ABSTRACT

Objective: To analyze the clinical phenotype and detect the pathogenic gene in a Chinese pedigree with autosomal dominant polycystic kidney disease(ADPKD). Methods: The proband of this study was hospitalized in Dongguan City People's Hospital on October 10, 2017, due to "left maxillary apical cyst". Clinical phenotypes were noted, imaging examinations and determination of biochemical indicators were carried out for the clinical diagnosis of the proband. Genomic DNA was extracted from peripheral venous blood. Whole-exome genotyping of the proband was performed with the next generation sequencing technology, and the candidate mutation site of the patient and his family members was verified by PCR and Sanger sequencing technology. The mutation site was further screened in 150 unrelated healthy Chinese controls. Mutation frequency within human populations and bioinformatics analysis were predicted with softwares including ExAC, dbSNP, HGMD, 1000 genomes, ClinVar, PKDB, Mutation Taster and PhyloP. Results: The proband, a 46-year-old male, was diagnosed with hypertension, positive urine occult blood and elevated blood creatinine. B-ultrasound and CT examinations showed that he had bilateral polycystic kidney with left kidney stones and polycystic liver. The gene analysis showed that the c.11017-10C>A heterozygous splice mutation in PKD1 gene was identified in the proband, his second younger brother, younger sister, daughter and niece, but absent in 150 healthy controls. Bioinformatics analysis showed it has been reported in the dbSNP, ClinVar, HGMD, PKDB and Mutation Taster databases. Some databases predicted it has a harmful function for probably leading to production of a truncated polycystin1(PC1) protein. Conclusion: c.11017-10C>A underlies the Chinese ADPKD pedigree and expands mutation spectrum of PKD1.

ABSTRACT

Technology has contributed significantly to the adoption of EVs (EVs) in the era of industry 4. 0. However, consumer intentions for EVs have been elusive, and the pace of adoption has been confined. Therefore, this study aims to investigate the influence of external factors in promoting customer behavioral intention for EVs. The study also investigates the mediating role of perceived ease of use (PEU) and perceived usefulness (PU) between external factors and consumer intentions for EVs. It also examined the moderating role of gender on the study variables. A study approach based on the expanded version of the technology acceptance model (TAM) was utilized to analyse data from 203 customers in China. The model was tested using structural equation model (SEM) and multigroup analysis (MGA) techniques. The results indicated that two external factors have a positive relationship with TAM constructs. The results also indicate that PEU and PU have a serial mediating relationship between external factors and behavioral intention. Finally, the study revealed gender-related differences in TAM for EVs. The study's findings help managers to design successful strategies by knowing the external factors impacting customer EV intentions and gender differences. Finally, this is a ground-breaking research that applies TAM to the automobile sector. As a result, examining both new and current factors and evaluating them in a new setting adds to the body of the literature on the adoption of technology.

ABSTRACT

OBJECTIVES: To compare the clinical effects among three kinds of treatments (external fixation, plate fixation, and non-operation) of 1/3 displaced midshaft clavicular fractures in adults. METHODS: Retrospective analysis was conducted on 306 patients with clavicular fractures in Trauma Orthopedic Department in Jishuitan Hospital from March 2010 to May 2013, and 89 of these patients met the inclusive criteria.Among them, there were 29 cases of external fixation (Group A), 30 cases of plate fixation (Group B), and 30 cases of non-operation with the clavicle sling in Outpatient and Emergency Department (Group C). The average follow-up period was 32 months. The results were evaluated from four perspectives, respectively, follow-up period, Constant Shoulder Score (CSS), Disability of the Arm Shoulder and Hand Score (DASH), and imaging. There was no significant difference in demography, severity of injuries, mechanism of injury, and fracture type among the patients in the three groups. RESULTS: Among 29 cases of external fixation, 25 cases were successfully followed up, 4 were lost.60 cases of plate fixationand non-operation were all successfully followed up.Based on the pairwise comparison of CSS and DASH among the three groups, there was no significant difference between external fixation [(94.0±6.4), (15.2±3.4)Score] and plate fixation[(92.9±8.4), (15.7±3.5)Score] (both P>0.05). Mean values of CSS and DASH of non-operation group [(86.0±3.6), (18.0±3.6)Score] were lower than those of the former two groups (P<0.001). The healing time of 3 kinds of treatments were as follows: external fixation 10.4±2.32 weeks; plate fixation 12.1±2.5 weeks; and non-operation 15.7±2.2 weeks. In terms of their complications, for the external fixation group, there was 1 case of nonunion (4%); and 3 cases of angulation deformity in the 12(th) week, whose external fixations were maintained till their unions in the 16(th) week.Shortening and displacement did not appear in any cases. In the plate fixation group, there was 1 case of nonunion (3%) and no malunion in this group. The biggest complications of plate fixation were implants stimulus and to remove the plate in the second operation, accounting for 60%.In the non-operation group, there were 3 cases of nonunion (10%), and 12 cases of malunion (40%) including 10 cases without symptoms and 2 cases with symptoms. The satisfaction rates for the appearance of shoulder after these 3 kinds of treatments among the patients were 96%, 93% and 77%, respectively. CONCLUSIONS: (1) In treating 1/3 displaced midshaft clavicular fractures in adults, external fixation and plate fixation are overall better than non-operation.And there is no significant difference in curative effects between the two. (2) Both external fixation and plate fixation have its advantages and disadvantages. The choice of treatment must be based on the clinical type of the fracture and the communication with patients.

