ABSTRACT

This study investigated the hypothesis that dietary supplementation of lignocellulose in broilers influences the gut bacterial population and bacterial fermentation, has anti-inflammatory effects, and increases mucin synthesis in the intestine, and, through these changes, influences broiler performance positively. Day-old male Cobb 500 broilers (n = 96) were allotted to 3 experimental groups and fed 3 different maize-wheat-soybean meal-based basal diets during days 1 to 10, 11 to 21, and 22 to 35. The basal diets were fed to the control group, and were supplemented with 0.8% of a standard lignocellulose (LCS) or a fermentable lignocellulose (LCF). Body weight and feed consumption were determined, and at slaughter (day 35), carcass and gizzard weights and gizzard content pH were recorded, and samples of jejunum, cecum, and colon mucosa and of cecum digesta were collected from 15 birds/group. Growth performance and feed intake were not influenced, but dressing percentage was higher in group LCF compared to the other groups. In group LCS and the control group, performance, gizzard weight and gizzard content pH, intestinal gene expression of pro-inflammatory cytokines and of the mucins 2, 5ac and 13, the cecal short-chain fatty acid (SCFA) profile, and bacterial diversity were similar, and relative abundance of bacterial groups (16S DNA sequencing) differed. Supplementation of LCF decreased the expression of the pro-inflammatory genes encoding interleukins 1ß and 17 (P < 0.05) and those of 2 and 8 (P < 0.10) in the jejunum only. The bacterial population differed, and the SCFA profile shifted toward acetate at the expense of butyrate in group LCF compared to the control group. For example, the abundance of Firmicutes and of Ruminococcaceae and Lactobacillaceae decreased, whereas those of Peptostreptococcaceae, Erysipelotrichaceae, and Enterobacteriaceae and that of members of the phylum Proteobacteria increased in group LCF compared to the control group. These data indicate that the susceptibility of lignocellulose to fermentation is crucial for mediating its effects on intestinal gene expression and the bacterial population in the cecum, which may also affect dressing percentage.

ABSTRACT

In dairy cows, administration of high dosages of niacin (nicotinic acid, NA) was found to cause antilipolytic effects, which are mediated by the NA receptor hydroxyl-carboxylic acid receptor 2 (HCAR2) in white adipose tissue (WAT), and thereby an altered hepatic lipid metabolism. However, almost no attention has been paid to possible direct effects of NA in cattle liver, despite evidence that HCAR2 is also expressed in the liver and is even more abundant than in WAT. Because of this, we hypothesized that feeding a high dosage of rumen-protected NA to dairy cows influences critical metabolic or signaling pathways in the liver by inducing changes in the hepatic transcriptome. To identify these pathways, we applied genome-wide transcript profiling in liver biopsies obtained at d 7 postpartum (p.p.) from dairy cows used in our recent study; cows received either no NA (control group, n = 9) or 79 mg of rumen-protected NA/kg of body weight daily (NA group, n = 9) from 21 d before calving until 3 wk p.p. Hepatic transcript profiling revealed that 487 transcripts were differentially expressed (filter criteria: fold change >1.2 or <-1.2 and P < 0.05) in the liver at d 7 p.p. between cows fed NA and control cows. Substantially more transcripts were downregulated (n = 338), whereas only 149 transcripts were upregulated by NA in the liver of cows. Gene set enrichment analysis for the upregulated transcripts revealed that the most-enriched gene ontology biological process terms were exclusively related to immune processes, such as leukocyte differentiation, immune system process, activation of immune response, and acute inflammatory response. Gene set enrichment analysis of the downregulated transcripts showed that the most-enriched biological process terms were related to metabolic processes, such as cellular metabolic process, small molecule metabolic process, lipid catabolic process, organic cyclic compound metabolic process, small molecule biosynthetic process, and cellular lipid catabolic process. In conclusion, hepatic transcriptome analysis showed that rumen-protected NA induces genes that are involved mainly in immune processes, including acute phase response and stress response, in dairy cows at d 7 p.p. Thus, supplementation of a high dosage of rumen-protected NA to dairy cows in the periparturient period may induce or amplify the systemic inflammation-like condition that is typically observed in the liver of high-yielding dairy cows in the p.p. period.

