiviral expression system, the CD44v16 gene was introduced into the sensitive K562 cell, and the result indicated it could remarkably improve the drug resistance index. Real-time PCR and western blot experiments showed the expressions of protein p65, Bcl-2, Snail, and IKKß in CD44v16-positive cell group were higher than the CD44v16-negative group. Therefore, we suggest the CD44v16 could induce the K562 cell acquired with resistance to adriamycin via NF-κB/Snail/Bcl-2 pathway, which paved the way for further study the function of CD44v16 in drug resistance.

ABSTRACT

Objective To analyse the expression and significance of CD44 variant (CD44v17) in the process of cervical intraepithelial neoplasia (CIN) to cervical cancer,and to explore the relationship of CD44v17 with CIN and cervical cancer.Methods The expressions of CD44v17,matrix metalloproteinase 9 (MMP9) and Ki67 in normal cervical tissue,CIN Ⅰ,Ⅱ,Ⅲ and cervical carcinoma tissues were detected by real-time quantitative RCR.The expressions of CD44v17,MMP9 and Ki67 in human cervical cancer cell lines HeLa and SiHa after transfecting with CD44v17 siRNA were detected,as well.The tumor-bearing nude mice were treated with lentivirus particles of CD44v17,and the influence of CD44v17 siRNA on tumorigenicity in nude mice was analysed.Results The expression level of CD44v17 increased gradually from CIN Ⅰ,Ⅱ,Ⅲ to cervical carcinoma tissues.Compared with normal cervical tissue,the expression levels of CD44v1 7,MMP9 and Ki67 were significantly increased in CIN Ⅱ,Ⅲ and cervical carcinoma tissues (P<0.05),while no significant difference was found between normal cervical tissue and CIN Ⅰ tissue (P>0.05).The expression levels of MMP9 and Ki67 in human cervical cancer cell lines HeLa and SiHa were decreased significantly after transfecting with CD44v17 siRNA (P<0.01).The tumor volume and weight were decreased by treatment with CD44v17 siRNA transfection (P<0.01),and the tumorigenicity in nude mice was decreased.Conclusion CD44v17 may promote the development of CIN to cervical cancer.It is expected to be an indicator in predicting malignant potential of CIN.

ABSTRACT

Objective To study the effects of core control training on gross motor function of children with spastic cerebral palsy. Meth-ods 60 children with spastic cerebral palsy were divided into control group (n=30) and treatment group (n=30). The control group accepted routine comprehensive rehabilitation therapy including physical therapy, acupuncture, electrotherapy, etc., while the treatment group accept-ed the core control training in addition. They were assessed with the Gross Motor Function Measure (GMFM-88) and the Trunk Control Test (TCT) before and 3 months after rehabilitation. Results Both groups improved in the scores of GMFM-88 and TCT after treatment (P<0.001), and improved more in the treatment group than in the control group (P<0.001). Conclusion The core control training can strengthen the trunk control and further improve the gross motor function in children with spastic cerebral palsy.

@#Objective To investigate the methods and effectiveness of the physical therapy team management mode.MethodsAccording to the type of cerebral palsy and a certain ratio of beds and staff, the therapists were divided into 3 groups, and relatively fixed. They treated children with differrent types of CP and different beds by turn. The implementation of treatment was 3 years.ResultsAfter the physical therapy team management mode was executed, the therapist's theoretical knowledge and technical skill were upgraded, patient satisfaction raised, and the occurrence of accidental injury reduced.ConclusionThe physical therapy team management mode is conducive to standardize therapist behavior, improve the capacity of theory and practice of therapists and promote patient recovery.

ABSTRACT

@#Objective To observe the effect of the body weight support training (BWST) on walking ability of children with cerebral palsy (CP). Methods 50 children with CP were divided into the BWST group (n=30) and control group(n=20). All cases were treated with physical therapy, but the BWST group were added with BWST. The scores of D and E dimensions of the Gross Motor Function Measure Scale (GMFM) were used to assess the walking ability before and after the treatment, respectively. Results The scores of D and E dimensions of GMFM improved in both groups after treatment (P<0.01), and that of the BWST group improved more than control group (P<0.05). Conclusion BWST can facilitate the recovery of the walking ability after cerebral palsy.