ABSTRACT

High glucose (HG) state is closely related to diabetic complications, and among these one main pathogenesis involves apoptosis of important functional cells. However, the mechanism of osteoblast (OB) apoptosis induced by HG state is not clear. Peroxynitrite (ONOO-), the strongest oxidant, can mediate apoptosis in various kinds of cells, including OB. Therefore, this study was aimed at investigating whether HG state could induce OB apoptosis through ONOO-. Cultured OB from rat calvariae explanted from E21 fetuses were treated with HG solution. The ratio of OB apoptosis and ONOOcontent was assayed by TUNEL, flow cytometry, and immunohistochemistry. The results showed that HG state could induce ONOO- overformation (p<0.01), and excessive OB apoptosis (p<0.001). However, the HG-induced OB apoptosis could be attenuated by peonol, a potent scavenger of ONOO-. In conclusion, HG state might trigger excessive ONOO- formation, mediating OB apoptosis.

ABSTRACT

In the present study, an adapted bacterium PF-11 with high 1,3-dichlorobenzene degradation capability was isolated from town-gas industrial effluent through continuous introducing of N-methyl-N'-nitro-N-nitrosoquanidine (NTG). In suitable condition, a degradation rate of 32 mg L(-1) d(-1) of 1,3-dichlorobenzene was obtained by strain PF-11 with effective chlorion release. Strain PF-11 was tentatively identified as gram-positive Bacillus cereus. The substrate specificity of the strain PF-11 was relatively low, and the degradation rate for different chlorobenzenes was in the order of monochlorobenzene > 1,3-dichlorobenzene > 1,2-dichlorobenzene. Initial oxidation step was molecular oxygen attacking chlorobenzene ring catalyzed by dioxygenase.

ABSTRACT

Telomerase is an important biomarker in cancer cells. It is active in germline cells, most of cancer tissues and cell lines, but not in most somatic tissues. Telomerase is composed of two components, and while hTER is present in normal and tumor cells, expression of hTERT appears to be highly regulated and correlates with telomerase activity. In order to detect the telomerase enzyme and hTERT protein, anti-hTERT polyclonal antibodies were produced in this study. A segment of hTERT cDNA was amplified by RT-PCR and cloned into the multi-cloning site of the GST gene fusion vector pGEX-5X-3. After the recombinant plasmid was expressed in E. coli BL21, the fusion protein was purified for immunization. Extracts from several cultured cells were analyzed by Western blot, and the results indicated that telomerase enzyme and hTERT protein could be specifically detected by this anti-hTERT antibod'. Thus, a simple and effective method was primarily established for the immunodetection of telomerase enzyme and hTERT protein.

ABSTRACT

Eighteen patients with acute hemorrhagic necrotizing pancreatitis who survived from operation were followed up for 12 to 43 months. Late complications including pancreatic external fistula, pseudocyst, hyperglycemia, etc were reviewed. The pancreatic juice was collected through endoscopic cannulation. The volume and HCO3- concentration of the pancreatic juice and three kinds of pancreatic enzymes were measured. The results demonstrated that inspite of anatomic abnormality or functional impairment of the pancreas, the pancreatic insufficiency rarely manifested to be marked because of the compensative ability of the viable exocrine pancreas.

ABSTRACT

We present a method of staining with Chinese-Ink or methylenum coeruleum under endoscope preoperatively and its clinical valve as a guide for radical resection of esophagus (cardia) cancer. There were 39 cases of esophageal cancer and 9 cases of gastric cardia cancer. 37 cases were stained with Chinese-ink and 11 cases with methylenum coeruleum. 268 of the 334 groups (427 of 592 lymph modes) were stained. In the 73 metastatic groups (107 lymph nodes) 65 groups (96 lymph nodes) were stained (88.1%, 96/107). In this series radical operation was achieved in 45 cases. The indication of staining, choice of marking materials and main points of manipulation practice were discussed.