ABSTRACT

Nicotinic acid (NA) has been shown to induce muscle fiber switching toward oxidative type I fibers and a muscle metabolic phenotype that favors fatty acid (FA) utilization in growing rats, pigs, and lambs. The hypothesis of the present study was that supplementation of NA in cows during the periparturient phase also induces muscle fiber switching from type II to type I fibers in skeletal muscle and increases the capacity of the muscle to use free FA, which may help to reduce nonesterified fatty acid (NEFA) flow to the liver, liver triglyceride (TG) accumulation, and ketogenesis. Thirty multiparous Holstein dairy cows were allocated to 2 groups and fed a total mixed ration without (control group) or with ∼55 g of rumen-protected NA per cow per day (NA group) from 21 d before expected calving until 3 wk postpartum (p.p.). Blood samples were collected on d -21, -14, -7, 7, 14, 21, 35, and 63 relative to parturition for analysis of TG, NEFA, and ß-hydroxybutyrate. Muscle and liver biopsies were collected on d 7 and 21 for gene expression analysis and to determine muscle fiber composition in the musculus semitendinosus, semimembranosus, and longissimus lumborum by immunohistochemistry, and liver TG concentrations. Supplementation of NA did not affect the proportions of type I (oxidative) or the type II:type I ratio in the 3 muscles considered. A slight shift from glycolytic IIx fibers toward oxidative-glycolytic fast-twitch IIa fibers was found in the semitendinosus, and a tendency in the longissimus lumborum, but not in the semimembranosus. The transcript levels of the genes encoding the muscle fiber type isoforms and involved in FA uptake and oxidation, carnitine transport, tricarboxylic acid cycle, oxidative phosphorylation, and glucose utilization were largely unaffected by NA supplementation in all 3 muscles. Supplementation of NA had no effect on plasma TG and NEFA concentrations, liver TG concentrations, and hepatic expression of genes involved in hepatic FA utilization and lipogenesis. However, it reduced plasma ß-hydroxybutyrate concentrations in wk 2 and 3 p.p. by 18 and 26% and reduced hepatic gene expression of fibroblast growth factor 21, a stress hormone involved in the regulation of ketogenesis, by 74 and 56%. In conclusion, a high dosage of rumen-protected NA reduced plasma ß-hydroxybutyrate concentrations in cows during early lactation, but failed to cause an alteration in muscle fiber composition and muscle metabolic phenotype.

ABSTRACT

This study hypothesized that plasma and tissue antioxidant status of broilers is positively influenced when dietary Met concentrations exceed, and negatively when they go below NRC recommendations. In addition, different Met sources are hypothesized to affect the antioxidant defence system differently. Day-old male Cobb-500 broilers (n = 336) were allotted to seven groups and phase-fed three wheat-soya bean meal-based basal diets during days 1-10, 11-21 and 22-35. The basal diets (Met- group, Met + Cys concentration 15% below NRC recommendations) were supplemented with 0.10%, 0.25% or 0.40% Met either as DL-Met (DLM) or DL-2-hydroxy-4-(methylthio) butanoic acid (DL-HMTBA) (equimolar comparison). Growth performance and carcass weights were lower in the Met- group compared to the groups whose diets met or exceeded Met requirements. The antioxidant defence system was not influenced by the Met source. However, in the liver, concentrations of glutathione increased with increasing dietary Met concentrations. Tocopherol concentrations in the liver at days 10 and 21 were lower in the Met- group than in the groups supplemented with Met. However, liver concentrations of thiobarbituric acid reactive substances (TBA-RS) and protein carbonyls (PC) were largely not influenced by dietary Met concentration. Plasma tocopherol concentrations at day 35 were lower, and those of TBA-RS and PC at day 35 were higher in Met- group than in the groups fed the Met-supplemented diets. In jejunum, but not in liver, relative mRNA abundances and activities of superoxide dismutase, catalase and glutathione peroxidase were higher in the Met- group than in the groups fed Met-supplemented diets. These data indicate that suboptimum supply of Met results in decreased antioxidant concentrations in plasma and body tissues, and increases oxidative stress in the jejunum mucosa. However, supplementation of Met in excess of the requirements (based on NRC) compared to diets adequate in Met + Cys did not influence the antioxidant defence system.

