ABSTRACT

Numerous molecular regulatory mechanisms are involved in the formation of mammalian oocytes, but many of their key proteins and molecules remain unknown. GPR50, the lone G protein-coupled receptor located in the X chromosome, is one of the superfamily members of the G protein-coupled receptor. GPR50 has been recently proved to play an important role in various physiological activities. However, the role of GPR50 in reproduction is currently unclear. In our previous research, we proved that the GPR50 receptor is present in yak oocytes. In the present study, the expression level and subcellular localization of GPR50 in the in vitro maturation process of yak oocytes were investigated to explore further its role in the maturation of yak oocytes. In the germinal vesicle (GV) stage, GPR50 was expressed and positioned in the cell membrane. By comparison, in the metaphase II (MII) stage, GPR50 had the highest expression level and was highly diffused in the cytoplasm. On the basis of these observations, the knockdown and overexpression of GPR50 yak oocyte models were constructed. Results showed that the expression level of GPR50 knockdown significantly reduced the excretion rate and maturity level of the yak oocyte PB1 (P < 0.01). However, the expression level of GPR50 overexpression exerted no significant influence on the excretion rate and maturity level of the yak oocytes (P > 0.05). This study verified that GPR50 plays an important role in the in vitro maturation process of yak oocytes.

ABSTRACT

INTRODUCTION: This meta-analysis aims to assess the effectiveness and safety of warm acupuncture therapy for treating Primary sciatica. METHODS: The following 9 databases will be search from their inception to December 6, 2020: MEDLINE, EMBASE, the Cochrane Central Register of Controlled Trials (CENTRAL), the Chinese Biomedical Literature Database (CBM), the Chinese Medical Current Content (CMCC), the Wan-Fang Database and the China National Knowledge Infrastructure (CNKI). Randomized controlled trials (RCTs) of warm acupuncture for treating Primary sciatica, Chinese or Japanese without restriction of publication status will be included. Two researchers will independently undertake study selection, extraction of data and assessment of study quality. Meta-analysis will be conducted after screening of studies. Data will be analyzed using risk ratio for dichotomous data, and standardized mean difference or weighted mean difference for continuous data. DISSEMINATION: This meta-analysis will be disseminated electronically through a peer-reviewed publication or conference presentations. CONCLUSION: This study will provide evidence to judge whether warm acupuncture. TRIAL REGISTRATION NUMBER: INPLASY2020120109.

ABSTRACT

The literature study and physical measurement were adopted to analyze the origin and relevant texture information of a box of French golden acupuncture needles stored in Yunnan University of CM. In 1901 the French vice-consul George Soulié de Morant went to China, based on his Chinese linguistic skills, he studied with several Chinese medicine masters during that time. After returning to France, he researched clinic acupuncture with western medical doctors and published articles and books on it, which led him to be a Nobel Prize nominee. He made a box of acupuncture needles, collected by Yunnan University of CM, in which nine needles made by the gold-copper and silver-zinc alloy, decorated with ruby and sapphire representing tonification and dispersion. The parameter of material, thickness and length match well to the description in Zhengjiufa (Chinese Acupuncture). Being the golden box with needles of its kind in the whole world, it has more value of medicine, education, historical and cultural relics than jewellery, which is the wisdom and innovation stimulated by ancient Chinese medical theory in the western world.
