ABSTRACT

Influenza A virus (IAV) infection often leads to influenza-associated fatalities, frequently compounded by subsequent bacterial infections, particularly Gram-negative bacterial co-infections. Lipopolysaccharide (LPS), a primary virulence factor in Gram-negative bacteria, plays a crucial role in influenza-bacterial co-infections. However, the precise pathogenic mechanisms underlying the synergistic effects of viral-bacterial co-infections remain elusive, posing significant challenges for disease management. In our study, we administered a combination of IAV and LPS to mice and examined associated parameters, including the lung function, lung index, wet/dry ratio, serum inflammatory cytokines, Nedd4L expression in lung tissue, and mRNA levels of inflammatory cytokines. Co-infection with IAV and LPS exacerbated lung tissue inflammation and amplified M1 macrophage expression in lung tissue. Additionally, we stimulated macrophages with IAV and LPS in vitro, assessing the inflammatory cytokine content in the cell supernatant and cytokine mRNA expression within the cells. This combined stimulation intensified the inflammatory response in macrophages and upregulated Nedd4L protein and mRNA expression. Subsequently, we used siRNA to knockdown Nedd4L in macrophages, revealing that suppression of Nedd4L expression alleviated the inflammatory response triggered by concurrent IAV and LPS stimulation. Collectively, these results highlight the pivotal role of Nedd4L in mediating the exacerbated inflammatory responses observed in IAV and LPS co-infections.

ABSTRACT

In this study, the seismic behavior of a recycled aggregate concrete-filled square steel tube (S-RACFST) frame under different design conditions was investigated. Based on previous studies, a finite element model for the seismic behavior of the S-RACFST frame was developed. Moreover, the axial compression ratio, beam-column line stiffness ratio, and yield bending moment ratio of the beam-column were regarded as the variation parameters. It was through these parameters that the seismic behavior of eight S-RACFST frame finite element specimens was discussed. The seismic behavior indexes, such as the hysteretic curve, ductility coefficient, energy dissipation coefficient, and stiffness degradation were obtained-which, in turn, revealed the influence law and the degree of the design parameters regarding seismic behavior. Moreover, the sensitivity of the various parameters with respect to the seismic behavior of the S-RACFST frame was evaluated via grey correlation analysis. The results show that the hysteretic curves of the specimens were fusiform and full with respect to the different parameters. Firstly, with the axial compression ratio increasing from 0.2 to 0.4, the ductility coefficient increased by 28.5%. In addition, the equivalent viscous damping coefficient of the specimen with the axial compression ratio of 0.4 was 17.9% higher than that of the specimen with the axial compression ratio of 0.2, which was 11.5% as well as that with an axial compression ratio of 0.3. Second, when the line stiffness ratio rises from 0.31 to 0.41, the specimens' bearing capacity and displacement ductility coefficient both get better. However, the displacement ductility coefficient gradually decreases when the line stiffness ratio is greater than 0.41. As a result, an optimal line stiffness ratio (0.41) thus exhibits good energy dissipation capacity. Thirdly, with the increase in the yield bending moment ratio from 0.10 to 0.31, the bearing capacity of the specimens improves. In addition, the positive and negative peak loads increased by 16.4% and 22.8%, respectively. Moreover, the ductility coefficients were all close to three, thus demonstrating good seismic behavior. The stiffness curve of the specimen with a large yield bending moment ratio with respect to the beam-column, is higher than those that possess a small beam-column yield moment ratio. In addition, the yield bending moment ratio of the beam-column possesses a significant influence on the seismic behavior of the S-RACFST frame. Furthermore, the yield bending moment ratio of the beam-column should be considered first in order to ensure the seismic behavior of the S-RACFST frame.