ABSTRACT

The changes taking place with age in energy turnover of dairy cattle are largely unknown. It is unclear whether the efficiency of energy utilization in digestion (characterized by faecal and methane energy losses) and in metabolism (characterized by urine and heat energy losses) is altered with age. In the present study, energy balance data were obtained from 30 lactating Brown Swiss dairy cows aged between 2 and 10 years, and 12 heifers from 0.5 to 2 years of age. In order to evaluate a possible dependence of age effects on diet type, half of the cattle each originated from two herds kept at the same farm, which were fed either on a forage-only diet or on the same forage diet but complemented with 5 kg/day of concentrate since their first calving. During 2 days, the gaseous exchange of the animals was quantified in open-circuit respiration chambers, followed by an 8-day period of feed, faeces, urine and milk collection. Daily amounts and energy contents were used to calculate complete energy balances. Age and feeding regime effects were analysed by parametric regression analysis where BW, milk yield and hay proportion in forage as consumed were considered as covariates. Relative to intake of gross energy, the availability of metabolizable energy (ME) increased with age. This was not the result of an increasing energy digestibility, but of proportionately lower energy losses with methane (following a curvilinear relationship with the greatest losses in middle-aged cows) and urine (continuously declining). The efficiency of utilization of ME for milk production (k l) increased with age. Potential reasons include an increase in the propionate-to-acetate ratio in the rumen because of a shift away from fibre degradation and methane formation as well as lower urine energy losses. The greater k l allowed older cows to accrete more energy reserves in the body. As expected, offering concentrate enhanced digestibility, metabolizability and metabolic utilization of energy. Age and feeding regime did not interact significantly. In conclusion, older cows seem to have digestive and metabolic strategies to use dietary energy to a certain degree more efficiently than younger cows.

ABSTRACT

The absorption and metabolism of vitamin A is linked with that of lipids. It is known that conjugated linoleic acid (CLA) affects the lipid metabolism in growing and lactating animals. In the present study, the hypothesis was investigated that dietary CLA influences vitamin A status of lactating rats and their pups during the suckling period. For this purpose, Wistar Han rats were fed either a control diet (control group, n = 14) or a diet containing 0.87% of cis-9, trans-11 and trans-10, cis-12 (1:1) CLA (CLA group, n = 14) during pregnancy and lactation. Vitamin A concentrations in various body tissues were determined 14 days after delivery in dams and 1, 7 and 14 days after birth in pups, and expression of selected genes involved in metabolism of retinoids was determined in dams. Vitamin A concentrations in liver, plasma and muscle were similar in control and CLA-fed dams. Expression of genes involved in retinoid transport, storage and degradation in liver and adipose tissue in dams was also not different between control and CLA-fed dams. Vitamin A concentrations in milk curd, sampled at d 1, 7 and 14 of lactation were not different between control and CLA-fed dams. Vitamin A concentrations in liver, lung and adipose tissue were also not different in pups from control dams and pups from CLA-fed dams. In conclusion, we show for the first time that dietary CLA has little effect on vitamin A concentrations and vitamin A metabolism in lactating rat dams and, moreover, does not influence tissue vitamin A concentrations in their newborn and suckling pups.

