ABSTRACT

BACKGROUND:Adipose stem cells can differentiate into bone cells, cartilage cells, smooth muscle cells, myocardial cells and nerve cells. Adipose-derived stem cells and mesenchymal stem cells have very similar biological properties, and they are also very close in terms of cellsurface marker expression profile. OBJECTIVE:To investigate the expression of human leukocyte antigen II in adipose-derived stem cells and its immunogenicity and immunomodulatory effect, and to explore the specific mechanism. METHODS:After isolation and culture, the adipose-derived stem cells were stimulated with interferon-γ, and the expression of human leukocyte antigen II was detected by flow cytometry, the expression of indoleamine 2,3-dioxygenase was determined by real-time PCR and western blot. Adipose-derived stem cells were used to stimulate lymphocytes, and the cellproliferation was observed. The lymphocytes were stimulated with phytohemagglutinin, and then 1×102-1×105 adipose-derived stem cells were added to inhibit the proliferation;meanwhile, adipose-derived stem cells, a third party, at a dose of 1×102-1×105 were added to the ongoing two-way mixed lymphocyte reaction, and the cellnumber was calculated as count per minute. RESULTS AND CONCLUSION:The expression of human leukocyte antigen II and indoleamine 2,3-dioxygenase was low in adipose-derived stem cells, but up-regulated with stimulation of interferon-γ. Adipose-derived stem cells could not stimulate lymphocyte proliferation, but could inhibit lymphocyte proliferation induced by phytohemagglutinin or al ogeneic lymphocytes, and interferon-γwas highly expressed in the supernatant of co-culture of adipose-derived stem cells and mixed lymphocytes. These results indicate that the adipose-derived stem cells have low immunogenicity and can inhibit cellproliferation of lymphocytes in vitro, and the addition of exogenous interferon-γcan enhance the immunosuppressive effects of adipose-derived stem cells.

ABSTRACT

Objective To study the indication,feasibility and short-term efficacy of combined thoracoscopic and laparoscopic radical esophagectomy for the treatment of esophageal cancer.Methods Retrospective medical records analysis was conducted for 139 esophageal cancer patients who underwent combined thoracoscopic and laparoscopic esophagectomy in our department from December 2009 to August 2011.The tumors were located in upper esophagus in 16 cases,middle esophagus in 107 cases,and lower esophagus in 16 cases.The surgery started with the thoracoscopic mobilization of thoracic esophagus and lymph nodes dissection,which were followed by the laparoscopic stomach mobilization and gastroesophageal anastomosis in left neck.Postoperative pathological staging identified stage Ⅰ esophageal cancer in 25 cases ( stage Ⅰ a:13 cases,stage Ⅰ b:12 cases),stage Ⅱ esophageal cancer in 71 cases,stage Ⅲ esophageal cancer in 31 cases ( stage Ⅲ a:16 cases,stage Ⅲ b:15 cases) and stage Ⅳ esophageal cancer in 12 cases.Results Except for open conversions in 4 cases (2.9%),all surgical operations were completed smoothly.Postoperative anastomotic leak was found in 6 cases(4.3% ),chylothorax in 1 case(0.7% ),arrhythmia in 4 cases(2.9% ),and dumping syndrome in 1 case( 0.7% ).All of these complicated cases fully recovered after conservative treatments.Postoperative lung infection was found 11 cases (7.9%),3 of whom required tracheotomy and assisted ventilation and 1 case died as a result of the infection (mortality rate:0.7% ).Ten cases(7.2% ) presented with hoarseness postoperatively.Out of the 139 cases,130 cases were successfully followed up with durations ranged from 1 to 20 months,during of time the esophageal cancer spread to liver in 2 cases,celiac lymph nodes in 4 cases,lung in 2 cases,and bone in 1 case.Ten cases died,and all remaining cases remained alive during the follow up.The one-year survival rate was 88.9% for these cases.Conclusion Combined thoracoscopic and laparoscopic radical esophagectomy is a technically safe and feasible treatment for esophageal cancer.The short-term efficacy results are satisfactory.This technique is indicated not only for early and middle stage esophageal cancer,but also for some of the advanced esophageal cancer cases.

ABSTRACT

The multidimensional transfer function is a flexible and effective tool for exploring volume data. However, designing an appropriate transfer function is a trial-and-error process and remains a challenge. In this paper, we propose a novel volume exploration scheme that explores volumetric structures in the feature space by modeling the space using the Gaussian mixture model (GMM). Our new approach has three distinctive advantages. First, an initial feature separation can be automatically achieved through GMM estimation. Second, the calculated Gaussians can be directly mapped to a set of elliptical transfer functions (ETFs), facilitating a fast pre-integrated volume rendering process. Third, an inexperienced user can flexibly manipulate the ETFs with the assistance of a suite of simple widgets, and discover potential features with several interactions. We further extend the GMM-based exploration scheme to time-varying data sets using an incremental GMM estimation algorithm. The algorithm estimates the GMM for one time step by using itself and the GMM generated from its previous steps. Sequentially applying the incremental algorithm to all time steps in a selected time interval yields a preliminary classification for each time step. In addition, the computed ETFs can be freely adjusted. The adjustments are then automatically propagated to other time steps. In this way, coherent user-guided exploration of a given time interval is achieved. Our GPU implementation demonstrates interactive performance and good scalability. The effectiveness of our approach is verified on several data sets.

Gastrointestinal stromal tumor (GIST) is the most common mesenchymal tumors of the diges-tive tract. Along with histology progress, the people have made the new progress regarding the mesenchy-moma understanding, also proposed the new viewpoint to its treatment, the therapy of GIST has changed from surgical resection alone to surgery after the application of tyrosine kinase inhibitor. The article reviews the therapy of GIST and proposes possible therapeutic regimen.

ABSTRACT

Objective To study the protective effect of total flavone from Rhododendron simsii(TFRS) on myocardial ischemia-reperfusion injury rats and its mechanism.Methods The ischemic model was made by occluding the anterior descending of the left artery(LAD) in rats.The change of ST segment and T wave of electrocardiograph(ECG) were observed,and the activity of lactate dehydrogenase(LDH),creatine kinase(CK),levels of the maleic dialdehyde(MDA),and nitric oxide(NO) in serum were measured.And by tetrazolium chloride(TTC) staining,the areas of myocardial infarction were observed.The expression of inducible nitricoxide synthase(iNOS) in rats was detected by emploring the reverse transcription-polymerase chain reaction(RT-PCR) technique.Results On the myocardial infarction model by occluding the anterior descending of the LAD in rats,TFRS(100 mg/kg) obviously reduced the height of ST segment after occluding 30 min and TFRS(25,50,and 100 mg/kg) obviously reduced the height of ST segment after reperfusion 30 min.TFRS(50 mg/kg) reduced by myocardial infarction area.TFRS(50 and 100 mg/kg) obviously reduced the activity of CK and LDH.TFRS(50 mg/kg) decreased the level of MDA in serum.By RT-PCR technique,it was found that the expression of iNOS mRNA in myocardium in IR rats pretreated with TFRS(100 mg/kg) was higher than that in IR and normal groups.Conclusions TFRS has the significant protection against myocardial ischemia-reperfusion injury via atte-nuating oxygen free radical and increasing the expression of iNOS mRNA and NO production.