ABSTRACT

In this study, the basalt fiber content (0%, 0.075%, and 0.15%) and replacement ratio of recycled coarse aggregate (0%, 50%, and 100%) were used as parameters, and the compressive strength of 15 cubes and 15 prisms was analyzed. The failure morphology of the specimens was characterized, and the cubic compressive strength, axial compressive strength, elastic modulus, Poisson's ratio, and other mechanical property indices of the specimens were measured. Upon increasing the replacement ratio, the degree of damage of the specimens gradually increased, whereas the cubic compressive strength, axial compressive strength, and elastic modulus gradually decreased. As the replacement ratio was increased from 50% to 100%, the cubic compressive strength and elastic modulus were noted to decrease the most by about 9.07% and 9.87%, respectively. On the other hand, the Poisson's ratio first decreased, followed by an increase. Upon increasing the fiber content, the degree of damage of the specimens was gradually reduced, whereas the cubic compressive strength, axial compressive strength, and elastic modulus gradually increased. As the fiber content increased from 0.075% to 0.15%, the axial compressive strength and elastic modulus increased the most by about 6.65% and 10.19%, respectively. On the other hand, the Poisson's ratio gradually decreased. Based on the test data, the functional relationships between the strength indices and different variables, as well as the conversion value of each strength index and different variables were established; after comparison and verification, the formula calculation results were found to be in good agreement with the test results. The microstructural changes in the basalt fiber reinforced recycled aggregate concrete were characterized by scanning electron microscopy (SEM), and the changes in the mechanical properties of the basalt fiber reinforced recycled aggregate concrete as well as the mechanism of fiber modification and reinforcement were explained from a micro perspective.

ABSTRACT

Based on Janus kinase 1/2-signal transducer and activator of transcription 1(JAK1/2-STAT1) signaling pathway, this study explored the immune mechanism of Maxing Shigan Decoction in alleviating the lung tissue and colon tissue damage in mice infected with influenza virus. The influenza virus infection was induced in mice by nasal drip of influenza virus. The normal group, model group, oseltamivir group, antiviral granule group, and Maxing Shigan Decoction group were designed. After intragastric administration of corresponding drugs or normal saline for 3 or 7 days, the body mass was measured, and lung index, spleen index, and thymus index were calculated. Based on hematoxylin-eosin(HE) staining, the pathological changes of lung tissue and colon tissue were observed. Enzyme-linked immunosorbent assay(ELISA) was used to detect serum levels of inflammatory factors interleukin-8(IL-8) and interferon-γ(IFN-γ), Western blot and real-time quantitative polymerase chain reaction(RT-qPCR) to determine the protein and mRNA levels of JAK1, JAK2, STAT1, interferon regulatory factor 9(IRF9), and IFN-γ in lung tissue and colon tissue. The results showed that after 3 and 7 days of administration, the body mass, spleen index, and thymus index were lower(P<0.05 or P<0.01), and the lung index was higher(P<0.01) in the model group than in the normal group. Moreover, the model group showed congestion, edema, and infiltration of a large number of lymphocytes and macrophages in the lung tissue, irregular structure of colon mucosa, ulceration and shedding of epithelial cells, and infiltration of a large number of inflammatory cells. The model group had higher levels of serum IFN-γ(P<0.01), higher protein and mRNA expression of JAK1, JAK2, STAT1, IRF9, IFN-γ in lung tissue(P<0.05 or P<0.01), higher level of JAK2 protein in colon tissue(P<0.01), and higher protein and mRNA levels of STAT1 and IRF9(P<0.05 or P<0.01) than the normal group. Compared with the model group, Maxing Shigan Decoction group had high body mass, spleen index, and thymus index(P<0.05 or P<0.01), low lung index(P<0.05 or P<0.01), and significant alleviation of pathological injury in lung and colon. Moreover, lower serum level of IFN-γ(P<0.05 or P<0.01), protein and mRNA levels of JAK1, JAK2, STAT1, IRF9, and IFN-γ in lung tissue(P<0.05 or P<0.01), JAK2 protein level in colon tissue(P<0.01), and protein and mRNA levels of STAT1 and IRF9(P<0.05 or P<0.01) were observed in the Maxing Shigan Decoction group than in the model group. After 3 days of administration, the level of serum IL-8 in the model group was significantly higher than that in the normal group(P<0.01), and the level in the Maxing Shigan Decoction group was significantly reduced(P<0.01). In conclusion, Maxing Shigan Decoction can significantly up-regulate body mass, spleen index, and thymus index, down-regulate lung index, reduce the levels of IL-8 and IFN-γ, and down-regulate protein and mRNA levels of JAK1, JAK2, STAT1, IRF9, and IFN-γ in lung tissue and protein and mRNA levels of JAK2, STAT1, and IRF9 in colon tissue, and alleviate pathological damage of lung tissue and colon tissue. The mechanism is the likelihood that it inhibits the activation of JAK1/2-STAT1 signaling pathway to alleviate the damage to lung and colon tissue damage.