ABSTRACT

In response to increasing efforts for reducing concentrate inputs to organic dairy production in grassland-rich areas of Europe, a long-term study was conducted, which assessed the impacts of concentrate reductions on cows' performance, health, fertility and average herd age. In total, 42 Swiss commercial organic dairy cattle farms were monitored over 6 years ('Y0', 2008/09 until 'Y5', 2013/14). In comparison with overall data of Swiss herdbooks (including conventional and organic farms), the herds involved in the project had lower milk yields, similar milk solids, shorter calving intervals and higher average lactation numbers. During the first 3 project years farmers reduced the concentrate proportion (i.e. cereals, oilseeds and grain legumes) in the dairy cows' diets to varying degrees. In Y0, farms fed between 0% and 6% (dietary dry matter proportion per year) of concentrates. During the course of the study they changed the quantity of concentrates to voluntarily chosen degrees. Retrospectively, farms were clustered into five farm groups: Group '0-conc' (n=6 farms) already fed zero concentrates in Y0 and stayed at this level. Group 'Dec-to0' (n=11) reduced concentrates to 0 during the project period. Groups 'Dec-strong' (n=8) and 'Dec-slight' (n=12) decreased concentrate amounts by >50% and <50%, respectively. Group 'Const-conc' (n=5 farms) remained at the initial level of concentrates during the project. Milk recording data were summarised and analysed per farm and project year. Lactation number and calving intervals were obtained from the databases of the Swiss breeders' associations. Dietary concentrate amounts and records of veterinary treatments were obtained from the obligatory farm documentations. Data were analysed with GLMs. Daily milk yields differed significantly between farm groups already in Y0, being lowest in groups 0-conc (16.0 kg) and Dec-to0 (16.7 kg), and highest in groups Dec-slight (19.6 kg) and Const-conc (19.2 kg). Milk yield decreases across the years within groups were not significant, but urea contents in milk decreased significantly during the course of the project. Milk protein, somatic cell score, fat-protein ratio, average lactation number, calving interval and frequency of veterinary treatments did not differ by group and year. In conclusion, 5 years of concentrate reduction in low-input Swiss organic dairy farms, affected neither milk composition, nor fertility and veterinary treatments. Milk yields tended to decline, but at a low rate per saved kilogram of concentrate.

ABSTRACT

The extensive protein degradation occurring during ensiling decreases the nutritive value of silages, but this might be counteracted by tannins. Therefore, silages from two legume species containing condensed tannins (CT) - sainfoin (SF) and birdsfoot trefoil (two cultivars: birdsfoot trefoil, cv. Bull (BTB) and birdsfoot trefoil, cv. Polom) - were compared for their in vitro ruminal fermentation characteristics. The effect of combining them with two CT-free legume silages (lucerne (LU) and red clover (RC)) was also determined. The supply of duodenally utilisable CP (uCP) in the forages was emphasised. The legumes were each harvested from three field sites. After 24 h of wilting on the field, the legumes were ensiled in laboratory silos for 86 days. Proximate constituents, silage fermentation characteristics, CT content and CP fractions were determined. Subsequently, silage samples and 1 : 1 mixtures of the CT-containing and CT-free silages were incubated for 24 h in batch cultures using ruminal fluid and buffer (1 : 2, v/v). Each treatment was replicated six times in six runs. The effects on pH, ammonia and volatile fatty acid concentrations, protozoal counts, and total gas and methane production were determined. uCP content was calculated by considering the CP in the silage and the ammonia in the incubation fluid from treatments and blanks. Statistical evaluation compared data from single plants alone and together with that from the mixtures. Among treatments, SF silage contained the least CP and the most CT. The non-protein nitrogen content was lower, favouring neutral detergent soluble and insoluble protein fractions, in the SF and RC silages. Absolute uCP content was lowest in SF and SF mixtures, although the ratio to total CP was the highest. In comparison with LU, the ammonia concentration of the incubation fluid was lower for SF, RC and BTB and for the mixture of SF with LU. The total gas and methane production was similar among the treatments, and the total volatile fatty acid production was decreased with the CT-containing legumes. Protozoal count was increased with the mixtures containing LU and either SF or BTB compared with single LU. In conclusion, compared with the other legumes, SF and RC have similar advantages as they show limited proteolysis during ensiling. In addition, SF supplies more uCP relative to total CP. The CT-containing legumes also differed in their effect on ruminal fermentation and ammonia formation, probably because of their different CT contents. Thus, SF and its mixtures appear promising for improving the protein utilisation of ruminants.

ABSTRACT

This study investigated the hypothesis that dietary concentrations of methionine (Met), as a precursor of cysteine which is a constituent of glutathione (GSH), affect tissue antioxidant concentrations and the antioxidant defence system in pigs. Forty-five piglets (DanZucht × Pietrain) were allotted to three groups of similar mean body weight (11.0 ± 0.9 kg). The basal diet was composed of barley, wheat, corn starch, soybean oil, sucrose, cellulose and a mineral supplement with suboptimal concentrations of Met and was supplemented with dl-Met to reach 0.16%, 0.20% and 0.24% of dietary Met and 0.40%, 0.44% and 0.48% of dietary Met and cysteine in groups 0.16, 0.20 and 0.24 respectively. After 3 weeks, at slaughter, samples of liver, jejunum mucosa and plasma were collected. Feed intake and weight gains increased and feed:gain ratio decreased when dietary Met concentrations increased. The Trolox equivalent antioxidant capacity (TEAC), concentrations of GSH and thiobarbituric acid reactive substances (TBA-RS) and the activity of the glutathione peroxidase (GPx) in liver and jejunum mucosa were similar in all groups (p > 0.05). Relative mRNA concentrations of selected target genes of the nuclear factor (erythroid-derived 2)-like 2 (Nrf2), the master regulator of the antioxidant response, and of the nuclear factor 'kappa-light-chain-enhancer' of activated B-cells (NF-κB), the master regulator of inflammation, were largely unaffected both in jejunum and liver. In conclusion, inflammation- and oxidative stress-related pathways on the molecular level, and concentrations of lipid peroxidation products, of antioxidants and of enzymes involved in the antioxidant defence system were mostly unaffected by dietary Met concentration in gut and liver. These findings suggest that suboptimal dietary Met concentrations did not influence the antioxidant defence system of gut and liver in healthy piglets.

ABSTRACT

Milk production strategies focusing on longevity and limited use of concentrate are receiving increasing attention. To evaluate such strategies, knowledge of the development with age of animal characteristics, particularly digestion, is indispensable. We therefore investigated the development of feed intake, chewing activity, and digestion in 30 lactating Brown Swiss cows (876-3,648 d old) and 12 heifers (199-778 d old). We also studied whether age effects were exhibited differently in animals selected from herds subjected for 11 yr either to a forage-only or to a forage-concentrate feeding regimen. Forages consisted of grass hay (the only feed for heifers), corn silage, and grass pellets. Measurements lasted for 8 d, where amounts and composition of feeds, feces, and milk were recorded and analyzed. Ruminal pH data and eating and rumination activity were assessed by pH sensors put into the rumen and halter-mounted noseband sensors. The mean retention time of feed particles was assessed using Cr-mordanted fiber and data were used to calculate dry matter gut fill. Data were subjected to regression analyses with age and feeding regimen as explanatory variables, and body weight, milk yield, and proportion of hay in forage as covariates. This allowed separating age-related changes of body weight and milk yield from independent age effects and correcting for differences in preference for individual forages. In cows, organic matter intake increased with age (from slightly below to above 20kg/d), as did mean retention time and gut fill. Digestibility of organic matter did not show a clear age dependency, but fiber digestibility had a maximum in cows of around 4 to 6 yr of age. Ruminal pH and absolute eating and rumination times did not vary with cow age. Young and old cows chewed regurgitated boluses more intensively (60-70 times) than middle-aged cows (about 50 times). Effects of feeding regimen were small, except for fiber intake and rumination time per unit of intake, owing to the different fiber content of the diets. No significant interactions between age and feeding regimen were found. Heifers spent more time eating and ruminating per unit of feed than cows, which resulted in a high fiber digestibility. Irrespective of the feeding regimen tested, older cows maintained intake and digestion efficiency with longer retention times and chewing rumination boluses more intensively. The results support efforts to extend the length of productive life in dairy cows.