ABSTRACT

Since graphene oxide­silver nanoparticles (GO-AgNPs) have special affinities to DNA, it become increasingly important in fields of biological analysis in which GO-AgNPs nanocomposites universally functioned as a quencher. In this paper, GO-AgNPs nanocomposites with different GO to AgNPs ratios were synthesized as a fluorescence quencher to interact with DNA labeled by methylene blue (MB). The results showed that the fluorescence intensity of DNA-MB system decreased with the increasing of GO-AgNPs nanocomposites concentration. The quenching phenomenon of DNA-MB by AgNPs and GO was not a simple additive effect but a synergistic effect. The quenching efficiency of synthesized GO-AgNPs nanocomposites with different ratios (1:1, 1:3, 1:5, 1:10) increased with the decrease of GO/Ag ratio. Thermodynamic analysis was employed to investigate the interaction of GO-AgNPs and DNA-MB, it can be concluded that the intermolecular force between GO-AgNPs and DNA-MB was hydrogen bonding. Our works will provide important theoretical and experimental bases for fluorescence sensing of DNA.

ABSTRACT

Liposome mediated DNA transport possesses a number of preventing diseases in clinical trials, thus, the study of interaction between DNA and liposomes has become a hot research direction. In this paper, the adsorption behavior of DNA onto two representative lipids had been studied by the fluorescence spectrum measurement, Ultraviolet absorption spectrum and Langmuir-Blodgett technology. The results of fluorescence spectrum measurement indicated that the fluorescence liposomes were quenched statically by DNA at all three temperatures. Thermodynamic analysis displayed that the intermolecular forces between DNA and liposomes were van der Waals forces and Hydrogen bonding. The experimental results of Ultraviolet absorption spectrum and Langmuir-Blodgett technology further verified these mechanisms. This work provides useful theoretical basis for the development of novel DNA delivery materials.

ABSTRACT

A simple and fast spectrofluorimetric method coupled with carbon quantum dots (CQDs) modified ZnO/CdS nanoparticles was developed for the detection of Ferric iron (Fe(III)). The fluorescence of CQDs/ZnO/CdS NPs was effectively quenched by Fe(III) due to the strong interaction between the CQDs/ZnO/CdS NPs and Fe(III). In addition, the detection limit of Fe(III) was about 1.72×10-7M. The effect of foreign ions on the fluorescence intensity of CQDs/ZnO/CdS NPs showed that the interference response in detecting of Fe(III) ions was low. Moreover, the quenching of Fe(III) and CQDs/ZnO/CdS NPs was discussed to be a static quenching procedure, which was proved by quenching constant KSV and fluorescence lifetime τ. The study of thermodynamics showed that the values of entropy change (ΔS) and enthalpy change (ΔH) were both positive, and the value of free energy (ΔG) was negative, which implied that the weak interaction of the molecular between CQDs/ZnO/CdS NPs and Fe(III) was hydrophobic force, and the quenching process was endothermic and spontaneous.

ABSTRACT

This paper describes experimental results of degradation of Rhodamine B (RB) by vortex scattering ultrasound in aqueous solution. Vortices are produced by a high-speed agitator. The effects of different factors on the degradation of RB solution were studied, such as different reaction container, the initial concentration of RB, stirring speed and ultrasonic frequencies. Ultraviolet-visible spectrophotometer (UV) was used to measure the absorbance value of RB to assess degradation rate. The optimal experimental condition was three-necked bottle, stirring speed of 700 r/min, initial concentration of 10 mg/L and ultrasonic frequency of 40 kHz. The optimal degradation rate of RB can reach 98% within one hour. The combination of ultrasonic irradiation and mechanical stirring was discovered that can degrade the RB efficiently in aqueous solution. The investigation of mechanism demonstrates that the cavitation bubbles produced by agitation play a major role in promoting degradation. COMSOL Multiphysics (Version 5.3a) software was used to simulate this process. And the method of combination of ultrasonic irradiation and mechanical stirring has also been shown to be effective in degrading methylene blue, bromophenol blue and Congo red. So the combination of ultrasonic irradiation and mechanical stirring can be used as an option for treating organic wastewater in the future. This study established a new way to degrade other organic pollutants.

ABSTRACT

OBJECTIVE: To study the chemical constituents of Wedelia prostrata. METHODS: Chromatography and spectroscopic analysis were employed to isolate and elucidate the chemical constituents in the plant. RESULTS: Seven compounds were obtained and identified as ent-kaur-16-en-19-oic acid (I), ent-kaur-16alpha, 17-dihydroxy-19-oic acid (II), ent-kaur-15alpha-senecioyloxy-16-en-19-oic acid (III), beta-sitosterol (IV), stigmasterol (V), stigmasterol-3-O-beta-D-glucopyranoside (VI) and n-hexacosanol (VII). CONCLUSION: Compounds I-III are isolated from this plant for the first time.