ABSTRACT

Previous studies indicated that absolute CH4 emissions and CH4 yield might increase and that milk production efficiency might decrease with age in cattle. Both would make strategies to increase longevity in dairy cattle less attractive. These aspects were experimentally determined in Brown Swiss cattle distributed continuously across a large age range. Thirty lactating dairy cows (876-3,648 d of age) received diets consisting of hay, corn silage, and grass pellets supplemented with 0 or 5kg of concentrate per day. Twelve heifers (199-778 d of age) received hay only. Cows and heifers were members of herds subjected to the 2 different feeding regimens (with or without concentrate) for the past 10 yr. Methane emissions were measured individually for 2 d in open-circuit respiration chambers, followed by quantifying individual feed intake and milk yield over 8 d. Additional data on digestibility, rumination time, and passage time of feed of all experimental animals were available. Regression analyses were applied to evaluate effects of age and feeding regimen. Body weight, milk yield, and the hay proportion of forage dry matter intake were considered as covariates. Methane emissions per unit of intake, body weight, and milk yield were significantly related to age. Their development in the cows with age was characterized by an increase to maximum at around 2,000 d of age, followed by a decline. This response was not accompanied by corresponding age-related changes in intake, chewing activity, digesta passage time, and digestibility of organic matter, which would have explained shifts in CH4. However, fiber digestibility showed a similar change with age as methane emissions, resulting in quite stable methane emissions per unit of digestible fiber. As expected, methane emissions intensity per unit of milk produced was greater by 8% without concentrate than with concentrate, but no difference was noted in the response to age when the animals were subjected to different feeding regimens. The efficiency of milk production was only marginally influenced by age and diet, and no different response was observed for age in the 2 dietary regimens. In conclusion, life cycle analyses of milk production systems focusing on longevity should consider changing methane yields with age in addition to the variation in environmental costs for replacements of culled cows.

ABSTRACT

AIMS: To investigate the relationship between the protonation of medium-chain fatty acids (MCFA) and their inhibitory effect on a ruminal methanogen species. METHODS AND RESULTS: Cell suspensions of Methanobrevibacter ruminantium M1 in 1 mg dry matter (DM) ml(-1) were supplemented with lauric acid (C12 ) and myristic acid (C14 ) at a concentration of 8 µg ml(-1) with different pH levels of the potassium-free buffer, where the calculated degrees of protonation of C12 and C14 varied from 0·3 to 50% and from 1 to 76% respectively. Methane formation, ATP efflux, potassium leakage and cell viability were monitored 15, 30 and 45 min after the reaction started. Declining methane formation rate, increasing ATP efflux and potassium leakage, and decreasing survival of M. ruminantium were observed with increasing degrees of protonation, i.e. with decreasing pH. CONCLUSIONS: The inhibition of methanogenesis by C12 and C14 is more efficient at a pH of 5-6 as compared to pH 7. SIGNIFICANCE AND IMPACT OF THE STUDY: Methane mitigation strategies in ruminants which use supplementation of feed with MCFA such as C12 and C14 may be more effective in a low rumen pH environment. This finding is helpful in designing diets to effectively decrease methane emissions by ruminants.

ABSTRACT

This study investigated the hypothesis that dietary fats rich in lauric (C12) and myristic acid (C14) increase broiler performance and that the underlying mechanism involves antimicrobial effects on gut bacteria and changes in gut morphology. One hundred eighty 1-day-old Cobb-500 broilers were allotted to 3 groups. All groups received a basal diet consisting of maize, wheat, soybean meal, and a fat source (4.5, 7.0, 7.6, and 8.0% of fat product in the diet during d 1 to 9, 10 to 17, 18 to 27, and 28 to 35, respectively) until 35 d of age. The diet of the control group contained a fat with 67% of oleic and linoleic acid and 1.4% of C12 and C14 of total fatty acids, that of the esterified lauric and myristic acid (ELA) group a fat with 33% of esterified C12 and C14 and that of the free lauric and myristic acid (FLA) group a fat with 31% of both esterified and free (1:1) C12 and C14 (6 replicates/treatment, 10 birds/replicate). Gain and feed consumption did not differ between groups, but feed:gain was lower in FLA group as compared to the control group (P < 0.05). Carcass weight, liver weight, triglyceride content of liver and muscle, and muscle cholesterol were similar between groups; however, breast muscle weight was higher in the FLA than in the control group (P < 0.05). The villus height:crypt depth ratio of the duodenal wall did not differ between groups, but in the jejunum, it was lower in the FLA group as compared to the control group (P < 0.05). DNA copy numbers of Lactobacillus, Bifidobacteria, Enterobacteria, Escherichia coli, and Campylobacter jejuni in jejunal digesta were similar among groups. The study shows that dietary fats rich in free C12 and C14 improved feed:gain and breast muscle yield, but the observed effects could not be conclusively explained based on the parameters measured. The decreased jejunal villi:crypt ratio may point to changes in gut protein or cell turnover.

ABSTRACT

Conjugated linoleic acids (CLA) are well known as milk fat-reducing feed supplements in diets for lactating ruminants. However, their effects on milk concentrations of fat-soluble vitamins are unknown. This study was performed to investigate the hypothesis that CLA affect the concentrations of retinol and tocopherol in ewe milk. For that purpose, group-housed Merino ewes (101 ± 13.7 kg) nursing twin lambs and fed with a hay:concentrate diet were supplemented with either 45 g of a rumen-protected CLA supplement containing 3.4 g of cis-9,trans-11-CLA and 3.4 g of trans-10,cis-12-CLA (CLA group, n=11) or with 45 g of a hydrogenated vegetable fat (control group, n=12) per ewe per day during the first 6 wk of lactation. Feed intake was recorded daily (concentrate) or weekly (hay) per group. Milk spot samples were collected at the beginning of the experiment (5 ± 2.4 d postpartum) and then weekly after lambs had been separated for 2 h from their mothers. The milk fat content was determined and feed and milk were analyzed for concentrations of α-, γ-, and δ-tocopherol and for retinol by HPLC. Dietary intake of tocopherol and retinol was similar in both groups. Feeding CLA decreased milk fat concentration by 23% on average, and during the first 3 wk of the study milk tocopherol concentration tended to be increased by feeding CLA (+17%), but retinol concentrations were not influenced. When related to milk fat, CLA feeding significantly increased both milk tocopherol (+40%) and retinol (+32%) and these effects were evident during the whole experimental period corresponding to the first half of lactation.

ABSTRACT

The effects of non-starch-polysaccharide-degrading enzymes, added to a maize silage- and grass silage-based total mixed ration (TMR) at least 14 h before feeding, on the rumen bacterial population were investigated. Six non-lactating Holstein Friesian cows were allocated to three treatment groups using a duplicate 3 × 3 Latin square design with three 31-day periods (29 days of adaptation and 2 days of sampling). Treatments were control TMR [69% forage and 31% concentrates on a dry matter (DM) basis] or TMR with 13.8 or 27.7 ml/kg of feed DM of Roxazyme G2 liquid with activities (U/ml enzyme preparation) of xylanase 260 000, ß-glucanase 180 000 and cellulase 8000 (DSM Nutritional Products, Basel, Switzerland). The concentrations of 16S rDNA of Anaerovibrio lipolytica, Fibrobacter succinogenes, Prevotella ruminicola, Ruminococcus flavefaciens, Selenomonas ruminantium and Treponema bryantii, and their relative percentage of total bacteria in rumen samples obtained before feeding and 3 and 7 h after feeding and from two rumen fractions were determined using real-time PCR. Sampling time had only little influence, but bacterial numbers and the composition of the population differed between the transition layer between rumen fluid and the fibre mat (fraction A) and the rumen fluid (fraction B) highlighting the importance to standardize sampling. The 16S rDNA copies of total bacteria and the six bacterial species as well as the population composition were mainly unaffected by the high levels of exogenous enzymes supplemented at all sampling times and in both rumen fractions. Occasionally, the percentages of the non-fibrolytic species P. ruminicola and A. lipolytica changed in response to enzyme supplementation. Some increases in the potential degradability of the diet and decreases in lag time which occurred collaterally indicate that other factors than changes in numbers of non-particle-associated bacteria are mainly responsible for the effects of exogenous enzymes.
